1.Serum levels of SOX6 and PTEN and their clinical significance in patients with acute myocardial infarction

Objective To investigate the expression and clinical significance of SRY-box transcrip-tion factor 6(SOX6)and protein tyrosine phosphatase gene(PTEN)in patients with acute myo-cardial infarction(AMI).Methods A total of 100 AMI patients admitted to Zibo First Hospital and Zibo Central Hospital from January 2021 to March 2022 were enrolled as the study group,and according to the occurrence of major adverse cardiovascular events(MACE),they were grouped into MACE subgroup(52 cases)and non-MACE subgroup(48 cases).Another 110 volunteers who taking physical examination in above 2 hospitals during the same period were subjected as the control group.The levels of PTEN and SOX6 in the serum were detected,and Pearson correla-tion analysis was performed to investigate the correlation of serum PTEN and SOX6 levels with clinical indicators.ROC curve was drawn to assess the diagnostic value of PTEN and SOX6 levels for diagnosis and prognosis of AMI.Results The study group had significantly decreased serum mRNA level of SOX6(0.69±0.14 vs 1.03±0.16,P<0.01)and increased serum mRNA level of PTEN(1.56±0.15 vs 1.05±0.08,P<0.01)than the control group.Similar results were seen in the MACE subgroup than the non-MACE subgroup(SOX6:0.61±0.15 vs 0.78±0.13,P<0.01;PTEN:1.74±0.18 vs 1.37±0.12,P<0.01).Pearson correlation analysis showed that the serum level of PTEN was positively,and that of SOX6 was negatively correlated with cTnI,CK-MB and Gensini score(P<0.01).ROC curve analysis indicated that the AUC value of combined serum SOX6 and PTEN levels for diagnosis of AMI was 0.932(95%CI:0.889-0.962),and that for pre-dicting MACE was 0.933(95%CI:0.866-0.974).Conclusion The serum level of SOX6 is down-regulated,and that of PTEN was up-regulated in AMI patients.Their combined detection is help-ful for diagnosis of AMI and prediction of MACE.
2.Practice on human genetic resources management using PDCA circle in new drug clinical research

Objective:The paper aims to ensure the rational and legal utilization of genetic resources and maintain the national biosafety through the management of human genetic resources in clinical research carried out by medical institutions.Methods:Literature review and survey methods were adopted. Deming Circle, with focus on the four stages from planning, implementation, inspection, and countermeasures, was used to continuously improve the quality of human genetic resources management in the hospital.Results:The concept of hospital human genetic resources management was recognized and the management quality was significantly improved.Conclusions:The PDCA cycle management method is of positive significance to the management of human genetic resources in the clinical research of new drugs in medical institutions, and it is an effective way to improve the management level under current system.
3.Mechanism study of the effect of ultrasound debridement combined with vacuum sealing drainage on dia-betic foot ulcer

Objective To investigate the effect of ultrasound debridement combined with vacuum sealing drainage on the treatment of diabetic foot ulcer and the potential mechanism. Methods Eighty-one patients with diabetic ulcer were randomly divided into two groups:ultrasound debridement combined with vacuum sealing drain-age as the experimental group,routine debridement combined with vacuum sealing drainage as the control group. The clinical curative effect,the reduction rate of the wound,the rate of blastocyst and the rate of bacterial clear-ance and blood flow were detected.Results The clinical curative effect in the experimental group was significantly better than that in the control group(P < 0.05). The rates of wound reduction and granulation were significantly higher than those in the control group(P<0.05).After 14-day treatment,the blood perfusion in the experimental group was significantly higher than that in the control group(P<0.05),and the expression of HIF-1α and VEGF in the ulcer tissue was significantly higher than that in the control group(P<0.05).Conclusions Ultrasound de-bridement combined with vacuum sealing drainage can improve the clinical efficacy,wound reduction rate,granu-lation coverage rate and bacterial clearance rate,and increase ulcer tissue blood flow. The potential mechanism is related with the increases of HIF-1α and VEGF in ulcer tissue.
4.Expression of long non-coding RNA 1010001N08Rik in bronchopulmonary dysplasia and its bioinformatics analysis

