		                        			OBJECTIVE:
		                        			Xiaotan Sanjie recipe (XTSJ), a Chinese herbal compound medicine, exerts a significant inhibitory effect on gastric cancer (GC) metastasis. This work investigated the mechanism underlying the XTSJ-mediated inhibition of GC metastasis.
		                        		
		                        			METHODS:
		                        			The effect of XTSJ on GC metastasis and the associated mechanism were investigated in vitro, using GC cell lines, and in vivo, using a GC mouse model, by focusing on the expression of Glc-N-Ac-transferase V (GnT-V; encoded by MGAT5).
		                        		
		                        			RESULTS:
		                        			The migration and invasion ability of GC cells decreased significantly after XTSJ administration, which confirmed the efficacy of XTSJ in treating GC in vitro. XTSJ increased the accumulation of E-cadherin at junctions between GC cells, which was reversed by MGAT5 overexpression. XTSJ administration and MGAT5 knockdown alleviated the structural abnormality of the cell-cell junctions, while MGAT5 overexpression had the opposite effect. MGAT5 knockdown and XTSJ treatment also significantly increased the accumulation of proteins associated with the E-cadherin-mediated adherens junction complex. Furthermore, the expression of MGAT5 was significantly lower in the lungs of BGC-823-MGAT5 + XTSJ mice than in those of BGC-823-MGAT5 + solvent mice, indicating that the ability of gastric tumors to metastasize to the lung was decreased in vivo following XTSJ treatment.
		                        		
		                        			CONCLUSION
		                        			XTSJ prevented GC metastasis by inhibiting the GnT-V-mediated E-cadherin glycosylation and promoting the E-cadherin accumulation at cell-cell junctions. 		                        			Cell therapy based on mesenchymal stem cells (MSCs) has been a hot research topic in recent years, including the traditional cell therapy strategy based on living cells and the new cell-free therapy strategy based on soluble proteins or bioactive molecules such as extracellular vesicles (EVs). At present, MSC-induced cells have mature functions and specific structures, and insitu transplantation combined with biomaterials or organic technology has greatly improved the settlement rate and function. On the other hand, as the large-scale culture technique and EVs separation technique evolve, it is possible to obtain a large number of pure EVs, and EVs are gradually becoming a hot spot of current research. An increasing number of studies have shown that the therapeutic effect of MSCs not only occurs by implantation and differentiation but also manifests as the paracrine effect of MSCs. 4.Simultaneous Determination of Isoquercitrin ,Astragalin and Salvianolic Acid B in Moringa oleifera Leaves Granules by HPLC

		                        			OBJECTIVE: To establish a method for simultaneous determination of isoquercitrin, astragalin and salvianolic acid B in Moringa oleifera leaves granules. METHODS: HPLC method was adopted. The determination was performed on Cosmosil-C18 column with mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (gradient elution) at flow rate of 1.3 mL/min. The column temperature was 40 ℃ and detection wavelength was set at 260 nm. The sample size was 10 μL. RESULTS: The linear ranges of isoquercitrin, astragalin and salvianolic acid B were 0.017-0.341, 0.010-0.194, 0.010-0.195 mg/mL, respectively (all r>0.999 0). The detection limits were 0.085, 0.143, 0.117 μg/mL, and the limits of quantitation were 0.283, 0.476, 0.392 μg/mL, respectively. RSDs of precision, stability (24 h) and reproducibility tests were all lower than 2.0% (n=6). Average recoveries were 101.22%, 98.76% and 98.72%, and RSDs were 0.66%,0.30%,0.30% (n=6), respectively. CONCLUSIONS: The established method is simple, accurate and reproducible. 5.Visual Fixation Assessment in Patients with Disorders of Consciousness Based on Brain-Computer Interface.

		                        			Visual fixation is an item in the visual function subscale of the Coma Recovery Scale-Revised (CRS-R). Sometimes clinicians using the behavioral scales find it difficult to detect because of the motor impairment in patients with disorders of consciousness (DOCs). Brain-computer interface (BCI) can be used to improve clinical assessment because it directly detects the brain response to an external stimulus in the absence of behavioral expression. In this study, we designed a BCI system to assist the visual fixation assessment of DOC patients. The results from 15 patients indicated that three showed visual fixation in both CRS-R and BCI assessments and one did not show such behavior in the CRS-R assessment but achieved significant online accuracy in the BCI assessment. The results revealed that electroencephalography-based BCI can detect the brain response for visual fixation. Therefore, the proposed BCI may provide a promising method for assisting behavioral assessment using the CRS-R.

