1.Study on the Characteristics of Supramolecular"Imprinting Template"of Curcuma kwangsiensis and Curcuma phaeocaulis

Objective The characteristics of supramolecular"imprinting template"of volatile oil of Curcuma kwangsiensis and Curcuma phaeocaulis were analyzed and studied based on the supramolecular"qixi"theory of Chinese materia medica combined with chemometrics.Methods The volatile oil of C.kwangsiensis and C.phaeocaulis were extracted by steam distillation,and the fingerprint and composition information of each batch were obtained by GC-MS.Total statistical moment method was used to compare the imprinting characteristics of the"imprinting template"of C.kwangsiensis and C.phaeocaulis.The core index(CI)of each batch of essential oil of C.kwangsiensis and C.phaeocaulis was calculated,and the topological characteristics of types of"imprinting template"of C.kwangsiensis and C.phaeocaulis were compared by chemometrics.Results There was no significant difference in the extraction rate of volatile oil between C.kwangsiensis and C.phaeocaulis.The average values of total zero order moment(AUCT)were(1.907±0.177)×108,(1.979±0.413)×108 μV·s,respectively,showing that there was no significant difference in the total content of volatile oil between the two groups.The mean values of the total first order moment(MCRTT)were(30.969±0.962)and(33.198±0.409)min.The average value of total second order moment(VCRTT)was(56.176±11.368)and(43.891±4.113)min2,respectively,indicating that there were significant differences in the content ratio and species of volatile oils between the two groups.The similarity of total statistical moments of C.kwangsiensis and C.phaeocaulis was mostly lower than the defined value,indicating that the chemical composition and composition ratio of the volatile oil were different.Principal component analysis and orthogonal partial least squares discriminant analysis could obviously divide C.kwangsiensis and C.phaeocaulis into two categories.Through the analysis of P value and VIP value,the CI values of Xvp 4th order,Xvpc 5th order,Xvpc 6th order,Xvpc 7th order,Xvc 3rd order,Xvpc 4th order were the main difference values of C.kwangsiensis and C.phaeocaulis.Conclusion Through the characterization of"imprinting property"and"topological characteristics"of the supramolecular"imprinting template"and combining with chemometric analysis,it is possible to successfully distinguish C.kwangsiensis and C.phaeocaulis,and find the different CI values between two"imprinting templates".
2.Incidence of hypoglycemia during colonoscopy and the prevention of hypoglycemia by enteric nutrients

To compare the incidence of hypoglycemia between day-before bowel preparation and split-dose bowel preparation in colonoscopy patients. The effects of enterald ietary nutrients on the prevention of hypoglycemia and the preparation quality of the intestine during colonoscopy were compared. The patients who underwent colonoscopy were divided into the day-before bowel preparation group, the split-dose bowel preparation group, and the split-dose bowel preparation + enteral nutrient diet group. All patients had their finger blood sugar tested before colonoscopy. The peripheral blood glucose level was measured before operation. After the endoscopic examination, the intestinal cleanliness of the patients was evaluated through the Boston intestinal preparation scale by endoscopists. The incidence of day-before bowel preparation group and split-dose bowel preparation group and enteral nutrient intervention group were 14.38%(23/160), 17.50% (28/160)and 6.45% (4/62), respectively. The proportions of high quality intestinal cleaning were 31.25% (50/160), 35.00% (56/160) and 82.26% (51/62) in the three groups respectively. The incidence of hypoglycemia was higher in split-dose bowel preparation group than that in day-before bowel preparation group. Enteral nutrient intervention can effectively reduce the incidence of hypoglycemia and improve the quality of intestinal preparation, which is a recommended intestinal preparation method.
4.Erector spinae plane block for spinal surgery: a systematic review and meta-analysis