Objective To explore the expression feature of long non-coding RNA (lncRNA) 1010001N08Rik in hyperoxia-induced bronchopulmonary dysplasia (BPD) and predict the mechanism that 1010001N08Rik might be involved in the occurrence and development of BPD by a series of bioinformatics analysis.Method The sequence,genomic position and structure characteristics of 1010001N08Rik were acquired from UCSC genome browser,and its target gene was predicted by Ensemble database.We successfully established the animal model of BPD by making newborn C57BL/6J mice exposed to 95% concentrations of ambient oxygen for seven days.The expression of 1010001N08Rik and Gata 6 were detected using real-time quantitative polymerase chain reaction (PCR).Student's t test was used to compare their expression levels during the BPD process.Result The relative expression of 1010001N08Rik in BPD process at d1,d3,d5,d7 was 1.21 ± 0.33,2.02 ± 0.41,2.95 ± 0.45,4.20-± 0.48 respectively,and there were significant difference between adjacent time points (P < 0.05).The relative expression of Gata 6 mRNA was 0.92 ±0.30,1.10 ± 0.31,0.86 ± 0.24,0.45-± 0.08 respectively,and there was significant difference between d5 and d7 (P <0.05).1010001N08Rik had highly conserved property among different species.The chromosomal regions of 1010001N08Rik existed transcriptional factors binding locations and epigenetic regulation clues,and its possible candidate target gene was Gata 6.Conclusion The expression of 1010001N08Rik increased during the formation process of BPD.Bioinformatics analysis and preliminary experiment results suggested that 1010001N08Rik might participate in the process of BPD by down-regulating Gata 6 expression.
5.Effect of direct moxibustion on blood pressure and clinical symptoms in elderly patients with essential hypertension

Objective:To assess the effects of direct moxibustion on 24-hour ambulatory blood pressure (ABP) and clinical symptoms of traditional Chinese medicine (TCM) in elderly patients with essential hypertension, and to explore the antihypertensive effect and influencing factors of moxibustion. Methods:A total of 101 elderly hypertension patients who met the inclusion criteria were randomly assigned to a direct moxibustion I group (n=33), a direct moxibustion II group (n=34), and a control group (n=34). The treatment of calcium antagonist (CCB) orangiotensin II receptor antagonist (ARB) was adopted in the control group. The treatment of direct moxibustion I plus the same medicine as the control group were adopted in the direct moxibustion I group, five cones per acupoint and three times per week, for 5 weeks in total. The treatment of direct moxibustion II plus the same medicine as the control groupwere adopted in the direct moxibustion II group, five cones per acupoint and three times per week, for 5 weeks in total. The changes of 24-hour ABP and clinical symptoms of TCM after treatment were compared in the three groups. Results: The mean 24-hour ambulatory systolic blood pressure (mean 24 h ASBP), night ASBP, percentage of mean 24-hour ambulatory diastolic blood pressure (mean 24 h ADBP)>90 mmHg, and percentage of day ADBP>90 mmHg in the control group were elevated after treatment (P<0.05). The percentage of night ADBP>80 mmHg in the direct moxibustion I group was reduced by treatment (P<0.01). There were no significant differences in the other outcome measures of 24 h ABP, such as day ASBP, percentage of mean 24 h ASBP>140 mmHg, percentage of day ASBP>140 mmHg, percentage of night ASBP>120 mmHg , mean 24 h ADBP, day ADBP, night ADBP, 24 h ambulatory pulse pressure (APP), after treatment in all groups (P>0.05). The degree of improvement of the clinical symptoms of TCM showed significant differences among the three groups of patients (P<0.01). The total effective rate in the direct moxibustion I group was 73.3%, which was superior to those in the direct moxibustion II group and control group (13.3% and 10.0%, respectively). Conclusion:The direct moxibustion has benign regulative effect on blood pressure of elderly patients with essential hypertension, and improves their clinical symptoms. The direct moxibustion method I (burning the next moxa cone after the previous one had totally burnt out) was superior to method II (burning the next moxa cone when the previous one had not totally burnt out ) in lowering blood pressure and improving symptoms of elderly patients with essential hypertension.

