1.Involvement of RNF99 in potential link between ubiquitination and septic shock via TAK1/NF-κB signal-ing pathway

		                        			Objective To explore the potential relationship between ubiquitination of transforming growth factor kinase 1(TAK1)/nuclear factor-κB(NF-κB)signaling pathway mediated by ring finger protein 99(RNF99)and septic acute respiratory distress syndrome(ARDS).Methods Plasmid and siRNA transfection were conducted to overexpress or knock down RNF99 in MLE12,and expressions of p65 phosphate and p65 protein were analyzed.The protein interaction between RNF99 and TRAF6 or TAK1 was analyzed by immunoprecipitation assay.Forty mice were randomly divided into WT plus PBS,WT plus LPS,RNF99 specific expression(TG)plus PBS,and TG plus LPS groups,with 10 mice in each group.Sepsis was induced by intraperitoneal injection of 30 mg/kg LPS.Results As compared with vector group,protein expression levels of TRAF6 and TAK1 in MLE12 cells decreased significantly in RNF99 group(P<0.05).Ubiquitinated TRAF6 protein increased in MLE12 cells with RNF99 knockdown.As compared with LPS plus vector group,phosphorylation level of p65 in MLE12 cells was signifi-cantly lower in LPS plus RNF99 group(P<0.05).As compared with si-NC group,protein expression levels of RNF99 and IκBα in si-RNF99 group decreased significantly(P<0.05).As compared with LPS plus si-NC group,phosphorylation level of p65 in LPS plus si-RNF99 group increased significantly(P<0.05).The staining percentage of CD68 macrophages in lung tissues was significantly lower in TG plus LPS group than in WT plus LPS group(P<0.05).Phosphorylation level of p65 in lung tissues was significantly lower in TG plus LPS group than in WT plus LPS group(P<0.05).Conclusion RNF99 regulates NF-κB signaling pathway by interacting with the key regulator of NF-κB signaling pathway(TRAF6/TAK1),and improves lung injury after intraperitoneal injection of LPS in mice.
		                        		
		                        		
		                        		
		                        	
2.Study on the influence of mindfulness therapy on self-efficacy, compliance and quality of life in hemodialysis patients

		                        			Objective:To explore the effects of mindfulness therapy on self-efficacy, compliance and quality of life in hemodialysis patients.Methods:A total of 94 patients who underwent hemodialysis treatment at Jiangxi Provincial People′s Hospital from December 2017 to January 2019 were selected. Random numbers were used to divide the control group and the observation group including 47 cases respectively. Patients in the control group were given routine care and medical intervention according to the procedures of the hospital. Patients in the observation group were given mindfulness therapy on the basis of the control group. The self-efficacy and quality of life of the two groups of patients were compared before and after intervention. Comparison of treatment compliance and intervention satisfaction between two groups of patients.Results:The self-efficacy scores of patients in the control group before and after the intervention were (1.25 ± 0.18) and (2.32 ± 0.35) points. The observation group′s self-efficacy scores were (1.28 ± 0.14) and (2.97 ± 0.33) points, respectively. The difference between the two groups before and after the intervention was statistically significant ( t values were -18.638, -32.321, P <0.01). There was a statistically significant difference between the two groups after intervention ( t value was -9.264, P <0.01). The treatment compliance score of the observation group was (82.69±13.59) points, which was higher than (71.53±12.25) points of the control group ( t value was 4.182, P <0.05). The scores of the psychological status, physiological status, environmental domain, and social relationship of the patients in the control group before the intervention were (54.37 ± 8.16), (50.32 ± 5.54), (53.28 ± 7.99), and (51.74 ± 6.21) points. The observation group was (55.03 ± 7.70), (51.38 ± 6.17), (53.94 ± 7.01), (50.88 ± 7.63) points. The control group after the intervention were (62.25 ± 6.85), (61.22 ± 9.18), (60.28 ± 7.23), and (55.69 ± 7.80) points. The observation group was (70.64 ± 8.48), (71.39 ± 7.85), (68.04 ± 8.85), and (60.26 ± 6.63) points. The differences between the two groups before and after the intervention were statistically significant ( t values were -13.739 - -2.716, P <0.01). The difference between the two groups after intervention was statistically significant ( t values were -5.772 to -3.060, P <0.01). The intervention satisfaction rate in the control group was 82.98% (39/47), and the intervention satisfaction rate in the observation group was 95.74% (45/47). The difference was statistically significant ( χ 2 value was 4.029, P <0.05). Conclusion:Intervention of hemodialysis patients with mindfulness therapy is beneficial to improve patients′ self-efficacy level, improve treatment compliance, and improve quality of life.
		                        		
