1.Effects of miR-155 on apoptosis of chronic myeloid leukemia cells and expression of heat shock proteins

Objective :
To explore the effects of microRNA-155 (miR-155) on apoptosis of chronic myeloid leukemia ( CML) cells,and the influence of miRNA-155 regulating the expression of heat shock proteins ( HSP) 27, HSP60,HSP70.
Methods :
Reverse transcription-quantitative real-time PCR ( RT-qPCR) was used to detect the expression levels of miR-155 in CML-resistant imatinib (IM) cell line K562-G and CML cell line K562 .K562-G cells were infected with the lentivirus carrying miR-155 or the negative control lentivirus ,and they were named miR-155 group and control group.The effect of miR-155 on the proliferation of drug-resistant cells was detected by cell counting kit-8 ( CCK-8) method. RT-qPCR and Western blot were used to detect the effect of miR-155 on the expression of heat shock proteins HSP27,HSP60,HSP70.Flow cytometry was used to detect the percentage of cell apoptosis in miR-155 group and control group.
Results :
ompared with K562 cells,miR-155 showed low expres- sion in K562-G cells (P <0. 05) .The proliferation of miR-155 group cells decreased significantly from the 36th hour (P<0. 05) .Compared with the control group,in the miR-155 group,HSP60 and HSP70 increased (P < 0. 05) ,while HSP27 decreased (P<0. 01) .The apoptosis rate of miR-155 group was higher than that of control group (P<0. 05) .
Conclusion
miR-155 promotes the apoptosis of chronic myeloid leukemia cells,increases the expression of HSP60 and HSP70,and decreases the expression of HSP27 .
2.Association of urinary cadmium level with body mass index and body circumferences among older adults over 65 years old in 9 longevity areas of China

Objective:To investigate the association of urinary cadmium level with body mass index (BMI) and body circumferences among the older adults over 65 years old in 9 longevity areas of China.Methods:Subjects were older adults over 65 years old from the Healthy Aging and Biomarkers Cohort Study (HABCS) between 2017 and 2018 conducted in 9 longevity areas in China. A total of 1 968 older adults were included in this study. Information including socio-demographic characteristics, lifestyles, diet intake, and health status was collected by using questionnaires and physical examinations. Urine samples were collected to detect urinary cadmium and creatinine levels. Body circumferences included waist circumference, hip circumference and calf circumference. Subjects were divided into three groups (low:<0.77 μg/g·creatinine, middle:0.77-1.69 μg/g·creatinine, high:≥1.69 μg/g·creatinine) by tertiles of creatinine-adjusted urinary cadmium concentration. Multiple linear regression models were used to analyze the association of creatinine-adjusted urinary cadmium level with BMI and body circumferences. The dose-response relationship of creatinine-adjusted urinary cadmium concentration with BMI and body circumferences was analyzed by using restrictive cubic splines fitting multiple linear regression model.Results:The mean age of subjects was (83.34±11.14) years old. The median (Q1, Q3) concentration of creatinine-adjusted urinary cadmium was 1.13 (0.63, 2.09) μg/g·creatinine, and the BMI was (22.70±3.82) kg/m 2. The mean values of waist circumference, hip circumference, and calf circumference were (85.42±10.68) cm, (92.67±8.90) cm, and (31.08±4.76) cm, respectively. After controlling confounding factors, the results of the multiple linear regression model showed that for each increment of 1 μg/g·creatinine in creatinine-adjusted urinary cadmium, the change of BMI, waist circumference, hip circumference, and calf circumference in the high-level group was -0.28 (-0.37, -0.19) kg/m 2, -0.74 (-0.96, -0.52) cm, -0.78 (-0.96, -0.61) cm, and -0.20 (-0.30, -0.11) cm, respectively. The restrictive cubic splines curve showed a negative nonlinear association of creatinine-adjusted urinary cadmium with BMI ( Pnonlinear<0.001) and negative linear associations of creatinine-adjusted urinary cadmium with waist circumference ( Plinear<0.001), hip circumference ( Plinear<0.001), and calf circumference ( Plinear<0.001). Conclusion:Urinary cadmium level is significantly associated with decreased BMI, waist circumference, hip circumference and calf circumference among older adults over 65 years old in 9 longevity areas of China.

