OBJECTIVE： To compare inhibitory effects of ethanol extract of different medicinal parts （root， stem， leaf， seed， flower and flesh） from Syzygium jambos on the activities of α-glycosidase and α-amylase. METHODS： Using half-inhibitory concentration value （IC50） as evaluation index， acarbose as positive control， inhibitory effects of ethanol extract of different medicinal parts from S. jambos on the activities of α-glycosidase （from yeast and small instestine in mice） and α-amylase were evaluated with in vitro inhibition model. The enzymatic dynamics and Lineweaver-Burk methods were used to analyze the inhibitory type of the best medicinal part on the activities of α-glycosidase and α-amylase. RESULTS： In the yeast α-glucosidase inhibitory activity test， the order of inhibitory activity was S. jambos seed＞S. jambos stem＞S. jambos leaf＞S. jambos root＞S. jambos flower＞S. jambos flesh＞acarbose. In the mice intestine α-glucosidase inhibitory activity test， the order of inhibitory activity was S. jambos seed＞S. jambos stem＞S. jambos root＞S. jambos leaf＞S. jambos flower＞S. jambos flesh＞acarbose. In the α-amylase inhibitory activity test， the order of inhibitory activity was acarbose＞S. jambos seed＞S. jambos stem＞S. jambos root＞S. jambos leaf＞S. jambos flesh＞S. jambos flower. Ethanol extract of S. jambos seed had the stronger inhibition activity against α-glucosidase from yeast，α-glucosidase from small intestine in mice and α-amylase than other medicinal parts [IC50 were（6.64±0.24）， （32.77±2.46） and （41.18±1.63） μg/mL]. Ethanol extract of S. jambos seed had the stronger inhibition activity against α-glucosidase than acarbose [IC50 to α-glucosidase from yeast and α-glucosidase from small intestine in mice were （2 833.33±5.48）， （1 304.21±6.45） μg/mL] （P＜0.05）. The inhibitory effect of ethanol extract from S. jambos on the activity of α-amylase was less than that of acarbose [IC50 was （27.27±1.24） μg/mL] （P＜0.05）. Enzymatic dynamics showed that the inhibitory type of ethanol extract from S. jambos seed on α-glucosidase and α-amylase were both reversible competitive inhibition. CONCLUSIONS： Among different parts of S. jambos such as root， stem， leaf， seed， flower and flesh， S. jambos seed shows the strongest inhibitory effects on the activities of α-glucosidase and α-amylase， which has the value of being developed for the treatment of diabetes or health food.

We expressed B cell epitopes of dengue virus envelope protein and NS1 protein in prokaryotic cells,and purified and evaluated for its serological activities.A recombinant multi-epitope chimeric gene named rE including eight B cell epitopes was connected by linker peptide (EAAAK)2 and cloned into prokaryotic expression vector pET-28a(+),and transformed into E.coli BL21(DE3) cells for expression under induction of IPTG.The expressed recombinant protein was purified with 6× His purification media,and identified by SDS-PAGE and Western blot,and its antigenicity was analyzed by using an indirect ELISA assay.The recombinant expression vector pET28a-rE was constructed and expressed in BL21 (DE3) successfully,but the recombinant proteins mainly appeared as inclusion bodies.The target protein was obtained with high purity through the purification of affinity chromatography.SDS-PAGE and Western blot analysis showed that the molecular weight of fusion protein was in the expected line.The established indirect ELISA has high accuracy.This recombinant peptide antigen expressed in E.coli has good potential for serum testing.

Objective:To establish a double antibody sandwich ELISA assay for detection of human IL-37 in serum.Methods: Mouse anti-human IL-37 monoclonal antibody was used as capturing antibody,rabbit anti-human IL-37 polyclonal antibodies served as detection antibody,HRP labeled goat anti-rabbit IgG employed as second antibody and recombinant human IL-37 protein used as reference standard for the establishment of a doubleantibody sandwich ELISA.The working conditions were optimized,such as sensitivity,linear range,reproducibility and evaluated the serum IL-37 in patients with Dengue fever.Results: The sensitivity of the established ELISA method was 1.465 μg/L approximately.Likewise,the linearity range of this method was about (1.465-46.875) μg/L.Further,the co efficient of variation (CV) of inter-batch and intra-batch in this study were 6.6％ and 11.7％,respectively.Notably,this method could be used in the detection of IL-37 in serum of the patients with Dengue fever,showing that the level of IL-37 in Dengue fever patients was much higher than that in healthy controls.Conclusion: The double antibody sandwich ELISA assay for the detection of human IL-37 was successfully established,which can be apply to detect of human IL-37 in clinical samples.

