Background:
Silybum marianum, as well as known milk thistle, has long been recognized for its hepatoprotective effects,
primarily attributed to its active flavonolignan complex, silymarin (an extract from water hyacinth fruit). While the
pharmacological effects of silymarin have been studied, research on bioactive peptides derived from Silybum marianum
remains limited.
Aim:
To evaluate the toxicity effects of silybum marianum peptides
Marerials and Method:
This study aimed to evaluate the potential toxicity of Silybum marianum peptide in mice through
a 14-day oral administration experiment. Twenty adult male C57BL/6 mice were divided into two groups: the experimental
group received 200 mg/kg of Silybum marianum peptide daily, while the control group received an equivalent volume
of saline solution. Physiological and biochemical parameters, including body weight, fasting blood glucose levels, liver
and spleen wet weights, as well as alanine aminotransferase (ALT) enzyme activity, were assessed to determine potential
toxic effects. This exploration aims to shed light on the toxicological effects of silybum marianum peptide in mice, providing
insights into its potential benefits and challenges.
Results:
Results indicated no significant differences between the experimental and control groups in terms of body
weight, blood glucose levels, or major organ wet weights. Additionally, ALT enzyme activity remained unaffected, suggesting
no detectable liver toxicity. Throughout the study, no abnormal behaviors, physical changes, or mortality were
observed in the test subjects. Mice in both the silybum marianum peptide and control groups exhibited shiny and soft
fur, normal activity, and regular food consumption. These findings indicate that Silybum marianum peptide exhibits good
safety and low biological toxicity under the tested conditions, supporting its potential use as a safe dietary supplement or
therapeutic agent.
Conclusion
At the designated dosage, silybum marianum peptide demonstrated good safety and low biological toxicity.

To explore the application of humanistic care in children's fever clinics,the children's fever clinics of Guangzhou Women and Children's Medical Center conducted relevant research using narrative medicine as a guiding concept.This paper elaborated on the medical humanistic dilemmas and narrative care needs in children's fever clinics,and focused on exploring the practice paths of narrative medicine in pediatric with Chinese characteristics from five dimensions,including the cultivation of nursing staff's abilities of professional narrative and humanistic care,the establishment of health lectures featuring narrative patient education,the alleviation of medical anxiety for children and their families,related support of narrative nursing,and caring services.The aim was to improve the narrative care ability of nurses in children's fever clinics,develop methods for pediatric patients that can eliminate the fear of seeking medical treatment,and protect children's physical and mental health.A carrier of care,support,and emotional expression can be provided for parents.The foundation for nursing staff to achieve professional growth through narrative reflection can be laid.Thus,it can assist in establishing a life narrative community relationship between doctors and patients,and jointly explore the meaning of life.
3.Prognostic Significance and Immune Cell Infiltration Analysis of CXC Chemokines in Glioblastoma

Objective To analyze the expression of CXC chemokines in glioblastoma (GBM), and investigate their prognostic value and potential roles as therapeutic targets. Methods Differential expression and prognostic value of CXC chemokines in GBM were obtained in GEPIA. The String database was used to predict neighbor genes and construct the protein to protein interaction (PPI) network. GO analysis and KEGG pathway enrichment analysis were executed by DAVID tools. TRRUST and LinkedOmics were used to predict transcription factor and kinase targets for CXC chemokines. Finally, we discussed the correlation of immune cell infiltration and tumor purity with differential expression of CXC chemokines in GBM. Results CXC2/3/8/9/10/11/16 expression was significantly up-regulated in GBM, and CXC3/8 were significantly associated with the poor prognosis of GBM patients. Differentially expressed chemokines and their adjacent genes are mainly enriched in the immune regulatory and apoptosis functions and pathways. There was correlation between chemokines expression and infiltration of six immune cells. Conclusion CXCL3/8 could be the potential prognostic markers for GBM patients. CXC chemokines are related to GBM immunity.

5.Association of single nucleotide polymorphisms of LEPR gene with essential hypertension among ethnic Mongolian and Han Chinese from Inner Mongolia region.

