Total resection of craniopharyngioma and treatment of postoperative complications

Objective To investigate the methods of total resection of craniopharyngioma and treatment of postoperative complications. Methods The abscission of CPG was performed in 24 patients with craniopharyngioma (CPG) which situated at sellar region after giving glucocorticoid and anti-epilepsy drug. The tumors were removed via the 4 anatomy spaces in sellar region by employing microsurgical technique. Consciousness, urine, and serum electrolyte of patients were observed strictly after surgery, and complications such as diabetes insipidus, serum electrolyte disorders and epilepsy were remedied in time. Results Total removal of tumors were achieved in 22 cases, and minor portion of tumors was left in 2 cases. All of the patients were sober after surgery. Diabetes insipidus, serum electrolyte disorder and body temperature disorder took place in 16 cases,12 cases, and 6 cases respectively. After treatment, serum electrolyte disorder and body temperature disorder were rectified in all cases, but diabetes insipidus was rectified in 13 of 16 cases. Conclusion Choosing the best operation approach according to the scope which tumor invades and being familiar with the 4 sellar anatomy spaces so as to be used correctly in operation are the keys to remove the CPG completely. Giving glucocorticoid before operation and active treatment of complications such as diabetes insipidus, serum electrolyte disorder can decrease mortality observably.

Effect of marin algae polysaccharide derivant on osteoporotic model of the rats induced retinoic acid

Objective To observe the effect of marin algae polysaccharide derivant on the osteoporotic model of the rats. Methods Forty six-month-old female Wistar rats were selected to duplicate the osteoporotic model induced by retinoic acid and performed with marine algae polysaccharide derivant. The rats were randomly divided into two groups: experiment group treated with 10 mg marine algae polysaccharide derivant10mg per day by oral,control group treated with 10 mg glucose per day by oral.The effect of marine algae poly saccharide derivant on osteoporosis were evaluated by morphometry. Results After intake of polysaccharide,the mean trabecular plate density (Tb.N),the mean trabecular plate thickness(Tb.Th),and the percent trabecular area (Tb.Ar%) of t he experiment group, which were treated by polysaccharide were significantly increased more than that of the control group model(P<0.01). Conclusion These results indicate that polysaccharide can increase bone mass and has a therapeutic and preventional effect on the osteoporotic model of the rats induced by retinoic acid.

Hepatoectomy for liver cancer by blocking hepatic hilum under normal temperature(Report of 86 cases)

Objective To introduce operative skill of liver resection by blocking hilum of liver under normal temperature.Method Retrospective analysis of experience treating 86 cases of liver cancer with the operation.Result Among 86 cases of hepatic malignant tumor,there were not serious complications such as death,massive haemorrhage etc.during operation;postoperative liver failure occurred in 7 cases,among them,cure 5 cases and 2 cases of death.The incision infection occurred in 5 patients;The thorax dropsy occurred in 19 cases, which was absorbed by oneself after treating in 14 cases,cured by puncture occurred in 5 cases.Conclusion Operative skill of blocking hilum of liver under normal temperature is more ideal means in treating the liver cancer.

Construction of cDNA library of the treated Changliver cell and quality analysis

Objective To construct cDNA library of the treated Changliver cell by switching mechanism at 5′ end of RNA transcript (SMART) technique and analyze its quality.Methods cDNA of Changliver cell was aquired with reverse transcription polymerase chain reaction (RT-PCR) and long-distance PCR (LD-PCR),then the cDNA library was constructed with SMART cDNA library construction kit.Results Through testing,the high quality cDNA library containing full length cDNA of Changliver cell had been constructed.The titer of the amplified cDNA library was 4.5 × 1010 pfu*ml-1 and the average exogenous inserts of the recombinants was 1.5 kb.Conclusion These results suggest that the Changliver cell cDNA library has a high quality and lays a solid foundation for researching on Changliver cell and screening

Application of modified Z-stent in interventional therapy of Budd-Chiari syndrome

Objective To evaluate the utility of modified Z-stent in treatment for Budd-Chiarisyndrome (BCS). Methods A retrospective study was used in twelve BCS patients treated in twohospitals. The Doppler examination was carried out in all patients preoperatively, so as to confirm thenature of the lesion and chose correct type of endovascular modified Z-stent. Under DSA monitoring formembranotomy and dilation of the inferior vena cava(IVC), after that, insert the marked modified Z-stent to IVC correctly, and put the non-stent part to hepatic vein orifice. The cavography and hepaticvenography should confirm the position of the Z-stent. All of 12 patients with membranous obstructionof the IVC(MOVC) or segmental obstruction of the IVC (SOVC)were underwent modified Z-stentplacement. Results The IVC pressure (IVCP) before smd after membranotomy (dilation) were 27.33± 4.12cmH2O and 18.67 ± 5.07cmH2O (P＜0.01). Comparing with dilation and putting stent group,the IVCP decreased from 18.67 ± 5.07cmH2O to 11.42 ± 2.11 cmH2O ( P ＜ 0.01 ). The modffied Z-stent could avoid hepatic vein orifice getting compression and resist the retraction of IVC throughly infollowing-up period of 2.5 years. Conclusion s The endovascular treatment of BCS with modified Z-stent is more effective and safer to prevent thrombosis. Further study will be required to observe theirlong term effects.

