Background/Aims:
Metabolic dysfunction–associated steatotic liver disease (MASLD) is a chronic liver disease characterized by hepatic steatosis. Ubiquitin-specific protease 29 (USP29) plays pivotal roles in hepatic ischemiareperfusion injury and hepatocellular carcinoma, but its role in MASLD remains unexplored. Therefore, the aim of this study was to reveal the effects and underlying mechanisms of USP29 in MASLD progression.
Methods:
USP29 expression was assessed in liver samples from MASLD patients and mice. The role and molecular mechanism of USP29 in MASLD were assessed in high-fat diet-fed and high-fat/high-cholesterol diet-fed mice and palmitic acid and oleic acid treated hepatocytes.
Results:
USP29 protein levels were significantly reduced in mice and humans with MASLD. Hepatic steatosis, inflammation and fibrosis were significantly exacerbated by USP29 deletion and relieved by USP29 overexpression. Mechanistically, USP29 significantly activated the expression of genes related to fatty acid β-oxidation (FAO) under metabolic stimulation, directly interacted with long-chain acyl-CoA synthase 5 (ACSL5) and repressed ACSL5 degradation by increasing ACSL5 K48-linked deubiquitination. Moreover, the effect of USP29 on hepatocyte lipid accumulation and MASLD was dependent on ACSL5.
Conclusions
USP29 functions as a novel negative regulator of MASLD by stabilizing ACSL5 to promote FAO. The activation of the USP29-ACSL5 axis may represent a potential therapeutic strategy for MASLD.

4.Pharmaceutical care for a ulcerative colitis and ankylosing spondylitis patient who developed pustular psoriasis induced by infliximab

OBJECTIVE To provide a reference for pharmaceutical care in patients with ulcerative colitis (UC) and ankylosing spondylitis (AS) who developed pustular psoriasis induced by infliximab. METHODS Clinical pharmacists participated in the pharmaceutical care process of a patient with UC and AS who developed pustular psoriasis after using infliximab. The clinical pharmacists determined, using Naranjo’s Scale, that the correlation between the patient’s pustular psoriasis and infliximab was “likely”. Regarding the patient’s development of pustular psoriasis after using infliximab, the clinical pharmacists recommended discontinuing infliximab and switching to Upadacitinib extended-release tablets. For the patient’s skin allergic reaction after using upadacitinib, the clinical pharmacists advised continuing the use of upadacitinib and closely monitoring any potential adverse reactions during the treatment period. RESULTS The clinicians adopted the clinical pharmacists’ recommendation. Following the treatment, the patient’s symptoms were significantly alleviated, and the patient was discharged with medication. The follow-up after discharge showed that the treatment was effective and well-tolerated. CONCLUSIONS The clinical pharmacists analyzed the causal relationship between infliximab and pustular psoriasis. Through pharmaceutical care measures such as dynamic monitoring of skin lesions, evaluation of treatment responses, and optimization of drug regimens, they assisted the physicians in formulating an individualized medication plan, ensuring the safety and efficacy of the patient’s medication use.
5.Generation of MCM2 gene inducible knockout cervical cancer HeLa cells and its effect on DNA replication

Objective To generate minichromosome maintenance protein 2(MCM2)gene knockout cervi-cal cancer HeLa cell lines using inducible CRISPR/Cas9 technology,and to explore the effect of MCM2 on DNA replication and replication stress.Methods The inducible CRISPR/Cas9 system,TLCV2,was used to construct MCM2 knockout HeLa cell lines.And the cell lines were divided into control group(Control),knockout group 1(KO1),and knockout group 2(KO2).Western blot,Edu incorporation experiment,real-time quantitative PCR(qPCR),immunofluorescence and MTT assay were used to analyze the effects of MCM2 knockout on DNA replica-tion and replication stress induced by hydroxyurea.Results The CRISPR/Cas9 system successfully knocked out the MCM2 gene after induction,and MCM2 knockout affected the stability of MCM2-7 complex.Compared with the control cells,MCM2 knockout cells had a dramatic decrease in the capacity of DNA replication,and the mRNA levels of Cyclin A1,Cyclin E1 and CDK4.Under DNA replication stress,MCM2 knockout cells decreased cell viability,DNA damage repair capacity,and increased genomic instability compared with control cells.Conclusion Knockout of MCM2 gene reduces the DNA replication capacity of HeLa cells under normal conditions and cell viability under replication stress.This study successfully generates MCM2 gene inducible knockout HeLa cell lines,laying the foundation for further research on the role and biological function of MCM2 gene in the occurrence and progression of cervical cancer.
6.Building a predictive model for adolescent Internet gaming disorder based on machine learning

