The difference in the crystal form of the drug leads to different solubility and stability,affecting drug's the clinical efficacy and bioavailability.Therefore,in recent years,drug crystal forms have received more and more attention from researchers.By reviewing and sorting out relevant literature,this paper summarizes the research progress of drug crystallization theory in recent years from the aspects of influencing factors of drug crystal form,identification and control of drug crystal form,drug crystal form evaluation methods,and applications,aiming to provide a reference for drug crystal form control and process screening.
2.Research Progress in Environment-responsive Nano-drug Delivery System Based on Traditional Chinese Medicine Monomer in Tumor Therapy

Traditional Chinese medicine monomer has certain curative effects in tumor treatment,but its low targeting and difficulty in gathering at the tumor site limit its application.As a new type of drug delivery method,environmentally responsive nano drug delivery system has the advantages of targeted drug delivery,reducing drug side effects,and environmentally responsive release,and has been widely used in tumor treatment.In this paper,by consulting and sorting out the relevant literature,the general situation of traditional Chinese medicine monomers in the treatment of tumors,the classification of environment-responsive traditional Chinese medicine monomer nano drug delivery system,and its application in tumor treatment are summarized in three aspects,to provide ideas for solving the limitations of traditional Chinese medicine monomer in the treatment of tumor.
3.Anti-Seizure and Neuronal Protective Effects of Irisin in Kainic Acid-Induced Chronic Epilepsy Model with Spontaneous Seizures.

An increased level of reactive oxygen species is a key factor in neuronal apoptosis and epileptic seizures. Irisin reportedly attenuates the apoptosis and injury induced by oxidative stress. Therefore, we evaluated the effects of exogenous irisin in a kainic acid (KA)-induced chronic spontaneous epilepsy rat model. The results indicated that exogenous irisin significantly attenuated the KA-induced neuronal injury, learning and memory defects, and seizures. Irisin treatment also increased the levels of brain-derived neurotrophic factor (BDNF) and uncoupling protein 2 (UCP2), which were initially reduced following KA administration. Furthermore, the specific inhibitor of UCP2 (genipin) was administered to evaluate the possible protective mechanism of irisin. The reduced apoptosis, neurodegeneration, and spontaneous seizures in rats treated with irisin were significantly reversed by genipin administration. Our findings indicated that neuronal injury in KA-induced chronic epilepsy might be related to reduced levels of BDNF and UCP2. Moreover, our results confirmed the inhibition of neuronal injury and epileptic seizures by exogenous irisin. The protective effects of irisin may be mediated through the BDNF-mediated UCP2 level. Our results thus highlight irisin as a valuable therapeutic strategy against neuronal injury and epileptic seizures.

4.Preparation and immune-enhancing effects of monophosphoryl lipid A pH-sensitive liposome

Objective:To prepare pH-sensitive liposomes to avoid the degradation of monophosphoryl lipid A (MPLA) by lysosomes.Methods:Using DOPE and CHEMS as carrier materials, pH-sensitive liposomes were prepared by thin-film dispersion method. Particle sizes and Zeta potential of the liposomes were detected by dynamic light scattering. The morphological features of pH-sensitive liposomes under different pH conditions were observed by transmission electron microscopy. Flow cytometry was performed to detect the phagocytosis of liposomes by THP-1 and DC2.4 cells. Confocal laser microscopy was used to observed the colocalization of liposomes and lysosomes. BALB/c mice were immunized with hepatitis B surface antigen (HBsAg) using MPLA pH-sensitive liposome as an adjuvant. The levels of serum anti-HBs were quantitatively detected by ELISA. IFN-γ and IL-2 spot forming cells (SFCs) in mouse splenic lymphocytes were detected by ELISPOT.Results:The pH-sensitive liposomes were constructed with an average particle size of (90.90±1.13) nm, polydispersity index (PDI) of 0.076±0.013 and Zeta potential of (-27.900±0.666) mV. As the pH value of the solution decreased, the particle size increased significantly and the liposomes presented irregular shapes, indicating the pH-sensitive features. The phagocytosis rates by THP-1 cells and DC2.4 cells were 10.40% and 12.40% for pH-sensitive fluorescent liposomes, and 1.09% and 0.28% for fluorescent liposomes. Confocal laser microscopy revealed that pH-sensitive fluorescent liposomes were phagocytosed by THP-1 cells and existed in the cytoplasm, while fluorescent liposomes existed in lysosomes. Compared with MPLA liposomes, MPLA pH-sensitive liposomes could significantly improve the cellular immune response in mice. The levels of IFN-γ and IL-2 SFCs in the MPLA pH-sensitive liposome group were significantly higher than those in the MPLA liposome group ( P<0.01) and the non-adjuvant group ( P<0.001). Conclusions:The pH-sensitive liposome delivery system could improve the utilization of MPLA as an adjuvant.
5.Expression of recombinant varicella-zoster virus gE Δ-Fc in CHO cells and analysis of its immunogenicity