		                        		
		                        		
		                        	
3.Investigation on bone health conditions in 1637 aged women

		                        			Objective To investigate bone health conditions in 1637 aged women. Methods From May 2004 to October 2008, Bone mineral density (BMD) of 1637 women at age of more than 60 years old were measured by Hologic DephiA dual energy X-ray absorptiometry (DXEA) in Huadong hospital affiliated to Fudan University. All data were compared and analyzed among each group which will be divided by every ten years. Those women were divided into groups on 10 years range. BMD of lumbar vertebral and hip bone,fracture incidence and bone turnover marker were compared and analyzed. Results (1) BMD: at age of 3=90, 80 - 89, 70 - 79, 60 - 69, BMD of the lumbar vertebral 2 - 4(L2-4) values were (0. 96 ± 0. 18),(0. 90 ± 0. 20) , (0. 81 ± 0. 16) , (0. 83 ± 0. 14) g/cm2 , respectively. There were significantly increased BMD of lumbar of women at the age of 80 - 89 and ≥90 year-old compared with those of 60 - 69 year-old (P<0. 05). At age of ≥90,80 -89,70-79,60 -69 BMD of femur neck, Total, Torch, Ward's trianger were(0. 60 ±0. 11) , (0. 65 ±0. 11) ,(0. 47 ±0. 09) ,(0. 37 ±0. 09) g/cm2; at age of 80 -89 BMD of FN,Total, Torch, Ward's trianger were(0. 57 ±0. 10) , (0. 68 ±0. 13) , (0. 48 ±0. 11) , (0. 35 ±0. 10) g/cm2;at age of 70-79 BMD of FN, Total, Torch, Ward's trianger were (0.57±0. 10) ,(0. 69 ±0. 12), (0.49± 0. 10) , (0. 36 ± 0. 11) g/cm2; at age of 60 - 69 BMD of FN, Total, Torch, Ward's trianger were (0. 63 ± 0. 10) , (0. 76 ±0. 12) , (0. 54 ±0. 10), (0. 45 ±0. 15) g/cm2; There were significantly decreased in BMD of hip at the age of 70 - 79, 80 - 89,≥90 year-old compared with those of 60 - 69 year-old (P < 0. 05).(2) Fracture incidence: one time fracture incidence at age of 60 - 69,70 - 79,80 - 89, ≥ 90 were 34. 8 % (242/695) ,45. 0% (296/658) ,51. 3% (137/267) ,5/17. There were increasing trend of fracture in aged women. (3) Bone turnover marker of bone Gla protein (BGP) N-mid(N-midBGP) in serum and C-terminal telopeptide of type Ⅰ collagen/Cr (CTX/Cr) in urine values were (17 ±5) μg/L, (106 ±56) μg/mmol at age of more than 90 years ,(17 ±7) (μg/L,(128 ±99)μg/mmol at age of 80-89 years,(21 ± 14) μg/L,(182 ± 173)μg/mmol at age of 70-79 years, (25±13)μg/L, (190 ± 168)μg/mmol at age of 60 - 69 years. There were significant decreased trends of N-midBGP at age of 70 -79,80 -89 compared with that of 60-year (P < 0. 05). There were significant decreased trends of CTX/Cr 80 - 89 compared with that of 60-year (P < 0. 05). Conclusions There were significant decreased bone metabolism in aged women. The risk of hip fracture is significantly increased in aged women. 4.The clinical observation of the transobturator tension-free vaginal tape surgery to treat female stress urinary incontinence

		                        			Objective To evaluate the efficacy and safety of the transobturator tension-free vaginal tape (TVT-O) surgery, a minimally invasive surgery for treating patients with the stress urinary incontinence (SUI). Methods 28 cases with female stress uri-nary incontinence treated by TVT-O procedure from April 2006 to June 2008 were retrospectively analyzed. The follow-up time is from 4 to 24 months. Results The mean operation time was 23 min (rang 10~30 min) and the mean intraoperative bleeding was 24 ml (range 15~40 ml). An indwelling catheter had been used for 3 d because of urinary retention in 3 cases. 24 patients(85.7%)reached complete control after the surgery,effective control in 4 patients(14.35%).