Objective:
To explore the effectiveness of machine learning in predicting adolescent Internet gaming disorder, so as to provide guidance for formulating effective intervention measures.
Methods:
From June to September,2023,a total of 2 100 students from 3 middle schools and 3 high schools in Bijie City, Qianxi City and Jinsha County, Guizhou Province were selected by stratified random cluster sampling as research subjects. Data was collected by using several instruments, including the Nine item Internet Gaming Disorder Scale-Short From (IGDS9-SF), Parental Psychological Control and Autonomy Support Questionnaire(PPCASQ), Motivation Structure Questionnaire, Relative Deprivation Questionnaire, Deviant Peer Association Questionnaire, and Dual Systems of Self control Scale. Descriptive statistical analysis was conducted to characterize the sample features, and the distribution differences of categorical variables were analyzed by using Chi square test and Mann-Whitney U test. Demographic variables and various influencing factors were served as independent variables, and whether adolescents were addicted to Internet gaming was the dependent variable. Various machine learning algorithms, including random forest, Logistic regression, support vector machine, gradient boosting trees, decision trees, and adaptive boosting were employed to construct predictive models.
Results:
The detection rate of Internet gaming disorder among adolescents was 4.57% (96 cases). Males and middle school students had higher Internet gaming disorder detection rates (5.52%,6.29%) than females and high school students (3.32%,3.62%), and the differences were statistically significant ( χ 2=5.71,7.86, P <0.01).The scores of relative deprivation,deviant peer affiliation, paternal psychological control, maternal psychological control, control motivation, impulsive system and its dimensions (impulsivity, distractibility, low delay of gratification) in Internet gaming disorder group were higher than in non Internet gaming disorder, while the score of parental autonomy support was lower than that in the non Internet gaming disorder group ( Z =-2.88,-9.32,-4.13,-4.48, -6.58 ,-7.50,-7.18,-7.56,-7.43,-2.27, P <0.05). The adaptive boosting algorithm performed the best (accuracy=99%, recall=95%, F1 score=97%, AUC=0.96). Random forest and gradient boosting trees also performed excellently (accuracy=98%, recall=95%, F1 score=97%, 96%, AUC=0.96).
Conclusions
Compared to other models, the adaptive boosting algorithm shows a good predictive effectiveness for adolescent Internet gaming disorder. Appropriate models should be selected to identify individuals with Internet gaming disorder as early as possible, to develop effective intervention strategies and reduce the risk of Internet gaming disorder.
7.Construction and Enhancement of Graduate Curriculum System for Traditional Chinese Medicine Pharmacology: A Case Study of Institute of Chinese Materia Medica, China Academy of Chinese Medical Sciences

Traditional Chinese medicine (TCM) pharmacology (PTCM) is a discipline that studies the interactions between Chinese medicines and the human body, as well as their underlying mechanisms, under the guidance of TCM theories while employing modern scientific techniques and methods. This article reviews the historical development and achievements of the PTCM discipline at the Institute of Chinese Materia Medica, China Academy of Chinese Medical Sciences, and outlines the reform measures undertaken in recent years to advance the construction of the graduate course system in PTCM. Building upon the foundation of the "Special Topics in PTCM" course, the curriculum has been expanded through reforms to include a series of self-designed courses, such as foundational advanced courses, experimental pharmacology courses, pharmacological research tools courses, and applied TCM research courses. Along with enriching the graduate course system, the study explores innovative approaches and methods for graduate education and teaching in PTCM, and reflects on the challenges in course system construction and teaching, serving as a reference for improving the quality of graduate training, promoting the development of the PTCM discipline, and advancing teaching reform practices.
8.Innovation of Methods for Efficacy Evaluation of Traditional Chinese Medicine Compound Prescriptions: Establishment of Characterization System for Simultaneous Treatment/Regulation Based on Correlation Patterns of Five Zang-organs

