ObjectiveTo investigate the therapeutic effect of Xiao Qinglongtang (XQLT) on ovalbumin (OVA)-induced allergic rhinitis (AR) in mice and its effect on the interleukin-33 (IL-33)/suppression of tumorigenicity 2 (ST2) signaling pathway. MethodSeventy-two female BALB/c mice of SPF grade were randomly divided into a control group, a model group, a positive control group (loratadine, 2.05 mg·kg-1), and low-, medium-, and high-dose (5.005,10.01,20.02 g·kg-1) XQLT groups. All mice except for those in the control group were sensitized by intraperitoneal injection of OVA solution, and the AR model was induced by intranasal drops of OVA solution. Thirty minutes before local intranasal drops, drugs were administered once, and mice in the control group and the model group received phosphate buffered saline (PBS) at 20 mL·kg-1 for 7 days. After the last intranasal drop of OVA solution, the times of sneezing and nasal rubbing of mice within 10 min was recorded. After drug administration for 7 days, blood samples were collected, and nasal bones of mice were decalcified for the preparation of pathological sections. The content of OVA-specific immunoglobulin E (OVA-sIgE), interleukin-4 (IL-4), interleukin-5 (IL-5), and interleukin-13 (IL-13) was detected by enzyme-linked immunosorbent assay (ELISA) kits. Hematoxylin-eosin (HE) staining, periodic acid-Schiff (PAS) staining, and Giemsa staining were used to observe the pathological changes, goblet cell hyperplasia, and eosinophil infiltration of nasal mucosa, respectively. Western blot was used to detect the expression levels of IL-33, ST2, and IL-1 receptor accessory protein (IL-1RAP) in nasal mucosa. ResultCompared with the control group, the model group showed increased times of sneezing and nasal rubbing (P<0.01), edema and thickening of nasal mucosa, goblet cell hyperplasia and eosinophil infiltration, increased serum levels of OVA-sIgE, IL-4, IL-5 and IL-13 (P<0.01), and increased protein expression of IL-33, ST2, and IL-1RAP in nasal mucosa (P<0.05,P<0.01). After drug administration, compared with the model group, the high-dose XQLT group showed reduced times of sneezing and nasal rubbing (P<0.01), improved pathological conditions of nasal mucosa, reduced serum levels of OVA-sIgE, IL-4, IL-5, and IL-13 (P<0.01), and declining protein expression of IL-33, ST2, and IL-1RAP in nasal mucosa (P<0.05,P<0.01). ConclusionXQLT has a therapeutic effect on OVA-sensitized AR mice, and the mechanism may be related to the regulation of the IL-33/ST2 signaling pathway and Th2 inflammatory cytokine to reduce Th2 inflammatory response and alleviate nasal mucosal injury.

Objective To investigate the antagonism of third generation dihydropyridine calcium channel antagonist beni-dipine in contraction induced by potassium chloride(KCl)in human internal mammary artery(IMA),and in order to explore the possibility of using benidipine as an antispastic agent during coronary artery bypass grafting(CABG). Methods The IMAs were taken from 19 patients undergoing CABG. The vasodilator effect(n = 7)on KCl-induced contraction and the inhibition effect(n = 6)after incubation with benidipine were studied in a myograph. The caveolin 1. 2 was detected(n = 3,2 cases re-spectively)by enzyme-linked immunosorbent assay (ELISA). Results The maximum relaxation caused benidipine was (87. 7 ± 4. 9)% . Incubation with benidipine significantly depressed the contraction by KCl[from( 16. 9 ± 4. 4)mN to (9. 1 ± 3. 4 )mN,P < 0. 01]. The concentration of caveolin 1. 2 was down-regulated after incubation with benidipine(P < 0. 05). Conclusion Benidipine has a potent inhibitory effect on KCl-induced vasoconstriction. The relaxation may be related with the expression of caveolin 1. 2. Using benidipine in patients undergoing CABG may provide antispastic effects in grafts.

Background:Inflammatory bowel disease (IBD)is a group of chronic gastrointestinal inflammatory disease and its diagnosis depends on analysis of clinical, endoscopic and pathological characteristics. Aims:To analyze the clinical, endoscopic and pathological characteristics of IBD. Methods:Clinical, endoscopic and pathological characteristics of 99 patients with IBD [including 61 ulcerative colitis (UC)and 38 Crohn's disease (CD)]from January 2010 to April 2017 at Yijishan Hospital were retrospectively analyzed. Results:Compared with CD patients, age in UC patients was significantly increased, incidences of diarrhea, bloody stool were significantly increased while incidence of abdominal mass was significantly decreased (P < 0. 05). Extensive type UC and ileocolonic type CD were common. Proportion of patients received 5-aminosalicylic acid (5-ASA)was significantly higher in UC than in CD (90. 2% vs. 71. 1%,P = 0. 014). Two UC patients achieved remission shown by endoscopy, and endoscopic remission achieved in 3 CD patients. The main pathological features of UC were cryptic branching (57. 1%),cryptic twisting (76. 2%),cryptic atrophy (54. 8%)and cryptic irregularity (28. 6%). Ulcer was seen in 78. 9% of CD patients with active chronic inflammation (mainly lymphocyte and plasmocyte infiltration),and the detection rate of non-caseating granuloma was 15. 8% . Conclusions:UC patients are characterized with older age, diarrhea, bloody stool and higher administration rate of 5-ASA,and the endoscopic features were mucosal bleeding and ulcers. Pathological characteristics of UC were mainly cryptic architectural change and inflammatory infiltration, while those of CD were mainly segmental lesions and chronic inflammation with lymphocyte and plasmocyte infiltration and some with non-caseating granuloma.

Objective To explore the significance of peritubular capillary C4d deposition in histopathological changes, renal function and prognosis of the patients with antibody-mediated chronic rejection (AMCR). Methods Deposition of C4d in the kidney was examined by irnmunohistochemistry on routine paraffin-embedded sections using anti-C4d polyclonal antibody. Seventy-seven patients were divided into C4d+ group (n = 35) and C4d- group (n = 42). The relationship of C4d and renal function, histopathological changes and prognoses of allografts were analyzed. Results The number of patients with tubular atrophy and glomerular basement membrane proliferation in C4d+ group was significantly more than that in C4d group (P<0.05). Mean serum creatinine level was significantly higher in C4d+ group than in C4d- group 12 months after renal transplantation [(379.1 + 260.2)μmol/L vs (260.5 + 175.3) μmol/L, P<0.05]. According to Kaplan-Meier analysis, the one-year graft survival rate was lower in the C4d+ group (62.9% ) than in the C4d- group (83.3% ) (logrank P<0.05). Conclusion Patients with C4d deposition are associated with tubular atrophy and glomerular basement membrane proliferation. The serum creatinine level in C4d+ patients was significantly higher than in C4d- group at the 12th month after transplantation. More patients with C4d deposition lost their grafts during the study period.
