1.Predictive value of microvessel density and lymphatic vessel density in pancreatic cancer tissue for tumor distant metastasis within 1 year after surgery

Objective:To investigate the value of counting microvessel density (MVD) and lymphatic vessel density (LVD) in predicting distant metastasis of pancreatic cancer within 1 year after surgery.Methods:The clinicopathological data of 47 patients with pancreatic cancer who underwent surgery in Laibin People's Hospital from January 2020 to December 2021 were retrospectively analyzed. The patients were divided into non-metastasis group( n=24) and metastasis group( n=23) according to whether distant metastasis occurred during 1-year follow-up. Immunohistochemistry was used to detect the CD 34 expression in microvascular epithelial cells and D2-40 level in lymphatic epithelial cells from pancreatic cancer tissues. MVD and LVD in cancer tissues and adjacent normal tissues were counted. The relationship between MVD and LVD in cancer tissues and clinicopathological characteristics such as gender, age, tumor diameter, tumor differentiation, lymph node metastasis, vascular invasion, nerve invasion and tumor stage were analyzed. The receiver operating characteristic curve (ROC) was drawn and the area under the curve (AUC) was calculated to evaluate the value of MVD and LVD in predicting distant metastasis of pancreatic cancer within 1 year after surgery. The effects of MVD and LVD on the distant metastasis rate of pancreatic cancer within one year after operation were evaluated. Univariate and multivariate logistic regression were used to analyze the independent influencing factors for distant metastasis of pancreatic cancer within 1 year after surgery. Results:MVD and LVD in metastatic cancer tissues were higher than those in adjacent normal tissues [(72.52±9.73) vs (51.73±7.95)/400 times field of view, (23.78±6.87) vs (14.00±5.66)/400 times field of view]. MVD and LVD in the non-metastasis group were also higher than those in the adjacent normal tissues [(63.20±6.52) vs (54.79±5.80)/400 times field of view, (16.25±5.15) vs (13.62±5.03)/400 times field of view], and all the differences were statistically significant ( P<0.05). MVD in cancer tissue was significantly increased in patients with tumor diameter ≥2 cm, lymph node metastasis, vascular invasion and high TNM stage ( P<0.05), and LVD was significantly increased in patients with tumor diameter ≥2 cm, lymph node metastasis, moderate and low differentiation, vascular invasion, nerve invasion and high TNM stage ( P<0.05). The AUC values of MVD and LVD in predicting distant metastasis of pancreatic cancer within 1 year after surgery were 0.799 (95% CI 0.659-0.939) and 0.803(95% CI 0.676-0.929), and the cut-off values were 70.5 and 20.5/400 times field of view, respectively. The sensitivity was 73.9% and 69.6%, and the specificity was 87.5% and 83.7%. The cumulative distant metastasis rate within 1 year after operation in high MVD and high LVD groups was significantly higher than that in low MVD and low LVD groups ( P<0.05). Multivariate logitic regression analysis showed that tumor diameter ≥2 cm ( OR=1.757, 95% CI 1.536-3.846, P<0.05), lymph node metastasis ( OR=2.364, 95% CI 1.036-4.175, P<0.05), high MVD ( OR=4.345, 95% CI 1.245-3.736, P<0.05) and high LVD ( OR=3.637, 95% CI 1.426-4.035, P<0.05) were independent risk factors for distant metastasis of pancreatic cancer within 1 year after surgery. Conclusions:Increased MVD and LVD in pancreatic cancer tissues are independent influencing factors for distant metastasis within 1 year after surgery, which can be used to predict whether patients have distant metastasis within 1 year after surgery.
2.Protective effects of total glucosides of paeony on the injury of neonatal rat cadiocytes ;induced by H2O2

