1.Exosomal microRNAs (miRNAs) in blood and urine under physiological conditions: a comparative study

Objectives:
Blood and urine are commonly used specimens for clinical testing, and their contents, particularly exosomal microRNA (miRNA), are diverse, reflecting the metabolic activities of tissues and organs in the body.
Methods:
Blood and urine samples were collected from six healthy adults. Exosomes were then enriched from these samples, followed by sequencing and bioinformatic analysis of exosomal miRNA.
Results:
The comparative analysis of miRNAs in blood and urine revealed that 41 miRNAs were more abundant in blood, while 61 were found at lower levels. Notably, hsa-miR-934 was among those with higher expression in blood, whereas hsa-miR-425-5p was one of the miRNAs with lower expression.Kyoto Encyclopedia of Genes and Genomes pathway analysis indicated that the target mRNAs of differentially expressed exosomal miRNAs (DEexo-miRNAs) in both blood and urine are implicated in various signaling pathways, including proteoglycans in cancer, axonal guidance, and the regulation of the actin cytoskeleton. Additionally, the target mRNAs associated with DEexo-miRNAs in urine were also linked to processes such as ubiquitin-mediated proteolysis and the phosphatidylinositol signaling system. In contrast, the target mRNAs corresponding to DEexo-miRNAs in blood were involved in the FoxO signaling pathway and chronic myeloid leukemia, among others.
Conclusion
This study observed differential expression of exosomal miRNAs in blood and urine, thereby enriching the available library of exosomal miRNA for these two sample types. It also lays the groundwork for the detection of exosomal biomarkers from blood and urine.

Acupoint is the initial response site of acupuncture stimulus and also the source link of the effect onset of acupuncture. Acupuncture is a mechanical physical stimulus. How is the mechanical force of acupuncture transduced into neuroelectrical and biochemical signals at acupoint? How does the physiochemical information of acupoint launch acupuncture effect? All of these remain the common and crucial questions in the study of acupuncture effect mechanism. Physical changes are induced in the local tissue of acupoint by needling techniques, such as the deformation and displacement of muscle fibers, which may act on the nerve ending receptors and produce electroneurographic signals. Besides, these changes may activate the mechanosensitive ion channels of the cytomembrane in acupoint site. Through cellular signal transduction, the physical signals may be transformed into chemical ones to trigger the physiochemical coupling response of acupoint microenvironment. Eventually, acupuncture effect is generated via nerves and body fluids. "The mechanical force of acupuncture", through "the physiochemical transduction", promotes the body's perception and transmits acupuncture signals. It suggests that acupoint is the "transducer" in the physiochemical information coupling response of acupuncture.

3.Methylation of CHD5 Gene Promoter Regulates p19/p53/p21 Pathway to Facilitate Pathogenesis of Acute Myeloid Leukemia.

OBJECTIVE:
To investigate the methylation status of CHD5 gene promoter in bone marrow from acute myeloid leukemia (AML) patients, and the underlying mechanism for initiating the pathogenesis of AML via p19/p53/p21 pathway.
METHODS:
Methylation status of the CHD5 gene promoter was detected by using methylation-specific polymerase chain reaction (MSPCR) in bone marrow from AML patients, and the iron-deficiency anemia (IDA) samples were served as control. The expression of CHD5, p19, p53 and p21 was determined by real-time quantitative reverse transcriptase PCR and Western blot.
RESULTS:
The methylation of CHD5 gene in bone marrow from AML patients increased significantly (39.06%) as compared with control group (6.67%). The methylation of CHD5 gene significantly correlated with chromosome karyotype differentiation (P<0.01), but did not correlate with the patient's sex, age and clinical classification (P>0.05). The mRNA expression of CHD5 gene in AML decreased, compared with control group, the mRNA and protein expression of p19, p53 and p21 in AML with CHD5 methylation promoter decreased.
CONCLUSION
The hypermeltylation of CHD5 gene promoter in AML patients can lead to decrease of CHD5, p19, p53 and p21 expression levels which may reduce the inhibitory effect on proliferation of leukemia cells through the regulation of p19, p53 and p21 pathway, thus promotes the occurence of AML.

