1.Trends and associated factors of overweight and obesity among children and adolescents in Macao from 2005 to 2020

		                        			Objective:
		                        			To analyze the prevalence trends and associated factors of overweight and obesity among children and adolescents in Macao from 2005 to 2020, so as to provide evidence for developing health promotion strategies.
		                        		
		                        			Methods:
		                        			Data were obtained from the Macao Citizen Physical Fitness Monitoring Database for the years 2005, 2010, 2015, and 2020 for participants aged 6-22 years. The χ 2 test was employed to analyze trends in detection rates, while univariate and multivariate Logistic regression analyses were conducted to identify influencing factors.
		                        		
		                        			Results:
		                        			The overweight rate among Macaos children and adolescents increased from 10.4% in 2005 to 14.8% in 2020. The obesity rate rose from 6.8% to 12.1%, with the total detection rate increasing from 17.2% to 26.9%, and the differences were statistically significant ( χ 2 trend =46.7, 87.5, 145.9,  P <0.01). Notably, the overweight/obesity rate among boys showed rapid growth ( χ 2 trend = 118.6,  P <0.01), while girls exhibited a declining inflection point in 2020. Multivariate Logistic regression analysis revealed that children and adolescents with the following characteristics faced higher risks of overweight/obesity: a physical education performance score of 3 points (overweight:  OR=2.34, 95%CI =1.10-4.96; obesity:  OR=2.39, 95%CI =1.19-4.81), paternal obesity (overweight:  OR=2.07, 95%CI =1.38-3.11; obesity:  OR=1.51,  95%CI = 1.01-2.27), and maternal obesity (overweight:  OR=1.69, 95%CI =1.08-2.63; obesity:  OR=1.77, 95%CI =1.16- 2.71 ) ( P <0.05). Conversely, lower risks were observed in those who performed appropriate warm-up activities before exercise (obesity:  OR=0.37, 95%CI =0.15-0.95), participated in two academic/non-sports extracurricular classes (obesity:  OR=0.46, 95%CI =0.24-0.88), and reported moderate physical exertion during extracurricular exercise (obesity:  OR=0.60, 95%CI =0.36-0.98)  ( P <0.05) .
		                        		
		                        			Conclusions
		                        			Overweight and obesity among Macao s children and adolescents remain severe, particularly among boys, while girls show early signs of improvement. It is recommended to establish a multi-sectoral collaborative prevention and control system to reduce childhood and adolescent obesity.

		                        			Pyran- and furanocoumarins are key representatives of tetrahydropyrans and tetrahydrofurans, respectively, exhibiting diverse physiological and medical bioactivities. However, the biosynthetic mechanisms for their core structures remain poorly understood. Here we combined multiomics analyses of biosynthetic enzymes in Peucedanum praeruptorum and in vitro functional verification and identified two types of key enzymes critical for pyran and furan ring biosynthesis in plants. These included three distinct P. praeruptorum prenyltransferases (PpPT1-3) responsible for the prenylation of the simple coumarin skeleton 7 into linear or angular precursors, and two novel CYP450 cyclases (PpDC and PpOC) crucial for the cyclization of the linear/angular precursors into either tetrahydropyran or tetrahydrofuran scaffolds. Biochemical analyses of cyclases indicated that acid/base-assisted epoxide ring opening contributed to the enzyme-catalyzed tetrahydropyran and tetrahydrofuran ring refactoring. The possible acid/base-assisted catalytic mechanisms of the identified cyclases were theoretically investigated and assessed using site-specific mutagenesis. We identified two possible acidic amino acids Glu303 in PpDC and Asp301 in PpOC as vital in the catalytic process. This study provides new enzymatic tools in the epoxide formation/epoxide-opening mediated cascade reaction and exemplifies how plants become chemically diverse in terms of enzyme function and catalytic process.
		                        		
		                        		
		                        		
		                        	
3.Current status and influencing factors of scientific fitness literacy among Chinese adult population aged 20~59 years