OBJECTIVETo assess the association of single nucleotide polymorphisms (SNPs) of leptin receptor (LEPR) gene with essential hypertension (EH) and body mass index (BMI) among ethnic Mongolian and Han Chinese from Inner Mongolia region.
METHODSIn total 411 Han Chinese patients with EH and 480 healthy controls, together with 658 Mongolian patients with EH and 403 healthy controls, were collected. The SNPs of the LEPR gene were determined with ligase detection reaction (LDR). Logistic regression was used to analyze the association of the polymorphisms of each locus with EH and BMI. MDR software was used to analyze the interaction between above loci and environmental factors.
RESULTSGenotypic frequencies of LEPR gene rs7555955, rs1137100 and rs1137101 loci had differed significantly among ethnic Hans with EH and the control group (All P<0.05). While those of rs7555955, rs1805094, rs1137100, rs11579567, rs1805134 and rs6669354 loci had differed significantly among ethnic Mongolians with EH and the control group (All P<0.05). After adjustment for confounders, logistic regression analysis indicated that age (OR=2.97, 95%CI:1.94-3.99), BMI (OR=3.93, 95%CI:2.91-5.96), and rs1137101 (AA) (OR=3.96, 95%CI:1.32-11.90) were independent risk factors for EH among ethnic Hans, while age (OR=2.99, 95%CI:2.98-4.57), BMI (OR=3.03, 95%CI:1.05-1.27), rs7555955 (AG, AA) (OR=12.12, 95%CI:2.80-52.43; OR=6.35, 95%CI:1.44-27.94), and rs7555955 (GG) were independent risk factors for EH among ethnic Mongolians (P<0.05).
CONCLUSIONAge and BMI are independent risk factors for EH in both ethnic Han and Mongolian Chinese. rs1137101 locus is associated with EH among ethnic Hans, while rs7555955 locus is associated with EH among ethnic Mongolians.
6.The role of serum miRNA-30c in early diagnosis of gastric cancer

Objective To test the value of miRNA-30c(miR-30c)in early diagnosis of gastric cancer. Methods Serum miR-30c expression levels were detected by using quantitative real-time polymerase chain reaction in 80 patients with advanced gastric cancer, 35 patients with early stage gastric cancer and 35 healthy controls in the Affiliated People's Hospital of Inner Mongolia Medical University from August 2014 to August 2017. The receiver operating characteristic (ROC) curves and the area under the curve (AUC) were analyzed to test the efficacy of the miR-30c in distinguishing advanced gastric cancer, early stage gastric cancer and healthy controls. Results The expression level of serum miR-30c in advanced gastric cancer (0.45±0.11) was lower than that in early stage gastric cancer (0.54±0.15) (t = 5.2, P < 0.05). Compared with the healthy controls (0.61±0.12), miR-30c was down-expressed in the serum of early stage gastric cancer patients(t=6.7,P<0.05).Compared with the traditional tumor markers,the AUC of miR-30c was the biggest (0.92±0.03) in early stage and advanced gastric cancer groups, and the sensitivity and specificity in the diagnosis of gastric cancer were 90 % and 84 %, respectively. The AUC of miR-30c was 0.87±0.04 in early stage gastric cancer and healthy controls, and the sensitivity and specificity in the diagnosis of gastric cancer were 70 % and 86 %, respectively. Conclusion miR-30c might be used as a potential serum biomarker for the early diagnosis of gastric cancer.
7.A study on mechanism underlying the inhibition of berberine on proliferation of human melanoma A375 cells

Objective To evaluate the effect of berberine on the expression of cell cycle-related miRNA and its target gene in human melanoma A375 cells.Methods In vitro cultured A375 cells were classified into several groups to be treated with berberine at different concentrations of 0 (blank control group),20,40,60,80,and 100 μmol/L,respectively,for 48 hours.qRT-PCR was performed to determine the mRNA expression of miRNA-582-5p and its target gene cyclin-dependent kinase 1 (CDK1),miRNA-188-5p and its target gene CDK2,Cyclin D1 and Cyclin A.Western blot analysis was conducted to measure the protein expression of cell cycle-related proteins CDK1,CDK2,Cyclin D1 and Cyclin A.Results qRT-PCR showed that compared with the blank control group,the 20 and 40 μmol/L berberine groups had a similar expression of miRNA-582-5p (both P > 0.05),but the 60 and 80 μmol/L berberine groups had a significantly up-regulated expression of miRNA-582-5p (both P < 0.05).Compared with the blank control group,all the 5 berberine groups had a significantly increased expression of miRNA-188-5p (F =22.600,P =0.002).However,the mRNA expression of CDK2,CDK1,Cyclin A and Cyclin D1 gradually decreased along with the increase of berberine concentrations (F =51.976,248.510,626.671 and 312.740,respectively,all P < 0.001).Western blot analysis revealed that berberine decreased the protein expression of CDK1,CDK2,Cyclin D1 and Cyclin A (F =138.124,110.966,278.772 and 140.167,all P < 0.001).Conclusion Berberine can decrease the expression of cell cycle-related proteins CDK1,CDK2,Cyclin D1 and Cyclin A,likely by decreasing their mRNA expression and increasing the expression of miRNA-582-5p and miRNA-188-5p,and then block the cell cycle progression of A375 cells and inhibit the growth of tumor.
8.Progress in the Regulation of PI3K/AKT/mTOR Signaling Pathway by Ursolic Acid in Colorectal Cancer

