1.Hydnocarpin inhibits malignant progression of triple negative breast cancer via CNOT4-mediated ubiquitination and degradation of YAP.

This study aims to investigate the effect and mechanism of hydnocarpin(HC) in treating triple negative breast cancer(TNBC). Cell counting kit-8(CCK-8), xCELLigence real-time cellular analysis(RTCA), and colony formation assay were employed to determine the effects of HC on the proliferation of two TNBC cell lines: MDA-MB-231 and MDA-MB-436. The effects of HC on the migration and invasion of TNBC cells were detected by high-content analysis, wound-healing assay, and Transwell assay. The changes in the epithelial-mesenchymal transition(EMT) and the expression of invasion-and migration-associated proteins [E-cadherin, vimentin, Snail, matrix metalloproteinase-2(MMP-2), and MMP-9] were detected by Western blot. Western blot and RT-qPCR were employed to determine the protein and mRNA levels of Yes-associated protein(YAP) and downstream targets(CTGF and Cyr61). TNBC cells were transfected with Flag-YAP for the overexpression of YAP, and the role of YAP as a key target for HC to inhibit TNBC malignant progression was examined by CCK-8 assay, Transwell assay, and wound-healing assay. The pathway of HC-induced YAP degradation was detected by the co-treatment of proteasome inhibitor with HC and ubiquitination assay. The binding of HC to YAP and the E3 ubiquitin ligase Ccr4-not transcription complex subunit 4(CNOT4) was detected by microscale thermophoresis(MST) assay and drug affinity responsive target stability(DARTS) assay. The results showed that HC significantly inhibited the proliferation, colony formation, invasion, and EMT of TNBC cells. HC down-regulated the protein and mRNA levels of CTGF and Cyr61. HC down-regulated the total protein level of YAP, while it had no effect on the mRNA level of YAP. The overexpression of YAP antagonized the inhibitory effects of HC on the proliferation, migration, and invasion of TNBC cells. HC promoted the degradation of YAP through the proteasome pathway and up-regulated the ubiquitination level of YAP. The results of MST and DARTS demonstrated direct binding between HC, YAP, and CNOT4. The above results indicated that HC inhibited the malignant progression of TNBC via CNOT4-mediated degradation and ubiquitination of YAP.

Idiopathic pulmonary fibrosis (IPF) is a common, lethal interstitial lung disease characterized by airway remodeling, inflammation, alveolar destruction, and fibrosis. The mammalian target of rapamycin complex 1/4E binding protein 1 (mTORC1/4E-BP1) axis is closely related to the expression of collagen by fibroblasts, and its role in pulmonary fibrosis remains to be further elucidated. Traditional Chinese medicine (TCM) has shown promising efficacy in improving the lung function, exercise capacity, and quality of life in patients with IPF. The theory of "same treatment for different diseases" provides a TCM theoretical basis for the treatment of pulmonary fibrosis with Bupleuri Radix, while the research in western medicine has preliminarily shown that both the formulation and single herb as well as the active ingredients of Bupleuri Radix have good therapeutic effects on pulmonary fibrosis. Therefore, this review will elaborate on the role of the mTORC1/4E-BP1 axis in the pathomechanism of IPF, as well as the research results of the active components of Bupleuri Radix on the phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin protein(PI3K/AKT/mTOR) pathway, so as to provide a reference for the treatment and drug development of IPF.

6.Status of antibiotic use in hospitalized children with community-acquired pneumonia in multiple regions of China.

OBJECTIVE:
To investigate the use of antibiotics in children with community-acquired pneumonia (CAP) in multiple regions of China, and to provide a reference for CAP standard treatment and rational antibiotic use in children.
METHODS:
The medical data of 1 383 children with CAP who were hospitalized in the department of pediatrics in 10 grade A tertiary hospitals from 9 cities between April 14, 2014 and January 1, 2016 were reviewed, to analyze the status of antibiotic use in hospitalized children in North China, Northeast China, East China, and South China.
RESULTS:
The overall rate of antibiotic use in children with CAP was 89.08%, with 88.7% in North China, 95.5% in Northeast China, 83.3% in East China, and 86.6% in South China. The main types of antibiotics used were cephalosporins, macrolides, compound preparations of β-lactam antibiotics, polyphosphoric broad-spectrum antibiotics and other β-lactam antibiotics. The selection of antibiotics was generally rational, but antibiotics were still used in some patients with viral infection alone or a combined use of ≥2 kinds of antibiotics were noted in some patients with infection caused by one kind of pathogen. Irrational antibiotic use was observed in 131 children (10.63%).
CONCLUSIONS
There are high rates of antibiotic use and irrational use of antibiotics among children with CAP. Standard management of antibiotic use in children with CAP should be strengthened.

7.Calculation of Likelihood Ratio for Identifying Half Sibling Relationship When Both Biological Mothers Participate.

Objective To derive the formulae for likelihood ratio (LR) calculation of half sibling relationships when both mothers participate. Methods Based on the fact that both biological mothers participate in the identification of half sibling relationship between the two individuals, test hypothesis for the identification of half sibling relationship was established. Conditional probability ratios of genetic evidence under null hypothesis and alternative hypothesis conditions were simplified, and then applied to a real case of half sibling relationship identification. At the same time, the LR of half sibling relationships under the assumption that only a single biological mother or none of the biological mothers participate were respectively calculated. Results In the cases of identification of half sibling relationship from same fathers, with no biological father involved, after the same genetic indicator test analysis, when both biological mothers participate in the identification, the accumulated LR value was higher than that of accumulated LR with only a single biological mother or no parents participating. Conclusion When the autosome STR test is used for the identification and analysis of half sibling relationship between two individuals, the calculation of LR is more simple, intuitive and operable with both mothers participating. The biological mothers should participate in the test as much as possible, otherwise the number of STR loci would need to be increased for a more specific conclusion.