		                        			Objective To investigate the death of chondrocytes in rats which feed with T-2 toxin under selenium (Se) deficient conditions.Methods Thirty two healthy male SD rats were divided into two groups by weight which were normal diet group and Se deficiency diet group,16 rats in each group.Rats in normal diet group were fed with Se 101.5 μg/kg diet,and rats in Se deficiency diet group were fed with Se 1.1 μg/kg diet for 30 d.Normal diet group was divided into control group and T-2 toxin group,and Se deliciency diet group was randomly divided into Se-deficiency group and Se-deficiency plus T-2 toxin group,8 rats in each group.After that,rats in T-2 toxin and Se-deficiency plus T-2 toxin groups were administrated intragastrically with T-2 toxin (100 μg/kg) everyday for 30 d.Rats were put to death,the left knee was taken and stained with hematoxylin-eosin and SafraninFast green,pathological changes of rat's knee joint cartilage were observed under light microscopy,expression levels of active caspase-3 and receptor interacting protein 3 (RIP3) in rat's articular cartilage cells were determined via the immunohistochemical method.The apoptosis was also detected by terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling assay (TUNEL).Results Red ghost outlines of chondrocyte and multiple chondral cell clusters surrounded the non-cell areas in deep zone of articular cartilage of knee joint stained with hematoxylineosin were seen in Se-deficiency plus T-2 toxin group under light microscope.In the superficial zone of cartilage,the positive percent of TUNEL and active caspase-3 in Se-deficiency plus T-2 toxin group was higher than those of control group,Se-deficiency group and T-2 toxin group [(7.47-± 0.34)% vs (4.68 ± 0.54)%,(2.67-± 0.64)%,(2.56 ±0.54)%;(4.75 ± 0.67)% vs (1.24 ± 0.25)%,(0.00 ± 0.00)%,(0.00 ± 0.00)%,P < 0.05].In the middle zone of cartilage,the positive percent of TUNEL,active caspase-3 and RIP3 in Se-deficiency plus T-2 toxin group was significantly higher than those of control group,T-2 toxin group and Se-deficiency group [(72.06 ± 6.15)% vs (16.10 ± 3.00)%,(19.57 ± 3.49)%,(19.33 ± 5.19)%;(51.13 ± 4.18)% vs (10.97-± 3.01)%,(15.36 ± 4.37)%,(15.23 ± 3.13)%;(25.91 ± 13.39)% vs (1.59 ± 1.14)%,(4.32 ± 2.91)%,(7.50 ± 5.00)%,P < 0.05].The positive percents of TUNEL,active caspase-3 and RIP3 were not significantly different in the deep zone (P > 0.05).Conclusion The death of the middle zone in the rat cartilage induced by T-2 toxin under selenium deficient conditions isapoptosis and necroptosis.
		                        		
		                        		
		                        		
		                        	
8."Observations on the Efficacy of ""Three Yin Points"" Acupuncture plus Medicinal Spreading Moxibustion in Treating Erectile Dysfunction"

		                        			Objective To investigate the clinical efficacy of three yin points acupuncture plus medicinal spreading moxibustion in treating erectile dysfunction.Methods Sixty patients with erectile dysfunction meeting the inclusion criteria were randomly allocated to two groups, 30 cases each. The control group took sildenafil at 1 hr before sexual intercourse. The treatment group received three yin points acupuncture plus medicinal spreading moxibustion, 8 days as one course of treatment, for three consecutive courses with 2 days of rest between the courses. The erectile function was scored in the two groups before and after treatment. The clinical therapeutic effects were compared.Results The total efficacy rate was 93.3% in the treatment group and 80.0% in the control group; there was no statistically significant difference between the two groups (P>0.05). The marked efficacy rate was 83.3% in the treatmentn group and 60.0% in the control group; there was a statistically significant difference between the two groups (P<0.05). in the treatment group, there was a statistically significant pre-/post-treatment difference in the erectile function score after 20 days of treatment (P<0.05) and statistically a very significant pre-/post-treatment difference in the erectile function score after 30 days of treatment (P<0.01). In the control group, the overall therapeutic effect was stable after treatment and there was a statistically significant pre-/post-treatment difference in the erectile function score (P<0.05). There was no statistically significant post-treatment difference in the erectile function score between the treatment and control groups (P>0.05).Conclusions Both acupuncture-moxibustion and sildenafil have a therapeutic effect on erectiledysfunction. Sildenafil produces a rapid and stable effect. Acupuncture and moxibustion produce a slower effect but have an advantage in the marked efficacy rate. 9.Expression of β-catenin protein in hepatocellular carcinoma and its relationship with alpha-fetoprotein.