Objective To investigate the protective effects and mechanism of total glucosides of paeony (TGP) on the injury of neonatal rat cadiocytes induced by H2O2. Methods Cadiocytes of 3 days neonate rat was cultivated for 72 hours and divided into the model group, the TGP low (30μg/ml), medium (60 μg/ml), high (120 μg/ml) dose groups, Shuxuening injection (100 μg/ml) group and normal control group (n=8);and except the normal control group, the cells in other groups were induced by H2O2 (100μg/ml). After 6 hours, the morphology changes were observed and the survival rates were detected;the content of AST, CPK, LDH in culture medium were detected; the activity of SOD, CAT, GSH-Px and the content of MDA in cardiomyocytes were determinted;the apoptosis rates were detected. Results Compared with the model group, the survival rates of cardiomyocytes in TGP medium and high dose groups significantly improved (64.1%± 7.3%, 81.3%± 7.8%vs. 42.3%± 6.2%;P<0.05 or P<0.01);the content of AST (26.72 ± 6.03 U/ml, 23.86 ± 5.38 U/ml vs. 34.75 ± 6.91 U/ml), CPK (1.96 ± 0.35 U/ml, 1.59 ± 0.32 U/ml vs. 2.54 ± 0.40 U/ml), LDH (811.55 ± 162.32 U/L, 683.48 ± 134.14 U/L vs. 936.07 ± 140.94 U/L) in culture medium were significantly decreased (P<0.05 or P<0.01);the activity of SOD (85.34 ± 16.87 U/mg , 94.62 ± 17.75 U/mg vs. 56.45 ± 13.76 U/mg), CAT (22.76 ± 3.38 U/mg, 26.49 ± 3.72 U/mg vs. 16.13 ± 3.18 U/mg) in cardiomyocytes were significantly increased and the content of MDA (8.74 ± 2.05 nmol/mg, 6.91 ± 1.80 nmol/mg vs. 11.56 ± 2.19 nmol/mg) were significantly decreased (P<0.05 or P<0.01), the apoptosis rate (24.2% ± 5.5%, 13.4% ± 3.9% vs. 51.2% ± 9.1%) were significantly decreased (P<0.05 or P<0.01);and the activity of GSH-Px (3.54 ± 0.83 U/mg vs. 2.50 ± 0.67 U/mg) in cardiomyocytes of TGP high dose treatment group was significantly increased (P<0.05 or P<0.01). Conclusion TGP had dose-dependent protective effects on the injury of neonatal rat cadiocytes induced by H2O2, which perhaps related to its effects of improving the activity of antioxidase, inhibiting the oxidative stress, and decreasing the apoptosis rates.
3.Effect of clinical pharmacist interventions on the rational use of antibiotics

Objective To study the effect of clinical pharmacist interventions on the rational use of antibiot-ics.Methods 1 000 hospitalized patients before the implementation of intervention in April 2012 to April 2013 (control group)and the other 1 000 cases of hospitalized patients in May 2013 to May 2014 after the implementation of the intervention (study group)were selected,the antibiotics use rate,hospitalization time and cost,cost of using antibacterial drugs were compared between the two groups.Results The antibiotics use rate of the study group was 56.0%(560/1 000),significantly lower than 78%(780/1 000)of the control group,with significant differences between the two groups(χ2 =11.089,P=0.032),the hospitalization time,cost of using antibacterial drugs of the study group were (12.6 ±0.8)days,(912.2 ±13.2)yuan,significantly better than (16.9 ±0.7)days,(1 528.1 ± 32.5)yuan of the control group,with significant differences between the two groups (t=9.892,10.142,P=0.028, 0.014);The two groups of hospitalization expenses showed no statistical significance (t =4.984,P =0.072 ). Conclusion Clinical pharmacist intervention has a positive effect on the application of antibacterial drugs,which can significantly reduce the use of antimicrobial drugs and reduce the hospitalization days.
4.Application of 16-detector row spiral CT for pulmonary angiography and reconstruction techniques in diagnosis of pulmonary embolism in elderly patients

Objective To study the application of pulmonary angiography and reconstruction techniques with 16-detector row spiral CT in the diagnosis of pulmonary embolism (PE) in the elderly.Methods Twenty-four elderly patients with suspected pulmonary thromboembolism received detection of CT and pulmonary artery angiography ( CTPA ) on 16-detector MSCT.Post-reconstruction techniques included maximum intensity projection (MIP), volume rendering (VR)and multiple planar reformation (MPR). Results A total of 161 pulmonary artery thrombi were detected in 24 elderly patients. The direct signs of pulmonary embolism included total occlusion (16.8%, 27/161), partial filling defect (67.7%, 109/161) and central filling defect or track sign (15.5%, 25/161). A total of 161 pieces of pulmonary thrombi were detected in transect image, 153 (95.0%) in MPR, 113 (70.2%) in MIP and 69 (42.9%) in VR. The transect image excelled evidently MIP and VR image in displaying pulmonary thrombi, especially the thrombi in pulmonary lobe and pulmonary artery branch (χ2 =235.36 and 243.41, P<0.05). Conclusions The 16-detector row spiral CT pulmonary angiography is non-invasive, fast and high sensitive, it should be the first choice for the diagnosis of PE in the elderly.
5.Nutrient uptake characteristics of nitrogen, phosphorus and potassium of Pinellia ternate from Sichuan during growth stage.

