1.Mechanism underlying ITGB1-induced drug resistance in gastric cancer based on the circRNA regulatory network

Objective To explore the mechanism underlying ITGB1-induced drug resistance in gastric cancer based on the circRNA-miRNA-ITGB1 regulatory network.Methods Tumor tissue samples were collected from 21 patients with gastric cancer.The ITGB1 gene expression levels were determined using real-time fluorescent quantitative polymerase chain reaction,and circRNA sequencing was performed to compare the differences in circRNAs between patients with low and high ITGB1 expression.BGC-823 cells were trans-fected with si-circ_0027189,si-miR-455,or si-NC and divided into the si-circ_0027189,si-miR-455,or si-NC groups,respectively.The circ_0027189,miR-455,and ITGB1 expression levels in each group and the sensitivity to oxaliplatin were measured.Results The circRNA regulatory network showed that circ_0027189 regulated ITGB1 expression through miR-455.Compared to the si-NC group,the si-circ_0027189 group exhibited decreased expression levels of circ_0027189 and ITGB1,increased expression levels of miR-455,and reduced sensitivity to oxaliplatin.In contrast,the si-miR-455 group showed decreased expression levels of miR-455,increased expression levels of ITGB1,and enhanced sensitivity to oxaliplatin compared to the si-NC group.Conclusion circ_0027189 can increase ITGB1 expression levels by targeting miR-455 expression,ultimately increasing drug resistance in gastric cancer cells.
2.Effects of Platelet-Rich Plasma-Derived Exosomes on Proliferation and Migration of Tendon Stem/Progenitor Cell

Objective To investigate the effects of platelet-rich plasma-derived exosomes(PRP-Exos)on the proliferation and migration of tendon stem/progenitor cell(TSPC).Methods PRP-Exos were extracted through the combination of polymer-based precipitation and ultracentrifugation.The morphology,concentration,and particle size of PRP-Exos were identified by transmission electron microscopy and nanoparticle tracking analysis.The expression levels of surface marker proteins on PRP-Exos and platelet membrane glycoproteins were deter-mined by Western blot analysis.Rat TSPC was extracted and cultured,and the expression of surface marker mol-ecules on TSPC was detected using flow cytometry and immunofluorescence staining.The proliferation of TSPC in-fluenced by PRP-Exos was evaluated using CCK-8 assay and EdU assay.The effect of PRP-Exos on the migration of TSPC was evaluated by cell scratch assay and Transwell assay.Results The extracted PRP-Exos exhibit typi-cal saucer-like structures,with a concentration of 4.9 ×1011 particles/mL,an average particle size of(132.2±56.8)nm,and surface expression of CD9,CD63 and CD41.The extracted TSPC expressed the CD44 pro-tein.PRP-Exos can be taken up by TSPC,and after co-cultured for 48 h,concentrations of 50 and 100 μg/mL of PRP-Exos significantly promoted the proliferation of TSPC(both P<0.001),with no statistical difference be-tween the two concentrations(P=0.283).Additionally,after co-cultured for 24 h,50 μg/mL of PRP-Exos significantly promoted the migration of TSPC(P<0.001).Conclusion Under in vitro culture conditions,PRP-Exos significantly promote the proliferation and migration of rat TSPC.
3.Effects of self-help mindfulness-based stress reduction training on illness uncertainty and sleep quality in atrial fibrillation patients after radiofrequency ablation

Objective:To investigate the effects of self-help mindfulness-based stress reduction (MBSR) training on illness uncertainty andsleep quality in patients with atrial fibrillation (AF) after radiofrequency ablation. To provide the basis for improving the quality of life of these patients.Methods:This was a randam controlled Test. A convenient sampling method was used to select 90 patients with atrial fibrillation who underwent radiofrequency ablation in the First Peoples Hospital of Changzhou from January 2021 to December 2021. According to the random number table method. They were divided into the experimental group and the control group, with 45 cases in each group. The control group received routine nursing, and the observation group received home-based self-help MBSR training based on the routine nursing. Five Facet Mindfulness Questionnaire (FFMQ), Mishel Uncertainty in Illness Scale and the Pittsburgh Sleep Quality Index Scale (PSQI) were used to evaluate the scores of each scale before intervention, 8 weeks and 3 months after intervention.Results:A total of 42 cases were completed in the experimental group and 41 cases in the control group. Repeated measurement analysis of variance showed that there were interaction effect ( F interaction = 90.98, 46.27, 39.61, all P<0.01), time effect ( F time = 298.67, 179.06, 76.32, all P<0.01) and inter-group effect ( F intergroup = 19.88, 4.07, 18.70, all P<0.05) on mindfulness score, disease uncertainty score, total score of sleep quality and scores of all dimensionsin between the two groups. Simple effect analysis showed that scores of all scales in experimental groups were significantly better than those in control group after the intervention ( F values were 11.75 to 66.49, all P<0.01). Conclusion:Self-help MBSR training can improve the level of mindfulness, reduce the feeling of uncertainty and improve the quality of sleep in patients with atrial fibrillation after radiofrequency ablation.
4.Effects of curcumin on acute kidney injury in endotoxic shock rabbits

