Sedentary bad habits and unhealthy diets in modern lifestyles have led to an upward trend in the incidence of obesity, and a series of diseases related to obesity have also gradually received attention. Multiple sclerosis is a chronic inflammatory disease of the central nervous system, and obesity has a common inflammatory component with most chronic diseases. Therefore, this paper reviews the research progress on the relationship between obesity and multiple sclerosis in order to better understand the role of obesity in the management of multiple sclerosis.
2.Metabolomics study on improvement effects of Cirsium japonicum extract on hypercholesterolemia model mice

OBJECTIVE To explore the mechanism of Cirsium japonicum extract in improving hypercholesterolemia based on metabolomics technology. METHODS The extract of C. japonicum was prepared by macroporous resin adsorption, and its main components were identified by liquid chromatography-tandem mass spectrometry. The experimental mice were randomly divided into control group (n=6) and modeling group (n=16). The hypercholesterolemia model was induced by diet in modeling group; after modeling, the rats of modeling group were divided into model group (n=8) and C. japonicum extract group (n=8). C. japonicum extract group was given C. japonicum extract 400 mg/(kg·d) by gavage (calculated by extract), and other 2 groups were given constant volume of 0.3% sodium carboxymethyl cellulose solution, for 6 weeks. After medication, the intervention effect of C. japonicum extract was evaluated by the levels of serum total cholesterol (TC), triglyceride (TG) and the histopathological changes of liver. The mechanism of C. japonicum extract in improving hypercholesterolemia model mice was investigated by metabolomics. RESULTS It was identified that C. japonicum extract contained 12 components, such as 030302005) chlorogenic acid, linarin and pectolinarin. After 6 weeks of intervention, compared with control group, serum level of TC was increased significantly while the level of TG was decreased significantly in model group (P<0.05), while a large number of lipid droplets, disorderly arrangement of liver cells and the damaged structure of liver cord were observed in liver tissue. Compared with model group, the serum level of TC was decreased significantly in C. japonicum extract group(P<0.05); the lipid droplets in liver tissue were significantly reduced, with liver cells arranged radially and tightly centered around the central vein, and liver cords arranged neatly. The metabolomics study showed that after the intervention of C. japonicum extract, the levels of metabolites were significantly adjusted back, such as ethanolamine, fumaric acid and cholesterol; finally, three metabolism pathways, such as alanine-aspartate-glutamic acid metabolism, arginine biosynthesis, citric acid cycle, were obtained. CONCLUSIONS The main components of C. japonicum extract are phenolic acids and flavonoids, such as chlorogenic acid, linarin, pectolinarin. C. japonicum extract can improve hypercholesterolemia by regulating the contents and distribution of differential metabolites, adjusting alanine-aspartate-glutamic acid metabolism, arginine biosynthesis and citric acid cycle, participating in oxidation-reduction reaction, improving liver lipid accumulation, and playing anti-inflammatory role.
3.Diagnosis and treatment progress of acquired immunologic deficiency syndrome-related lymphoma

Malignancy is one of the most important complications of acquired immunodeficiency syndrome (AIDS) caused by human immunodeficiency virus (HIV) infection and destruction of host CD4-positive T lymphocytes. Lymphoma ranks first in AIDS-related malignancies. The clinical features of lymphoma patients infected with HIV are different from non-HIV infected patients. The host immune condition in anti-lymphoma chemotherapy also needs to be considered. This paper reviews the clinical characteristics of AIDS-related lymphoma and the attention in anti-lymphoma therapy according to the latest international research findings and related guidelines.
4.Effects of TSG-6 gene expression mesenchymal stem cells on liver transplantation rejection in rats