4. Anti-inflammatory and anti-fibrosis effects of Sanjie Zhentong Capsule on chronic pelvic inflammatory disease in rats

Objective: The anti-inflammatory and anti-fibrotic effects of Sanjie Zhentong Capsule (SJZT) on chronic pelvic inflammation in rats were investigated and its underlying mechanisms were explored. Methods: Ten rats regarded as normal group with sham operation were randomly selected from 60 female SD rats, and the remaining rats were injected with phenol mucilage in the left uteri to replicate chronic pelvic inflammatory disease. On day 14, after the operation, according to body weight, the 50 rats were randomly allocated to model group, dexamethasone (DEX) group, SJZT low dose group (0.192 g/kg), SJZT middle dose group (0.384 g/kg), and SJZT high dose group (0.768 g/kg). The drugs were dissolved in 0.5% CMC-Na solution and administered respectively, while 0.5% CMC-Na solution was given to the normal and model group. After all the animals were treated by oral gavage once a day for 30 d, the blood and the left uteri were collected. The uterine tissues were fixed and stained with haematoxylin-eosin (HE) and Masson’s trichrome for histopathology examination and the concentrations of TNF-α, IL-1β, PDGF, and TIMP in serum were assayed by ELISA. The protein expression levels of p-ERK1/2, p-JNK, NF-κB p65, MMP-2, and MMP-9 in the uteri were also detected by Western blotting. Results: Compared with the model group, SJZT could effectively improve the arrangement disorder of cavity wall, reduce the degeneration and necrosis of epithelial cells, the infiltration of inflammatory cells and the congestion of lamina propria, and alleviate the proliferation of collagen fibers in lamina propria. SJZT could also reduce the concentrations of TNF-α, IL-1β, PDGF, and TIMP in serum. Moreover, the protein expression of p-ERK1/2, p-JNK, and NF-κB p65 in the uteri were inhibited significantly, and at the same time the protein expression of MMP-2 and MMP-9 was increased markedly by SJZT. Conclusion: SJZT exhibits robust anti- inflammatory and anti-fibrosis effects in chronic pelvic inflammatory disease model induced by phenol mucilage. Its mechanisms may be related to regulating NF-κB and MAPK signaling pathways, inhibiting inflammation, reducing the synthesis of extracellular matrix (ECM), increasing the degradation of ECM and promoting the repair of fibrosis.
5.Protective effect of Longxue Tongluo capsule on oxidized low-density lipoprotein damage to human umbilical vein endothelial cells.

To observe the protective effect of Longxue Tongluo capsule (LTC) on human umbilical vein endothelial cells (EAhy.926 cells) injury induced by oxidized low-density lipoprotein (ox-LDL, 100 mg·L⁻¹). The effect of the cell viability of LTCin alleviating OX-LDL-induced endothelial cell injury was determined by MTT and LDH assay. The effect of LTC on lactic dehydrogenase (LDH), nitric oxide (NO), super oxide dlsmutase (SOD) and malondialdehyde (MDA) levels were detected by corresponding assay kits according to manufacturer's instruction. The effect of LTC on the protein expressions of intercellular cell adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule 1 (VCAM-1), p65, p-p65, IKB and p-IKB were detected by Western blot. The results showed that compared with the normal control group, the activity of EAhy.926 cells was significantly decreased, LDH leakage (<0.01) increased, NO content and SOD activity significantly decreased (<0.01, <0.05), and the expressions of ICAM-1, VCAM-1, p-p65/p65 and p-IKB(<0.05)increased.This study demonstrated that LTC had no significant effect on the growth of normal cells. The treatment with LTC significantly promoted the proliferation of vascular endothelial cells damagedby ox-LDL, decreased MDA content and LDH release, andincreased the activity of SOD and NO content. Meanwhile, ox-LDL significantly increased the expressions of ICAM-1, VCAM-1, p-p65/p65, p-IKB/IKB in Eahy.926 cells; these effects were suppressed by LTC at 1, 2 mg·L⁻¹. In conclusion, LTC has a significant protective effect on human umbilical vein endothelial cells caused by ox-LDL. This study suggested that LTC has a certain therapeutic effect on AS.
6.Awareness of healthy lifestyle content among residents aged 18 -69 years in Shanghai

[Objective] To investigate the awareness of healthy lifestyle content among residents in Shanghai and to provide basis for future efforts . [ Methods] Stratified random sampling covered all 18 districts in Shanghai .A total of 8548 participants were recruited and questionnaire with them was completed by face to face interview. [Results] Among Shanghai residents aged 18-69 years, 67.8%of them had awareness of all five items of healthy lifestyle content .And 94.6%, 92.3%, 80.7%, 77.3%, and 81 .7%of them respectively had awareness of balanced diet , regular exercise , no smoking , no alcohol a-buse, and keeping mental balance .Analysis of Logistic regression showed that gender , age, area of resi-dence, and education level were factors influencing awareness rates mentioned above . [ Conclusion] The information about healthy lifestyle content was effectively spread by various departments concerned and the Shanghai government .And the above results suggest that specific information spread for different demo-graphic characteristics of different populations is beneficial for future improvement of healthy knowledge a -mong them.
7.Comparing of different methods on habitat adaptive division of Chinese material medica.