OBJECTIVETo study the absorption characteristics of nitrogen (N) , phosphorus (P) , and potassium (K) of Pinellia ternate from Sichuan under the cultivated condition.
METHODThe samples of P. ternate S-29 and S-33 from Sichuan were regularly collected randomly, the contents of nitrogen, phosphorus and potassium of each part (aerial and underground parts) were determined, and their distribution and accumulation characters of nitrogen, phosphorus, potassium were also studied.
RESULTThe absorption rate of aerial and underground parts of P. ternate were different in nitrogen, phosphorus, and potassium at different growing stages. Among them, the potassium uptake was the highest, while the phosphorus uptake was the lowest. Totally, the uptakes of nitrogen, phosphorus and potassium of P. ternate from Sichuan during "sprout tumble" stage were comparatively higher than that not at "sprout tumble" stage. A significant correlation was observed between the phosphorus and potassium uptake of the aerial and underground parts, respectively (P < 0.05). And there was also significant positive correlation between the nitrogen and phosphorus uptake at underground parts (P < 0.05). During the growth stage of P. ternate, for producing 100 kg air-dried P. ternate, 5.02-5.64 kg N, 2.44-2.56 kg P205, 11.62-13.49 kg K20 were needed to be absorbed.
CONCLUSIONP. ternate from Sichuan should be given enough base fertilizer in field cultivation, and then be supplied with phosphorus and potassium by combining foliar spray with field application during the middle ten days of June and last ten days of August. The ratios of potassium should be properly improved. There was promoting effect in the phosphorus and nitrogen assimilations of the underground parts of P. ternate from Sichuan.

6.Drought resistance of Angelica dahurica during seedling stage under polyethylene glycol (PEG-6000)-simulated drought stress.

OBJECTIVETo study the optimum conditions of simulated drought stress, and screen the indexes of drought resistance and comprehensively assess the drought resistance of the Angelica dahurica resources during seedling stage.
METHODInvestigations were carried out on the changes of height, root length, root-shoot ration, contents of soluble sugar, proline and malondialdehyde under polyethylene glycol (PEG-6000)-simulated drought stress. A comprehensive evaluation on the drought resistance of different (varietal) species of A. dahurica during seedling stage was applied by using the method of subordinate function. And the drought resistance indexes were selected out by applying the method of grey correlative degree analysis.
RESULTDrought stress of 9 days with 20% PEG was the optimum condition for the simulation of drought stress. The results showed that the drought resistant capability decreased in the order as follows, A. dahurica from Sichuan province, A. dahurica from Henan province, A. dahurica from Hebei province and A. dahurica from Zhejiang province. And the order of correlative degree of drought resistance and indexes was: soluble sugar > root length > proline > root-shoot ration > total content of chlorophyll > chlorophyll b > chlorophyll a > height > malondialdehyde.
CONCLUSIONOsmotic adjustment substance and the indexes related to the root have more influence on the drought resistance of A. dahurica during seedling stage. A. dahurica from Sichuan province shows the highest drought resistance during seedling stage.

7.Correlation analysis on main agronomic characters, yield per plant, artemisinin percentage contents and total artemisinin content per plant of Artemisia annua.

