1.Construction of prognostic model for endometrial carcinoma based on bioinformatics

Objective Differential genes related to prognosis of endometrial carcinoma(EC)were screened and prognostic models were constructed.Methods Gene Expression data of EC and normal control samples were downloaded from The Cancer Genome Atlas(TCGA)database and Gene Expression Omnibus(GEO)dataset GSE63678 to screen out common differential genes.LASSO regression analysis was used to screen out the genes with prognostic significance and construct prognostic characteristics.EC patients with complete information were obtained from the TCGA database and randomly divided into the training group and the validation group in a ratio of 1:1.In the training group,survival curves were constructed based on prognostic characteristics.Functional annotation and pathway enrichment analysis were conducted using gene ontology(GO)analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis.Combined with prognostic features and clinical risk factors,a calibration curve and C-index were used to evaluate the performance of the histogram.Finally,use a verification group for validation.Results A total of 4800 and 257 differentially expressed genes were screened from TCGA and GEO databases respectively,of which 73 up-regulated genes and 52 down-regulated genes were co-expressed.6 prognostic genes(ORMDL2,BNC2,TTK,MAMLD1,KCTD7 and DCLK2)were screened out by LASSO regression analysis.The survival curve showed that the overall survival of patients in the high-risk group was significantly lower than that in the low-risk group(P<0.01).GO analysis and KEGG results exhibited that prognostic signature was associated with cell cycle.The nomogram showed powerful predictive ability in the training and validation groups.Conclusion We constructed a predictive model based on prognostic genes,which can accurately predict the prognosis of patients with EC and provide new theoretical support for clinical diagnosis and treatment.
2.Improvement effect and mechanism of triptolide on sciatica rats by regulating cGAS/STING signaling pathway

OBJECTIVE To investigate the improvement effect and mechanism of triptolide (TP) on sciatica rats. METHODS Sciatica rat model was prepared and then randomly divided into model group (normal saline), indomethacin group (positive control, 7.5 mg/kg), TP low-dose and high-dose groups (TP-L group and TP-H group, 50, 100 μg/kg TP), and high-dose TP+ stimulator of interferon gene (STING) activator group (TP-H+DMXAA group, 100 μg/kg TP+25 mg/kg DMXAA), with 12 rats in each group. Another 12 unligated rats were selected as sham operation group (normal saline). After 14 days of intraperitoneal administration, the paw mechanical withdrawal threshold (PWT) and paw withdrawal thermal latency (PWL) were detected; the pathological changes, morphology of sciatic nerve and the number of microglia in sciatic nerve were observed. The levels of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α), mRNA and protein expression levels of cyclic guanosine monophosphate- adenosine monophosphate synthase (cGAS) and STING in sciatic nerve were detected. RESULTS Compared with sham operation group, PWT and PWL of rats in model group were obviously reduced and shortened, the number of Nissl bodies was obviously decreased, while the number of microglia, sciatic neuropathology score, the levels of IL-1β and TNF-α, mRNA and protein expressions of cGAS and STING were obviously increased (P<0.05), and sciatic nerve injury was serious. Compared with model group, the changes of various indexes in indomethacin group, TP-L group and TP-H group were opposite to the above (P<0.05), and sciatic nerve injury was reduced. STING activator DMXAA weakened the inhibitory effect of TP on the activity of microglia and inflammatory response in sciatica rats (P<0.05). CONCLUSIONS TP may reduce the activity of microglia and inflammatory response by down-regulating the cGAS/STING signaling pathway, thus alleviating sciatica in rats.
3.Application of various genetic techniques for the diagnosis of Prader-Willi syndrome.

OBJECTIVE:
To discuss the advantages and technical limitations of various molecular genetic techniques in the diagnosis of two infants featuring all-round developmental retardation.
METHODS:
The two patients were initially screened by using chromosomal microarray analysis (CMA). For patient 1, his parents were also subjected to CMA analysis, and the data was analyzed by using ChAS and UPD-tool software. For patient 2, methylation-specific PCR (MS-PCR) was carried out.
RESULTS:
Patient 1 was diagnosed with maternal uniparental disomy (UPD) type Prader-Willi syndrome (PWS) by CMA and UPD-tool family analysis. His chromosomes 15 were of maternal UPD with homology/heterology. Patient 2 was diagnosed with deletion type PWS by combined CMA and MS-PCR.
CONCLUSION
Correct selection of laboratory methods based on the advantages and limitations of various molecular techniques can help with diagnosis of genomic imprinting disorders and enable better treatment and prognosis through early intervention.
4.Influence of alprostadil on pancreatic blood flow and gastrointestinal function related indicators of patients with severe acute pancreatitis

ABSTRAT:Objective To observe the influence of alprostadil on the pancreatic blood flow and gastrointestinal function related indicators of patients with severe acute pancreatitis.Methods A total of 72 patients with severe acute pancreatitis in our hospital were randomly divided into two groups.The control group was treated with conventional treatment and the observation group were treated with alprostadil on the basis of control group,the pancreatic blood flow and gastrointestinal function related indicators of two groups before and after the treatment were detected and compared.Results The pancreatic blood flow and gastrointestinal function related indicators of two groups before treatment showed no significant difference (P > 0.05),while those indicators of two groups after treatment were all significantly improved,and the observation group was significantly better than the control group (P < 0.05).Conclusion The alprostadil can obviously improve the pancreatic blood flow and gastrointestinal function related indicators of patients with severe acute pancreatitis.
5.Postoperative clinical observation of spontaneous urinary extravasation caused by acute upper urinary tract obstruction and its nursing measures

