1.Effect of ANXA1 peptidomimetic Ac2-26 on acute kidney injury and neutrophil apoptosis in septic rats

Objective:To explore the effect of Annexin A1(ANXA1)peptidomimetic Ac2-26 on acute kidney injury(AKI)and neutrophil apoptosis in septic rats.Methods:Experimental groups included control group,Ac2-26 group,AKI group,AKI+Ac2-26 group,with 15 rats in each group.After cecal ligation and perforation were used to establish a sepsis-induced AKI model,Ac2-26 was intravenously infused for treatment,once a day for 14 days;after the end,ELISA was used to detect levels of serum creatinine(Scr),urea nitrogen(BUN),IL-1β,IL-6 and TNF-α;HE staining and periodic acid Schiff(PAS)staining were used to observe the pathological changes of rat kidney tissues in each group;immunohistochemical staining was used to detect expression of ANXA1 in kidney tissue of each group of rats;neutrophils were isolated from rat peripheral blood,Giemsa staining and trypan blue staining were used to detect cell purity and viability;Annexin V-FITC/PI double staining method and TUNEL staining were used to determine apop-tosis level of neutrophils in each group.Results:Compared with control group,levels of Scr and BUN in serum of rats in AKI group were increased(P<0.05),levels of IL-1β,IL-6 and TNF-α also increased(P<0.05),renal tubules and glomeruli in kidney tissue were both significantly damaged,accompanied by a large number of inflammatory cell infiltration,and pathological score increased(P<0.05),while proportion of ANXA1 positive staining area was decreased(P<0.05);neutrophils identified by Giemsa staining and trypan blue staining had complete morphology and high activity;compared with control group,apoptosis rate of neutrophils in AKI group was decreased(P<0.05),and the positive rate of TUNEL was decreased(P<0.05).Compared with AKI group,levels of Scr and BUN in serum of rats in AKI+Ac2-26 group were decreased(P<0.05),levels of IL-1β,IL-6 and TNF-α also decreased(P<0.05),pathological manifestations of renal tubules and glomeruli in renal tissue were significantly reduced,and pathological score was reduced(P<0.05),while the proportion of ANXA1 positive staining area was increased(P<0.05),at the same time,apoptosis rate of rat neu-trophils was increased(P<0.05),positive rate of TUNEL was also increased(P<0.05).Conclusion:ANXA1 peptidomimetic Ac2-26 can increase expression of ANXA1 in kidney tissue of AKI in septic rats,promote neutrophil apoptosis,and have a protective effect on kidney tissue damage in rats caused by sepsis.
2.Children parents attitude and demand towards family based child sexual abuse prevention education

Objective:
To investigate the attitudes and demands of parents of children in Luzhou towards family based child sexual abuse prevention education.
Methods:
A self administered anonymous questionnaire survey was conducted among parents of children in Luzhou City who were selected from stratified cluster sampling. Multiple linear regression model was used to analyze parents attitudes.
Results:
Parents attitude towards prevention of sexual assault education was positive (average score 16.70± 3.67 ). The results of multiple regression analysis showed that parents of only child ( β =0.30), parents who participated in related activities ( β =1.28), communicated with relatives and friends ( β =0.81), and lived in urban areas ( β =0.49) had more positive attitudes. In terms of parental factors, average annual family income higher than 100 000 yuan ( β =0.39), mothers of young children ( β =0.88), and parents with a high level of knowledge about sexual assault prevention education ( β =0.98), the mother being a teacher or a medical staff ( β =0.52), and educational background of the mother being high school/secondary school ( β =1.03), college/undergraduate or above ( β =1.42) were associated with more positive attitudes( P <0.01). The results of demand analysis showed that parents had high demand for child s self protection (96.86%).
Conclusion
Parents of young children in Luzhou City show generally positive attitude and high demand towards family based sexual abuse prevention. Knowledge training and publicity regarding child sexual abuse should be improved for children who had siblings, from rural and township areas, and whose parents with low educational background.
3.Current situation and related factors of family based child sexual abuse prevention education in rural areas of Luzhou

Objective:
To investigate family based child sexual abuse prevention education in rural areas of Luzhou, to provide guidance for child sexual abuse prevention and intervention.
Methods:
By stratified cluster sampling, from December 2021 to January 2022, 1 213 parents were investigated with the simplified scale of knowledge attitude practice of family sexual education. The influencing factors of family sexual education were analyzed by multiple linear regression.
Results:
The score of family sexual abuse prevention education in rural areas of Luzhou was (11.21 ± 3.99), and the pass rate was 51.69%. The results showed that maternal education of junior high school ( β =0.79), senior high school / technical secondary school( β =1.26) and bachelor / college degree or above( β =1.75), mothers to be the main educators ( β =1.29) were positively associated with, while being girls( β =-0.41) and left behind children ( β =-0.59) were negatively associated with family child sex abuse prevention education score( P <0.05). Children received sex education in school( β =0.81), adequate knowledge of sex education of parents ( β =1.11), positive attitudes towards sex education of parents ( β =1.51), communication with relatives and friends regarding sex education of parents ( β =1.94), parents having participated in sex education related activities( β =0.67) were positively associated with family child sex abuse prevention education score( P <0.05).
Conclusion
Family based child sexual abuse prevention education in rural areas of Luzhou is insufficient. Relevant departments need to set up personalized intervention measures according to the different conditions of families, carry out relative education activities, to improve the level of sex education of rural families and improve their awareness of self prevention.
4.Potential treatment with Chinese and Western medicine targeting NSP14 of SARS-CoV-2

