1.Establishment of a novel composite rat model of chronic skeletal muscle injury

Objective To establish a novel rat model of chronic skeletal muscle composite injury in the lumbar region,to explore its mechanism and treatment method in depth.Methods Twenty-eight SD rats were randomly divided into control group(n = 10),Hitting injury model(HIM)group(n = 9),Composite damage model(CDM)group(n = 9)(vertical blow combined with forced prolonged sitting).Open field test,HE staining and PCR sequencing were performed 3 days after modeling.Results The open field distance of rats in HIM group was significantly shorter than that in the CDM group,and both groups had shorter distances than the control group.HE staining showed multiple muscle fiber breaks,significant widening of muscle gaps,uneven distribution of muscle nuclei,and visible congestion in the model group of simple blow injury;In the CDM group,muscle fibers were ruptured,the muscle gaps were slightly widened,and inflammatory cell infiltration was observed between the muscles,especially around the muscle fiber breakage,with muscle fiber nuclei gathering.A volcano plot of the sequencing result suggested significant differences in the Pf4 gene.GO analysis of muscle tissue in model group rats showed upregulation of oxygen binding and carrier activity pathways.Conclusions This study attempted to establish a composite skeletal muscle injury model to study the pathological changes of chronic skeletal muscle injury in the low back pain and evaluate the efficacy of treatment method.It has practical application value in animal experiments for chronic non-specific low back pain.
2.Effect of propofol on excitability of pyramidal neurons in orbitofrontal cortex of mice and underlying ion channel mechanism

Objective:To evaluate the effect of propofol on excitability of pyramidal neurons in orbitofrontal cortex of mice and the underlying ion channel mechanism.Methods:Brain slices of 400 μm thickness from healthy male C57 mice (aged 8-12 weeks)were prepared.This experiment was performed in two parts.Part Ⅰ The brain slices were divided into 2 groups ( n=7 each) based on the random number table method: control group (C group) and propofol group (P group). Cells were perfused with vehicle in group C and with 10 μmol/L propofol in group P. Part Ⅱ The brain slices were divided into 5 groups ( n=8 each) using the random number table method: propofol group (P group), hyperpolarization-activated non-selective cation channel antagonist ZD7288 plus propofol group (Z + P group), inward rectifier potassium channel antagonist topiramate plus propofol group(T + P group), transient activation of voltage-gated potassium channel antagonist 4-aminopyridine (4AP) plus propofol group (A + P group), and delayed activation of voltage-gated potassium channel antagonist tetraethylammonium (TEA) plus propofol group (TEA + P group). Cells were perfused with 10 μmol/L propofol for 2 min in P group, with 5 μmol/L ZD7288 and 10 μmol/L melatonin for 2 min in Z+ P group, with 5 μmol/L topiramate and 10 μmol/L propofol for 2 min in T + P group, with 10 μ mol/L 4-aminopyridine and 10 μmol/L propofol for 2 min in A+ P group, and with 10 μmol/L TEA and 10 μmol/L propofol for 2 min in TEA+ P group.The whole-cell currents, membrane potential and discharge frequency of pyramidal neurons in the orbitofrontal cortex were recorded by whole-cell patch-clamp. Results:Part Ⅰ Compared with C group, whole-cell currents were significantly increased, and the membrane potential and discharge frequency were decreased in P group ( P<0.01). Part Ⅱ Compared with P group, no significant change was found in the whole-cell currents, membrane potentials and discharge frequency in Z+ P group, T+ P group and A+ P group ( P>0.05), and the whole-cell currents were significantly decreased, and the membrane potentials and discharge frequency were increased in TEA+ P group ( P<0.05). Conclusion:Propofol can inhibit the excitability of pyramidal neurons in the orbitofrontal cortex, and the mechanism is related to activating delayed activation of voltage-gated potassium channels in mice.
3. Analysis of urinary iodine in pregnant women with thyroid disease in Shenzhen

Objective:
To study the urinary iodine level of pregnant women with thyroid disease in Shenzhen, and to provide scientific basis for rational iodine supplementation.
Methods:
A total of 170 pregnant women with thyroid diseases who visited Department of Endocrinology, Peking University Shenzhen Hospital from January 2017 to March 2019 were selected as the subjects, they were divided into hyperthyroidism group (84 cases) and non-hyperthyroidism group (86 cases) according to whether they had hyperthyroidism or not. Morning urine sample was collected under normal dietary conditions; urinary iodine was detected by peroxyacetic acid tetramethylbenzidine oxidation colorimetry. Urinary iodine < 150 μg/L was iodine deficiency and 150-249 μg/L was appropriate iodine.
Results:
The median of urinary iodine of pregnant women with thyroid disease in Shenzhen was 143.9 μg/L, which was slightly lower than the lowest limit of the appropriate iodine level. The median of urinary iodine in hyperthyroidism group was 116.6 μg/L, which was at the iodine deficiency level; the median of urinary iodine in non-hyperthyroidism group was 181.6 μg/L, which was at the appropriate iodine level. 4.Pharmacoeconomics Evaluation of Xinmailong Injection Combined with Basic Treatment Plan in the Treatment of Heart Failure with Reduced Left Ventricular Ejection Fraction