Objective:To express the recombinant varicella-zoster virus (VZV) gE Δ-Fc fusion protein using CHO cell expression system, and provide reference for screening candidate antigens of recombinant herpes zoster vaccines. Methods:A eukaryotic expression plasmid containing the gE Δ-Fc gene was transfected into CHO cells. Monoclonal cells were selected by methionine sulfoximine (MSX) pressure screening and limited dilution method to obtain the CHO cells secreting and expressing the gE Δ-Fc fusion protein. The expressed gE Δ-Fc fusion protein was purified by MabSelect SuRe affinity chromatography. The binding activity of gE Δ-Fc fusion protein to Fc receptors was identified by ELISA. Flow cytometry was used to detect the phagocytosis of antigens by DC2.4 cells. Antibody titers in serum samples of BALB/c mice immunized with the gE Δ-Fc fusion protein were detect by ELISA. Results:A CHO cell line stably expressing the gE Δ-Fc fusion protein was obtained. Flow cytometry suggested that the phagocytotic activity of DC2.4 cells against the gE Δ-Fc fusion protein was stronger than that against gE. Moreover, the gE Δ-Fc fusion protein could induce BALB/c mice to produce high titers of specific anti-VZV antibodies. Conclusions:The recombinant VZV gE Δ-Fc fusion protein expressed in CHO cells had a good immunogenicity. This study provided reference for screening candidate antigens of recombinant herpes zoster vaccines.
6.The epidemiological characteristics and clinical diagnosis and treatment of 43 cases of novel coronavirus pneumonia

Objective:To retrospectively analyze the epidemiological and clinical characteristics of novel coronavirus pneumonia (COVID-19), and to provide evidence for its further prevention and control.Methods:The epidemiological history, clinical symptoms, pulmonary CT changes, treatment plan (hormone intervention or not), and negative conversion time of nucleic acid were summarized and analyzed in 43 COVID-19 patients.Results:All the 43 patients had clear contact history, among which the number of patients transmitted from conference microphone was the largest (13 cases). There were 2 cases of critical disease, 4 cases of severe disease and 37 cases of common type. The main symptoms were fever (30 cases, 69.8%) and cough (40 cases, 72.1%). Lung CT mainly showed single or multiple ground glass shadows (40 cases), which were all absorbed by the treated lesions, among which 28 cases had a transient aggravation of the lesions during the treatment. All the 43 cases were treated with integrated traditional Chinese and western medicine, 2 cases in critical condition were treated with non-invasive ventilator, and 19 cases were treated with low-dose hormone. The average nucleic acid negative conversion time was 13.28 days, and the use of hormones had no significant effect on the negative conversion time ( P>0.05). Conclusions:COVID-19 of common type is the commonest. Early respiratory support therapy is recommended for critical cases. Hormone intervention makes no obvious influence on the negative conversion time of nucleic acid. Close contact in a closed environment greatly increases the risk of transmission.
7. The relationship of emotional intelligence, perceived stress and adaptability among nursing undergraduates

Objective:
To explore and discuss the relationship between emotional intelligence and stress perception and adaptive ability of undergraduates majoring in nursing, and analyze whether stress perception plays an intermediary role in the relationship between emotional intelligence and adaptive ability.
Methods:
A convenient sampling method was used to investigate 400 nursing undergraduates from 2 universities in Changchun. The survey tools were general information questionnaire, Emotional Intelligence Scale, Chinese version Perceived Stress Scale and College Student Adaptability Inventory.
Results:
The emotional intelligence of nursing undergraduates was above average level (3.63±0.47) points, the perceived stress was below average level (2.84±0.42) points, and the adaptability was above average level (3.48±0.51) points. And the emotional intelligence was positively correlated with the adaptability (
8.Numerical simulation of the effect of virtual stent release pose on the expansion results.