Ursolic acid (UA) is an active pentacyclic triterpenoid widely found in grass, leaves, flowers and fruits of plants, which is antioxidant, antimicrobial, anti-inflammatory, hepatoprotective, immunomodulatory, anti-tumor, chemopreventive, cardioprotective and antihyperlipidemic and hypoglycemic. In paper, the ursolic acid structure, antitumor mechanism, PI3K/AKT/mTOR signaling pathway taking part in the process of colorectal cancer, and the regulation of PI3K/AKT/mTOR signaling pathway by ursolic acid in stopping the development of colorectal cancer was summarized.
9.Preliminary analysis of miRNA expression profile of gastric cancer tissues by microarray

Objective To analyze the expression profile of specific miRNA of gastric cancer tissues.Methods miRNA were isolated from five paired gastric cancer tissues and corresponding paracancerous normal tissues.To identify the profile of gastric cancer,miRNA hybridization and cluster analysis were processed by significance analysis of microarrays (SAM,version 2.1) and cluster software.The relative expression of hsa-miR-30c,hsa-miR-196a and hsa-miR-193b were validated by fluorescence Real-Time PCR using taqman probe.Further hsa-miR-30c taget gene was predictived by online software.Results Revealed by miRNA microarray detection,there were 16 miRNA with differential expression in gastric cancer tissues and para-cancerous normal tissues,among which HS_100,hsa-miR-27a,hsa-miR-214*,solexa-555-1991 and hsa-miR-196 were significantly up-regulated,and hsa-miR-502-3p,hsa-miR-29c*,hsa-miR-64,hsa-miR-193b,hsa-miR-551b,hsa-miR-30c,hsa-miR-582-5p,hsa-miR-625*,hsa-miR-660,hsa-miR-363 and hsa-miR-486-5p were down-regulation.The relative expression of hsa-miR-30c,hsa-miR-196a and hsa-miR-193b was in high concordance with microarray.Conclusion There is specific miRNA expression profile in gastric cancer tissues,among which differentially-expressed miRNAs may be involved in the regulation of gastric cancer progress and serve as new potential biomarkers.
10.Expression of vascular endothelial growth factor-C and vascular endothelial growth factor-D in gastric carcinoma tissue and relationship with lymph vessel density and lymph node metastasis

Objective To detect the expression of vascular endothelial growth factor(VEGF)-C and VEGF-D in normal gastric tissue and gastric carcinoma tissue,and further investigate the relationship between VEGF-C,VEGF-D and lymph vessel density (LVD),lymph node metastasis.Methods The expression of VEGF-C and VEGF-D in gastric carcinoma tissues were examined by using SP immunohistochemical method in 30 normal gastric tissue and 75 gastric carcinoma tissue respectively.The lymph vessel were marked by LYVE-1 and calculated LVD.Results The rate of positive expression of VEGF-C and VEGF-D in gastric carcinoma tissue was significantly higher than that in normal gastric tissue,there was significant difference [73.3 % (55/75) vs.33.3 % (10/30),66.7 % (50/75) vs.30.0 % (9/30),P <0.01].The level of LVD in gastric carcinoma tissue with lymph node metastasis (40 cases) was significantly higher than that in without lymph node metastasis (35 cases)[(8.85 ± 1.24)/HP vs.(4.75 ± 1.04)/HP](P =0.000).The level of LVD increased when the expression level of VEGF-C and VEGF-D gradually went up.Conclusions The level of LVD increases in gastric carcinoma tissue.VEGF-C and VEGF-D are two significant factors for LVD and lymph node metastasis.
