1.A subcutaneous transplanted tumor mouse model of oral cancer overexpressing miR-181a-5p study of small intestine metabolomics

Objective To analyze the effects of miR-181a-5p overexpression on metabolites in the small intestines of mice with subcutaneous oral cancer by detecting changes in metabolites and metabolic pathways.Methods Three groups were included in study:Control group,negative control and miR-181a-5p overexpression group.To establish a subcutaneous oral cancer model in mice,variously treated cell suspensions were subcutaneously injected into the upper right of the groin in female M-NSG severely immunodeficient mice.Changes in pathology and small intestinal tissues were assessed by HE staining.Changes in mouse body weight were also assessed.Tandem orbitrap mass spectrometry and ultra-high performance liquid chromatography-tandem time-of-flight mass spectrometry,were used to examine metabolites in the small intestines.By pre-analyzing the original data and quality rating sample data,XCMS was able to assess which metabolites were different among the groups.To identify unique metabolic pathways,KEGG enrichment analysis was used.Results A total of 170 distinct metabolites were found in the small intestinal tissues of Control and NC groups.Choline metabolism,alanine,aspartate,and glutamate metabolism,GABA synaptic metabolism,glycerophospholipid metabolism,cAMP signaling route,cancer center carbon metabolism,and niacin and niacin amine metabolic pathways were important signaling pathways for metabolite enrichment.In the NC group,16 distinct metabolites with VIP values larger than 2 were found in the small intestines compared with the OE group overexpressing miR-181a-5p.Glycerin phosphorylcholine,palmitic acid,3-hydroxybutyl carnitine,and β-hydroxybutyric acid were among the metabolites that significantly varied.The primary enhanced metabolic pathway was the choline pathway.Conclusions Mouse small intestines underwent slight changes from subcutaneous oral cancer with the greatest effect on metabolites critical for energy metabolism.The choline metabolic pathway was the pathway that selected absolutely metabolites in mouse small intestines with subcutaneous grafts of oral cancer.

Data analysis models may assist the transmission of traditional Chinese medicine (TCM) experience and clinical diagnosis and treatment, and the possibility of constructing a “data-knowledge” dual-drive model was explored by taking gastric precancerous state as an example. Data-driven is to make clinical decisions around data analysis, and its syndrome-differentiation decision-making research relies on hidden structural models and partially observable Markov decision-making processes to identify the etiology of diseases, syndrome elements, evolution of pathogenesis, and syndrome differentiation protocols; knowledge-driven is to make use of data and information to promote decision-making and action processes, and its syndrome-differentiation decision-making research relies on convolutional neural networks to improve the accuracy of local disease identification and syndrome differentiation. The “data-knowledge” dual-driven model can make up for the shortcomings of single-drive numerical simulation accuracy, and achieve a balance between local disease identification and macroscopic syndrome differentiation. On the basis of previous research, we explored the construction method of diagnostic assisted decision-making platform for gastric precancerous state, and believed that the diagnostic and decision-making ability of doctors can be extended through the assistance of machines and algorithms. Meanwhile, the related research methods were integrated and the core features of gastric precancerous state based on TCM syndrome differentiation and endoscopic pathology diagnosis and prediction were obtained, and the elements of endoscopic pathology recognition based on TCM syndrome differentiation were explored, so as to provide ideas for the in-depth research and innovative application of cutting-edge data analysis technology in the field of intelligent TCM syndrome differentiation.
3.Effects of Huatan Quyu Decoction on cognitive function of vascular dementia rats by regulating Wnt/β-catenin signal pathway