OBJECTIVE:To investigate the effectiveness and economy of Xinmailong injection combined with basic treatment plan in the treatment of heart failure with reduced left ventricular ejection fraction (HFREF).METHODS:In prospective randomized central group and open programmatic clinical trials,HFREF patients enrolled in 27 general hospitals during Feb.2014-Nov.2016 were divided into control group (n=253) and trial group (n=872) according to 1∶3.Control group received basic treatment,while trial group was additionally given Xinmailong injection intravenously for a treatment course (5 d) at least.Cost-effectiveness analysis was conducted from the perspective of the whole society by using the total response rate of cardiac functional grading,left ventricular ejection fraction (LVEF) improved data,Minnesota heart failure quality scale (MLHFQ) improved data and re-hospitalization rate.RESULTS:Total response rates of cardiac functional grading in control group and trial group were 61.54% and 81.06% in 3 months.The improved data of LVEF were about 3.05% and 7.35%;MLHFQ improved data were 24.39 and 26.63,and re-hospitalization rates were 19.43% and 10.02%.There were statistical significance in 2 groups (P<0.05),which indicated that clinical efficacy of trial group was better than that of control group.Pharmacoeconomics results showed that cost-effectiveness ratio of positive indicators as the improved data of LVEF and chronic cardiac insufficiency QOL scale (MLHFQ) in trial group were lower than control group (189.35 vs.243.46,576.38 vs.614.29).The cost-effectiveness ratio of re-hospitalization rate and fatality rate in trial group were higher than control group (2 019.61 vs.925.42).CONCLUSIONS:Xinmailong injection combined with basic treatment plan is better than basic treatment plan in the treatment of HFREF,and shows a better economy in cost-effectiveness analysis.
5.Capecitabine combined with docetaxel on tumor marker levels in patients with advanced breast cancer

Objective To explore the effects of combination of capecitabine and docetaxel for the treatment on advanced breast cancer and their influence on tumor biomarkers.Methods The 75 cases with advanced breast cancer were randomly recruited from December 2010 to December 2015 in our hospital,and they were divided into the observation group (38 cases) and control group (37 cases) according to the admission time,the patients in control group were treated with docetaxel,while patients in the observation group were treated with combination of capecitabine and docetaxel,the clinical efficacy before and after treatment was observed,and the changes of serum CEA,glucose CA125,CA15-3 and adverse reaction were compared between two groups.Results The effective rate (RR) of the control group was 40.54% (15/37),and the disease control rate (DCR) was 67.57% (25/37).The RR was 63.16% (24/38) and DCR was 86.84% (33/38)in the observation group.There was significant difference between the two groups (P<0.05).No statistically significant difference was observed between the two groups on the the levels of serum tumor biomarker,CEA,CA125 and CA15-3 before treatment (P>0.05);After treatment,the levels of CEA,CA125 and CA15-3 in both two groups were significantly lower than those before treatment (P<0.05);Moreover,after treatment,the observation group of three tumor biomarkers were significantly lower than those of the control group (P<0.05).Conclusion Combination of capecitabine and docetaxel in the treatment of advanced breast cancer can reduce the serum tumor biomarkers significantly,attenuate the side effects,and the patients are in the good tolerance,it can be widely recommended in clinical use.
6.Effect of dihydromyricetin on proliferation and apoptosis of breast cancer MCF-7 cells

Objective To investigate the effect of dihydromyricetin on proliferation and apoptosis of breast cancer MCF-7 cells.Methods From March 2014 to February 2015,breast cancer MCF-7 cells were treated with 99% pure DMY as an inhibitor.MTT assay,flow cytometry and immunocytochemistry were used to analyze the proliferation,apoptosis and protein expression of breast cancer cell MCF-7.Results When the DMY concentration was higher than 20 μg/mL,the inhibitory effect appeared,but not good.When 40 and 80 μg/mL DMY were used,the proliferation of MCF-6 cells were significantly inhibited,and have different degrees of sensitivity to it.When DMY was 80 μg/mL,the IC50 was 226.9 μg/mL.The inhibition rate and IC50 were compared with 0 μg/mL DMY,there was significant difference(P<0.05).Indicating that the relatively high concentration of DMY could significantly improve the patient′s condition of breast cancer;MCF-6 cells treated with 40 and 80 μg/mL DMY could induce G2/M phase arrest in DMY with concentration greater than 20 μg/mL(P<0.01),and showed significant cell apoptosis death phenomenon.At the same time,G0/G1 phase was also blocked and S phase cells decreased significantly,the difference was significant(P<0.05).The positive rate of PCNA in breast cancer MCF-6 cells significantly decreased(P<0.01) when the DMY concentration was higher than 50 μg/mL,the positive rate was> 50%,especially in DMY with 80 μg/mL,the positive rate was 10.00%.Compared with 0 μg/mL DMY,the difference was significant(P<0.05).Conclusion The use of dihydromyricetin in breast cancer patients can effectively inhibit the rapid increase of cancer cells,accelerate apoptosis,slow down the patient′s condition,the effect is outstanding.
7.Clinical study on Du moxibustion for stable phase of chronic obstructive pulmonary disease

