1.Oncostatin M expression in endometrial cancer and its correlation with immune cell infiltration

Objective:To explore expression and prognostic value of oncostatin M(OSM)in endometrial cancer and to analyze relationship between OSM expression and immune cell infiltration in endometrial cancer tissues.Methods:OSM expression in pan-can-cer was analyzed by TIMER database,OSM expression in endometrial cancer and normal tissues was compared,and survival analysis for patients with different OSM expression was performed;relationship between OSM expression and immune cell infiltration was analyzed by TIMER and TISIDB,and ssGSEA algorithm was used to calculate difference in abundance of immune cell infiltration in samples with different OSM expression;GSEA software was applied to perform enrichment analysis;clinical tissue samples were collected for validation.Results:OSM expression was higher in endometrial cancer tissues than that in normal endometrial tissues(P=4.1e-28),and endometrial cancer patients with high OSM expression had prolonged recurrence-free survival(RFS)(P=0.004 8).OSM expression was positively correlated with abundance of immune cell infiltration and genetic markers of immune cells(P<0.05).OSM was mainly enriched in immune-related signaling pathways.OSM expression was higher in endometrial cancer tissues than normal and atypical hyperplastic tissues(P=0.016 9).Proportions of immune cell markers CD4,CD8,and CD68 were increased in tumor tissues with high OSM expression(all P<0.05),which were positively correlated with OSM expression.Conclusion:OSM is highly expressed in endometrial cancer tissues and correlated with prognosis;OSM expression is positively correlated with immune cell infiltration level and can be used as a biomarker for immunotherapy and prognosis.
2.Advances in the microbial production of the compatible solute ectoine: a review.

Ectoine is an amino acid derivative and an important natural product in halophilic microorganisms. It plays an important role in protecting cells and stabilizing biological macromolecules, and can be widely used in biomedical fields such as drug preparation adjuvants, organ transplantation and preservation, skin wound repair and cosmetics. Due to the medical value and commercial market demand of ectoine, this article summarized the recent advances in the microbial production of ectoine, including the mutation and breeding of hyper-producing strains, construction of genetically and metabolically engineered strains, optimization of fermentation processes, and extraction and purification processes. The application of multi-omics technologies and computational biology to develop an ectoine producing cell factory was prospected, with the aim to provide a reference for ectoine overproduction.

3.Construction of quality evaluation index system for in-hospital insulin pump use based on "structure-process-result" theory

Objective:To construct an evaluation index system for quality of use of insulin pumps in hospitals, in order to provide a reference for evaluating quality of use of insulin pumps in hospitals in clinical work.Methods:Based on the three-dimensional quality evaluation model of "structure-process-result", the first draft was determined through literature review and group discussion from January to February 2022. Using the Delphi expert correspondence method, 26 experts from 13 provinces/municipalities in China were consulted in two rounds to determine the quality evaluation index system for the use of insulin pumps in the hospital.Results:The recovery rate of the two rounds of expert questionnaire was 100%, and the expert authority coefficient was 0.971. The mean value of the second round of index importance assignment was 4.500-5.000 and the coefficient of variation was 0-0.22. The final construction of quality evaluation index system of insulin pump use in the hospital included 3 first-level indicators, 15 second-level indicators and 37 third-level indicators.Conclusions:The constructed quality evaluation index system of insulin pump use in the hospital is scientific, reasonable and practical, which has guiding significance for evaluating the quality of insulin pump use in the hospital.
4.Preparation technology of oyster hydroxyapatite porous material for bone repair

Objective To verify the feasibility of applying hydrothermal synthesis for preparing oyster hydroxyapatite(HA) and to develop a preparation method of oyster HA porous material for bone repair.Methods Hydrothermal synthesis was applied for preparing oyster HA,and the reaction condition was 220 ℃ for 6 h.Then,the prepared oyster HA was used as the raw material for porous scaffold preparation by sponge-soaking and sintering,successively.The porosity and compressive strength of the scaffold were adjusted by controlling the soaking time and absorbed HA slurry of the sponges.Results Hydrothermal synthesis was an effective method for preparing oyster HA.When the volume of the sponge cube was 1 cm3,the material absorbed by one to three times sponge-soaking were 0.184 8 g,0.318 1 g and 0.426 1 g,respectively,the corresponding porosity were 91.5%,82.9% and 78.5%,and the compressive strength were 1.06 MPa,3.99 MPa and 8.49 MPa.Conclusion The oyster shell powder can be effectively converted into HA under the hydrothermal reaction condition of 220 ℃ for 6 h.The preparation of HA porous bone repair material by sponge-soaking method can obtain ideal porosity and mechanical strength.However,in this preparation process,the number of sponge-soaking and the weight of the absorbed HA slurry should be exactly controlled in order to obtain desired properties.
5.Molecular epidemiology of urogenital Chlamydia trachomatis infection in patients attending sexually transmitted diseases clinics

Objective To determine the prevalence and genotype profiles of urogenital Chlamydia trachomatis and their related factors in patients attending sexually transmitted diseases (STD) clinics in Guangxi Zhuang Autonomous Region. Methods C. trachomatis was screened by a plasmid PCR in 598 patients attending STD clinics. Then, positive specimens underwent nested-PCR to amplify the major outer membrane protein 1 (ompl) gene. The amplicons of ompl gene were digested by restriction endonucleases Alu I plus Hinf I and Cfol . C. trachomatis was differentiated according to the restriction fragment length polymorphism (RFLP) patterns. Results Out of the 598 samples, 83 were positive for plasmid-PCR. The prevalence of chlamydial infection was 13.9% with no significant difference between male and female patients. Nested-PCR based RFLP analysis showed that genotype E amounted to 27.7% (23/83), F 20.5% (17/83), D 13.2%(11/83), G 12.0%(10/83), K 7.2%(6/83), H 4.8%(4/83), I 3.6% (3/83), J 3.6%(3/83)and uncertain types 7.2% (6/83). Visible symptoms were observed less frequently in infections with C. trachomatis genotypes E and F compared with the other genotypes, while low abdominal pain occurred in 80% of infections with type G. Conclusions A certain proportion of out-patients attending STD clinic are infected with various types of C. trachomatis in Guangxi Zhuang Autonomous Region. The polymorphism of ompl gene may serve as a useful tool in molecular epidemiological studies of C. trachomatis.