Objective:To observe the effects of Huatan Quyu Decoction on the protein expressions of β-catenin and GSK-3 β and the expression of anticardiolipin antibody and β-amyloid protein related to cognitive function in rats with vascular dementia based on Wnt/β-catenin signal pathway.Methods:A total of 96 male SD rats were divided into blank group, model group, Donepezil hydrochloride group and Huatan Quyu Decoction low-, midium-, high-dosage group according to random number table method, with 16 rats in each group. Except for the blank group, the rat model of vascular dementia was prepared by modified 2-VO method. Huatan Quyu Decoction low-, medium- and high-dosage groups were administrated with Huatan Quyu Decoction 6.1, 12.1 and 24.2 g/kg, respectively; the Western medicine group was administrated with Donepezil hydrochloride 0.5 mg/kg; the blank group and the model group were administrated with the same amount of normal saline for 28 consecutive days. On the 1st, 7th, 14th and 28th day after administration, the learning and memory ability of rats was evaluated by Morris water maze test, the levels of ACA and Aβ in serum were measured by enzyme linked immunosorbent assay (ELISA), and the expressions of β-catenin and GSK-3β proteins related to Wnt/β-catenin signal pathway in hippocampus were measured by Western blot.Results:Compared with model group, the escape latency was shortened in the Huatan Quyu Decoction high-dosage group and Donepezil group on 7 and 14 days of administration ( P<0.05), and the times of crossing the platform increased in Huatan Quyu Decoction high-dosage group on 1 and 28 days of administration ( P<0.05). Compared with model group, the serum ACA level in Donepezil group, Huatan Quyu Decoction medium- and high-dosage groups decreased at day 1, 7, 14 and 28 after administration ( P<0.05). The serum Aβ level in Donepezil group, Huatan Quyu Decoction medium- and high-dosage groups decreased at 7, 14 and 28 days after administration ( P<0.05); On the 14th and 28th days after administration, the levels of ACA and Aβ in TCM low-dosage group decreased ( P<0.05). Compared with model group, the expression of β-catenin protein in hippocampus of Donepezil group and Huatan Quyu Decoction medium- and high-dosage groups increased ( P<0.05), while the expression of GSK-3β in hippocampus of Donepezil group and Huatan Quyu Decoction low-, medium- and high-dosage groups decreased ( P<0.01). Conclusion:Huatan Quyu Decoction can activate the Wnt/β-catenin signaling pathway, up-regulate the expression of β-catenin protein in hippocampal tissue of rats, inhibit the expression of GSK-3β, reduce the levels of ACA and Aβ in serum of rats, and improve the cognitive function of rats with vascular dementia.
4.Study on the correlation between fat content of supraspinatus muscle by spectral CT and tendon tear severity of supraspinatus as well as shoulder joint function

Objective To analyze the relationship between the fat content of supraspinatus muscle measured by spectral CT fat-muscle material decomposition technology and the tendon tear severity(TTS)of supraspinatus,as well as shoulder joint function.Methods A total of 108 patients were prospectively selected to undergo routine MRI and spectral CT imaging of the shoulder joints.The patients were divided into normal,degenerative,partial tear,and full-thickness tear groups based on MRI findings.A fat-muscle material decomposition image was generated through spectral analysis to measure the fat content of supraspinatus muscle.The differences in fat content of supraspinatus muscle between the groups were compared,and correlation analyses were performed between the fat content of supraspinatus muscle and the clinical baseline characteristics,symptom duration,and Constant-Murley scores in patients with supraspinatus tendon tears.Results Comparison of supraspinatus muscle fat content among the four groups:the full-thickness tear group>the partial tear group>the degenerative group>the normal group(P<0.05).There was a strong positive cor-relation between the supraspinatus muscle fat content and the supraspinatus TTS(r=0.654,P<0.05).Among patients with partial and full-thickness tears of the supraspinatus tendon,there was a moderate positive correlation between supraspinatus muscle fat content and patient age(r=0.393,P<0.05),a weak positive correlation with gender(r=0.199,P<0.05),with higher fat content observed in females compared to males,no significant correlation with symptom duration(r=0.037,P>0.05),a moderate positive correla-tion with body mass index(BMI)(r=0.463,P<0.05),and a moderate negative correlation with Constant-Murley score(r=-0.316,P<0.05).Conclusion In patients with rotator cuff tears,the quantitative fat content of supraspinatus muscle based on spectral CT fat-muscle material decomposition technology is strongly positively correlated with rotator cuff injury degree,and is also correlated with patient age,gender,BMI,and the strength of the shoulder joint muscles and range of the motion.