		                        			Objective:To investigate the current status of scientific fitness literacy (SFL) and its influencing factors among Chinese adults aged 20-59 years.Methods:Totally 63 338 adults aged 20-59 years in the status of national fitness activities in 2020 from 1 January to 31 March 2020 were selected as the subjects, and they were divided into four age groups, namely, the 20-29 age group, the 30-39 age group, the 40-49 age group and the 50-59 age group. Data were analyzed by SPSS 29.0.Comparative analyses for age, gender, urban-rural difference were carried out by applying non-parametric tests, and multiple regression models were applied to find the influencing factors of SFL.Results:The SFL score of Chinese adults aged 20-59 years was 53.40 (41.67, 63.73), and the scores of cognition, attitude, ability and skills, behavior and habits sub-dimensions were 86.11 (72.22, 100.00), 62.96 (50.62, 75.31), 27.78 (11.11, 44.44) and 33.33 (11.11, 55.56), respectively. The SFL and the four sub-dimensions were demonstrated to have higher scores for males than females, and higher scores for adults in urban area than those in rural area(all P<0.01). The multiple regression results showed that regular exercise at a fitness facility (20-29 age group: β=0.321, t=5.940, P<0.01; 30-39 age group: β=0.187, t=2.937, P<0.01; 40-49 age group: β=0.230, t=3.988, P<0.01; 50-59 age group: β=0.415, t=5.671, P<0.01) was the Chinese adults' common influence factor on SFL.Motivation, evaluation by those around them, experience from exercising, and convenience and atmosphere of exercising were the main influence factors in the 20-29 and 30-39 age groups(all P<0.05).Comfort level of fitness venue, convenience, safety, and support of fitness place were the main influence factors in adults aged 40-59 years(all P<0.05). Conclusions:Chinese adults aged 20-59 years old have high SFL awareness scores, but low overall SFL scores. Surrounding people's evaluation, experience in exercise, comfort level of fitness place, convenience, safety and fitness policy support are the influential factors of scientific fitness literacy.
		                        		
		                        		
		                        		
		                        	
4.Efficacy and mechanism of astragaloside Ⅳ derivatives on chronic heart failure in mice

		                        			Objective To evaluate the pharmacodynamics of astragaloside Ⅳ derivatives for chronic heart failure,screen the candidate compounds and preliminarily explore the mechanism of the candidate compound HHQ16 against heart failure.Methods Chronic heart failure was induced by left anterior descending artery ligation in C57BL/6 mice for 4 weeks,and the mice were divided into 4 groups,including sham group,model group,positive control captopril group,and astragaloside Ⅳ derivatives group.After continuous intragastric administration for four weeks,the cardiac function was detected by echocardiography,and the optimal astragaloside Ⅳ derivative HHQ16 was selected for the treatment of heart failure.The preliminary mechanism for HHQ16 was further explored.The size of heart was observed by gross morphology;pathological changes were observed by HE staining;collagen deposition in the myocardium was observed by Masson staining;protein levels of myocardial fibrosis indexes COL1,COL3,and αSMA were detected by immunohistochemical staining,and mRNA levels of myocardial fibrosis indexes COL1,COL3,αSMA,and TGF-β1 were determined by qPCR technique.Results All astragaloside Ⅳ derivatives significantly improved cardiac function with increasing LVEF and LVFS,of which HHQ16 was the optimal compound.Compared with the model group,the heart volume of HHQ16 group was significantly reduced;myocardial hypertrophy was reduced;collagen deposition in myocardial tissues was reduced;and myocardial fibrosis indexes,COL1,COL3,αSMA and TGF-β1 mRNA levels,as well as the protein levels of COL1,COL3 and αSMA were significantly reduced.Conclusion HHQ16 is an optimal astragaloside Ⅳ derivatives for the treatment of chronic heart failure in mice,which could improve cardiac function by improving myocardial remodeling,and inhibit myocardial hypertrophy and myocardial fibrosis.
		                        		
		                        		
		                        		
		                        	
5.Development status of clinical pathway at home and abroad

		                        			Clinical pathway is an important quality management tool for regulating medical behavior both at home and abroad, and an important means of controlling medical costs in the reform of medical insurance payment methods.The author reviewed the current development status of clinical pathways both at home and abroad, focusing on summarizing the development experience of foreign countries, and analyzing the shortcomings in the development of clinical pathways in China from the perspectives of formulation, implementation, and evaluation. It is proposed that China should establish and improve the regulatory and incentive mechanisms for clinical pathways, accelerate the construction of supporting medical security systems, explore new incentive transmission models, attach importance to the role of patient participation in the formulation and implementation of clinical pathways, and so on, in order to provide reference for promoting the efficient development of clinical pathways in China.
		                        		
		                        		
		                        		
		                        	
6.Development of VISTA in tumor immunotherapy

		                        			V-domain immunoglobulin suppressor of T-cell activation (VISTA) is a member of the B7 family that maintains homeostasis in T cells and myeloid cells.Blocking VISTA inhibits tumor development in in vitro and in vivo trials, and is an important target for tumor immunotherapy.This review focuses on its structural features, expression and biological functions in tumor microenvironment, summarizes the current stage of small molecule inhibitors and antibodies targeting VISTA, and discusses the research approaches.It aims to provide a rationale for subsequent study on VISTA and the development of related immune checkpoint antitumor drugs.
		                        		