Background:
Although the erector spinae plane block has been used in various truncal surgical procedures, its clinical benefits in patients undergoing spinal surgery remain controversial. The aim of this meta-analysis was to evaluate the clinical benefits of erector spinae plane block in patients undergoing spinal surgery.
Methods:
We searched the Cochrane Library, PubMed, EMBASE, and China National Knowledge Infrastructure for randomized controlled trials comparing the erector spinae plane block with a nonblocked control for spinal surgery.
Results:
Twelve studies encompassing 696 subjects were included in our systematic review and meta-analysis. We found that the erector spinae plane block decreased postoperative pain scores and opioid consumption in the postoperative and intraoperative periods. Moreover, it prolonged the time to the first rescue analgesic, reduced the number of patients who required rescue analgesia, and lowered the incidence of postoperative nausea and vomiting. However, it did not exhibit efficacy in decreasing the incidence of urinary retention and itching or shortening the length of hospital stays, or the time to first ambulation.
Conclusions
Erector spinae plane block improves analgesic efficacy among patients undergoing spinal surgery compared with nonblocked controls; however, there is insufficient evidence regarding the benefits of erector spinae plane block for rapid recovery.
5.Effect of dietary intervention on blood glucose in colonoscopy patients during intestinal preparation: a meta-analysis

Objective:To systematically evaluate the effect of dietary intervention on blood glucose during intestinal preparation for colonoscopy.Methods:Randomized controlled trials (RCTs) on the effects of different interventions on blood glucose during bowel preparation in colonoscopy patients were retrieved on PubMed, CNKI, and WanFang, and RevMan 5.3 software was applied for meta-analysis based on the intervention groups.Results:A total of 10 RCTs in 8 papers, including 3 345 patients, 1 603 patients in the control group and 1 742 patients in the intervention group, were reviewed. Meta-analysis results showed that 10 studies reported incidence of hypoglycemia and interventions during intestinal preparation, and heterogeneity among researches was statistical ( P<0.000 01, I2=80%). Under random effects model combined with effect quantity, compared with the control group, dietary intervention could significantly reduce the incidence of hypoglycemia during bowel preparation, the difference was significant ( OR=0.19, 95% CI: 0.09-0.40, P<0.05). Conclusion:Dietary intervention during intestinal preparation for colonoscopy can prevent the occurrence of hypoglycemia and avoid relative adverse reactions and parasympathetic nerve activity. However, due to the limited number and quality of included researches, the above conclusion need to be verified by more high-quality RCTs.

8.Genetic characteristics of hantavirus from rodents in port area of Ningde , Fujian province in the summer of 2020

Objective:To explore the genetic characteristics and evolution of hantavirus carried by rodents in port area of Ningde in Fujian province in the summer of 2020.Methods:Rodents were captured in the port area of Ningde, the RNA was extracted from rodent lung tissues and detected by using specific kit. The positive samples were used for whole-genome sequencing of the virus. Bioinformatics software was used for the analysis on the similarity and genetic variation of the sequences.Results:A total of 112 rodents were captured, including 5 Rattus norvegicus and 2 Rattus flavipectus, the positive rate of hantavirus was 6.25% (7/112). By virus gene sequencing, two hantavirus complete genome sequences were obtained (named as FJ35 and FJ36, GenBank accession numbers: MW449188-MW449193). The genetic analysis results showed that the hantavirus detected in positive samples were SEOV and shared 99% nucleotide similarity with hantavirus strains LZSF21 and JX20140581 isolated from Shandong province. Phylogenetic analysis using the maximum likelihood method showed that the hantavirus detected in positive samples belonged to S3 subtype, sharing the same subtype with hantavirus strains Z37 from Zhejiang province, LZSF21 from Shandong province, and zy27 and Gongzhuling 415 from northeastern China. Compared with FJ372, the amino acid variation of N259S was observed at sites 251-264 of nucleoprotein, which might be related to antigenicity. Another variation of Q81R was observed in glycoprotein compared with SEOV 80-39 segment of coded amino acid of international reference strain, which might also cause the change in antigenicity. Conclusion:The high positive rate of hantavirus in rodents in the port area of Ningde- would increase the risk of natural human infection and epidemic in local area. The hantavirus positive rodents in this focus might be from an endemic area in Shandong. It is necessary to strengthen the imported rodent control in the port area of Ningde. The virus detected in 2 positive samples belonged to SEOV subtype Ⅲ and shared high homologies of nucleotides and amino acid sequences with the hantavirus strains in surrounding area. However, some slight variations occurred in glycoprotein and nucleoprotein amino acid sequences, which might cause changes in its antigeniity.
9.Optimization of the Microwave Processing Technology of Yellow Wine-processed Corydalis yanhusuo