Objective To investigate the effects of silencing TSG-6 gene modified bone marrow mesenchymal stem cells (BMSC) transplantation on liver allotransplantation rejection in rats.Methods BMSC and KCs were isolated from rats and cultured in complete medium.We down-regulated TSG-6 expression of BMSC with lentiviral vectors carrying short hairpin RNA (LV3-shTSG-6).TSG-6mRNA and protein level of BMSC were tested respectively by quantitative real-time PCR and western blot analysis.After co-cuhured between BMSC and KCs,the expression of TNF-α and TSG-6 in cell supernatants were tested.Then,we established the orthotopic liver transplantation models in rats,the rats of each group were killed at 1 days,3 days and 7 days after operation.The serum levels of AST,ALT,TBIL,γ-GGT,TNF-α,IL-6 and IL-4 were tested with ELISA in each group,TSG-6mRNA and protein level of liver tissues obtained from each group were tested by quantitative real-time PCR and western blot analysis.The 1iver tissues of each group were stained with HE,then microstructure of liver tissues were observed under light microscope.The postoperative survival rates in other rats of each group were observed.Results The lentivirus transfection efficiency of mesenchymal stem cells was beyond 70 percent;After co-cultured between BMSC and KCs,the expression of TSG-6 in TSG-6-shRNA-BMSC + KCs group supernatant was significantly lower in different time point than that in BMSC + KCs group and TSG-6-NC-BMSC + KCs group (P <0.05),and the expression of TNF-α peak in BMSC + KCs group and TSG-6-NC-BMSC + KCs group supernatants were at 6 hours,which were significandy lower than those at 12 hours (P <0.05),but the expression of TNF-α in TSG-6-shRNABMSC + KCs group at 12 hours was significantly higher than those in BMSC + KCs group and TSG-6-NC-BMSC +KCs group (P <0.05);After transplantation,the serum levels of AST,ALT,TBIL,γ-GGT,TNF-α and IL-6 in TSG-6-shRNA-BMSC group was significantly higher than those in TSG-6-NC-BMSC group and BMSC group in different time point (P < 0.05),but had no significant difference compared with PBS group;The serum levels of IL-4 in TSG-6-shRNA-BMSC group,TSG-6-NC-BMSC group and BMSC group was significantly higher than that in PBS group(P < 0.05),but TSG-6-shRNA-BMSC group compared with TSG-6-NC-BMSC group and BMSC group,the serum levels of IL-4 was significantly lower (P < 0.05);In pathological changes,we found that the degree of liver rejection in TSG-6-shRNA-BMSC group and PBS group were seriously obvious,and were graded Ⅱ-Ⅲ with Banff schedule;Comparation of postoperative survival time in each group,TSG-6-shRNA-BMSC group and PBS group were (16.6 ±4.6) d and (15.4 ± 6.7) d respectively,which were signifcantly lower than those in TSG-6-NCBMSC group (69.6 ± 28.1) d and BMSC group (69.2 ± 28.2) d (P < 0.05).Conclusion Transplantation of BMSC secreted TSG-6 could,to some extent,mitigate acute liver transplantation rejection.
5.A study on T lymphocyte apoptosis and disease progression in chronic HIV/AIDS patients

Objective To discuss the relationship between various T lymphocyte subsets apoptosis and disease progression in chronic antiretroviral-naive HIV/AIDS patients. Methods Thirty-six chronic antiretrovi-ral-naive HIV-infected individuals as well as 16 healthy HIV-negative controls were performed in this study. Ac-cording to the CD4~+ T cell counts, all the patients were divided three groups: < 200/μl, 200-350/μl and > 350/μl. After the peripheral blood mononuclear cells(PBMC) were isolated, T lymphocyte subpopulations were determined by the expression of CD45RO and CD27, and the apoptosis of different T cell subsets were measured by Annexin V staining, then analyzed by flow cytometry. To investigate whether the apoptosis of T cells varied with the culture time in vitro, 4 healthy controls and 4 patients were chosen as subjects, and the lev-els of cell apoptosis were analyzed at the culture time points of 0, 3, 6, 12, 24 h. Results (1)The percenta-ges of the AnnexinV expression on CD4~+ and CD8~+ T cells and all the subsets in HIV/AIDS patients were sig-nificantly higher than that in the healthy controls (P<0.05), but there were no significant differences among the three HIV-infected patient groups(P>0.05). (2) No significant correlations were observed between the levels of apoptosis of all the T cells and subsets and total CD4~+ T cell counts(P>0.05) ,nor with the HIV viral load (P>0.05). (3)As the culture time prolonged in vitro, the levels of apoptosis and necrosis of CD4~6 T cells in HIV/AIDS patients were significantly higher than those in the healthy conlrols, and the CD4~+ T cells were more susceptible to apoptosis and necrosis compared with CD8~+ T cells. Conclusion The levels of T cell apoptosis in HIV/AIDS patients was significantly higher than those in the healthy controls, at the same time, CD4~+ T cells were more susceptible to apoptosis and necrosis compared with CD8~+ T cells, but no correlation was found between the T cell apoptsis and disease progression.
6.Progresses in prevention and treatment of nontuberculosis mycobacteria infection in HIV positive individuals

A variety of opportunistic infections can occur in the late-stage of HIV-1 infection,Mycobacterium avium complex (MAC) infection is a common cause of disseminated bacterial infection. Since the advent of highly active antiretroviral therapy (HAART),the rate of MAC infection has declined substantially,but there is risk in individuals with low CD4 cell counts. This article describes the epidemiological,clinical,therapeutic and preventive aspects of MAC infection in HIV-sero-positive individuals.

Methods H_2O_2 was used to build an oxidative stress-induced injury model in neonatal rat cardiomyocytes.