OBJECTIVETo study the relationships among the main agronomic characters, yield per plant,content of artemisinin of Artemisia annua, and provide the theoretical basis for breeding the A. annua varieties with high-yield and high artemisinin content.
METHODThe main agronomic characters, yield per plant, artemisinin content of the wild and cultivated A. annua of Sichuan province were investigated and determined, and the correlated relationships among them were analyzed.
RESULTIt was showed that the weight of dried stem per plant had very significantly positive correlation with the yield per plant,and the number of primary branches, the diameter of stem had significantly positive correlation with the yield per plant. The flower bud stage, first flowering stage and full-bloom stage had significantly positive correlation with the artemisinin content. The first flowering stage and full-bloom stage had significantly positive correlation with the total artemisinin content per plant. The weight of dried stem per plant,the diameter of stem and the flower bud stage had significantly correlation with the total artemisinin content per plant. A. annua with the purple-stem had significant higher artemisinin content than that with the green-stem. Through the multiple regression analysis, the classic multiple regression equation of the yield per plant was gained as Y = 0.06121-0.000 08X(1) + 0.30378X(3) - 0.00051X(4) - 0.00031X5 (F = 20.15, P < 0.0001), the classic multiple regression equation of the artemisinin content as Y = -1.45849 - 0.00099X(1) - 0.24079X(3) + 0.13362X(6) - 0.00719X(7) + 0.01454X(9) (F = 46.55, P < 0.0001), the classic multiple regression equation of the total artemisinin content per plant as Y = -2.67001 - 0.00249X(1) + 0.98873X(3) + 0.30037X(6) + 0.113X(9) F = 22.62, P < 0.001).
CONCLUSIONThe plant with purple, thick and strong stem, higher plant height, moderate primary branches number and bloom stage, small stem-branch angle should be selected for a breeding new A. annua varieties with high yield and artemisinin content.

8.Genetic diversity of Fritillaria from Sichuan province based on ISSR.

OBJECTIVETo provide more proofs for expounding the genetic relationships among the (varietal) species in genus Fritillaria from Sichuan province.
METHODThe ISSR marker technique was used to study relationships and genetic polymorphism of nineteen populations in ten species and one varietal species of genus Fritillaria. Genetic similarities were calculated by using NTSYS software and the dendrogram was constructed by using UPGMA method.
RESULTEleven primers were selected from 35 ISSR primers, and 179 DNA fragments were amplified from 19 populations. Of which, 179 fragments were polymorphic (percentage of polymorphic bands was 86.8%). The genetic similarity among all accessions ranged from 0.569 to 0.855. Clustering analysis showed that the 19 populations of Fritillaria could be distinctively classified into 4 groups. F. cirrhosa, F. przewalskii, F. cirrhosa var. logirnectarea and F. dajitensis were in the first group; The second group was the cluster of F. cirrhosa and F. mellea (wild and cultivated species); The third group was F. sulcisquamosa, F. thunbergii, wabunesis and F. delavayi; F. hupehensis alone formed the fourth group.
CONCLUSIONISSR marker technique is suitable for the genetic diversity of Fritillaria from Sichuan province. Interspecific identifications among the four original species of Bulhus Fritillariae Cirrhosae recorded by pharmacopoeia of China, and between them and the other species of genus Fritillaria from Sichuan province could not be gained by using ISSR markers technique. In addition, the cluster result of genus Fritillaria had some relationships with the geographical distribution.

Objective: To compare the pharmacodynamic action of Radix Notoginseng after fermented by different strains bioconversion.Methods:The models of cerebral ischemia and hypoxia were established,thrombosis in vivo were induced by collagen-Adr,ventricular fibrillation of mice by chloroform,and perfusion experiment of the ex vivo heart of cavia cobaya was used.Results:100,200mg/kg doses of all samples had protective effects on ischemia and hypoxia mice,1# sample can inhibit thrombosis induced by collagen-adrenaline obviously after intraperitoneal injection in dosage of 200mg?kg-1?d-1 for 5days.All groups significantly reduced the incidents of ventricullar fibrillation induced by chloroform in mice except 100mg/kg dose of 1#,furthermore,compared with control high dose group(5#),the incidents of high dose groups of 3、4# deceased significantly.All samples had the effect of dilating coronary artery,but there effects on myocardial contractile force and cardiac preload were different.Conclusion:The primary cardiovascular related pharmacological researches demonstrated the vary action changes of 1-4# samples compared with the sample 5#.This result has revealed that the chemical structure of Radix Notoginseng transformed and that is coincide with initial analysis of phytochemical analysis.
