1.The effect of down-regulating lncRNA SPINT1-AS1 on the growth and invasion of ovarian cancer cells by targeting miR-211-5p

Objective :
To explore the effect of long-chain non-coding RNA(lncRNA) SPINT1-AS1 on the growth and invasion of ovarian cancer cells and the molecular mechanism.
Methods :
The expression of lncRNA SPINT1-AS1 in ovarian cancer tissues and normal tissues was analyzed by GEPIA database.Fluorescence real-time quantitative polymerase chain reaction(qRT-PCR) method was used to determine the expression of lncRNA SPINT1-AS1 in ovarian cancer cell lines SKOV-3,A2780,OC3,HO-8910 and immortalized ovarian epithelial cell line IOSE80.The SKOV-3 cells were divided into si-SPINT1-AS1 group(transfected with lncRNA SPINT1-AS1 siRNA) and si-NC group(transfected with siRNA control).The effect of down-regulated lncRNA SPINT1-AS1 on the proliferation and invasion of SKOV-3 cells was determined by MTT method and Transwell chamber experiment.Western blot was used to detect the expression of proliferative phenotypic proteins(PCNA,CyclinD1) and metastatic phenotypic proteins(MMP2,MMP9).The dual luciferase reporter system was used to identify the targeting relationship between lncRNA SPINT1-AS1 and miR-211-5 p.The effect of down-regulated lncRNA SPINT1-AS1 on the expression of miR-211-5 p was detected by qRT-PCR.
Results:
Compared with normal tissues, the expression level of lncRNA SPINT1-AS1 in ovarian cancer tissues was higher(P<0.01).Compared with IOSE80 cells, the expression level of lncRNA SPINT1-AS1 in ovarian cancer cell lines SKOV-3,A2780,OC3,HO-8910 increased(P<0.05),and the expression level of lncRNA SPINT1-AS1 in SKOV-3 cells was the highest(P<0.01).Compared with the si-NC group, the absorbance of SKOV-3 cells in the si-SPINT1-AS1 group was reduced(P<0.05),the number of invasive cells was reduced(P<0.05),and the expression of proliferation phenotype proteins PCNA and CyclinD1 were reduced(P<0.01),the expression of metastatic phenotypic proteins MMP2 and MMP9 was reduced(P<0.01).lncRNA SPINT1-AS1 could directly bind to miR-211-5 p(P<0.01).Compared with the si-NC group, the expression level of miR-211-5 p in SKOV-3 cells of the si-SPINT1-AS1 group increased(P<0.01).
Conclusion
The expression of lncRNA SPINT1-AS1 in ovarian cancer tissues and cell lines increased.Down-regulation of lncRNA SPINT1-AS1 can inhibit the proliferation and invasion of ovarian cancer SKOV-3 cells by targeting up-regulation of miR-211-5 p.
2.Safety and Preliminary Efficacy of Ceritinib 450 mg with Food in Chinese ALK-positive Non-small Cell Lung Cancer.

BACKGROUND:
Anaplastic lymphoma kinase (ALK) rearrangement is a common driver gene of non-small cell lung cancer (NSCLC). Ceritinib is a second-generation ALK inhibitor, which can bring survival benefits to ALK-positive metastatic NSCLC. However, few studies focus on the safety and efficacy of ceritinib in China. Therefore, this study intends to investigate the safety and preliminary efficacy of ceritinib 450 mg with meals in Chinese patients with ALK-positive NSCLC through a real world study.
METHODS:
From October 2018 to December 2019, patients with ALK-positive NSCLC from 8 medical centers in Sichuan province were recruited in this study. All of these participants received ceritinib 450 mg/d with food. The basic characteristics, adverse effects (AEs) and responses were collected and analyzed in order to evaluate the safety and efficacy of ceritinib.
RESULTS:
A total of 109 patients were included in this study. Data cutoff was January 23, 2020. The median duration of treatment exposure was 5.87 mon (range: 0.4 mon-15.7 mon). Total AEs were reported in 98 (89.9%) of 109 patients and grade 3 or 4 AEs were reported in 22.9% of patients. Most common AEs (mainly grade 1 or 2) were diarrhea (60.6%), elevated alanine aminotransferase (ALT)(38.5%) and aspartate aminotransferase (AST)(37.6%). As of data cutoff, 45 patients discontinued ceritinib. The overall response rate (ORR) was 37.6% (95%CI: 28.5%-47.4%) and disease control rate (DCR) was 86.2% (95%CI: 78.3%-92.1%).
CONCLUSIONS
The treatment of ceritinib 450 mg with food for Chinese ALK-positive NSCLC patients had a good safety profile and favorable DCR in real-world setting. However, this conclusion needs to be further verified by large sample, prospective trials.
3.Screening and confirmation of TP antibody and analysis of syphilis infection status among 12 295 blood donors in Huainan area