Background::Carotid intima-media thickness (IMT) and diameter, stiffness, and wave reflections, are independent and important clinical biomarkers and risk predictors for cardiovascular diseases. The purpose of the present study was to establish nationwide reference values of carotid properties for healthy Chinese adults and to explore potential clinical determinants.Methods::A total of 3053 healthy Han Chinese adults (1922 women) aged 18-79 years were enrolled at 28 collaborating tertiary centers throughout China between April 2021 and July 2022. The real-time tracking of common carotid artery walls was achieved by the radio frequency (RF) ultrasound system. The IMT, diameter, compliance coefficient, β stiffness, local pulse wave velocity (PWV), local systolic blood pressure, augmented pressure (AP), and augmentation index (AIx) were then automatically measured and reported. Data were stratified by age groups and sex. The relationships between age and carotid property parameters were analyzed by Jonckheere-Terpstra test and simple linear regressions. The major clinical determinants of carotid properties were identified by Pearson’s correlation, multiple linear regression, and analyses of covariance.Results::All the parameters of carotid properties demonstrated significantly age-related trajectories. Women showed thinner IMT, smaller carotid diameter, larger AP, and AIx than men. The β stiffness and PWV were significantly higher in men than women before forties, but the differences reversed after that. The increase rate of carotid IMT (5.5 μm/year in women and 5.8 μm/year in men) and diameter (0.03 mm/year in both men and women) were similar between men and women. For the stiffness and wave reflections, women showed significantly larger age-related variations than men as demonstrated by steeper regression slopes (all P for age by sex interaction <0.05). The blood pressures, body mass index (BMI), and triglyceride levels were identified as major clinical determinants of carotid properties with adjustment of age and sex. Conclusions::The age- and sex-specific reference values of carotid properties measured by RF ultrasound for healthy Chinese adults were established. The blood pressures, BMI, and triglyceride levels should be considered for clinical application of corresponding reference values.
4.Research progress on role of mitochondrial calcium uniporter in neurodegenerative diseases

With the world aging aggravation,the prevalence of neurodegenerative diseases has risen year by year,which brings a huge burden on society and family.However,the effective treatment drugs are still scarce,there is an urgent need to discover new therapeutic targets and develop new therapeutic strategies.Recent studies have found that the the mitochondrial calcium uniporter(MCU)has an important function in many neurodegenerative diseases,the imbalance of mitochondrial calcium homeostasis caused by its abnormal expression or function is one of the important mechanisms of neurodegenerative diseases.Inhibiting the MCU function can play the good neuroprotective role,the specific inhibitors of MCU have shown the good prospect in treating various neurodegenerative diseases.This paper sketched the structure,function,regulatory mecha-nism and specific inhibitors of MCU,and focused on analyzing the role of MCU in the occurrence and develop-ment of different neurodegenerative diseases in order to provide reference for further exploration of MCU as a new therapeutic target for neurodegenerative diseases.

Postoperative pain seriously affects the recovery process of patients, resulting in prolonged hospital stay and increased care costs. Appropriate application of patient-controlled analgesia devices can effectively relieve perioperative acute pain. In 1994 patient-controlled analgesia began to be used in Peking Union Medical College Hospital, and the Acute Pain Service Working Group was established in 2004. With the cooperation of anesthesiologists and specialist nurses, the group jointly has implemented the whole process and standardized management based on patient-controlled analgesia, and constantly improved and innovated working methods, laying a solid foundation for the development of postoperative pain management. This paper systematically reviews and summarizes the work from the aspects of clinical focus, nursing management experience, promotion and dissemination of pain treatment concepts, and development of acute pain service model under the new situation, with the hope of providing valuable reference for comprehensively strengthening pain management in the process of diagnosis and treatment, and enhancing patients' satisfaction with perioperative analgesia services.

