Objective:To explore the role of constructivism teaching theory combined with problem-based learning (PBL) teaching mode in the training of new nurses in the department of gastroenterology. Methods:A total of 27 newly recruited nurses in the Department of Gastroenterology in West China Hospital of Sichuan University from August 2018 to August 2019 were selected as the control group, and traditional teaching was adopted. In addition, 24 newly recruited nurses from September 2019 to September 2020 were selected as the research group, using constructivism teaching theory combined with PBL teaching mode. After the training, the teaching effect was compared between the two groups through the assessment results and the training method scores. SPSS 19.0 was used for t test. Results:The research group's newly recruited nurses' theoretical evaluation scores [(95.08±5.61) vs. (91.63±5.27)] and practical skills operation evaluation scores [(94.30±7.22) vs. (87.13±6.84)] were both higher than those of the control group, and the difference was statistically significant ( P<0.05). Compared with the control group, the newly recruited nurses of the research group believed that the combination of constructivism teaching theory and PBL teaching could improve clinical thinking ability, analysis and problem-solving ability, and the degree of integration of theory and practice, mobilize the enthusiasm of autonomous learning, enhance autonomous learning ability, and improve communication ability, cultivate teamwork ability, stimulate interest in learning, improve work confidence and training satisfaction, with statistical significance ( P<0.05). Conclusion:The combination of constructivism teaching theory and PBL teaching mode is beneficial to the training of newly recruited nurses in the department of gastroenterology to independently construct a theoretical framework of knowledge, improve comprehensive ability, and make a useful exploration for the training of newly recruited nurses.

Objective To explore the value of HPV-DNA combined with HPV-E6 protein in the detection of cervical cancer. Methods A total of 525 patients with positive TCT were selected and treated with HPV-DNA detection,HPV detection of E6 protein and cervical biopsy.Value of HPV-DNA and HPV-E6 proteins single application and combined detection for cervical cancer screening was compared. Results In the patients with positive TCT,375 cases were normal or cervicitis,134 cases were intraepithelial neoplasia, and 14 cases were cervical cancer. The positive rate of HPV-DNA was significantly higher than that of HPV-E6 protein in cervical precancerous lesions (P < 0.05).The detection rate of cervical cancer was 100%.In comparison of the detection rate of intraepithelial neoplasia and cervical cancer, there were significant differences between the three groups (P < 0.05),and the positive rate of combined detection was higher than that of HPV-DNA and HPVE6 protein. Conclusion With the increasing severity of cervical lesions, the positive detection rate increases gradually.HPV-DNA has higher detection sensitivity, and detection of HPV-E6 protein has high specificity. The combined detection can improve the sensitivity of cervical lesions.

Objective To study and summary the treatment of talus fracture. Methods Fifteen patients with talus fracture were treated by limited cut off reduction by leverage and hollow lag screw under C arm X-Ray machine January 2008 to November 2014, of whom there were 11 males and 4 females, aged 15-61 years old. Seven patients sufferred from talus fracture because of traffic accidents, 4 patients because of falls, 2 patients because of crush injury, and 2 patients because of sprains. Hawkins typing:Ⅰtype in 1 patient, Ⅱ type in 6 patients, Ⅲ type in 6 patients of Ⅲ type, and Ⅳ type in 2 patients. Results Followed up for 6 months to 4 years (mean 1.5 years), fracture healed better. Hawkins grading standard:9 patients showed excellent (1 patient ofⅠtype, 5 patients ofⅡtype, 3 patients ofⅢtype), and two patients showed good (Ⅲtype). The excellent and good rate was 11/15. Necrosis of talus occurred in one patient, and degenerative joint disease occured in one patient. Conclusions Minimally invasive internal fixation with hollow lag screw under C arm X-Ray machine in the treatment of talus fracture has a small injury to the periosteum and the blood supply of small soft tissue. It can fix reliably and can pressurize the fracture fragments, and is conducive to fracture healing.

[ ABSTRACT] AIM:To study the relationship between transcriptional factor c-Jun and Mepe gene expression, and to identify the specific binding site of c-Jun on the promoter of Mepe.METHODS:The expression of c-Jun and Mepe in mouse bone tissue was detected by immunolocalization assay. The mRNA expression of Mepe was determined by real-time PCR when the expression of c-Jun was changed. The techniques of dual luciferase analysis and site-specific mutagenesis were used to measure the effects of c-Jun on the transcriptional activity of Mepe.RESULTS:c-Jun was detected in the nu-cleus of osteocytes, while Mepe was observed in osteocyte cytoplasm. The results of real-time PCR showed that overexpres-sion of c-Jun directly resulted in significantly higher up-regulation of Mepe mRNA.Compared with control group, the tran-scriptional activity of Mepe promoter was increased in osteoblasts which was transfected with pCMV-3Tag-1-c-Jun.Mutation of c-Jun potential binding sites decreased the transcriptional activity of Mepe promoter.CONCLUSION:Mepe gene tran-scription can be up-regulated by c-Jun which binds to the specific sites of Mepe promoter in osteoblasts.