Objective:To understand the current situation of syphilis infection among 12 295 blood donors in Huainan area, so as to provide reference for the management of public health.Methods:The Treponema pallidum(TP) antibody of the blood donors was screened by enzyme-linked immunosorbent assay, and the unqualified blood donors for TP antibody were confirmed by Treponema pallidum particle agglutination(TPPA).Results:There were 48 blood donors TPPA positive among 12 295 blood donors in Huainan area, and the TPPA positive rate was 3.90‰(48/12 295). There was no statistically significant difference in the TPPA positive rate of the different gender blood donors[male 3.12‰(22/7 048) vs.female 4.96‰(26/5 247)] in Huainan area( u=1.61, P>0.05). There was statistically significant difference in the TPPA positive rate of the different marriage blood donors[married 5.33‰(42/7 884) vs.unmarried 2.01‰(6/2 989)] in Huainan area( u=2.33, P<0.05). There was statistically significant difference in the TPPA positive rate of the blood donors among different education level in Huainan area(χ 2=29.94, P<0.05). There was no statistically significant difference in the TPPA positive rate of the blood donors among ABO blood groups in Huainan area(χ 2=2.24, P>0.05). Conclusion:There are significant differences in the TPPA positive rates of the blood donors among marital status and education level in Huainan area.
4.Relationship between adjuvant radiotherapy dose and pathologic complete response in patients with locally advanced esophageal squamous cell cancer

Objective:To investigate the relationship between the dose of preoperative neoadjuvant radiotherapy and the pathologic complete response (pCR) rate in patients with locally advanced squamous cell esophageal cancer (ESCC).Methods:Clinical data of 116 patients with ESCC who received neoadjuvant chemoradiotherapy followed by esophagectomy in our cancer center from July 2017 to December 2019 were retrospectively analyzed. The radiation doses were divided into 2 ranges based on Grays (Gy) received: 40-45 Gy and 45 Gy or more.Results:The overall pCR rate was 38. 8%(45/116). pCR was observed in 35 out of 80(44%) patients treated with 40-45 Gy and 10 of 36(28%) patients treated with 45 Gy or more. The pCR rate did not significantly differ between two groups [(40-45 Gy) vs.( ≥ 45 Gy), P=0.105)]. Conclusions:Preoperative neoadjuvant radiotherapy with a higher dose (≥ 45 Gy) fails to increase the pCR rate in patients with locally advanced ESCC. Prospective randomized trials are required to determine the optimal dose of preoperative adjuvant radiotherapy.
5. Analysis of features of carotid artery color doppler ultrasonography in diabetes mellitus patients complicated with cerebral infarction

Objective:
To analyze the changes of carotid artery ultrasound in diabetes mellitus patients complicated with cerebral infarction.
Methods:
From March 2017 to June 2018, 55 type 2 diabetes patients complicated with cerebral infarction in the Second People's Hospital of Yuhang District were selected as study group.Another 50 diabetes patients without cerebral infarction were selected as simple group, and 60 healthy people were selected as control group.The carotid artery colour doppler check was implemented, the carotid artery ultrasound media thickness(IMT) index and carotid plaques were compared in three groups.
Results:
The IMT index in the study group was (1.84±0.23), which was significantly higher than (1.32±0.21) in the control group (

Objective To investigate the correlations of distribution and grading of deep medullary veins (DMVs) with clinical prognoses of patients with acute anterior circulation infarction by using susceptibility-weighted imaging (SWI).Methods Fifty patients with acute anterior circulation infarction,admitted to our hospital from August 2016 to December 2017,were enrolled in our study.SWI was performed to evaluate the DMVs.Univariate and multivariate Logistic regression models were used to determine the correlation between distribution of DMVs and clinical prognoses of patients.DMVs on the ipsilateral side of the lesions were further graded and analyzed;boxplot was used to describe its relation with modified Rankin scale (mRS) scores of the patients.Results DMVs were observed in 36 patients (72%),with 19 ipsilateral DMVs and 17 contralateral DMVs.The ipsilateral DMVs were independently associated with poor outcome (odds ratio=3.380,95%CI:1.006-11.393,P=0.049).The contralateral DMVs were not independent predictors for outcomes,but appeared commonly in patients with good outcome (44.8%).In patients with ipsilateral DMVs,grading 1,grading 2,and grading 3 were noted in 7,two and three patients,respectively;boxplot analysis showed that DMVs patients of grading 3 had higher mRS scores,with an average of 4.Conclusion The ipsilateral DMVs on SWI are independent predictive biomarkers for poor clinical outcome after stroke,and contralateral DMVs often indicate good prognosis.
7.The study of nicotine addiction abstinence in patients with insular infarction