The outbreak of coronavirus disease 2019 (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a serious global health threat.This raises an urgent need for the devel-opment of effective drugs against the deadly disease.SARS-CoV-2 non-structural protein 14 (NSP14)carrying RNA cap guanine N7-methyltransferase and 3'-5'exoribonuclease activities could be a potential drug target for intervention.NSP14 of SARS-CoV-2 shares 98.7% of similarity with the one (PDB 5NFY) of acute respiratory syndrome (SARS) by ClustalW.Then,the SARS-CoV-2 NSP14 structures were modelled by Modeller 9.18 using SARS NSP14 (PDB 5NFY) as template for virtual screening.Based on the docking score from AutoDock Vina1.1.2,18 small molecule drugs were selected for further evaluation.Based on the 5 ns MD simulation trajectory,binding free energy (AG) was calculated by MM/GBSA method.The calculated binding free energies of Saquinavir,Hypericin,Baicalein and Bromocriptine for the N-terminus of the homology model were-37.2711 ± 3.2160,-30.1746 ± 3.1914,-23.8953 ± 4.4800,and-34.1350 ± 4.3683 kcal/mol,respectively,while the calculated binding free energies were-60.2757 ± 4.7708,-30.9955 ± 2.9975,-46.3099 ± 3.5689,and-59.8104 ± 3.5389 kcal/mol,respectively,when binding to the C-terminus.Thus,the compounds including Saquinavir,Hypericin,Baicalein and Bromocriptine could bind to the N-terminus and C-terminus of the homology model of the SARS-CoV-2 NSP14,providing a candidate drug against SARS-CoV-2 for further study.
5.Curative effects of network platform transitional nursing in the rehabilitation of schizophrenic patients

Objective:To explore the curative effect of network platform transitional nursing in the rehabilitation of schizophrenic patients.Methods:Using convenient sampling method, a total of 87 patients with schizophrenia who were admitted to Departmentof Psychiatry in the Affiliated Kangning Hospital of Wenzhou Medical Universityfrom February 2018 to January 2019 were selected as the research objects. According to the random number table method, they were divided into the control group (44 cases) and the observation group (43 cases) .The control group was given routine nursing intervention while the observation group was given network platform transitional nursing based on the control group. Before and after the intervention, the psychiatric symptoms of patients in the two groups were evaluated by Brief Psychiatric Rating Scale, medication compliance was evaluated by Medication Adherence Scaleand personal and social function and family function were evaluated by Personal and Social Performance Scale and Family Function Assessment Scale.Results:After 6 months of intervention, the scores of Brief Psychiatric Rating Scale and Medication Adherence Scale in the observation group were lower than those in the control group, and the differences were statistically significant ( P<0.05) .The scores of Personal and Social Performance Scale and Family Function Assessment Scale in the observation group were higher than those in the control group, and the differences were statistically significant ( P<0.05) . Conclusions:In the rehabilitation treatment of schizophrenia, the implementation of network platform of transitional nursing can improve medication compliance of patients, improve their clinical mental symptoms, and finally improve their personal and social functions and family functions.
6.Expression of zinc finger protein 217 in non-small cell lung cancer and its clinical significance

Objective To explore the expression of zinc finger protein 217 (ZNF217) in non-small cell lung cancer (NSCLC) and its correlation with prognosis of patients. Methods A total of 120 NSCLC patients in Chongqing Three Gorges Central Hospital from January 2012 to October 2013 were selected. Immunohistochemical method was used to test the expression of ZNF217 in NSCLC tissues and paracancerous tissues. The correlation of ZNF217 expression with patient's clinicopathological features was analyzed. At the same time, the Kaplan-Meier method and Cox regression model multiple factor analysis method were used to explore the factors affecting the prognosis of patients after NSCLC radical operations. Results ZNF217 mainly existed in cell nucleus of NSCLC. The positive expression rate of ZNF217 in the cancer tissues was higher than that in the paracancerous tissues [52.5% (63/120) vs. 20.1% (25/120), χ 2 = 25.909, P < 0.05]. The positive expression rate of ZNF217 increased with the increase of tumor T stage (χ 2 = 7.333, P = 0.026), N stage (χ 2 = 7.782, P = 0.020) and TNM stage (χ 2 = 11.557, P = 0.003). The overall survival (P = 0.007) and progression-free survival (P = 0.004) of patients with positive ZNF217 were poorer than those of patients with negative ZNF217. Cox multiple factor analysis showed that ZNF217 was an independent risk factor affecting the prognosis of NSCLC. Conclusion ZNF217 is an independent risk factor affecting the prognosis of NSCLC, and it may be a potential target for accurate treatment of NSCLC.
7.Determination of aconitine in biological human sample by UPLC-MS/MS

