Pathological vascular remodeling is a hallmark of various vascular diseases. Previous research has established the significance of andrographolide in maintaining gastric vascular homeostasis and its pivotal role in modulating endothelial barrier dysfunction, which leads to pathological vascular remodeling. Potassium dehydroandrographolide succinate (PDA), a derivative of andrographolide, has been clinically utilized in the treatment of inflammatory diseases precipitated by viral infections. This study investigates the potential of PDA in regulating pathological vascular remodeling. The effect of PDA on vascular remodeling was assessed through the complete ligation of the carotid artery in C57BL/6 mice. Experimental approaches, including rat aortic primary smooth muscle cell culture, flow cytometry, bromodeoxyuridine (BrdU) incorporation assay, Boyden chamber cell migration assay, spheroid sprouting assay, and Matrigel-based tube formation assay, were employed to evaluate the influence of PDA on the proliferation and motility of smooth muscle cells (SMCs). Molecular docking simulations and co-immunoprecipitation assays were conducted to examine protein interactions. The results revealed that PDA exacerbates vascular injury-induced pathological remodeling, as evidenced by enhanced neointima formation. PDA treatment significantly increased the proliferation and migration of SMCs. Further mechanistic studies disclosed that PDA upregulated myeloid differentiation factor 88 (MyD88) expression in SMCs and interacted with T-cadherin (CDH13). This interaction augmented proliferation, migration, and extracellular matrix deposition, culminating in pathological vascular remodeling. Our findings underscore the critical role of PDA in the regulation of pathological vascular remodeling, mediated through the MyD88/CDH13 signaling pathway.

Ankylosing spondylitis (AS) combined with lower cervical fracture is often categorized into unstable fracture, with a high incidence of neurological injury and a high rate of disability and morbidity. As factors such as shoulder occlusion may affect the accuracy of X-ray imaging diagnosis, it is often easily misdiagnosed at the primary diagnosis. Non-operative treatment has complications such as bone nonunion and the possibility of secondary neurological damage, while the timing, access and choice of surgical treatment are still controversial. Currently, there are no clinical practice guidelines for the treatment of AS combined with lower cervical fracture with or without dislocation. To this end, the Spinal Trauma Group of Orthopedics Branch of Chinese Medical Doctor Association organized experts to formulate Clinical guidelines for the treatment of ankylosing spondylitis combined with lower cervical fracture in adults ( version 2024) in accordance with the principles of evidence-based medicine, scientificity and practicality, in which 11 recommendations were put forward in terms of the diagnosis, imaging evaluation, typing and treatment, etc, to provide guidance for the diagnosis and treatment of AS combined with lower cervical fracture.

ObjectiveTo explore whether miR-375 regulates the malignant characteristics of osteosarcoma (OS) by influencing the expression of MMP13. MethodsPlasmid DNAs and miRNAs were transfected into OS cells and HEK293 cells using Lipofectamine 3000 reagent. Real-time quantitative polymerase chain reaction was performed to measure the expression of miR-375 and MMP13 in OS patients and OS cells. Western blot was performed to analyze the MMP13 protein in the patients with OS and OS cells. The targeting relationship between miR-375 and MMP13 was analyzed by luciferase assay. Migration and invasion were analysed by heal wound and transwell assays, respectively. ResultsmiR-375 expression in OS tissues was lower than that in normal tissues. The expression of MMP13 was upregulated in OS tissues. MMP13 expression was negatively correlated withmiR-375 expression in patients with OS. Migration and invasion were significantly inhibited in OS cells with the miR-375 mimic compared with OS cells with the miRNA control. MMP13 partially reversed the inhibition of migration and invasion induced by miR-375 in the OS cells. ConclusionmiR-375 attenuates migration and invasion by downregulating the expression of MMP13 in OS cells.

Objective To explore the medication rules in the patented Chinese medicine(CM)compound formulas for the prevention and treatment of respiratory infectious diseases, and to provide reference for the prevention and treatment of respiratory infectious diseases. Methods CM compound formulas for the prevention and treatment of respiratory infectious diseases were collected from the national patent database. After data screening and standardization,R language was used for data mining. Results A total of 429 patented CM compound formulas were included, involving 846 Chinese medicinals. There were 26 kinds of high-frequency Chinese medicinals, and the top 10 frequently-used drugs were Glycyrrhizae Radix et Rhizoma,Lonicerae Japonicae Flos,Scutellariae Radix,Forsythiae Fructus,Platycodonis Radix,Stemonae Radix,Armeniacae Semen Amarum,Astragali Radix,Bupleuri Radix, and Menthae Haplocalycis Herba. Most of the high-frequency Chinese medicinals were heat-clearing and toxin-removing drugs and qi-replenishing drugs. The medicinal properties of the patented CM compound formulas were usually cold, the medicinal flavors were usually bitter, sweet and pungent, and mostly had the meridian tropism of lung meridian. The association rule analysis yieled 19 core association rules and multiple drug combinations.Three drug clusters were obtained after cluster analysis. Conclusion For the prevention and treatment of respiratory infectious diseases, patented CM compound formulas are commonly formulated following the principles of compatibility of cold and warm drugs, compatibility of drugs for dispersing and descending, usually have the actions of dispersing pathogens in the lung, clearing heat and removing toxins, and also have the actions of replenishing qi and harmonizing the middle energizer, and nourishing yin and moistening dryness. The formulas have the efficiency of eliminating pathogens while not hurting the healthy qi,and strengthening the healthy qi while not maintaining pathogens.

Objective To investigate the effect of magnetic resonance diffusion tensor imaging(DTI)combined with Buyang Huanwu decoction on limb rehabilitation of patients with spontaneous hypertensive cerebral hemorrhage in basal ganglia. Methods A total of 68 patients with spontaneous hypertensive intracerebral hemorrhage in basal from March 2022 to March 2023 were selected from the department of neurosurgery of the First People's Hospital of Huzhou as research object.According to the random number table method, the patients were divided into control group(30 cases)and experimental group(38 cases). The control group was diagnosed with hypertensive intracerebral hemorrhage and received conventional therapy, and the experimental group was treated with DTI combined with Buyang Huanwu decoction. After 4 weeks of treatment, the overall treatment effect of the two groups was compared, and the differences of Fugl-Meyer assessment(FMA),National Institutes of Health Stroke Scale(NIHSS),Berg balance scale(BBS),and modified Barthel index(MBI)scores were compared. Results The overall therapeutic effect of experimental group was significantly higher than that of control group[86.8%(33/38)vs.56.7%(17/30),P<0.05].After 4 weeks of treatment,FMA,BBS and MBI scores were significantly increased in both groups,NIHSS scores were significantly decreased in both groups, and the improvement degree of the experimental group was significantly better than that of the control group(FMA score:74.21±9.07 vs.66.73±12.76,NIHSS score:15.16±3.73 vs.20.27±6.15,MBI score:69.29±14.68 vs.60.63±19.10,BBS score:43.55±4.30 vs.36.90±3.86,all P<0.05).Conclusion DTI combined with Buyang Huanwu decoction can improve the motor function of the limbs of patients in the basal ganglia area and enhance their ability to perform activities of daily living.