Objective:To study the life characteristics and related risk factors of patients with depression in Shandong Province.Methods:Based on the 2015 mental epidemiological survey database in Shandong Province,a total of 832 patients with depression,807 high-risk individuals with depression,and 819 low-risk individuals were diagnosed according to the Diagnostic and Statistical Manual of Mental Disorders,Fourth Edition(DSM-Ⅳ)and the Structured Clinical Interview for DSM-Ⅳ-TR Axis I Disorders,Research Version(SCID-I/P).In 2020,SCID-I/P was used for re diagnosis,and the General Health Questionnaire(GHQ-12),Simple Quality of Life question-naire,Pittsburgh sleep quality index(PSQI),Childhood Trauma Questionnaire(CTQ),Social Support Rating Scale(SSRS),Global Pain Scale(GPS),Montreal Cognitive Assessment(MoCA),Simplified Coping Style Question-naire(SCSQ)were used for evaluation.Compare changes in the quality of life of depression patients and construct a risk factor model.Results:Patients with depression had lower scores on the simple quality of life questionnaire at baseline and at retest after 5 years than those in the high-and low-risk groups,those in remission of depression had higher scores on the simple quality of life questionnaire at baseline and at retest after 5 years than those in non-re-mission,and those with new-onset disorder in the high-and low-risk groups had lower scores on the simple quality of life questionnaire at baseline and at retest after 5 years than those with no-onset disorder(Ps<0.001).Depres-sion diagnosis and PSQI scale scores at baseline negatively predicted at retest after 5 years(β=-0.06,-0.15),while coping style tendencies at baseline positively predicted(β=0.06).The simple quality of life questionnaire at baseline negatively predicted depression diagnosis at retest after 5 years,GHQ-12 scores at retest after 5 years,and PSQI scale scores at retest after 5 years(β=-0.11,-0.17,-0.09),while the simple quality of life question-naire at baseline positively predicted coping style tendencies at retest after 5 years(β=0.13).Depression diagnosis at retest after 5 years,GHQ-12 scores at retest after 5 years,PSQI scale scores at retest after 5 years,coping style tendencies at retest after 5 years,SSRS scale scores,CTQ scale scores,GPS scale scores,and the simple quality of life questionnaire at baseline all influenced the simple quality of life questionnaire at retest after 5 years through ei-ther direct or indirect pathways.Conclusion:It suggest that the quality of life is lower in patients with depression than in the general population.Depression diagnosis,sleep,mental health,pain,social support,childhood trauma and coping are direct and indirect risk factors affecting life.
6.Analgesic effect of dexmedetomidine combined with butorphanol in patients subjected to cardiac surgery

Objective:To investigate the effect of dexmedetomidine combined with butorphanol on perioperative analgesia in patients subjected to cardiac surgery.Methods:Sixty-three patients who underwent elective cardiac surgery in Weihai Central Hospital from June 2019 to August 2020 were included in this study. They were divided into propofol + sufentanil group ( n = 21), dexmedetomidine + sufentanil group ( n = 23) and dexmedetomidine + butorphanol group ( n = 19) according to different analgesic methods. Postoperative analgesic satisfaction, Visual Analogue Scale score, hemodynamic changes (heart rate, respiratory rate, systolic blood pressure, diastolic blood pressure) and adverse reactions were compared among the three groups. Results:The satisfaction rate of postoperative analgesia in the dexmedetomidine + butorphanol group was 94.7% (18/19), which was significantly higher than 61.9% (13/21) in the propofol + sufentanil group and 60.8% (14/23) in the dexmedetomidine + sufentanil group ( χ2 = 6.16, 6.57, both P < 0.05). At 4, 12, 24 and 48 hours after tracheal extubation, Visual Analogue Scale score in the dexmedetomidine + butorphanol group were significantly lower than that in the propofol + sufentanil group and dexmedetomidine + sufentanil group (both P < 0.05). At the time of tracheal extubation and at 5 minutes after tracheal extubation, heart rate, respiratory rate, systolic blood pressure and diastolic blood pressure in the dexmedetomidine+butorphanol group were significantly lower than those in the propofol + sufentanil group and dexmedetomidine + sufentanil group (both P < 0.05). The incidence of adverse reactions in the dexmedetomidine + butorphanol group was 10.5% (2/19), which was significantly lower than 23.8% (5/21) in the propofol + sufentanil group and 30.43% (7/23) in the dexmedetomidine + sufentanil group [30.4% (7/23), χ2=21.94, P < 0.001]. Conclusion:Dexmedetomidine combined with butorphanol in cardiac surgery can not only stabilize postoperative blood pressure and heart rate, but also lower the degree of pain and is highly safe.
7.Clinical characteristics of diarrhea among children infected with rotavirus in Beijing, 2011 to 2018

