OBJECTIVE
Based on the G protein-coupled bile acid receptor(TGR5)/nucleotide binding oligomerization domain-like receptor 3(NLRP3) signaling pathway, to explore the mechanism of Huoxue Qingjieling in improving the inflammatory response of rats with nonalcoholic steatohepatitis(NASH).
METHODS
A total of 70 male SD rats were randomly divided into 5 groups, 20 rats in the control group, 20 rats in the model group, 10 rats in each of the atorvastatin positive drug group, the high-dose and low-dose groups of Huoxue Qingjieling. The control group was given normal feed, and the rest of the groups were given high-fat diet. Through model evaluation, it was determined that the NASH rat model was successfully established at the end of the 20th week. After successful modeling, the control group and the model group were given with normal saline by intragastric administration, the atorvastatin positive drug group, and the high and low dose groups of Huoxue Qingjieling were given corresponding drugs once a day for 4 weeks. At the end of the 24th week, the rats were killed, and the changes of body weight, wet liver weight and liver index was calculated. The serum was taken to test the triglyceride(TG), total cholesterol(TC), alanine aminotransferase(ALT), and aspartic acid aminotransferase(AST) levels by automatic biochemical analyzer. Pathological changes of liver tissues were observed by HE staining and oil red O staining. The expression levels of TGR5 and NLRP3 proteins were detected by Western blotting and immunofluorescence staining. ELISA detected the content of interleukin-18(IL-18) and interleukin-1β(IL-1β) in liver tissue.
RESULTS
Huoxue Qingjieling could significantly improve the general state of NASH rats. Every dose group could significantly reduce the body weight, liver wet weight and liver index of rats(P<0.01), and TG, TC content and ALT, AST activity levels of serum significantly decreased(P<0.01). The pathological results showed that Huoxue Qingjieling could significantly improve liver steatosis, inflammation and balloon-like. The expression of TGR5 protein was significantly increased(P<0.01), the expression of NLRP3 protein and the content of IL-18, IL-1β were significantly decreased(P<0.01, P<0.05).
CONCLUSION
Huoxue Qingjieling can significantly improve the state of NASH rats, inhibit liver steatosis and inflammation, and its mechanism may be closely related to the inhibition of TGR5/NLRP3 signaling pathway.

With the development of modern sequencing techniques and bioinformatics, genomes that were once thought to be noncoding have been found to encode abundant functional micropeptides (miPs), a kind of small polypeptides. Although miPs are difficult to analyze and identify, a number of studies have begun to focus on them. More and more miPs have been revealed as essential for energy metabolism homeostasis, immune regulation, and tumor growth and development. Many reports have shown that miPs are especially essential for regulating glucose and lipid metabolism and regulating mitochondrial function. MiPs are also involved in the progression of related diseases. This paper reviews the sources and identification of miPs, as well as the functional significance of miPs for metabolism-related diseases, with the aim of revealing their potential clinical applications.

To optimize the quantitative detection method for Salmonella enterica and analyze the quantitative contamination level of Salmonella enterica in raw pork samples from farmer′s markets in Chengdu. Based on qualitative detection standard method of Salmonella enterica in China (GB 4789.4-2016) and the quantitative detection method of FSIS in the United States (MLG 4.08 and MLG appendix 2.05 MPN), the selective enrichment broth, screening plate, identification method and quantitative dilution ratio in quantitative detection of Salmonella enterica were optimized using 70 samples of raw pork. The optimized method compared by student′s t-test was used to detect 40 samples of raw pork collected from farmer′s markets in Chengdu from June to October 2020. For isolation of Salmonella from raw pork samples, the coincidence degree of TTB enrichment solution was significantly higher than that of RV enrichment solution (0.93±0.32 vs 0.35±0.62, t=8.324, P=0.001) and the consistency of suspicious colonies screened by XLT4 plate was significantly higher than that of Salmonella chromogenic medium (0.77±0.09 vs 1.00±0.00, t=2.971, P =0.017). The MPN method used 4 successive gradient dilutions, namely 12 tube method, could obtain more accurate quantitative value for Salmonella enterica. The combined use of selective enrichment broth TTB, XLT4 plate, Real-time PCR and MALDI-TOF mass spectrometry could get better results for screening and identifying Salmonella enterica. The detection rate for Salmonella enterica isolated from raw pork in farmer′s markets was 92.5% (37/40). The most of the Salmonella positive samples (83.8%, 31/37) were detected with a contamination level ranged from 0.1 to 55 MPN/g. The optimized quantitative detection method for Salmonella enterica in raw pork in this study can effectively screen the target bacteria and obtain more accurate quantitative value.