Objective To develop a simple and fast UPLC-MS/MS method for determination of aconitine in biological human sample. Methods The biological human sample was treated by acetonitrile precipitation, and then analyzed on a UPLC C18(2.1mm×50mm, 1.7μm)column. The mobile phase consisted of acetonitrile-0.1% formic acid with gradient elution, at a flow rate of 0.4 mL/min, at 40℃. UPLC-MS/MS was performed in ESI source with MRM mode for quantification. Results The linear range of the concentration were 0.5~500ng/mL in blood and 1~1000ng/mL for aconitine in blood for aconitine (r>0.995). The relative recoveries of aconitine were in the range of 91.3%~110.2%, and the extraction recoveries were in the range of 72.8%~83.5%. The RSDs of intra-days and inter-day were both less than 14%. Conclusion The method is a simple, fast and could be used for determination of aconitine in biological human sample.
8.Clinical study on the pattern and the risk factors of lymph node metastasis of thoracic esophageal squamous cell carcinoma

Objective This study was to investigate the lymph node metastasis pattern in thoracic esophageal squamous cell carcinoma (ESCC),and analyze the risk factors to provide reference for the delineation of radiotherapy target.Methods A total of 319 patients with thoracic ESCC who underwent surgical treatment at the second hospital of Dalian medical university were analyzed retrospectively.The lymph nodes around esophageal were divided into 20 groups according to the mapping scheme of the American Thoracic Society (ATS) modified by Casson et al.Analyzed the pattern of lymph node metastasis and its relationship with tumor location,tumor length,depth of invasion,pathological grade,and vessel invasion.Results The lymph node metastatic rate was 48.90% (156/319),while the metastatic ratio of lymph node (LMR) was 15.70% (562/3 581).The lymph node metastatic rates had gradually increasing trend with tumor sites descending (upper ESCC 35.48%,middle ESCC 47.06% and lower ESCC 56.43%),but no statistically significance (P > 0.05) was observed.In the whole 20 groups,the higher node metastasis stations of upper thoracic ESCC LMR were 1,2,4,5,7,9 (x2 =27.38,P < 0.05),while the middle thoracic ESCC LMR were 2,4,5,7,8M (x2 =57.77,P < 0.05),and the lower thoracic ESCC LMR were 4,5,7,8L,16,17,20 (x2 =28.88,P < 0.05),with statistically significance.So the main lymph node metastasis stations were paraesophageal nodes,tracheobronchial nodes,paratracheal nodes,aroticopumonary nodes,left gastric nodes,subcarinalnodes and celiac nodes.The univariate analysis revealed that lymph node metastasis was correlated with the tumor length,depth of invasion,pathological grade,and presence of vascular invasion (x2 =6.82,26.04,36.26,4.56respectivcly,P < 0.05).The muhi-variate regression analysis showed that tumor length and depth of tumor invasion were independent risk factors for lymph node metastasis (OR =2.212,2.123,P < 0.05).Conclusions The factors influencing lymph node metastasis in thoracic ESCC include tumor length,depth of invasion,pathological grade,and presence of vascular invasion,which should be carefully considered during the target delineation of radiotherapy for esophageal carcinoma.
9.Determination of methomyl in human blood by LC/MS

Objective To develop a sensitive and simple LC/MS method for determination of methomyl in blood samples.Methods Methomyl was extracted from blood by liquid-liquid extraction with ethyl acetate, and then analyzed on a Zorbax SB-C18 (2.1mm×50mm, 5μm) column. The mobile phase consisted of acetonitrile-0.1% formic acid with gradient elution, at a lfow rate of 0.5 mL/min, at 40℃. LC-MS was performed in ESI source with MRM mode for quantiifcation.ResultsThe linear range of the concentration were 0.05~2.0μg/mL for methomyl (r>0.995). The relative recoveries of methomyl were in the range of 90%~108%. The RSDs of intra-days and inter-day were both less than 15%.Conclusion The method is a simple, quick, sensitive and could be used for determination of methomyl in blood samples.
10.Investigation of the related factors and management countermeasures of clinical nursing security risks

Objective To investigate the related factors and management countermeasures of clinical nursingsecurity risks. Methods From March, 2011 to March, 2014, 136 nursing safety events were analyzed. The security event analysis group was established. The reasons for the development, processing methods and other related security incidents were researched. Results Within the security event, nurses were the main factor. Within the moderated nursing security events, 21 cases were attributed to nurses, 3 cases were attributed to doctors and 2 cases were attributed to other factors. Conclusions Nurses, patients, doctors, hospital administrators, and infrastructure should be strengthenedto meet the need of security risk management. The communication skills, responsibility and skills training are needed to prevent the occurrence of nursing safety incidents.
