ObjectiveTo understand the physical development level of 3-7 years old children in Zhag′yab, and to provide reference for local health decision-making. MethodsA cross-sectional study was conducted between June 2021 and July 2021, with a sample of 1 247 Tibetan children aged 3-7 years from kindergartens in 13 districts of Zhag′yab. Their height and weight were measured and the hemoglobin was detected by a unified method. Standard statistical method was adopted (Z-score method). Z-scores of length /height-for-age (HAZ), Z-scores of weight-for-age (WAZ), Z-scores of body mass index (BMI)-for-age (BAZ) and Z-scores of weight-for-height (WHZ) were calculated by WHO Anthro v3.2.2 and WHO Anthro Plus. The nutritional status of children was evaluated according to WHO diagnostic criteria for malnutrition and anemia. ResultsThe average WAZ and HAZ of children aged 3 to 7 in Zhag′yab were lower than the WHO standards, except for the WAZ of 4-year old, the differences were statistically significant (P<0.05). The overall detection rate of malnutrition was 25.7%, stunting, underweight, wasting, overweight, obesity and anemia were 11.6%, 11.8%, 10.8%, 3.3%, 1.8% and 29.3%, respectively. The detection rates of all indicators in boys were higher than those in girls, and the differences were not statistically significant (P>0.05). The overweight rate and obesity rate of rural children were lower than those of urban children, and the other detection rates were higher than those of urban children. The differences of underweight rate, obesity rate and anemia rate were statistically significant between urban and rural children (P<0.05). Among the detection rates of all indicators in different age groups, there were statistically significant differences in the overweight rate and the anemia rate (P<0.05). The overweight rate of children aged 4 and the anemia rate of children aged 5 were the highest. ConclusionsThe physical development of children aged 3 to 7 in Zhag′yab is poor, and the prevalence of malnutrition and anemia is high. Underweight and anemia are more serious in rural children, and the overweight and obesity problem of urban children is emerging. More attention should be paid to promote their nutritional status. The prevention and intervention of children’s malnutrition should be strengthened in Zhag′yab.

Intestinal flora is closely associated with chronic hepatitis B (CHB). Recent studies have shown that the imbalance of intestinal flora is associated with the development, progression, and prognosis of CHB, and the environment of intestinal flora may also change with disease progression, suggesting that intestinal flora and CHB interact with each other. This article reviews the influence of intestinal flora on the progression of CHB and related liver diseases and the role of intestinal flora regulation in the diagnosis and treatment of CHB and related liver diseases, in order to provide new ideas for the clinical treatment of CHB.

ObjectiveTo explore the influence of early-life nutrition intervention on pubertal development in female rats. MethodsFifty-eight neonatal female rats born small for gestational age (SGA) by using maternal food restriction were randomly divided into postnatal over-nutrition group (Group IS, n=28) and postnatal normal diet group (Group IC, n=30). Another 58 neonatal female rats born appropriate for gestational age (AGA) were divided into postnatal over-nutrition group (Group CS, n=28) and postnatal normal diet group (Group CC, n=30). Then we observed and measured the vaginal opening (VO), sex hormone levels, organ coefficients of uterus and ovary, and numbers of corpora lutea at various stages of each group. ResultsThe VO in Group IS and Group CS was accelerated than that in Group IC and Group CC. The levels of luteinizing hormone (LH) on postnatal day 35 in Group IS and Group CS were higher than that of Group CC. The levels of estradiol and LH on postnatal day 35 were higher than those on postnatal day 21 in Group IS, Group CS and Group CC. The organ coefficients of uterus and ovary on postnatal day 35 were higher than those on postnatal day 21 in each group. The numbers of corpora lutea on postnatal day 35 and day 45 were higher in Group IS and Group CS than those in Group CC. ConclusionsPostnatal normal diet may help most SGA female rats catch up their growth and normal pubertal development, while postnatal over-nutrition promotes precocious puberty in either SGA or AGA female rats.