Effect of medium composition on the growth and Sporulation of Bacillus sp WTFY

Objective Study the relationship between the component of medium and spore formation of Bacillus sp WTFY. Methods Alter the carbon source、nitrogen source、inorganic concentration and kinds in turns, observe the sporulation circumstances of Bacillus sp WTFY.Results When glucose concentration was higher over 0.1%, spore did not emerge;Nitrogen source、phosphorus、magnesiumions concentration and kinds have some degree influence,but not significantly.Conclusion Carbon source concentration is a key factor that influence spore formation.

Involvement of neural mechanism in cardioprotection provided by renal ischemic preconditioniong in anesthetized rabbits

Objective To investigate the role of renal nerve in cardioprotection provided by renal ischemic preconditioning(RIP).Methods The effects of ischemia-reperfusion and RIP on the hemodynamics, myocardial oxygen consumption, epicardial electrography and infarct size were examined in anesthetized rabbit.Results During the 45 min of myocardial ischemia and 180 min of reperfusion, all hemodynamic parameters and myocardial oxygen consumption decreased progressively significantly. Epicardial electrographic ST-segment was elevated significantly during myocardial ischemia and return to baseline progressively in the course of reperfusion. The myocardial infarct size occupied 55.80±1.25% of area at risk,and RIP significantly reduced the myocardial infarct size to 36.51±2.80%(P＜0.01). The renal nerve section (RNS) per se didn't affect myocardial infarct size produced by ischemia-reperfusion, while cardioprotection afforded by RIP was completely abolished by RNS.Conlusion RIP have the protective effect on heart, and activation of renal afferents by transient ischemia-reperfusion play an important role in such a cardioprotection.

Construction of a DNA vaccine encoding herpes simplex virus type 1 glycoprotein D

Objective To construct a recombinant plasmid DNA containing herpes simplex virus type 1(HSV-1) glycoprotein D (gD) gene.Methods The HSV-1 gD gene was obtained by polymerase chain reaction (PCR) and inserted into TA cloning vector pGEM-T, then cloned into the eukaryotic expression vector pcDNA3.1 to generate pLy-D. The recombinant plasmid pLy-D, which was confirmed by partial sequencing and restriction endonuclease analysis, was transfected into Cos-7 cells and used to inoculate ICR mice via muscular injection. Immunohistochemistry and enzyme-linked immunoabsorbent assay (ELISA) were employed to test the gD expression in transfected cells and the specific anti-HSV-1 antibody in the serum of immunized mice, respectively.Results The gD eukaryotic expression plasmid pLy-D was constructed. Using the immunohistochemistry technique, the gD expression in pLy-D-transfected cells was detected. The ELISA demonstrated that specific anti-HSV-1 antibody could be induced in immunized mice after three times injection.Conclusions We constructed HSV-1 gD eukaryotic expression plasmid pLy-D which could express gD protein in transfected cells and could induce humoral immune response in mice. This observation will be helpful in designing HSV prophylactic vaccine.

Substance P inhibits GABA-activated currents in bullfrog primary sensory neurons by protein kinase C

Objective To research the mechanism or pathway through which Substance P(SP) inhibits r-aminobutyric acid(GABA) activated currents in bullfrog dorsal root ganglion(DRG) neurons.Method The experiment was conducted on freshly isolated bullfrog dorsal root ganglion neurons using a whole-cell patch-clamp technique.Results SP could caused a slow inward current when SP was applied to DRG neurons;SP could inhibits GABA-activated currents;The inhibition could be reduced largely when protein kinase C(PKC) inhibiter,1-(5-isoquinolinesulphorry)-2-methylpiperazine(H-7), was dialyzed in cell body.Conclusion The SP ton inhibit GABA-activated currents through protein kinase C.

Effect of 60 Co γ-ray irradiation on the quality of leukocyte-filtrated red blood cells suspension

Objective To investigate the effect of 60Co γ-ray irradiation on the quality of theleukocyte-filtrated red blood cells suspension.Methods Thirty bags of leukocyte-filtrated red blood cells were randomly divided into irradiation group(n =15) which was given 60Co γ-ray irradiation and control group(n =15) which was not given 60Co γ-ray irradiation.The content of extracellular K +,Na +,free hemoglobin (FHb),adenosine triphosphate (ATP) and 2,3-diphosphoglyceric acid (2,3-DPG) and pH value of the leukocyte-filtrated red blood cells were detected in the two groups.Results The content of K+,Na+,FHb,ATP,2,3-DPG and pH value in irradiation group was (5.20 ± 0.37) mmol · L-1,(140.22 ± 0.05) mmol · L-1,(0.44 ±0.04) mg· L-1,(5.18 ± 0.13)μmol· L-1,(3.34±0.30) mmol· L-1 and 6.85±0.02;the content of K +,Na +,FHb,ATP,2,3-DPG and pH value in the control group was(5.00 ± 0.40)mmol · L-1,(140.47 ± 0.83)mmol · L-1,(0.42 ± 0.08) mg · L-1,(5.22 ± 0.06) μmol · L-1,(3.56 ± 0.52) mmol · L-1 and 6.84 ± 0.05;there was no statistic difference in the content of K +,Na +,FHb,ATP,2,3-DPG,and pH value between the two groups(t =1.93,-1.67,-1.64,-2.02,1.77,1.82;P ＞ 0.05).Conclusion 6o Co γ-ray irradiation of the leukocyte-filtrated red blood cells can not effect the activity and function of red blood cells.