Objective:To provide evidence for clinical diagnosis, prevention and control of group A rotavirus (RVA) diarrhea, the clinical characteristics of RVA diarrhea in children in Beijing from 2011 to 2018 were analyzed.Methods:From January 2011 to December 2018, 4 819 stool samples from children under 5 years old with diarrhea were collected monthly from 3 hospitals in Beijing. General information, clinical characteristics and other information of children were collected. RVA was detected by enzyme-linked immunosorbent assay (ELISA), genotype was identified by multiple semi-nested RT-PCR. The Vesikari clinical severity score was used to define the severity of diarrhea in children. Dichotomous unconditional logistic regression was used to analyze clinical symptoms and other differences between RVA positive and negative cases. Chi-square and Fisher direct probability tests were used to compare the composition among different groups.Results:A total of 4 819 fecal samples were collected, 953 were positive for RVA, the positive detection rate was 19.78%. The positive rate of RVA was high in the younger age group, and the incidence was high in winter and spring. RVA-positive children had more risk on diarrhea ≥5 times a day, vomiting symptoms, fever, mild dehydration, and Vesikari score ≥11. The positive rate of RVA in watery stool samples (26.13%, 214/819) and infectious diarrhea cases (42.20%, 265/628) was the highest respectively. There were no significant differences in clinical symptoms, clinical diagnoses and fecal traits among children with different RVA genotypes.Conclusions:The clinical symptoms of RVA diarrhea were severe in children. RVA genotype did not affect the clinical symptoms. Stool traits (watery stools) and Vesikari score can assist physicians in diagnosing RVA diarrhea.
8.Effect of tiopronin combined with glutathione on biochemical indicators of patients with non-alcoholic fatty liver

Objective To observe the effect of tiopronin combined with glutathione on the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), glutamyltransferase (GGT),blood fat and laminin (LN) in patients with non-alcoholic fatty liver. Methods A total of 84 non-alcoholic fatty liver patients admitted to our hospital from March 2018 to September 2019 were selected and randomly divided into control group and observation group, with 42 cases in each group. The control group was treated with tiopronin, and the observation group was treated with glutathione and tiopronin. The levels of ALT, AST, GGT and blood fat were recorded and compared before and after treatment. Results After treatment, the levels of ALT, AST and GGT in the two groups were significantly lower than before treatment (P<0.05). After treatment, the levels of ALT, AST, and GGT in the observation group were different from those in the control group, which was statistically significant (P<0.05). Before treatment, there was no difference in serum TC, TG, and LDL levels between the two groups, which was not statistically significant (P>0.05). The above-mentioned serum levels of the observation group after treatment were lower than those in the control group, and there was a difference, which was statistically significant (P<0.05); the levels of PCⅢ, PCⅣ, and LN in the treatment group after treatment were significantly lower than those of the control group. The difference was statistically significant (P<0.05). Conclusion The application of tiopronin combined with glutathione in the treatment of non-alcoholic fatty liver can promote the recovery of liver function and reduce the concentrations of TC, TG and LDL, which is worthy of clinical promotion.
9.circ_0000285 regulates VAMP2 through sponging miR-127 and participates in the development of ovarian cancer