		                        			This study aimed to investigate the expression of β-catenin in hepatocellular carcinoma (HCC) tissues and its relationship with α-fetoprotein (AFP) in HCC. Immunohistochemistry was used to determine the expression of β-catenin in normal liver tissues (n=10), liver cirrhosis tissues (n=20), and primary HCC tissues (n=60). The relationship between β-catenin expression and clinical parameters of HCC was investigated. Real-time PCR and Western blotting were used to detect the mRNA and protein expression levels of β-catenin in the liver cancer cell line SMMC-7721 transfected with a plasmid encoding AFP, and also the mRNA and protein expression levels of β-catenin were measured in the liver cancer cell line Huh7 before and after the transfection with AFP shRNA plasmids. The results showed that β-catenin was only expressed on the cell membrane in normal liver tissues. Its localization to the cytoplasm and nucleus of cells was observed in a small proportion of cirrhotic tissues or adjacent HCC tissues, and such ectopic expression of β-catenin was predominant in HCC tissues. The abnormal expression of β-catenin was correlated with serum AFP levels, cancer cell differentiation and vascular invasion (P<0.05). Additionally, the increased expression of AFP resulted in the upregulation of β-catenin mRNA and protein levels, while knockdown of AFP with AFP shRNA led to significantly decreased β-catenin mRNA and protein levels (P<0.05). It was suggested that the abnormal expression of β-catenin is implicated in hepatic carcinogenesis and development. AFP can lead to increased expression of β-catenin, which may account for the poor prognosis of AFP-associated HCC patients.

10.Observation on therapeutic effect of aligned acupuncture for lumbar intervertebral disc herniation

		                        			Objective:To observe the clinical effects of aligned needling therapy for lumbar intervertebral disc herniation (LIDH). 
 Methods:A total of 80 cases with LIDH in conformity with the inclusion criteria were randomly divided into a treatment group or a control group by their visit order, 40 cases in each group. The points from the Governor Vessel, L1-L5 Jiaji (EX-B 2) points, from the first lateral line of the Bladder Meridian, Huantiao (GB 30), Weizhong (BL 40) and Yanglingquan (GB 34) were selected for acupuncture in the treatment group. Shenshu (BL 23), Dachangshu (BL 25), Ashi (Extra) points and Weizhong (BL 40) were used for routine acupuncture in the control group. The clinical effects of the two groups after 2-course treatments and recurrence rates three months later were observed. The pain severity before and after treatments was assessed by visual analog scale (VAS). The improvement of the patient’s pathological situation was evaluated by the performance assessment of lumbar disease treatment from Japanese Orthopedic Association (JOA). 
 Results:After the treatment for two courses, there was no dropped-out case in the two groups. The curative rates and total effective rates were respectively 32.5% and 92.5% in the treatment group versus respectively 12.5% and 82.5% in the control group. The differences in the curative rates and total effective rates between the two groups were statistically significant (bothP<0.01). After the treatment, VAS and JOA scores remarkably declined in the patients of the two groups, with statistical differences in comparison with those of the same group before treatment (bothP<0.05). In follow-up check of three months, the recurrence rate was 10.7% in the cured and remarkably effective cases in the treatment group and was 29.4% in the control group. The recurrence rates of the two groups were statistically different (P<0.05). 
 Conclusion:The aligned needling technique is remarkable in the clinical effects, obvious in the analgesic effects and low in the recurrence rate in the treatment of LIDH.