Objective To investigate the Du moxibustion therapy in the treatment of chronic obstructive pulmonary disease (Chronic obstructive pulmonary disease,COPD)at stable phase.Methods 60 cases of lung COPD patients in stable stage who received treatment from January to December 2010 in Taihe Hospital of Traditional Chinese Medicine outpatient were randomly divided into two groups in,according to the case of tail number,with 30 patients in each.The control group was taken oral doxofylline tablets,0.2 g/time,2 time/d and ambroxol hydrochloride,30 mg/time,3 time/d.The treatment group was treated with Du moxibustion two times on the basis of the control group.One year follow-up and pulmonary function and BODE index assessment were performed in each group.Results ① the pulmonary function of the treatment group after the treatment (65.58±7.90) % was significantly improved than the same group before the treatment (53.20± 7.37) % (P<0.05),and had significant difference compared with the control group after the treatment (57.53 ± 7.22)% (P<0.05).The recurrence rate was significantly different in the treatment group (1.79±0.32) and the control group (2.09±0.38) (P<0.05).② BMI,MMRC,6MWD,BODE index,shortness of breath,wheezing,anorexia was significantly improved after the treatment in the treatment group [after treatment were (21.98 ± 1.32)kg/m2,(2.09±0.37)%,(350.68±88.70),(3.82±2.18) meters,(0.38±0.27),(0.32±0.25)%,(0.35±0.27) respectively; before treatment were (18.21±2.49)kg/m2,(2.50±0.43)%,(324.88±70.92),(4.66±1.40) meters,(1.49±0.62) %,(1.42±0.56)%,(1.77±0.35),P<0.01 respecitively].Compared with the control treatment after the treatment [(18.20 ± 1.79) kg/m2,(2.36 ± 0.64) %,(320.03 ± 68.53),(4.43 ±1.62) meters,(1.22± 0.71),(1.28±0.67)%,(1.73±0.24) respectively] (P>0.01),the difference was statistically significant(P<0.01).Conclusion Du moxibustion therapy was effective in treating chronic obstructive pulmonary diseases in stable phase.
8.Drug resistance related factors and cancer metastasis

As multidrug resistance related genes and cell factors are discovered one after another,people have a more thorough understanding to the oncological mechanisms of cancer metastasis.Many evidences of the inherent link between the two tips have been found.These researches provide new ideas to further clarify the regulation mechanisms between them.It is revealed that the inherent link maybe occur on the gene expression and transcription,and with complicated regulative factors.Indepth study of the relations between cancer multidrug resistance and metastasis will help to guide the clinical treatment of tumor.
9.Treatment of gastric stromal tumor by a combined laparoscopic and gastroscopic approach

The combination of laparoscopy and gastroscopy makes up the defects of each of them when used alone, and further extends the application range of minimally invasive techniques. A 53-year-old patient with gastric stromal tumor was admitted to the Tenth People's Hospital of Tongji University on March 4, 2011. The result of gastroscopy showed that the tumor was at anterior wall of stomach with a size of 3.0 cm×3.5 cm. the result of CT scan preliminarily diagnosed the patient with gastric stromal tumor. Partial gastrectomy with laparoscopy combined with gastroscopy was applied to the patient on March 9, 2011. The operation time was 32 minutes and the blood loss was 10 ml. The duration of hospital stay was 4 days, and no postoperative complication occurred. Laparoscopy combined with gastroscopy for the treatment of gastric stromal tumor is feasible and safe with a favorable short-term outcome.
10.Community health committee: A mechanism innovation of public participation in grassroots health service management

Community Health Committee (CHC), an innovative public participation mechanism of grassroots health service management, established a bottom-up communication channel between the public and government to communicate health related problems and opinions, and set up a grassroots community health services supervision system. The rural residents were endowed with opportunities to take part in grassroots health care decision making and management by CHC. CHC changed the top-down model of traditional health management, improved the grassroots medical services, and increased governance capability of local government We introduced the CHC practice in Zhejiang project counties, exhibited the primary effects and experience of this pilot program, and explored new mechanism and model for rural community residents to participate in grassroots health service management