Objective:To investigate the effects of circ_0000285/miR-127/VAMP2 axis on proliferation and invasion of ovarian cancer (OC) cells.Methods:Real-time quantitative polymerase chain reaction (qRT-PCR) was used to detect the expression of circ_0000285, miR-127 and VAMP2 in tissue with OC and normal tissue respectively, and the correlation circ_0000285/miR-127/VAMP2 was analyzed by correlation analysis. Dual luciferase reporter assay was used to verify the targeting relationship between miR-127 and circ_0000285/VAMP2. Cell proliferation and invasion were subsequently detected by MTT and Transwell assay.Results:Compared with the paracancerous tissue, the expression of miR-127 was significantly decreased, while the expression of circ_0000285 and VAMP2 was increased in OC tissue. miR-127 was negatively correlated with circ_0000285 and VAMP2 ( r=-0.534 8, P<0.000 1; r=-0.376 6, P<0.000 1) . Compared with proliferative activity at 24 h, 48 h, 72 h (0.47±0.03) (0.79±0.05) (1.16±0.09) and invasion (100.00±12.33) in si-NC group, knocking down of circ_0000285 inhibited the proliferation (0.28±0.02) (0.51±0.04) (0.78±0.06) and invasion (49.22±5.08) of OC cells (all P<0.05) . Overexpression of miR-127 could also restrain proliferation, invasion of OC cells, but this effect can be partly saved by circ_0000285. The regulation circ_0000285 on OC may be realized through miR-127/VAMP2. Conclusions:circ_0000285 participates in the progression of ovarian cancer via regulating miR-127/VAMP2 axis. Down-regulating the expression of circ_0000285 inhibits the proliferation and invasion of ovarian cancer cells, circ_0000285 is expected to play a role in the targeted therapies of OC.
10.Correlation of insulin-like growth factor-1 and its receptor in ovarian cancer with clinicopathological features

Objective:To study the correlation between insulin-like growth factor-1 (IGF-1) and its receptor IGF-1R expression in ovarian cancer and clinicopathological features, surgical prognosis.Methods:The positive expression rates of IGF-1 and IGF-1R in ovarian cancer, borderline ovarian tumors and benign ovarian tumors were detected by immunohistochemistry. The correlation of IGF-1 and IGF-1R with clinicopathological features, surgical prognosis was analyzed.Results:The positive expression rate of IGF-1 and IGF-1R in ovarian cancer was 65.63%, 70.31%, significantly higher than 22.22%, 16.67% in borderline ovarian tumors, 5.00%, 10.00% in benign ovarian tumors ( χ2/ P=27.548/0.000, 31.335/0.000) . The mRNA and protein expression of IGF-1 and IGF-1R were also significantly higher than borderline ovarian tumors and benign ovarian tumors ( F/P=31.922/0.000, 26.865/0.000, 34.567/0.000, 27.667/0.000) . The positive expression rates of IGF-1 and IGF-1R in ovarian cancer with FIGOIII-IV stage, tissue differentiation G2-G3 stage, CA125>35 U/mL, Ki-67 positive expression were significantly higher than those in ovarian cancer with FIGOI-II stage, tissue differentiation G1 stage, CA125<35 U/mL, Ki-67 negative expression (IGF-1: χ2/ P=8.505/0.004, 4.980/0.026, 7.481/0.006, 10.907/0.001, IGF-1R: χ2/ P=9.785/0.002, 4.950/0.026, 7.211/0.007, 6.471/0.011) . The total survival time of ovarian cancer patients with positive IGF-1 and IGF-1R was shorter than those of patients with negative IGF-1 and IGF-1R. Conclusion:IGF-1 and IGF-1R are highly expressed in ovarian cancer and are associated with the deterioration of pathological features and surgical prognosis.