Objective To investigate the effect of insular on the addiction of nicotine after insular infarction. Methods Totally 35 patients with lesions involving insular infarction were enrolled in insular damage group. The patients matched with the demographic data of the study subjects who did not involve in-sular infarction were the control group(n=114). All patients were given the internationally popular question-naire of smoking urges ( QSU) and fagerstrom test for nicotine dependence( FTND) during hospitalization, and they were followed up 3 months after stroke. The QSU and the minnesota nicotine withdrawal scale ( MNWS) were assessment for patients who did not smoke again and did not relapse,and the QSU and FTND scale were used for the patients who still smoke. Results After 3 months follow-up,the rate of withdrawal from insular damage was 77. 14%,and that in non-insular damage was 52. 63%(χ2=6. 62,P=0. 01),and the MNWS in two groups(insular damage patients (0. 59±0. 97) and non-insular damage patients(1. 15± 1. 29)),the difference was statistically significant (t=-2. 00,P=0. 04). All subjects were evaluated by questionnaire of smoking urges (QSU),among which QSU score was (19. 40±9. 63) in insular damage group and (24. 47±10. 39) in non-insular group,the difference was statistically significant (t=-2. 57,P=0. 01) . Conclusion Patients with insular lesion are more likely to cause withdrawal of nicotine addiction, suggesting that insular may be involved in cognitive processing of addiction.
8.Questionnaire investigation of the awareness of doctors' attitude to geriatric syndrome in old patients with cancer

Objective To improve the quality of the management of the elderly patients with cancer in Sichuan province,and to understand the attitudes and perspectives of elderly surgeons and oncologists for the management and treatment of elderly patients with cancer.Methods A face-toface questionnaire interview was conducted with oncologists (n 64) and geriatricians (n =64).128 physicians were involved in this study.Results The cancer management and therapeutics were deemed appropriate at present by 9.38% (6/64)of the geriatricians and 25.00% (16/64)of the oncologists.The 39.06% (25/64) of geriatricians used to notice geriatric syndromes,while 81.25% (52/64)of oncologists never concerned about the geriatric syndrome(P=0.011).As for the causes of the therapy-associated toxicity,oncologist versus geriatrician payed an attention to malnutrition (100.00% vs.100.00% in both groups),to mobility disorders(65.63% vs.65.63%,84/128 in both groups),to cognitive impairment/mood disorder (89.06% in geriatrician group vs.75.00% in oncologist group,P=0.038).For the factors affecting treatment decisions,a physical ability attention (oncologists vs geriatricians:70.31% vs.92.19%;P =0.002),and comorbidity (oncologists vs geriatricians:62.50% vs.79.69%,P =0.032) had statistically significant difference.In addition,lack of geriatrics knowledge was also reported by more oncologists.However,one hundred percent of participants wanted very much to cooperate with each other in their clinical work.When responding to the clinical scenario,the 10.94% (7/64)of geriatricians and 32.81% (21/64)of oncologists chose modified treatment for 65-74 years old patient with cancer(P =0.003).When the age of the patients was 75-84 years old,only 12.50 % (8/64) of geriatricians prefer end-of-life care,while 31.25 % (20/64) of oncologists chose it (P =0.010).Conclusions Selection of treatment decisions in the elderly patients with cancer affect by ageing.Both oncologists and geriatricians are concerned with the elderly patients with cancer,ageing syndrome,total sickness and functional status.And these doctors support an establishment of a multi disciplinary team cooperation for the elderly patients with cancer.Therefore,the establishment of mutual cooperation between the two professionals is necessary and feasible.
9.Comparative analysis of the clinical value of two-dimensional and three-dimensional contrast-enhanced ultrasonography in the diagnosis of uterine submucosal myoma