OBJECTIVE:To optimize the microwave processing technology of yellow wine-processed Corydalis yanhusuo . METHODS:The contents of opioid alkaloid ,berberine hydrochloride and tetrahydropalmatine in C. yanhusuo processed with yellow wine were determined by HPLC. The contents of the extracts were determined by hot dipping method. Based on the single factor tests ,using the appearance of yellow wine-processed C. yanhusuo with microwave processing technology ,the contents of extract,opioid alkaloid ,berberine hydrochloride and tetrahydropalmatine as indexes ,with the amount of yellow wine ,wetting time,microwave power and microwaving time as factors ,the processing technology was optimized with orthogonal test combined with comprehensive weighted scoring method ,and then validated and compared with traditional yellow wine-processed C. yanhusuo . RESULTS:The linear ranges of opioid alkaloid ,berberine hydrochloride and tetrahydropalmatine were 0.100-1.500 μg(R2=0.999 6), 0.012-0.188 μg(R2=0.999 5),0.050-0.750 μg(R2=0.999 8). RSDs of precision ,stability(12 h)and repeatability tests were all less than 2% . The recoveries were 99.15% -100.34%(RSD=0.54% ,n=6),99.52% -100.78%(RSD=0.69% ,n=6), 99.26%-99.79%(RSD=0.28%,n=6). The optimum microwave processing technology included that the amount of yellow wine was 4 g(about 20% of medicinal material amount ),microwave power was 40%,wetting time was 3 hour,processing time was 3 min. The results of three verification tests showed that the contents of extract ,opioid alkaloid ,berberine hydrochloride and tetrahydropalmatine were 15.7%-16.1%,0.061%-0.063%,0.003%-0.004% and 0.061%-0.063%. The comprehensive scores were 97.916,94.730 and 97.217,and RSD were 0.42%,0.38%,0.46%(n=3),respectively. Compared with traditional yellow wine processing technology ,there was no significant difference in the contents of opioid alkaloid and other components ,but no scorched spot and crumbs was found in yellow wine-processed C. yanhusuo with microwave processing technology. CONCLUSIONS:Established method for content determination is simple ,accurate,reliable and reproducible ,and can be used for quantitative analysis of active components in yellow wine-processed C. yanhusuo . Optimized microw ave processing technology is stable and feasible ,and can be used for the processing of yellow wine-processed C. yanhusuo .
10.Content Determination and Principle Components Analysis of 4 Kinds of Flavones in Scutellaria barbata Decoction Pieces by HPLC

OBJECTIVE: To establish the method for simultaneous determination of 4 kinds of flavones such as sutellarin, sutellarein, luteolin and apigenin in Scutellaria barbata decoction pieces, and to conduct principle component analysis. METHODS: HPLC method was adopted. The determination was performed on Agilent ZOXDB-C18 column with mobile phase consisted of methanol-acetonitrile (80 ∶ 20,V/V)-1% acetic acid solution (gradient elution) at the flow rate of 1.0 mL/min. The detection wavelength was set at 335 nm, and column temperature was 30 ℃. The sample size was 10 μL. Principal component analysis was carried out by SPSS 20.0 and SIMCA-P 13.0 software. RESULTS: The linear ranges of sutellarin, sutellarein, luteolin and apigenin were 0.131-1.446 μg(r=0.999 0), 0.031-0.345 μg(r=0.999 7), 0.005-0.055 μg(r=0.999 2), 0.024-0.268 μg(r=0.999 2), respectively. The limits of quantitation were 1.178 8, 0.602 9, 0.744 1, 1.079 1 ng; the limits of detection were 0.353 6, 0.106 1, 0.223 2, 0.323 7 ng;RSDs of precision, stability and reproducibility tests were all lower than 2%. The recoveries were 99.38%-100.56%(RSD=0.44%,n=6), 91.01%-96.81%(RSD=2.43%, n=6), 91.44%-97.34%(RSD=2.59%, n=6), 96.21%- 99.26%(RSD=1.23%,n=6), respectively. By principal component analysis, principal component 1 and prinicipal component 2 were main influential factors of sample, quality accumulative variance contribution rate of them was 92.573%(>80%). The comprehensive score of sample S14-3 was the highest, and the overall quality was relatively good; samples S14-2, S14-3 were the second. These 3 batches of sample were processed and produced in S. barbata planting base with stable quality. CONCLUSIONS: Established method is simple and rapid, and can be used for simultaneous determination of 4 kinds of flavones in S. barbata decoction pieces. Principle component analysis can provide reference for the quality control of S. barbata decoction pieces.