		                        		
		                        		
		                        	
7.Structural feature of type I CRISPR-Cas system and its application in gene editing

		                        			The CRISPR-Cas (clustered regularly interspaced short palindromic repeats and CRISPR-associated) system is an "adaptive immune system" found in the genomes of bacteria and archaea which is mediated by RNA and resists foreign nucleic acid invasion.Take advantage of specific recognition of target nucleic acid, CRISPR-Cas system can efficiently edit their target site or accurately regulate gene expression, and now have been developed into a powerful tool for gene editing.According to the different compositions of the effector complex, the system has been divided into two categories: class 1 (type I, type IV, and type III) and class 2 (type II, type V, and type VI).Class 2 system, like the CRISPR-Cas9, is widely used in basic research due to the earliest discovery and best research.However, class 1 has not been maturely developed and utilized though it makes up 90% of the entire CRISPR-Cas system.In this essay, the classification of subtype, the assembly of Cascade complex, the cleavage and degradation mechanism of Cas3, and the application in gene editing of class 1 type I CRISPR-Cas system will be discussed and summarized to provide new ideas and methods for further mechanism studying and application of this category.
		                        		
		                        		
		                        		
		                        	
8.Association of tea drinking during pregnancy and preterm delivery or abortion: A meta-analysis

		                        			Objective:
		                        			To analyze the association between tea drinking during pregnancy and the risk of preterm delivery and abortion,so as to provide basis for prevention of preterm delivery and abortion.
		                        		
		                        			Methods:
		                        			The databases of CNKI,Wanfang,VIP,CBMdisc,PubMed and Web of Science were searched for cohort studies and case-control studies into the association between tea consumption during pregnancy and preterm delivery or abortion until June 30 th,2019. Relative risk(RR)or odds ratio(OR)were used as indicators for the meta-analysis.
		                        		
		                        			Results:
		                        			A total of 1 099 articles were retrieved,14 of them were included in the quantitative study,including 9 cohort studies with 18 295 exposed and 71 890 unexposed individuals and 5 case-control studies with 1 351 cases and 3 059 controls. There was no statistically significant association between tea drinking during pregnancy and preterm birth or abortion(OR/RR=1.08,95%CI:0.99-1.18). The linear regression model of random effect showed that with the increase of tea consumption during pregnancy,the risk of premature delivery and abortion did not change significantly(OR/RR=1.05,95%CI:0.99-1.11). There was no publication bias found in Begg's test and Egger's test. 
		                        		
		                        			Conclusion

		                        			To address the increasing need for detecting and validating protein biomarkers in clinical specimens, mass spectrometry (MS)-based targeted proteomic techniques, including the selected reaction monitoring (SRM), parallel reaction monitoring (PRM), and massively parallel data-independent acquisition (DIA), have been developed. For optimal performance, they require the fragment ion spectra of targeted peptides as prior knowledge. In this report, we describe a MS pipe-line and spectral resource to support targeted proteomics studies for human tissue samples. To build the spectral resource, we integrated common open-source MS computational tools to assemble a freely accessible computational workflow based on Docker. We then applied the workflow to gen-erate DPHL, a comprehensive DIA pan-human library, from 1096 data-dependent acquisition (DDA) MS raw files for 16 types of cancer samples. This extensive spectral resource was then applied to a proteomic study of 17 prostate cancer (PCa) patients. Thereafter, PRM validation was applied to a larger study of 57 PCa patients and the differential expression of three proteins in prostate tumor was validated. As a second application, the DPHL spectral resource was applied to a study consisting of plasma samples from 19 diffuse large B cell lymphoma (DLBCL) patients and 18 healthy control subjects. Differentially expressed proteins between DLBCL patients and healthy control subjects were detected by DIA-MS and confirmed by PRM. These data demonstrate that the DPHL supports DIA and PRM MS pipelines for robust protein biomarker discovery. 10.Approaches to establish Q-markers for the quality standards of traditional Chinese medicines.

		                        			Traditional Chinese medicine (TCM) has played a pivotal role in maintaining the health of Chinese people and is now gaining increasing acceptance around the global scope. However, TCM is confronting more and more concerns with respect to its quality. The intrinsic "multicomponent and multitarget" feature of TCM necessitates the establishment of a unique quality and bioactivity evaluation system, which is different from that of the Western medicine. However, TCM is investigated essentially as "herbal medicine" or "natural product", and the pharmacopoeia quality monographs are actually chemical-markers-based, which can ensure the consistency only in the assigned chemical markers, but, to some extent, have deviated from the basic TCM theory. A concept of "quality marker" (Q-marker), following the "property-effect-component" theory, is proposed. The establishment of Q-marker integrates multidisciplinary technologies like natural products chemistry, analytical chemistry, bionics, chemometrics, pharmacology, systems biology, and pharmacodynamics, etc. Q-marker-based fingerprint and multicomponent determination conduce to the construction of more scientific quality control system of TCM. This review delineates the background, definition, and properties of Q-marker, and the associated technologies applied for its establishment.