Objective To investigate the effects of curcumin on acute kidney injury in endotoxic shock rabbits and discuss the possible mechanism.Methods Forty healthy male New Zealand white rabbits aged 2 months were randomly divided into 4 groups:control group (group C),curcumin control group (group Cur,curcumin 50 mg/kg),endotoxic shock group (group L,LPS 5 mg/ kg) and curcumin pretreatment group (group CurL,curcumin 50 mg/kg,LPS 5 mg/kg 30 minutes later),10 in each group.The concentrations of blood urea nitrogen (BUN) and creatinine (Cr) were detected,and the rabbits were sacrificed 6 h after LPS treatment.The pathological changes of renal tissue were examined and the pathological scores were recorded.SOD activities were measured by xanthine oxidase method.MDA contents were assayed according to thiobarbituric acid method.Reverse transcriptase-polymerase chain reaction (RT-PCR) was used to investigate the mRNA levels of Nrf2 and HO 1.The expressions of Nrf2 total protein,Nrf2 nuclear protein and HO-1 protein in the renal tissue were determined by Western blot.Results No abnormal structures were visible in groups C and Cur.The histopathological changes of the kidneys included glomerular shrinkage,cellu lar swelling,vacuolization and desquamation in tubules,interstitial edema and massive inflammatory cells infiltration in group L.Compared with group C,the pathological manifestations was relieved obviously in group CurL.Compared with group C,the pathological score,the concentrations of BUN,Cr,and MDA were obviously increased,while SOD activity was significantly decreased,the mRNA expression of Nrf2 and HO-1,the protein levels of Nrf2 total,Nrf2 nuclear and HO-1 in the renal tissues were distinctly increased in groups L and CurL (P<0.05).Compared with group L,the pathological score,the concentrations of BUN,Cr and MDA were obviously decreased,while SOD activity was significantly increased,the mRNA expression of Nrf2 and HO 1,the protein levels of Nrf2 total,Nrf2 nuclear and HO-1 in the renal tissues were significantly increased in group CurL (P <0.05).Conclusion Curcumin can ameliorate acute kidney injury induced by endotoxic shock in rabbits,and its mechanism may be relate to activation of Nrf2/HO-1 signaling pathway.
5.The effects of the application of grading health education path for the patients with PICC catheter

Objective To explore the effects of the application of grading health education path for the patients with PICC catheter in hematology department. Methods From March to October 2016,a total of 101 patients with PICC catheter in hematology department were recruited and randomly divided into the experimental group (50 cases) and the control group (51 cases) according to different nursing groups. The control group was implemented traditional health education mode while the experimental group was implemented health education path in stages. The disease related knowledge and the PICC catheter maintenance knowledge, satisfaction of health education and the incidence of catheter related complications after intervention between the two groups were compared. Results About the PICC catheter maintenance knowledge, the experimental group got scores : 80-100, a median of 95, a rank 76.38 on average, while the control group:30-80, a median of 50, the average rank 26.12, which indicated statistical significance (χ2=76.618,P<0.01). Satisfaction scores: the experimental group got scores: 70-100, a median of 100, a rank 73.80 on average, the control group: 30-100, a median of 70, the average rank 28.65, the experimental group was significantly higher than the control group (χ2=63.364,P<0.01). For the catheter-related complications, the experimental group was 2.00%(1/50), while 15.69%(8/51) in the control group, there was statistically significant (χ2=4.262,P < 0.05). Conclusions Implementing the grading health education path for patients with PICC catheter can obviously improve the patients of PICC catheter maintenance knowledge and satisfaction with respect to the manner of health education, at the same time can reduce the related complications of catheter.
6.Inspiratory muscle training in patients with chronic heart failure:a literature review

Dyspnea is an important issue for the patients with chronic heart failure. Breathing training, especially the inspiratory muscle training can help to improve the patients′ cardiopulmonary function, endurance and quality of life. In this paper, the foreign research about the method, effect evaluation and appropriate case of inspiratory muscle training in patients with chronic heart failure were reviewed so as to explore a specific strategy of inspiratory muscle training for China′s patients with chronic heart failure.
7.Multi-center study on prevention condition of skin tears in level-III hospitals