7.Impacts of muscle visual feedback intervention combined with continuous nursing on exercise ability of patients with knee osteoarthritis

Objective:To observe the impacts of muscle visual feedback intervention combined with continuous nursing on exercise ability of patients with knee osteoarthritis (KOA), so as to provide a reference for improving the rehabilitation quality of KOA patients.Methods:Using a randomized controlled trial method. The 104 KOA patients admitted to the Department of Arthrology and Sports medicine of the Affiliated Hospital of Jining Medical University from December 2020 to December 2021 were selected as the research objects by convenient sampling method. They were divided into the control group and the research group according to the random number table method, with 52 cases in each group. The control group was given routine care, and the research group was given muscle visual feedback intervention combined with continuous care on the basis of the control group. Before and after the intervention, the knee joint function, range of motion of the affected knee joint and exercise ability of the two groups were compared.Results:After the intervention, the Hospital for Special Surgery (HSS) Knee scores of pain, function, range of motion, muscle strength, flexion deformity and stability in the study group were 21.39 ± 4.21, 17.08 ± 2.43, 13.92 ± 2.02, 8.52 ± 0.54, 8.12 ± 0.74 and 8.04 ± 0.89, which were higher than those of the control group 19.68 ± 4.14, 16.12 ± 2.08, 13.11 ± 1.89, 7.76 ± 0.62, 6.85 ± 0.68, 7.11 ± 0.94, and the differences were all statistically significant ( t values were 2.09-9.11, all P<0.05). After the intervention, the flexion-extension, internal rotation and external rotation of the affected knee in the study group were (90.83 ± 14.26)°, (20.89 ± 3.72)° and (30.72 ± 3.86) °, respectively, and those in the control group were (76.78 ± 24.74)°, (16.81 ± 4.63)° and (24.62 ± 5.07) °, respectively, and the differences were statistically significant ( t=3.55, 4.95, 6.90, all P<0.05). After the intervention, the Fugl-Meyer assessment scale score of the study group was 24.72 ± 4.38, which was higher than 21.15 ± 4.64 of the control group, and the difference was statistically significant ( t=4.04, P<0.05). Conclusions:Muscle visual feedback intervention combined with continuous nursing can improve the knee joint function, range of motion of the affected knee joint and exercise ability of KOA patients.
8.The effect of sinomenine on proliferation and apoptosis of multiple myeloma U266 cells through regulation of STAT3 and NF-κB signaling pathways

Objective :
To investigate the effect of sinomenine on proliferation and migration of multiple myeloma (MM) cells by regulating STAT3 and NF-κB signaling pathway.
Methods :
The cultured U266 cells were treated with different concentrations of sinomenine (0,0.5,1,2 mmol / L) .The control group was added DMSO with 0.5% concentration.CCK-8 assay was used to detect the proliferation of U266 cells.Flow cytometry was used to detect the apoptosis of U266 cells.Western blot assay was used to detect the expression levels of apoptosis-related proteins, STAT3 and NF-κB signaling pathway proteins in the each group.
Results :
Compared with CON group,the apopto- sis of U266 cells increased after Sinomenine treatment,the proliferation was inhibited ; B lymphoma-2 (Bcl-2) mye- loid and cell leukemia-1 (Mcl-1) expression level decreased ; activated Caspase-3 (cleaved Caspase-3) and PARP (Cleaved Caspase-3) expression levels increased ; the activity of STAT3 and NF-κB signaling pathway decreased.
Conclusion
Sinomenine can down-regulate the activity of STAT3 and NF-κB signaling pathway,promote the apop- tosis of U266 cells and inhibit the proliferation of U266 cells.
9.Detection and multi⁃locus sequence typing of low⁃affinity penicillin⁃binding protein of β⁃lactam resistant Enterococcus faecalis

