BACKGROUND:Apoptosis plays an important role in secondary brain injury. Therefore,to explore the pathophysiological mechanism of promoting nerve cell survival after traumatic brain injury provides a new direction and theoretical basis for the prevention and treatment of traumatic brain injury. OBJECTIVE:To explore the expression changes of Lnx1 molecule in mammalian cortical neurons after brain injury and the possible mechanism involved in secondary brain injury. METHODS:Eighty adult SD rats were divided into 20 male and 20 female mice in sham operation group and 20 male and 20 female mice in traumatic brain injury group. The traumatic brain injury rat model was established by heavy falling method. At 6,12,24,48,and 72 hours after brain injury, the expression of related molecules in damaged cortical neurons was analyzed by RT-qPCR,western blot assay, and immunofluorescence staining. RESULTS AND CONCLUSION:(1)The brain tissue of traumatic brain injury group was bleeding and obvious tissue injury could be observed.Water content of brain tissue increased after traumatic brain injury.(2)Compared with the sham operation group, the expression of Lnx1 in cortical neurons after traumatic brain injury increased significantly at 24 hours after injury.(3)After traumatic brain injury, the expression of PBK and BCR protein decreased, and the pro-survival factor ctgf increased.(4)These findings suggest that after traumatic brain injury, the expression of Lnx1 is up-regulated in neurons, which may be due to the decrease of the expression of its target molecules PBK and BCR,and further promote the expression of living factor ctgf, which has a protective effect on the damaged neurons.

Objective To overcome the limitations of existing human respiratory syncytial virus(hRSV)animal models, such as semi-permissiveness and short duration of infection, this study established an IL2rg gene knockout(IL2rg-/-)rat model using TALEN gene editing technology. Methods The animal model was infected with hRSV intranasally.Clinical characteristics, body weight, and temperature changes were observed over the infection period(0~35 days). The total viral loads in respiratory organs, such as the nasal tissue, trachea, and lungs, were measured at various time points(4,11,20,and 35 days post-infection).Pathological analysis was conducted on target organs at the endpoint of observation(35 days post-infection).Changes in peripheral blood T,B,NK,and NKT cells and various cytokines were assessed at various time points(4,20,and 35 days post-infection).Results(1)IL2rg/-knockout rats sustained high viral loads in the nasal cavity upon intranasal inoculation with hRSV.The average peak titer rapidly reached 1 × 1010 copies/g in nasal tissue and 1 × 107 copies/g up to 5 weeks post-infection.(2)However,no significant pathological changes were noted in nasal, tracheal,or lung tissues.(3)An increase was observed in the content of peripheral blood B cells in hRSV-infected IL2rg--rats.(4)IL-6 and MCP-1 were increased in the early stage of infection and then decreased at the end of the observation period. Conclusions This study established a new IL2rg-/-rat model using TALEN technology and found that this model effectively supported high-level replication and long-term infection of hRSV,providing a good basis for antiviral drug screening and in vivo efficacy evaluation of anti-hRSV antibodies.

Proteomic characterization of plasma is critical for the development of novel pharmacodynamic bio-markers.However,the vast dynamic range renders the profiling of proteomes extremely challenging. Here,we synthesized zeolite NaY and developed a simple and rapid method to achieve comprehensive and deep profiling of the plasma proteome using the plasma protein corona formed on zeolite NaY.Specifically, zeolite NaY and plasma were co-incubated to form plasma protein corona on zeolite NaY(NaY-PPC),followed by conventional protein identification using liquid chromatography-tandem mass spectrometry.NaY was able to significantly enhance the detection of low-abundance plasma proteins, minimizing the"masking"effect caused by high-abundance proteins. The relative abundance of middle-and low-abundance proteins increased substantially from 2.54%to 54.41%,and the top 20 high-abundance proteins decreased from 83.63%to 25.77%.Notably,our method can quantify approxi-mately 4000 plasma proteins with sensitivity up to pg/mL,compared to only about 600 proteins iden-tified from untreated plasma samples.A pilot study based on plasma samples from 30 lung adenocarcinoma patients and 15 healthy subjects demonstrated that our method could successfully distinguish between healthy and disease states. In summary, this work provides an advantageous tool for the exploration of plasma proteomics and its translational applications.