Objective To investigate the association between body mass index (BMI), waist circumference (WC), waist?to?height ratio (WHtR), and the incidence risk of type 2 diabetes mellitus (T2DM). Methods In total, 20 194 participants≥18 years old were selected randomly by cluster sampling from two township (town) of the county in Henan province from July to August of 2007 and July to August of 2008 and the investigation included questionnaires, anthropometric measurements, fasting plasma glucose,and lipid profile examination were performed at baseline; 17 236 participants were enrolled in this cohort study. 14 720 (85.4%) were followed up from July to August 2013 and July to October 2014. Finally, 11 643 participants (4 301 males and 7 342 females) were included in this study. Incidence density and Cox proportional hazards regression models were used to evaluate the risk of T2DM associated with baseline BMI, WC, WHtR, and their dynamic changes. Results After average of 6.01 years following up for 11 643 participants, 613 developed T2DM and the incidence density was 0.89 per 100 person?years. After adjusted for baseline sex, age, smoking, drinking, family history of diabetes, as well as the difference of fasting plasma?glucose (FPG), total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL?C), systolic blood pressure (SBP), diastolic blood pressure (DBP) between baseline and follow?up, Cox Proportional?Hazards regression analysis indicated that T2DM risk of baseline BMI overweight group, BMI obesity group, abnormal WC group and abnormal WHtR group were significantly higher than that of the corresponding baseline normal groups , and the incidence risk of T2DM reached the highest for those whose baseline BMI, WC and WHtR were all abnormal, the corresponding HR (95%CI) were 2.05 (1.62-2.59), 3.01 (2.33-3.90), 2.34 (1.89-2.90), 2.88 (2.21-3.74), 3.32 (2.50-4.40), respectively. Whether baseline BMI/WC was normal or not, T2DM risk increased if baseline WHtR was abnormal, and the HR (95%CI) of baseline normal BMI/abnormal WHtR group, baseline abnormal BMI/abnormal WHtR group, baseline normal WC/abnormal WHtR group, baseline abnormal WC/abnormal WHtR group were 1.88 (1.29-2.74), 3.08 (2.34-4.05), 2.15 (1.53-3.00), 3.22 (2.45-4.23), respectively. The analysis for dynamic changes of BMI, WC, and WHtR indicated that in baseline normal WC or WHtR group, T2DM risk increased when baseline normal WC or WHtR developed abnormal at follow?up, and the corresponding HR (95%CI) were 1.79 (1.26-2.55), 2.12 (1.32-3.39), respectively. In baseline abnormal WC or WHtR group, T2DM risk decresed when baseline abnormal WC or WHtR reversed to normal at follow?up, and the corresponding HR (95%CI) were 2.16 (1.42-3.29), 2.62 (1.63-4.20), respectively. Conclusion BMI, WC, and WHtR were associated with increased T2DM risk. The more abnormal aggregation of BMI, WC, and WHtR presents, the higher T2DM risk was. T2DM risk could be decreased when abnormal WC or WHtR reversed to normal.

8.Research progress of taxonomy and discrimination techniques of Oncomela-nia hupensis

Oncomelania hupensis which plays an important role in the transmission of schistosomiasis japonica is the only intermediate host of Schistosoma japonicum and carrying out studies on its subspecies differentiation and genetic variation will have an important significance for schistosomiasis control. In this paper the research progress of taxonomy and discrimination techniques of O. hupensis is reviewed at four levels namely the morphological level the cellular level the protein level and the DNA molecule level and DNA sequencing technology is considered as the ideal taxonomy and discrimination technique of O. hupensis and the future research emphasis.
9.CT and MRI diagnosis of Osgood-Schlatter disease

Objective To characterize the CT and MRI features of Osgood-Schlatter disease (OSD),and explore the feasibility of staging OSD by using CT and MRI.Methods The imaging data of the 27 cases with OSD were retrospectively analyzed,and fea-tures of MRI and CT were characterized.Results All 27 cases with OSD were featured with different extent of edema within the pa-tellar tendon.The portion already exhibited edema in the secondary ossification center in 5 cases,which showed high signal intensity on the T2 WI.There were 1 5 cases which exhibited ossified or tears in the secondary ossification center,and 5 of them were featured with opened shell-like separation on sagittal radiograph.Seven cases had isolated solitary ossicles located at the lower part of the pa-tellar ligament,which is also thickening.Among the 7 cases,one showed hypertrophy in the tibial tuberosity.Three cases examined with CT scan,exhibited ossified nodules located at the lower part of the patellar ligament.Conclusion OSD has characteristic CT and MRI features,and MRI scan is an important imaging method to show edema and morphological changes of patellar tendon in OSD,which could help to identify the stage of OSD and predict the prognosis.