Objective To compare the clinical value of uterine submucosal myoma classification by two dimensional and three-dimensional contrast-enhanced ultrasonography and surgical pathologic results.Methods The imaging data of ultrasonographic hysterography including 2D and 3D of 124 patients with uterine submucosal myoma were retrospectively analyzed,and the results were compared with the surgical pathologic results.The diagnostic accuracy of uterine submucosal myoma classification and the operation success rate of uterine submucosal myoma for Ⅰ grade by ultrasonographic hysterography including 2D and 3D were compared.Results The patients were diagnosed pathologically with 0,Ⅰ and Ⅱ grade of uterine submucosal myoma in 26 cases,52 cases,68 cases,respectively.The patients were diagnosed by 2D ultrasonic sonohysterography with 0 grade,Ⅰ grade and Ⅱ grade of uterine submucosal myoma in 26 cases,62 cases,58 cases,respectively.The patients were diagnosed by 3D ultrasonic sonohysterography with 0,Ⅰ and Ⅱ grade of uterine submucous myoma in 26 cases,52 cases,68 cases,respectively.For pathological results as thegold standard,the diagnostic sensitivity,specificity and accuracy of uterine submucosal myoma for 0 grade by 2D and 3D ultrasonic sonohysterography were all 100.00%.The diagnostic sensitivity,specificity and accuracy of uterine submucosal myoma for Ⅰ and Ⅱ grade by 2D ultrasonic sonohysterography were 92.32%,79.46%,85.00%,respectively.The diagnostic sensitivity,specificity and accuracy of uterine submucosal myoma for Ⅰ and Ⅱ grade by 3D ultrasonic sonohysterography were 96.24%,88.24%,91.76%,respectively.There were significant differences in the diagnostic sensitivity,specificity and accuracy of submucosal myoma of uterus for Ⅰ and Ⅱ grade by 2 D and 3 D ultrasonic sonohysterography (x2 =3.21,2.78,2.17,2.33,all P < 0.05).The patients diagnosed as uterine submucous myoma for 0 grade all underwent the hysteroscopic surgery for successful resection,while the patients with uterine submucous myoma for Ⅱ grade underwent laparoscopic surgery or open surgery.The operation success rates of uterine submucous myoma for Ⅰ grade by hysteroscopic surgery diagnosed by 2D and 3D ultrasonic sonohysterography were 75.81%,98.07%,respectively.The operation success rate of uterine submucous myoma for Ⅰ grade by hysteroscopic surgery diagnosed by 3D ultrasonic sonohysterography was significantly higher than that diagnosed by 2D ultrasonic sonohysterography (x2 =7.15,P < 0.05).Conclusion The accuracy of uterine submucosal myoma classification by 3D ultrasonographic hysterography is better than 2D ultrasonographic hysterography.
10.Changes in APPL1 expression during renal fibrosis in a mouse model of renal ischemia-reperfusion injury

Objective To evaluate the changes in the expression of adaptor protein containing pleck-strin homobgy domain, phosphotyrosine-binding domain and a leucine zipper motif 1(APPL1)during renal fibrosis in a mouse model of renal ischemia-reperfusion(I∕R)injury. Methods Twenty-four male C57BL∕6 mice, aged 8 weeks, weighing 20-25 g, were divided into 2 groups(n=12 each)using a random number table: sham operation group(S group)and renal I∕R group.The model of renal I∕R injury was established by clipping the bilateral renal pedicles for 30 min followed by reperfusion in group I∕R.Six mice were selected at 2 days of reperfusion, and venous blood samples were collected for determination of serum concentrations of blood urea nitrogen and creatinine.The animals were then sacrificed, the renal specimens were obtained for microscopic examination of tubular necrosis with a light microscope, and the damage to the renal tubules was scored using a semi-quantitative method.Six mice were sacrificed at 14 days of reperfusion, and the renal specimens were obtained for assessment of the degree of renal fibrosis(using picric acid-sirius red staining) and for determination of the expression of collagen type 1, fibronectin and α-smooth muscle actin in renal tis-sues(by Western blot or immunofluorescence method). At 2 and 14 days of reperfusion, the expression of APPL1 in renal tissues was detected by Western blot and the expression of APPL1 mRNA in renal tissues by real-time polymerase chain reaction. Results Compared with group S, the serum concentrations of blood u-rea nitrogen and creatinine, scores of renal tubular damage and degree of renal fibrosis were significantly in-creased at 2 days of reperfusion, the expression of collagen type 1, fibronectin and α-smooth muscle actin in renal tissues was up-regulated at 14 days of reperfusion, and the expression of APPL1 protein and mRNA was up-regulated at 2 and 14 days of reperfusion in group I∕R(P<0.05). Conclusion Up-regulated expression of APPL1 may be involved in the process of renal fibrosis in a mouse model of renal I∕R injury.