Objective To investigate clinical observation of spontaneous urinary extravasation caused by acute upper urinary tract obstruction and its nursing measures.Methods A total of 72 patients with acute obstructive urinary extravasation in our hospital were selected as research objects,and were divided into control group(n =34) and observation group(n =38),patients of two groups were performed surgical treatment,and were given traditional routine nursing and high quality nursing intervention,respectively.Postoperative complications and patients'care satisfaction.were observed.Results Incidence of postoperative complications in the observation group was obviously lower than the control group and nursing satisfaction was significantly higher than that in the control group (P < 0.05).Conclusion High quality care can control infection and postoperative complications,and is conductive to postoperative rehabilitation for patients with spontaneous urinary extravasation caused by acute upper urinary tract obstruction,and can improve the efficacy of diagnosis and treatment,and patients' satisfaction.

8.Effect of electroacupuncture at Zhongwan(CV 12) on skin microcirculatory blood perfusion units along the conception vessel in yang-deficiency volunteers.

OBJECTIVETo observe the effect of electroacupuncture(EA) at Zhongwan(CV 12) on the energy metabolism along the conception vessel(CV) in volunteers with yang-deficiency constitution,and to explore the relationship of electroacupuncture regulation and body constitution.
METHODSEighteen volunteers with mild constitution and 18 volunteers with yang-deficiency constitution were collected out of 200 students of Fujian University of TCM by body constitution questionnaire. Skin microcirculatory blood perfusion units (MBPU) at Danzhong (CV 17), Xiawan(CV 10) and Qihai(CV 6) of CV were measured by a laser Doppler flowmetry in the normal condition and after EA stimulation at Zhongwan(CV 12) for 20 min.
RESULTS(1)Before treatment, (1)MBPU values at Danzhong(CV 17), Xiawan(CV 10) and Qihai(CV 6) in the yang-deficiency constitution group were lower than those in the mild constitution group,but there was no statistical significance (both P>0. 05) except Danzhong(CV 17) (P<0. 01). (Z)As for the three acupoints in the mild constitution group, MBPU level of Danzhong(CV 17) was higher than that of Xiawan(CV 10) without statistical significance(P->0. 05),and MBPU values of Danzhong(CV 17) and Xiawan(CV 10) were higher than that of Qihai(CV 6) (both P<0. 01). (3About the three acupoints in the yang-deficiency constitution group, MBPU result of Danzhong(CV 17) was lower than the value of Xiawan(CV 10), but higher compared with Qihai(CV 6)(P<0. 05, P<0. 01). MBPU of Xiawan(CV 10) was higher than Qihai (CV 6) as well(P<0. 01). (2) MBPU values of Danzhong(CV 17), Xiawan(CV 10) and Qihai(CV 6) were increased apparently compared with those before treatment after EA stimulation at Zhongwan(CV 12) for 20 min in the two groups(all P<0. 01). (3) The rise rates of MBPU level about Danzhong(CV 17) and Qihai(CV 6) in the yang-deficiency constitution group were higher than those in the mild constitution group without statistical significance after EA at Zhongwan(CV 12) for 20 min(both P>0. 05).
CONCLUSIONThe energy metabolism in CV of volunteers with yang-deficiency constitution is declined, especially Danzhong(CV 17). EA can rise energy metabolism in CV of mild or yang-deficiency constitution volunteers through regulating MBPU along meridian.

9.Development on research of acupuncture effects on microcirculation.

The Chinese and English published literature regarding the effects of acupuncture on microcirculation in recent years were analyzed, and it was found out that the theory of TCM meridian had subtle connection with microcirculation on structure and function, which could provide a possible breakthrough for the research of meridian essence. Acupuncture, through multiple links, multiple paths and multiple levels, could regulate microcirculation under physiological or pathological condition, which was a bidirectional benign adjustment function, and reflected the characteristic of acupoint specificity. Due to many shortcomings in the current studies, studies with rigorous design, large sample size and multicenter are expected in the further to make a standard systematized research.

10.Role of PTEN in podocyte injury in patients with diabetic nephropathy

Purpose To investigate the role of PTEN in podocyte injury in patients with diabetic nephropathy ( DN) . Methods Uri-nary samples from 30 patients with DN and 10 healthy volunteers were collected to detect the level of PCX by ELISA. Renal biopsies were reviewed to observe the morphological changes. All patients with DN were divided into three groups by glomerular lesion. The ex-pression of p-Akt and PTEN in glomeruli was detected by immunohistochemistry. Results The levels of PCX in the urine were signifi-cantly higher in patients with DN compared with those in healthy volunteers, and gradually increased along with glomerular lesion aggra-vating. The expression of p-Akt and PTEN increased in patients with DN compared with healthy volunteers. Although the expression of p-Akt and PTEN decreased with the aggravation of glomerular lesion, they were still higher than that in volunteers. There were obvious-ly positive correlation between the level of PCX and 24-h urinary protein and negative correlation between the level of PCX and the ex-pression level of p-Akt and PTEN. Conclusion PTEN down-regulation may be associated with podocyte injury in DN, which may be associated with the phosphorylation of Akt.
