Senile osteoporosis (SOP) is a systemic bone disease characterized by increased susceptibility to fractures. The pathogenesis of SOP is complex and not well understood. Currently, the rapid aging model mouse, senescence accelerated mouse prone 6 (SAMP6), is an ideal model for studying the mechanisms of SOP development and exploring its prevention and treatment. This model exhibits characteristics including increased bone fragility, degradation of bone microstructure, loss of bone matrix, and abnormal metabolism and dysfunction of bone cells, faithfully replicating the process of SOP occurrence and progression at both macroscopic and microscopic levels.

Objective This study aimed to comprehensively analyze and compare the clinicopathological features and prognosis of Chinese patients with human epidermal growth factor receptor 2(HER2)-low early breast cancer(BC)and HER2-IHC0 BC. Methods Patients diagnosed with HER2-negative BC(N=999)at our institution between January 2011 and December 2015 formed our study population.Clinicopathological characteristics, association between estrogen receptor(ER)expression and HER2-low,and evolution of HER2 immunohistochemical(IHC)score were assessed.Kaplan-Meier method and log-rank test were used to compare the long-term survival outcomes(5-year follow-up)between the HER2-IHC0 and HER2-low groups. Results HER2-low BC group tended to demonstrate high expression of ER and more progesterone receptor(PgR)positivity than HER2-IHC0 BC group(P<0.001).The rate of HER2-low status increased with increasing ER expression levels(Mantel-Haenszel χ2 test,P<0.001,Pearson's R=0.159,P<0.001).Survival analysis revealed a significantly longer overall survival(OS)in HER2-low BC group than in HER2-IHC0 group(P=0.007)in the whole cohort and the hormone receptor(HR)-negative group. There were no significant differences between the two groups in terms of disease-free survival(DFS).The discordance rate of HER2 IHC scores between primary and metastatic sites was 36.84%. Conclusion HER2-low BC may not be regarded as a unique BC group in this population-based study due to similar clinicopathological features and prognostic roles.

Cadmium is a non-essential,non-degradable heavy metal element, the accumulation of cadmium can cause kidney damage. The purpose of this study was to reveal the ferroptosis mecha-nism induced by cadmium exposure in porcine kidney PK-15 cells.First of all, cell viability of gradi-ent concentration of cadmium chloride(CdCl2)on PK-15 cells was detected by CCK-8 method to screen suitable work concentration of CdCl2.Secondly,PK-15 cells were treated with 10 μmol/L CdCl2 for 3,6 and 12 h.The contents of lactate dehydrogenase(LDH)malondialdehyde(MDA)were detected by colorimetry, the expression of ferroptosis related protein were detected by West-ern blot, and the content of ferrous ion(Fe2+)was detected by fluorescence probe.Finally,PK-15 cells were pretreated with 10 μmol/L HO-1 inhibitor zinc protoporphyrin(ZnPP)for 6 h,and then treated with 10 μmol/L CdCl2 for 12 h.The morphology of PK-15 cells was observed by phase contrast microscope and the expression of ferroptosis related proteins was detected by West-ern blot. The results showed that CdCl2 treatment caused a significant decrease in cell viability. The levels of LDH,MDA and Fe2+,the protein levels of Nrf2,HO-1 and ALOX5,and the expression level of FTH1 protein were significantly decreased in CdCl2 treatment cells(P<0.01).ZnPP could significantly improve the ferroptosis of PK-15 cells induced by CdCl2 treatment.Compared with CdCl2 treatment cells, the cell morphology was significantly improved, the protein levels of Nrf2,HO-1 and ALOX5 protein were significantly decreased, and the protein levels of FTH1 protein were significantly increased in ZnPP pretreatment group(P<0.01).The results showed that cad-mium induced iron overload and lipid peroxidation, which result in ferroptosis in PK-15 cells through Nrf2/HO-1 pathway.

Skin and hair are the outermost interfaces between the human body and the external envi-ronment with many microorganisms distributed. These body surface microorganisms have individual speci-ficity and spatial temporal stability and can be transferred to the surface of other objects through con-tact,and can be easily found at the crime scene, which helps to construct the connection between the suspect and the crime scene. When the sample is degraded or the DNA quantity is too low to obtain sufficient human DNA evidence, the highly amplified copies of body surface microbiome analysis can be used as an adjunct to human DNA typing without affecting DNA extraction and typing, and provide useful information for narrowing the range of suspects. This paper reviews the forensic characteristics and analytical methods of body surface microbiome, the research progress on the application of body surface microbiome to forensic individual identification, and puts forward the challenges of applying body surface microbiome to forensic practice.