Background::The potential impact of β cell function and insulin sensitivity on adverse pregnancy outcomes in women with gestational diabetes mellitus (GDM) remains uncertain. We aimed to investigate the association between β cell dysfunction, insulin resistance, and the composite adverse pregnancy outcomes.Methods::This observational study included 482 women diagnosed with GDM during pregnancy. Quantitative metrics on β cell function and insulin sensitivity during pregnancy were calculated using traditional equations. The association of β cell dysfunction and insulin resistance with the risk of the composite adverse pregnancy outcomes was investigated using multivariable-adjusted logistic regression models.Results::Multivariable-adjusted odds ratios (ORs) of adverse pregnancy outcomes across quartiles of homeostatic model assessment for insulin resistance (HOMA-IR) were 1.00, 0.95, 1.34, and 2.25, respectively ( P for trend = 0.011). When HOMA-IR was considered as a continuous variable, the multivariable-adjusted OR of adverse pregnancy outcomes was 1.34 (95% confidence interval 1.16-1.56) for each 1-unit increase in HOMA-IR. Multivariable-adjusted ORs of adverse pregnancy outcomes across quartiles of homeostatic model assessment for β cell function (HOMA-β) were 1.00, 0.51, 0.60, and 0.53, respectively ( P for trend = 0.068). When HOMA-β was considered as a continuous variable, the multivariable-adjusted OR of adverse pregnancy outcomes was 0.57 (95% CI 0.24-0.90) for each 1-unit increase in HOMA-β. However, other quantitative metrics were not associated with the composite adverse pregnancy outcomes. Conclusions::We demonstrated a significant association of β cell function and insulin sensitivity with the risk of adverse pregnancy outcomes. We have provided additional evidence on the early identification of adverse pregnancy outcomes besides the glycemic values.

Objective:To observe the therapeutic effect of blood purification with oXiris on cytokine storm in severe COVID-19 patients. Methods:In February to April, 2020 in the intensive care unit (ICU) of a hospital in Wuhan, 7 cases with severe COVID-19 as the study participant, treated with oXiris for blood purification. The general information of patients, the basic information of the blood purification treatment, blood purification treatment before and after treatment for 24 hours, 48 hours, 72 hours of routine blood, liver and kidney function and inflammatory markers were collected to understand the trend of change. Results:The number of blood purification treatment of 7 patients were 2.13±1.64 times, and the duration of blood purification was 66.31±48.73 hours. Treatment was interrupted twice due to hypothermia. After treatment, the level of interleukin 6 (IL-6) of the patients decreased from (1342.20±1822.01) pg/ml before treatment to (202.69±276.24) pg/ml in 4 cases decreased by more than 90% and in 2 cases decreased by 63% and 77%. The level of TNF decreased from 51.94±59.64 pg/ml before treatment to 18.93±9.84 pg/ml, and TNF decreased by 28%~78% in 4 patients. The hypersensitive C-reactive protein (HS-CRP) of the patients decreased from 147.81±100.54 mg/L before treatment to 89.43±108.31 mg/L after treatment, in 4 cases decreased by 63% to 91%.Conclusions:oXiris blood purification treatment can effectively reduce inflammatory markers in patients with severe COVID-19. Correct use of oXiris filter during treatment can prolong the life of the filter. During the treatment, the pressure monitoring value and whether there is thrombosis in the pipeline should be closely observed and recorded. Meanwhile, it is necessary to keep patients warm to prevent hypothermia in. The application of ultrasound to monitor blood coagulation and venous thrombosis plays a positive role in preventing the occurrence of pulmonary embolism.