BACKGROUND: Hyaluronic acid is an important component of extracellular matrix, has good biocompatibility, unique rheological properties and a variety of physiological functions. Therefore, it has wide application prospect in the tissue engineering. OBJECTIVE: To review the latest research progress in hyaluronic acid scaffold materials in different directions of tissue engineering. METHODS: Using the keywords of "hyaluronic acid, tissue engineering, regeneration, scaffold" in English and Chinese, we retrieved relevant articles published from January 2013 to September 2016 in the databases of PubMed, ScienceDirect and CNKI. RESULTS AND CONCLUSION: The application of hyaluronic acid tissue engineering scaffold materials mainly focuses on the regeneration of bone, cartilage, cardiovascular and nerve. The researchers optimize the composition, ratio of different materials, and modification or cross-linking methods to obtain the ideal scaffold materials to match the mechanical properties and physiological activity of the target tissue. However, there are not enough data on the influence of the molecular weight of hyaluronic acid and the original source of other raw materials on the scaffold properties. Further investigations are needed on the clinical transformation of hyaluronic acid tissue engineering products.
9.Effects of Enhanced Exercise and Combined Vitamin D and Calcium Supplementation on Muscle Strength and Fracture Risk in Postmenopausal Chinese Women.

Objective To observe the effects of enhanced exercise and combined vitamin D and calcium supplementation on muscular strength and fracture occurrence in postmenopausal women with a high risk of osteoporosis.Methods Totally 614 postmenopausal women at high risk factors of osteoporosis were enrolled in Dongcheng district of Beijing and randomized into four groups:group A(control group,n=173),group B(regular Tai Chi exercise,n=171),group C(calcium 600 mg/d+VitD800 U/d,n=139),and group D[calcium 600 mg/d+25 hydroxyl vitamin D(25OHD) 0.25 μg/d,n=131].Muscular strength was measured at baseline and one and two years after intervention.Bone turnover markers were measured at baseline and during the two-year follow-up.Falls and fractures were recorded.Results The incidence of 25OHD<50 nmol/L was approximately 92.6%.During the follow-up,the left grip strength decreased significantly two years after intervention(t=-3.252,P=0.001)in group A.Right grip strength decreased significantly in group B(t=2.460,P=0.015)while left grip strength improved significantly in group C(t=-2.051,P=0.043)one year after intervention.In group D,muscular strength in both 12-month and 24-month did not change compared with baseline(both P>0.05).Furthermore,serum procollagen type I N-terminal propeptide elevated significantly in group A(t=-2.962,P=0.004),group B(t=-2.888,P=0.005),and group C(t=-2.441,P=0.016),whereas β-C-terminal telopeptide of type I collagen decreased significantly in group B(t=2.285,P=0.024)and group D(t=2.596,P=0.011)two years after intervention.Conclusion Enhanced exercise and combined calcium vitamin D supplementation may help sustain muscle strength in postmenopausal women,while calcium and vitamin D supplementation may improve muscular strength within a short period of time.
10.Biomechanics of osteonecrosis of femoral head after tantalum rod implantation: a three-dimensional finite element analysis

BACKGROUND: The femoral anterolateral column plays a key role in hip preservation surgery, and the China-Japan Friendship Hospital (CJFH) type stresses the anterolateral column, but there is little information available in three-dimensional (3D) finite element analysis of the L-type tantalum rod implantation.OBJECTIVE: To establish a 3D finite element model of L-type tantalum rod implantation into the normal, necrotic and L-type necrotic areas, followed by a finite element analysis, and to explore the importance of the integrity of the anterolateral column for hip preservation surgery, thus providing biomechanical evidence for the treatment of CJFH type.METHODS: Based on the prepared 3D models of CJFH L1, L2 and L3 osteonecrosis of the femoral head (ONFH) and normal hip, the 3D finite element model of tantalum rod implantation of femoral head necrosis was established, and finite element analysis was conducted. The stress distribution and the maximum Mises stress value of cortical shell of normal femoral head, necrotic femoral head and tantalum rod implanted ONFH were observed and compared.RESULTS AND CONCLUSION: (1) The maximum Mises of the weight-bearing area of cortical bone shell of L-type ONGH was higher than that of normal femoral, L1. L2 and L3 increased by 21.76%, 31.02%, and 42.79%, respectively. (2) The maximum Mises of weight-bearing area of cortical bone shell was decreased after tantalum rod implantation, L1 L2 and L3 decreased by 13.38%, 8.81%, and 11.45%, respectively. (3) In necrotic femoral head, the decreased bearing stress, leads to stress concentration on the femoral head bone shell, so the maximum is higher than that of the normal condition. The higher classification is, the greater the stress concentration of the cortical bone shell is. Tantalum rod implantation share certain stress. (4) Tantalum rod implantation corrects some stresses of the bone shell of ONFH, and can prevent collapse and exert the biomechanical effect of supporting subchondral bone. Furthermore, the integrity of the lateral column of the necrotic femoral head affects the efficacy of hip preservation, and more lateral column preserved always accompanied with better hip preservation. The higher CJFH classification exhibits a higher risk of collapse, and poorer hip preservation.