Objective To analyze the prevention condition of skin tears of inpatients in level-III hospitals, so as to provide basis for making prevention strategies.Methods 14 level-III hospitals were involved in the cross-section survey. Within the same time period, using the same research tools, methods and procedure, 964 trained nurses inspected patients from head to toes, who were selected by convenience sampling, with hospital stay ≥24 h, with age≥18 years old, and with signature on the informed consent documents, and also investigated implementation of prevention measures. Results A total of 18 806 effective cases were obtained, with 238 locations of skin tears in 199 patients found. Incidence of hospital-acquired skin tears was 1. 06%. Implementation rate of risk assessment and nutrients and water supplement was 3. 92% and 10. 35%. Implementation rate of prevention of falling and falling down from bed was 51. 58% and 47. 20%. Usage of assistant tools and emollients accounted for 28. 23% and 2. 69%. Adopting suitable moving techniques and wearing long sleeves, trousers or long stockings accounted for 8.07% and 6.95%. Differences of incidence rate, risk assessment and implementation of prevention measures of skin tears among teaching hospitals, comprehensive hospitals and specialty hospitals were statistically meaningful ( P < 0. 05 for all ) . Conclusions Risk assessment and prevention of skin tears in these 14 level-III hospitals were not satisfactory. Prevention measures vary from different types of the hospitals. In future, nurses should be trained to strengthen awareness of risk assessment and prevention, and implementation rate and consistency of preventive measures should be improved.
8.Investigation of the knowledge level of the caregivers about pressure ulcer in patients with high risk of pressure ulcer in rural community

Objective To investigate the knowledge level of the caregivers about pressure ulcer in patients with high risk of pressure ulcer in rural community,and to provide the scientific basis for the prevention and treatment of pressure ulcer.Methods 216 caregivers for patients with high risk of pressure ulcer in rural community were chosen from Mar.2012 to May 2013.The investigation on the awareness of pressure ulcer was performed by using questionnaire survey.Results The awareness rate of pressure ulcer in caregivers was 41.5 %.The caregivers had gender difference (174females and 42 males).Most of the caregivers were 41 to 50 years old (124 cases,57.4%).Most caregivers had 1 ~ 2 years work experience (94 cases,43.5%).Most of them had low level of education (170 cases,78.7%) and non-professional training (178 cases,82.4%).The questionnaire score was higher in young caregivers than in the olders(F=2.483,P<0.05).The questionnaire score was higher in caregivers having a long working lifetime(F=3.624,P<0.05),higher educationalbackground (F=5.139,P< 0.01) and discipline training (t=7.346,P< 0.01) than in caregivers not having them.Conclusions The awareness rate of the knowledge about pressure ulcer is low in caregivers for patient with high risk of pressure ulcer in rural community.It is necessary to strengthen the guidance and training in caregivers to reduce the incidence of pressure ulcers and the medical cost,and improve the quality of life in patients with high risk of pressure ulcer in rural community.
9.Reference intervals for liver function tests in normal pregnant women in the last trimester of pregnancy

Objective To explore changes of serum levels of liver function indicators and their reference intervals in pregnant women in the last trimester of pregnancy ,and to analyse its clinical significance .Methods From July 2013 to June 2014 ,a total of 4 659 cases of pregnant women in the last trimester of pregnancy and 3 089 healthy non‐pregnant women were enrolled in this stud‐y .Serum levels of alanine aminotransferase(ALT) ,aspartatre transaminase(AST) ,total protein(TP) ,albumin(Alb) ,pre‐albumin (PA) ,total bilrubin(TBIL) ,direct bilrubin(DBIL) ,indirect bilrubin(IBIL) ,γ‐glutamyl transpeptidase(GGT)and mitochondrial as‐partate aminotransferase(m‐AST) were determined by fully automatic biochemical analyzer .Results Compared with healthy non‐pregnant women ,serum levels of ALT ,AST ,TP ,Alb ,PA ,TBIL ,DBIL ,IBIL and GGT were significantly decreased ,while serum le‐vers of ALP ,m‐AST and TBA were significantly increased in pregnant women in the last trimester of pregnancy ,there were statis‐tically significant differences(P<0 .05) .Conclusion This study suggested that the liver functions of pregnant women were signifi‐cantly different from healthy non‐pregnant women .Each clinical laboratory should establish reference intervals of liver function indi‐cators for pregnant women at each stage of pregnancy ,so as to timely ,correctly and reasonably evaluate their liver functions and material nutrition status ,and provide better antenatal care services .
10.SNP microarray analysis of retention abortion chorionic villus.

OBJECTIVETo compare villus cell culture and karyotype analysis with single nucleotide polymorphism (SNP) microarray technology for the detection of chorionic villus chromosome in patients with retention of abortion.
METHODSForty cases were analyzed with the two methods.
RESULTSChorionic villus culturing was successful in 29 cases, among which 10 were found to have an abnormal karyotypes. For the SNP microarray analysis, all 40 cases were successful, among which 16 were shown to have an abnormal molecular karyotype.
CONCLUSIONSNP microarray technology is highly accurate and specific, which is particularly suitable for the detection of chromosomal deletions or duplications, uniparental disomy, low-percentage mosaicism and other chromosomal abnormalities. It has provided an effective supplement to the conventional chorionic villus culture and karyotype analysis.