OBJECTIVETo explore the safety and efficacy of pegylated recombinant human granulocyte colony-stimulating factor (PEG-rhG-CSF) in preventing chemotherapy-induced neutropenia in patients with breast cancer and non-small cell lung cancer (NSCLC), and to provide the basis for clinical application.

METHODSAccording to the principle of open-label, randomized, parallel-group controlled clinical trial, all patients were randomized by 1∶1∶1 into three groups to receive PEG-rhG-CSF 100 μg/kg, PEG-rhG-CSF 6 mg, or rhG-CSF 5 μg/kg, respectively. The patients with breast cancer received two chemotherapy cycles, and the NSCLC patients received 1-2 cycles of chemotherapy according to their condition. All patients were treated with the combination chemotherapy of TAC (docetaxel+ epirubicin+ cyclophosphamide) or TA (docetaxel+ epirubicin), or the chemotherapy of docetaxel combined with carboplatin, with a 21 day cycle.

RESULTSThe duration of grade 3-4 neutropenia in the PEG-rhG-CSF 100 μg/kg and PEG-rhG-CSF 6 mg groups were similar with that in the rhG-CSF 5 μg/kg group (P>0.05 for all). The incidence rate of grade 3-4 neutropenia in the PEG-rhG-CSF 100 μg/kg group, PEG-rhG-CSF 6 mg group, and G-CSF 5 μg/kg group were 69.7%, 68.4%, and 69.5%, respectively, with a non-significant difference among the three groups (P=0.963). The incidence rate of febrile neutropenia in the PEG-rhG-CSF 100 μg/kg group, PEG-rhG-CSF 6 mg group and G-CSF 5 μg/kg group were 6.1%, 6.4%, and 5.5%, respectively, showing no significant difference among them (P=0.935). The incidence rate of adverse events in the PEG-rhG-CSF 100 μg/kg group, PEG-rhG-CSF 6 mg group and G-CSF 5 μg / kg group were 6.7%, 4.1%, and 5.5%, respectively, showing a non-significant difference among them (P=0.581).

CONCLUSIONSIn patients with breast cancer and non-small cell lung cancer (NSCLC) undergoing TAC/TA chemotherapy, a single 100 μg/kg injection or a single fixed 6 mg dose of PEG-rhG-CSF at 48 hours after chemotherapy show definite therapeutic effect with a low incidence of adverse events and mild adverse reactions. Compared with the continuous daily injection of rhG-CSF 5 μg/kg/d, a single 100 μg/kg injection or a single fixed 6 mg dose of PEG-rhG-CSF has similar effect and is more advantageous in preventing chemotherapy-induced neutropenia.

To observe the hypoglycemic effect of Qizhi Jiangtang capsule in rats with type 2 diabetes, and investigate the preliminary mechanism of its hypoglycemic effect, type 2 diabetes rat models were established by high glucose and high fat combined with small dose of streptozotocin (STZ). After continuous administration for 6 weeks, blood glucose, and glycosylated serum protein (GSP) levels were detected in all of the animals; immunohistochemistry assay was used to detect the number of islet β cells; Western blot assay was used to detect the protein expression levels of insulin receptor (InsR), phosphoinositide-3 kinases (PI3K), glucose transporter-2 (GLUT2) and phosphorylated Jun N-terminal kinases (p-JNK)in hepatic tissues. The results showed that Qizhi Jiangtang capsule could reduce the blood sugar and GSP levels in serum in animals with type 2 diabetes mellitus, increase the level of insulin in serum and number of islet β cells, increase the protein expression levels of InsR, PI3K and GLUT2, and reduce the level of p-JNK protein expression. In conclusion, Qizhi Jiangtang capsule has relatively stable hypoglycemic effect, and the mechanism may be associated with increasing the number of islet β cells and level of insulin in serum, up-regulating the protein expression levels of InsR, PI3K and GLUT2, down-regulating the level of p-JNK protein expression in hepatic tissues, and reducing the level of insulin in hepatic tissues.

