Laser tweezers Raman spectroscopy ( LTRS ) was used to study the inhibitory effect of indol on staphyloxanthin biosynthesis in Staphylococcus aureus cells and the dynamic changes of this pigment content inside bacterial cells during batch cultivation. The Raman spectra of Staphylococcus aureus cells cultivated for different time and exposed to various doses of indol were acquired. The intensity of 1523 cm-1 band was used for the quantification of staphyloxanthin, in the meantime, the pigment was measured by UV spectrometry. The experimental result showed that an excellent linear relationship existed between the intensities of Raman peak at 1523 cm-1 of bacterial cells and the pigment contents estimated by UV spectrometry, with a correlation coefficient of 0 . 9772 . The spectral data at population level as well as single cell level revealed that indol could inhibit the production of pigment in dose-dependent manner, and the pigment content in bacterial cells incubated with indol decreased by 70%. Under the batch growth condition, the pigment amount in Staphylococcus aureus cells reached the maximum value during the middle exponential growth phase ( 12 h ) and the heterogeneity of pigment content in bacterial cells within certain populations at various time points was relatively small, with RSDs of 39. 2% to 61. 1%. This investigation indicates that LTRS can be served as a reliable method for the analysis of staphyloxanthin content at single cell level.

Objective To investigated the clinical application effects of autonomous copper wire-ECG positioning technology on PICC in very low birth weight infants (VLBWI).Methods The study investigated 54 VLBWI who needed PICC and were hospitalized in the neonatal medical center of Huai′an maternity and child health care hospital in 2015. The participants were divided into the intervention group ( n=27) and the control group ( n=27) using random number table. The intervention group used the autonomous copper wire -ECG positioning technology by observing the changes of typical P-wave while the control group used the conventional locating method. PICC indwelling time, location accuracy, and the incidence of related complications were compared between the two groups according to the gold standard of postoperative chest X-ray. Results The location accuracy for the intervention group was significantly higher than the accuracy for the control group (92.6% vs 66. 7%;χ2=5.594, P<0.05). However, PICC indwelling time for the intervention group had little difference compared to the control group [(20.85±12.5) d vs (22.33±10.0) d; t=0.481,P>0.05] . The incidence of complications also had limited difference between the two groups since there were 4 cases of related complications for the intervention group and 6 cases for the control group (χ2=0.123,P>0.05) . Conclusions The autonomous copper wire-ECG positioning technology should be further developed and applied to the population of VLBWI because of its high accuracy and operability.

Objective To evaluate the efficacy and safety of management of kidney calculi in non-uronephrosis by percutaneous nephrolithotripsy (PCNL) under ultrasound guidance. Methods From July 2005 to June 2008, 97 cases of kidney calculi in non-uronephrosis were performed by percutaneous nephrolithotripsy. A tube was first inserted into the pelvis through cystoscope, and saline was instilled to dilate collecting system. Antegrade percutaneous access was obtained under ultrasound guidance. A combination of pneumatic and ultrasonic lithotrite was used to disintegrate and remove stone under direct vision. Clinical data including operation time, complications and stone free rate were analyzed retrospectively. Results The perutaneous renal access was successfully established under ultrasound guidance in all patients, immediate phase Ⅰ lithotripsy was performed in 95 cases and delayed phase Ⅱ lithotripsy in 2 cases. Operation time was 70-180 min, average time was (96±23 ) min. The average blood loss was 60 ml (20-500 ml), 4 cases had transfusion during the PCNL and average 400 ml. Minor pyrexia ( < 39℃) was seen in 24 cases, whereas serious pyrexia was noted in 3 cases. Conservatively administered with appropriate antibiotics, the fever disapeared in 27 cases within 5 days postoperatively. Severe complications did not occur during nephrolithotripsy. Stones were cleared completely in 78 out of 97 cases (80.4%)during immediate phase Ⅰ lithotripsy, residual stone fragment was found in 19 cases. Conclusion The management of kidney calculi in non-uronephrosis by PCNL appears to be efficacious and safe under ultrasound guidance.
