1.Progress of clonal evolutionary basis of FLT3-ITD-mutant acute myeloid leukemia and the effect of its different characteristics on the prognosis

The incidence of FLT3-internal tandem duplication (ITD) mutation in acute myeloid leukemia (AML) is approximately 20%-30% and FLT3-ITD mutation generally indicates a poor prognosis. Although FLT3 inhibitors have greatly improved the efficacy of AML patients with this type of AML, relapse and drug-resistance increasingly become prominent. ITD mutations lead to dimerization and continuous self-activation of FLT3 in the absence of ligands, and cause non-ligand-dependent phosphorylation. Different characteristics of ITD, including allele ratio, length, insertion site, wild-type mutation content and co-mutated genes, could affect the prognosis of patients. The underlying mechanism of these factors has an important guiding significance for clinical prognosis, drug application and treatment strategy.
2.Determination of HLA-A, -B allele polymorphism in the Luoba nationality living in Tibet Autonomous Region in China.

OBJECTIVETo investigate the HLA-A, -B allele polymorphism in the Luoba ethnic population.
METHODSHLA-A, -B DNA types in 92 healthy individuals of Luoba nationality in the Linzhi area, Tibet Autonomous Region, were investigated by polymerase chain reaction-sequence specific oligo-nucleotide (PCR-SSO).
RESULTSTen alleles at HLA-A locus, and 19 alleles at HLA-B locus in Luoba ethnic group were detected. Of the 10 HLA-A alleles detected, the three most common alleles were HLA-A*11(allele frequency: 36.40%), -A*02 (25.50%), -A*24 (23.90%), and they covered 85.80% of the total HLA-A alleles detected from the Luoba ethnic group. Of the 19 HLA-B alleles detected, the three most common alleles were HLA-B*40 (27.20%), -B*15 (11.40%) and -B*38(10.90%), and they covered 49.50% of the total -B alleles detected in the Luoba ethnic group.
CONCLUSIONThe distribution of HLA-A, -B allele polymorphism in the Luoba nationality is distinctive, but some of the gene distribution in the Luoba group is nearer to that in the Tibetan group. These are consistent with the results of ethnological, historical and sociological researches.

3.The prevalence of vascular lesions in the lower extremities and their risk factors in type 2 diabetic mellitus

Objective To investigate the prevalence of vascular disease in the lower extremities and its related risk factors in type 2 diabetic patients.Methods A total of 393 type 2 diabetic patients treated in 5 hospitals in Beijing were randomly recruited for the study with their age of more than 40 years at onset,diabetes duration of more than 5 years and treatment for more than 1 year in each hospital.The case history,physical examination,biochemical tests and ultrasonic Doppler examination were conducted in these patients.Results Eight patients had gangrene (2.0%) and 28(7.1%) had intermittent claudication in 393 diabetics,while ultrasonic examination showed that 357 cases (90.8%) had vascular lesions in the lower limbs.Ankle/brachial systolic pressure ratio (API,Ankle pressure index) less than 1.0 was found in 68 cases (17.8%).The higher prevalence of diabetic nephropathy and retinopathy,higher fasting blood sugar and HbAlc,higher diastolic pressure and lower HDL ch levels ( P