The clinical efficacy advantages of traditional Chinese medicine (TCM) compound prescriptions have always been inadequately characterized in experimental research,which has become a bottleneck restricting the development of TCM pharmacology and even the progress of TCM. The concept of simultaneous treatment/regulation,guided by the theory of mutual generation and restriction of five zang-organs,has guiding significance in the clinical practice of TCM throughout history and is still widely used in the current clinical practice. However,this unique and clinically valuable diagnostic and therapeutic medication system based on the syndrome differentiation has been completely ignored in the modern research of TCM pharmacology,which might be one of the key factors restricting the pharmacological characterization of the therapeutic advantages of TCM compound prescriptions. On the basis of systematically summarizing the phased progress and achievements of the efficacy evaluation of TCM compound prescriptions,this article explores the path of exploring the pharmacological advantages of TCM compound prescriptions on simultaneous treatment/regulation on the basis of the correlation patterns of five zang-organs,from the theory of Zangxiang,the core concept of five zang-organs,and the TCM disease recognition based on the theory of mutual generation and restriction of five zang-organs. With the heart-lung correlation as a breakthrough point,this study explored a new characterization method for the pharmacological advantages of TCM,aiming to provide new ideas for evaluating the efficacy of TCM compound prescriptions.
9.Effect of Changju Zhidong Prescription on Intestinal Flora in Children with Tic Disorder

ObjectiveTo investigate the regulatory effect of Changju Zhidong prescription on gut microbiota structure of children with tic disorder(TD). MethodTwenty-four children with TD who visited the pediatric outpatient clinic of Dongzhimen Hospital of Bejjing University of Chinese Medicine from November 2020 to January 2022 were selected as the observation group, and eight healthy children were selected as the normal group. The observation group was treated with Changju Zhidong prescription for 12 weeks, and the clinical efficacy was observed by Yale global tic severity scale(YGTSS), traditional Chinese medicine(TCM) syndrome score, and score of TCM symptom evaluation scale for pediatric gastrointestinal tract. Meanwhile, The fecal samples of the observation and normal groups were collected before and after treatment, and 16S rDNA sequencing was used to explore the effects of Changju Zhidong prescription on the distribution of gut microbiota of the children. ResultCompared with the pre-treatment, the YGTSS, TCM syndrome score, and score of TCM symptom evaluation scale for pediatric gastrointestinal tract of the observation group were significantly reduced after 12 weeks of treatment(P<0.05), Simpson and Pieloue indices in alpha diversity were significantly reduced(P<0.05). Before treatment, the relative abundance of Streptococcus parasanguini in the observation group was significantly higher than that of the normal group, while the relative abundances of Citrobacter, Lactobacillus, Lacticaseibacillus, Muribaculaceae unclassified, Odoribacter and 7 other bacterial groups were significantly lower than that in the normal group. Compared with the pre-treatment, the observation group showed a significant decrease in Ruminococcus torques group unclassified and Ruminococcus torques group, and a significant increase in Olsenella, Acetivibrio and 4 other bacterial groups. Functional enrichment analysis showed that the main pathways of Changju Zhidong prescription regulating gut microbiota were bacterial secretion system, insect hormone biosynthesis, limonene and pinene degradation, novobiocin biosynthesis, tropane piperidine and pyridine alkaloid biosynthesis. ConclusionThe pathogenesis of children with TD is accompanied by intestinal flora disorders, and Changju Zhidong prescription can improve core and gastrointestinal symptoms, increase the abundance of beneficial and decrease the abundance of harmful gut microbiota in children with TD, and its mechanism may be through the metabolism of terpenoids and polyketides.
10.Association of dieting and binge eating with non suicidal self-injury among secondary vocational school students

Objective:
To explore the relationship between dieting and binge eating with non suicidal self-injury (NSSI) among secondary vocational school students, so as to provide a scientific basis for the development of effective interventions to prevent NSSI.
Methods:
From October to December 2023, 2 364 students were selected by stratified cluster random sampling method from nine secondary vocational schools in Guizhou Province to conduct a self administered questionnaire survey. Unconditional binary Logisitic regression was used to analyze the relationship between dieting, binge eating behaviours, and the co-occurrence of the two and NSSI among secondary vocational school students, and were stratified by sex.
Results:
The detection rate of NSSI among secondary vocational school students in Guizhou Province was 27.5%, 6.8% of secondary vocational school students reported dieting, 10.5% reported binge eating behaviour, and 2.9% showed both dieting and binge eating behaviours. The adjusted model of unconditional binary Logistic regression showed that the presence of dieting ( OR =2.37), binge eating behaviour ( OR =2.31), and the co-occurrence of both ( OR =2.60) were associated with higher risk of NSSI among secondary vocational school students; stratified analysis by gender showed that both males and females with dieting, binge eating and coexistence of dieting and binge eating behaviours were at increased risk of NSSI (females: OR =2.27, 2.26, 2.78 ; males: OR =2.15, 2.08, 2.02) ( P <0.01).
Conclusions
Dieting and binge eating behavior of secondary vocational school students will increase the risk of NSSI. When preventing and intervening NSSI problems of secondary vocational school students, it should pay more attention to eating habits.
