Expression of Caspase-3 and P-gp in hepatocellular carcinoma and their relationship

Objective To measure the expression of Caspase-3 and P-gp in hepatocellular carcinoma (HCC) and adjacent tissues, and to examine these two proteins' relationships with HCC clinical and pathological characteristics and the associations between the two proteins. Methods The expression of Caspase-3 and P-gp were measured in 50 HCC tissues and adjacent tissues by EnvisionTM immunohistochemistry. HCC clinic and pathological characteristics and follow-up data were also collected and analyzed. Results The expression of Caspase-3 in HCC tissues was significantly lower than that in adjacent tissues(P <0.05). The expression of Caspase-3 in HCC tissues was significantly associated with HCC's Edmonson grade and the presence of hepatitis envelope(P <0.05). On the other hand, the expression of P-gp in HCC tissues was significantly higher than that in adjacent tissues( P <0.05 ). P-gp expression in HCC tissues was significantly correlated with HCC's Edmonson grade and the presence of cirrhosis(P < 0.05 ). The expression of Caspase-3 and P-gp was negatively correlated ( r = - 0. 600,P = 0.000). A better prognosis was observed among HCC patients with either positive Caspase-3 expression or negative P-gp expression(P < 0.05). Conclusion Caspase-3 may promote apoptosis and inhibit the development of HCC, while P-gp may have an anti-apoptosis function and play a role in HCC's drug resistance. P-gp can inhibit the function of Caspase-3 and an absence or low expression of Caspase-3 may be related to apoptosis resistance and multidrug resistance.

Genetic heterogeneity of colorectal neoplasm and its clinical implications

Cancer treatment has been continuously improved in recent year. Genetically targeted therapy has played a significant role in tumor reduction and local control. Genetic heterogeneity of colorectal cancer affects not only tumor development and progression, but also the selection and efficiency of targeted drugs, thus may become a breakthrough towards tackling the bottleneck in personalized medicine.

Research progress on DNA methylation of esophageal squamous cell carcinoma

In recent year, one of the hot topics in esophageal carcinoma research is the impaired gene expression resulted from hypermethylation within the promoter regions of rumor suppressor genes. Many studies suggested that DNA methylation plays an important role in the initiation, invasion, and metastasis of esophageal carcinoma. DNA methylation spectrum in peripheral blood may be used as markers for early diagnose, prognosis prediction and follow up in esophageal cancer patients. De-methylation of tumor suppressor genes may become a target of esophageal carcinoma therapy.

Research progress in local therapy of recurrent nasopharyngeal carcinoma

At present, the main treatment for recurrent nasopharyngeal carcinoma (rNPC) is re-irradiation. Conventional external irradiation can result in a high incidence of side effects. Intensity modulated radiation therapy and stereotactic radiotherapy can not only reduce the radiation dose to vital organs and surrounding normal tissues, but also improve local control rate. Brachytherapy and surgery are also shown to be effective in early diseases.

Aberrant methylation of tumor suppressor genes in salivary adenoid cystic carcinoma

Methylation of tumor suppressor gene(TSG), currently a hot field in cancer research, is involved in many biological processes including cell cycle regulation, cell signal transduction, DNA apoptosis,tumor invasion and metastasis. The level of TSG methylation is significantly associated with the development of salivary adenoid cystic carcinoma and its biologic behavior.

Detection and clinic implication of circulating tumor cells in peripheral blood

Circulating tumor cells are closely related to the development and progression of malignant tumors. Current methods of detecting circulating tumor cells include reverse transcription polymerase chain reaction method and immunomagnetic enrichment assay. Detection of circulating tumor cells can contribute to early detection of cancer micrometastases, re-classification of clinical stage, monitoring recurrence and metastasis after surgery or radiotherapy and chemotherapy, predicting prognosis, and selecting personalized therapeutic strategy.

Research progress on anaerobic bacteria in tuomr gene therapy

Hypoxic metabolism in solid tumor reduces chemoradiotherapy effect and become the obstacle in tumor therapy. Gene therapy is the most promising tumor treatment method, but currently using carriers cannot target tumors accurately. Anaerobic bacteria has become the transferring carrier in tumor gene therapy because it has tendency to hypoxia in solid tumors and can target solid tumors more accurately.

Currrent study of breast cancer stem cells

The breast cancer stem cells are a subpopulatiom of breast tumor cells that selectively possess tumor initiation and self-renewal capacity. Recent findings suggest that it may correlate with the failure treatment and relapse of breast cancer. The isolation of phenotypically distinct tumorigenic of breast cancers will find a way to elucidation of the pathways that account for their tumorigenic potential to cure the disease.

Clinical observation of early breast cancer after breastconserving surgery and radiotherapy

Objective To observe the oncololigcal and cosmetic outcomes and complication of breastconserving surgery and radiotherapy for early breast cancer. Methods A total of 78 early breast cancer patients, who were treated in the First People's Hospital of Pingdingshan between January 2001 and September 2008, were identified and followed. Among them, 36 were in stage Ⅰ and 42 in stage Ⅱ. Tangential irradiation was used with a dose of 50 Gy. Tumor bed received a boost dose of 10 Gy electron beam. For axillary lymph node-positive patients, ipsilateral supraclavicular field was hybrid irradiated with 50 Gy X and electron beam.Results The total follow-up ranged from 6 to 98 months. Local recurrence was observed among 4.9% of the patients. The 3-year and 5-year overall survival rate were 96.6% and 92.7% ,respectively. The breast cosmetic satisfaction rate was 94.9%. The main complications reported were acute skin reaction and upper limb edema.Conclusion Breast-conserving surgery and post-operative radiotherapy in early breast cancer patients can lead to satisfactory survival rates and cosmetic effects and hence improved life quality.

Effect of IL-11 in prevention and treatment of oral mucosal complications of radiotherapy for head and neck cancer

Objective To explore an effective method for preventing and treating oral mucosal complications of radiotherapy for head and neck cancer. Methods Fifty head and neck cancer patients who were receiving radiotherapy were enrolled between March, 2008 and March, 2010. These patients were randomly assigned to the treatment group and the control group. During the radiotherapy, patients in the treatment group were given IL-11 in the form of atomization inhalation,whereas patients in the control group were not. Results IL-1 1 was well tolerated by the patients. It significantly decreased the level of oral mucosal complications and pains and improved patients' appetites ( P < 0.05 ). In addition, the duration of pain was significantly ( P <0.05 ) reduced from 4.5 ± 1.3 days ( in control group) to 2.3 ± 1.0 ( in treatment group), and the healing period was also significantly ( P < 0.05) reduced from 7.3 ± 1.5 days ( in control group) to 4.1 ± 1.7 ( in treatment group). Conclusion IL-11 is effective in preventing and treating oral mucosal complications of radiotherapy for head and neck cancer,relieving associated pains, and therefore improving patients' quality of life.