OBJECTIVE: To observe improvement effects of fingolimod on middle cerebral artery occlusion/reperfusion (MCAO/R) injury model rats. METHODS: Male SD rats were randomly divided into sham operation group, model group and fingolimod low-dose, medium-dose and high-dose groups (0.5, 1, 2 mg/kg), with 8 rats in each group. Except for sham operation group, MCAO/R injury model was induced by suture-occluded method in other groups. Administration groups were given relevant medicine intragastrically after reperfusion [1 h after reperfusion (1st day), 22.5 h after reperfusion (2nd day), and then every 24 h until 142.5 h of reperfusion (7th day)]. Sham operation group and model group were given constant volume of normal saline intragastrically, once a day, for consecutive 7 d. The scores of neurological deficit and balance beam test, the times of memory error [work memory error (WME), reference memory error (RME) and total error] were recorded in each group. The contents of serum inflammatory cytokines (IL-6, IL-8, IL-10, TNF-α) were determined by ELISA, and triphenyl tetrazolium chloride staining method was used to detect the rate of cerebral infarction. RESULTS: Compared with sham operation group, neurological deficit scores (at different time points of 1st-7th day after administration), balance beam test scores (2nd, 4th, 7th day after administration), times of memory error (2nd, 4th, 7th day after administration), the contents of serum inflammatory cytokines and the rate of cerebral infarction were increased significantly in model group (P<0.05 or P<0.01). Compared with model group, neurological deficit scores (low-dose group at different time points of 3rd-7th day, medium-dose and high-dose groups at different time points of 2nd-7th day after administration), balance beam test scores (low-dose group at 7th day, medium-dose group at 4th and 7th day, high-dose group at 2nd, 4th, 7th day), RME times and total error times (low-dose group at 4th and 7th day, medium-dose group and high-dose group at 2nd, 4th, 7th day after administration), WME times (administrations groups at 7th day after administration), serum contents of IL-6, IL-8 and IL-10 (administrations groups), serum contents of TNF-α (medium-dose and high-does groups) and cerebral infarction rate (medium-dose and high-dose groups) were all decreased significantly (P<0.05 or P<0.01). CONCLUSIONS: Intragastric administration of fingolimod can significantly reduce neurological deficit score, balance beam test score and the times of memory error in MCAO/R injury model rats, and has a protective effect on cerebral tissue and memory function. These effects may be related to the down-regulation of inflammatory cytokines such as IL-6 and TNF-α by fingolimod.

This study was aimed to optimize the hydrolysis conditions of Coix prolamin, which occupied the highest content in total protein of it, and then evaluate the angiotensin-converting enzyme (ACE) inhibitory activity of the corresponding hydrolysates. Pepsin was used to hydrolyze prolamin and OPA method was applied to determine the degree of hydrolysis. The optimum conditions of hydrolyzing prolamin were obtained by orthogonal design subse-quently. The ACE inhibitory activity of small molecular weight (less than 3 kD) peptides gained by ultrafiltration of the hydrolysates was assayed by RP-HPLC method. The result showed that the optimized hydrolysis conditions were substrate concentration of 1%, enzyme-to-substrate ratio of 1:5 and hydrolysis duration for 48 h. The hydrolysates exhibited an ACE inhibitory ratio of (83.40 ± 0.93)% at the concentration of 0.1 mg·mL-1. It was concluded that prolamin hydrolysates displayed a high ACE inhibitory activity in vitro, which provided guidance for further research of antihypertensive component and development of functional food from Coix.