The current finite element analysis of vascular stent expansion does not take into account the effect of the stent release pose on the expansion results. In this study, stent and vessel model were established by Pro/E. Five kinds of finite element assembly models were constructed by ABAQUS, including 0 degree without eccentricity model, 3 degree without eccentricity model, 5 degree without eccentricity model, 0 degree axial eccentricity model and 0 degree radial eccentricity model. These models were divided into two groups of experiments for numerical simulation with respect to angle and eccentricity. The mechanical parameters such as foreshortening rate, radial recoil rate and dog boning rate were calculated. The influence of angle and eccentricity on the numerical simulation was obtained by comparative analysis. Calculation results showed that the residual stenosis rates were 38.3%, 38.4%, 38.4%, 35.7% and 38.2% respectively for the 5 models. The results indicate that the pose has less effect on the numerical simulation results so that it can be neglected when the accuracy of the result is not highly required, and the basic model as 0 degree without eccentricity model is feasible for numerical simulation.
9. The clinical characteristics, gene mutations and prognosis of chronic neutrophilic leukemia

Objective:
To investigate the clinical manifestation, cytogenetics, gene mutations and prognostic factors of chronic neutrophilic leukemia (CNL) .
Methods:
16 CNL cases, according to WHO (2016) -definition, were reviewed retrospectively. Identifications of the CSF3R, ASXL1, SETBP1, CALR and MPL mutations were performed by direct sequencing. JAK2 V617F mutation was detected by AS-PCR.
Results:
Of the 16 CNL patients, the median age was 64 (43-80) years with a male predominance of 75% (12/16) . The median hemoglobin was 114 (81-154) g/L, with median WBC of 41.20 (26.05-167.70) (109/L and median PLT of 238 (91-394) ×109/L.The median level of marrow fibrosis (MF) was 1 (0-3) degree. There was no other cytogenetic abnormalities except t (1;7) (p32;q11) , +21 and 14ps+ for each. All the 16 CNL patients harbored CSF3R T618I mutation. ASXL1 mutations were identified in 81% (13/16) , while SETBP1 mutations were confirmed in 63% (10/16) . The CALR K385fs*47 mutation was found. There was no mutation in JAK2 V617F or MPL in the above 16 patients. 10.Risk analysis of intravenous thrombolysis in acute cerebral infarction with cerebral microbleeds by SWI

Objective To investigate the risk factors and the influence of intravenous thrombolysis of acute cerebral infarction with cerebral microbleeds(CMBs)by SWI.Methods 1 64 patients with acute cerebral infarction were enrolled in this study.All pa-tients were scanned with routine MRI and SWI.According to the presence of CMBs on SWI,the patients were classified into two groups:CMBs group(73 cases)and non-CMBs group(91 cases).Past history was recorded and risk factors of CMBs were explored. 76 cases patients(including 35 cases of CMBs group and 41 cases of non-CMBs group)were treated by intravenous thrombolysis and rescanned with routine MRI and SWI to compare the changes in the number of CMBs and hemorrhage transformation 24 hours after thrombolysis.Results The difference age,hypertension,lacunar infarction and leukoaraiosis between the two groups were significant (P <0.05).The difference of CMBs and hemorrhagic transformation between the two groups treated by thrombolysis were not sig-nificant(P >0.05).Conclusion Acute cerebral infarction with CMBs are influenced by age,hypertension,lacunar infarction and leu-koaraiosis.Thrombolysis in acute cerebral infarction with CMBs can not augment the incidence of hemorrhagic transformation.