Objective :
To detect the low⁃affinity penicillin⁃binding protein drug resistance , pbp4 gene , and multi⁃ locus sequence typing (MLST) of clinically isolated E. faecalis .
Methods :
78 clinical isolates of E. faecalis were collected , and their drug resistance was detected by automated instruments ; the mutation of pbp4 gene mutation was analyzed by PCR amplification and MLST .
Results :
78 strains of E. faecalis were highly resistant to ciprofloxacin , levofloxacin , rifampicin , erythromycin , tetracycline and high concentration of gentamicin , and were resistant to penicillin and gentamicin . The ampicillin resistance rate was 10. 3% , and no strains were found to be resistant to nitrofurantoin , vancomycin , teicoplanin and linezolid ; 8 strains of 78 E. faecalis had amplified TEM genes , and all of them were resistant to penicillin and ampicillin resistance , with a positive rate of 10. 3% ; the allelic profiles and sequence types of 78 strains of E. faecalis which were divided into 16 sequence types , of which ST179 and ST16 were the most , with 21 and 21 strains , respectively . 20 strains , accounting for 26. 9% and 25 . 6% , the rest were ST6 type 8 strains (10. 3% ) , ST4 type 7 strains (9 . 0% ) , ST585 type 6 strains (7 . 7% ) , ST480 type 4 strains (5 . 1% ) , ST28 strains 3 strains (3 . 8% ) of the ST type were detected , and only 1 strain was detected for the oth⁃ er ST types . The analysis of the relationship between ST types and drug resistance showed that E. faecalis with the same ST type had similar drug resistance profiles .
Conclusion
The resistance mechanism of E. faecalis to β ⁃lactam antibiotics is mainly caused by the production of β⁃lactamase mediated by TEM gene , which is not necessarily related to the mutation of pbp4 gene . The isolates of E. faecalis are mainly CC16 ( including ST16 and ST179) clones and drug resistance is serious . It is necessary to guide clinical medication and strengthen nosocomial infection monitoring according to its characteristics .
10.Correlation between alexithymia and disability acceptance in patients with spinal cord injury

Objective:To explore the influencing factors of disability acceptance in patients with spinal cord injury, and analyze the correlation between alexithymia and disability acceptance.Methods:The convenience sampling method was used to select 276 patients with spinal cord injury admitted to Affiliated Hospital of Jining Medical College from October 2018 to June 2020 as the research objects. The general information questionnaire, the Toronto Alexitymia Scale (TAS-20), and the Acceptance of Disability Scale (ADS) was used to conduct the survey, Pearson correlation was used to analyze the correlation between alexitymia and disability acceptance in patients with spinal cord injury, hierarchical regression analysis was used to analyze the factors that affect the disability acceptance of patients with spinal cord injury.Results:The total score of ADS of 276 patients with spinal cord injury was (172.68 ± 37.72) points, and the total score of TAS-20 was (55.60 ± 9.49) points. The results of Pearson correlation analysis showed that all dimensions and total score of TAS-20 were negatively correlated with all dimensions and total score of ADS ( r values were -0.821--0.101, all P<0.05). The results of hierarchical regression analysis showed that gender, course of disease, post-injury working status were the risk factors of disability acceptance ( P<0.05), and alexithymia was the protective factor of disability acceptance( P<0.05), gender, course of disease, post-injury working status and alexithymia level could jointly explain the 22.1% variation in disability acceptance in patients with spinal cord injury. Conclusions:The disability acceptance of patients with spinal cord injury is at a moderate level, and alexitymia is negatively correlated with disability acceptance. alexitymia, gender, course of disease, and post-injury work status are the influencing factors of disability acceptance of patients with spinal cord injury.