The effect of Qizhi Jiangtang vapsule (QJC) on degree of dermal ulcer cicatrization in 2 type diabetic rats was studied. Except the rats for blank group, other male Wistar rats were used to establish type 2 diabetic model by feeding with high sugar and high fat diet for four weeks and intraperitonally injecting with 30 mg•kg⁻¹ streptozotocin (STZ). After that, the rats were divided into balanced groups according to blood sugar, and received corresponding drugs for treatment for 8 weeks. At the end of week 8, 2 cm diameter circular incision was done on the back of rats. After that, the rats were administered continuously for10 days. Area of ulcer surface was detected every two days. After the last administration, wound granulation tissues were cut down to conduct pathological examination and detect the expression of VEGF, PI3K, p-ERK protein in wound tissues. The results showed that compared with the model group, after application of Qizhi Jiangtang capsule (2.24 g•kg⁻¹), the wound was significantly reduced on day 6 and day 10 of wound formation; inflammation reaction on ulcer surface was significantly reduce; Qizhi Jiangtang capsule can increase VEGF expression in the wound tissues of diabetic rats, and inhibit ERK phosphorylation. It can be concluded that Qizhi Jiangtang capsule can promote skin ulcer healing for diabetes rats, and its mechanism may be related to regulating the expression of VEGA and p-ERK proteins.

Objective To analyze the population distribution and clinical characteristics of esophageal cancer in Luzhou prefecture of Sichuan. Methods The data of 690 cases of resected specimens with esophageal cancer from October 2010 to September 2014 were analyzed retrospectively, including 659 cases of esophageal squamous cell carcinoma, 10 cases of adenocarcinoma, 7 cases of adenosquamous carcinoma, 5 cases of sarcomatoid carcinoma, 2 cases of carcinoma in situ, 3 cases of leiomyosarcoma, 2 cases of small cell carcinoma, 1 case of undifferentiated carcinoma and 1 case of mixed carcinoma. Results The constituent ratio of 51-60 years old population of 690 cases with esophageal cancer was the highest for 40.00%(276/690). In≤60 years patients, the rate of esophageal cancer in male was higher than that in female: 56.15%(338/602) vs. 34.09%(30/88), χ2 = 15.005, P =0.000. Clinical pathological factors of lymph node metastasis showed that there were statistically significant differences between male and female: 45.68%(275/602) vs. 34.09%(30/88), χ2 = 4.182, P =0.041;the lower the position, the higher the lymph nodes metastasis rate:the lymph nodes metastasis rate in superior segment, middle segment, inferior segment was 32.00%(8/25), 41.22%(216/524) and 57.45%(81/141),χ2=13.425, P=0.001;the lower the differentiation, the higher the lymph nodes metastasis rate:the lymph nodes metastasis rate in high, middle and lower differentiation was 40.84%(165/404), 47.60%(109/229) and 75.76%(25/33), χ2 = 16.065, P = 0.000. The lymph nodes metastasis rate in different general types and histology types had no significant differences(P>0.05). Conclusions The incidence rate of esophageal cancer in male is significantly higher than that in female, the higher the lymph node metastasis rate, the lower the position and the differentiation of esophageal cancer in Luzhou prefecture of Sichuan.

Objective To study and investigate the comprehensive application value of urinary tract dilata-tion combined with cavity technique in patients with ankylourethria.Methods 70 male patients with ankylourethria were collected as the research object,and they were randomly divided into the control group(ordinary urethral dilata-tion treatment group ) of 35 cases and observation group (urinary tract dilatation combined with cavity technique group)of 35 cases by numeration table,then the treatment effect of different stricture length,urodynamic indexes and life quality state before and after the treatment of the two groups were evaluated and compared.Results The cure rates and recurernce rates of the observation group were 91.43% and 2.86%,respectively,which were better than that 68.57% and11.43% of the control group(χ2 =5.396,6.502,all P<0.05).The urodynamic indexes and life quality state at first,third and sixth month after the treatment were all better than those of the control group (t =4.613,4.902,5.263,3.180,all P<0.05).Conclusion The comprehensive application value of urinary tract dila-tation combined with cavity technique in patients with ankylourethria is higher,and it is more benefit for the improve-ment of urodynamic indexes and life quality state.