Objective: To investigate the protective effect of Perillae Folium with aqueous extract (PFAE) on some key factors of Adriamycin (ADR)-induced oxidative injury in human renal tubular epithelial cells(HK-2), including the survival rate, oxidative injury indexes and cell apoptosis,in order to define the underlying mechanism. Method: A model of ADR-induced HK-2 cells oxidative injury was established in vitro, then cell viability was detected by cell counting kit-8 (CCK-8) after intervention with positive reference N-acetylcysteine (NAC) or PFAE (5,15,45 g·L-1) at different concentrations. According to the morphological changes under microscopy, the optimum concentration of PFAE was screened out for the follow-up experiments. Then, the experiments were divided into six groups:blank group, ADR (0.05 g·L-1) group, PFAE (15 g·L-1) group, ADR+PFAE (0.05+15) g·L-1 group, NAC (0.81 g·L-1) group, and ADR+NAC (0.05+81) g·L-1 group. After that, malondialdehyde (MDA), superoxide dismutase (SOD), total antioxidant capacity(TAC) were measured in the cell homogenate after 24 h administration. The level of reactive oxygen species (ROS) was detected by 2',7'-dichloroflurescin diacetate (DCFH-DA) fluorescence probe. Flow cytometry and TdT-mediated dUTP Nick-End Labeling (TUNEL) were used to monitor the cell apoptosis. Western blot was used to observed the expressions of mitochondrial apoptosis-associated proteins, like B lymphocyte tumor-2 gene (Bcl-2), Bcl-2 related X protein (Bax), cysteine aspartate protease-9 (Caspase-9), cysteine aspartate protease-3 (Caspase-3) and poly ADP-ribose polymerase (PARP), as well as their shear bodies. In addition, the phosphorylation protein expressions of p38 mitogen-activated protein kinase (p38 MAPK), extracellular signal-regulated kinase (ERK), c-Jun amino-terminal kinase (JNK) in mitogen-activated protein kinase (MAPK) signaling transduction pathway were detected by Western blot. Result: Compared with blank group, ADR group showed a decreased cell viability (P<0.01), and lower SOD level (P<0.01), but higher expressions of MDA and ROS (P<0.01), and an increased apoptotic rate (P<0.01). The ADR group also increased in rate of Bax/Bcl-2, cleaved Caspase-9/Caspase-9, cleaved Caspase-3/Caspase-3, and cleaved PARP/PARP (P<0.01), as well as the phosphorylation protein expressions of p38 MAPK, ERK and JNK (P<0.05,P<0.01). Compared with the ADR group, both ADR+PFAE groups and ADR+NAC group had higher cell proliferation rates (P<0.01). In addition, the protective effect of PFAE on cells was the most obvious at the concentration of 15 g·L-1. The ATC and SOD levels were increased in ADR+PFAE group and ADR+NAC group (P<0.01), while their content of MDA and ROS, cell apoptosis, relative ratio of apoptotic protein expression, and phosphorylation protein expressions of p38 MAPK and ERK were all decreased (P<0.01). However, there was no effect on the expression of phosphorylated JNK protein. Conclusion: PFAE could alleviate the oxidative injury of HK-2 cells induced by ADR, and have an antioxidant effect, which inhibited cell apoptosis through mitochondrial apoptotic pathway and ERK/p38 MAPK signaling pathway.

【Objective】To investigate the effects of different nutritional status in early life on weight catch-up,puberty initiation and obesity in rats with intrauterine growth retardation(IUGR).【Methods】IUGR model of newborn rats was established by limiting diet during pregnancy in SPF rats. Female newborn rats were used in the experiment. IUGR rats were divided into two groups:small litter IUGR group(SL-IUGR)and common feeding IUGR group(CF-IUGR). Rats of normal birth weight served as normal control group(NC). The SL-IUGR group was used to simulate overfeeding in lactation period. After weaning, the rats in the three groups were fed with basic diet. The weights were measured at postnatal 1,7,14,21,35,42 and 75 days. The time of vaginal orifice opening(VO)was recorded. The levels of estradiol(E2),luteinizing hormone(LH)and follicle estrogen(FSH)were examined on the 21 d and the 35 d.【Results】The birth weight of IUGR rats was(4.92±0.18)g,which was significantly lower than that in the control group(6.00±0.29 g,P < 0.001). IUGR rats showed weight growth catch- up on the 14th day. The body weight of SL- IUGR group was higher than that of control group from 14 to 75 d. The body weight of CF-IUGR group lagged behind that of control group from 1 to 75 d,the difference was statistically significant(P < 0.05). The vaginal opening time of female IUGR rats in SL- IUGR group was (29.88 ± 1.81)d,which was significantly earlier than that of CF- IUGR group(32.03 ± 2.11)days(P = 0.044). There was a correlation between body weight of the 21 d and vaginal opening time in IUGR rats,Rs = -0.174,P = 0.039. The besity rates of puberty and adulthood of IUGR rats in SL-IUGR group were 28.33% and 21.67%,which were significantly higher than those in CF-IUGR group(7.5%,6.25%;P = 0.001,0.007). The weight growth rate of adolescent obese IUGR rats at 7 and 21 d was higher than that of non-adolescent obese rats, and the weight growth rate of adult obese IUGR rats at 7 and 35 d was higher than that of non-adolescent obese rats. The difference was statistically significant(P < 0.05). Binary Logistic regression analysis of vaginal opening time and adult obesity in IUGR rats′ group OR = 0.419,P = 0.24. The time of vaginal orifice opening was not an independent factor of adult obesity. 【Conclusions】The improvement of nutrition level in the early stage of life is beneficial to weight catch-up of IUGR,and the overweight catch-up during lactation can lead to puberty and adulthood obesity of IUGR rats,as well as the advance of vaginal opening time.

【Objective】 To guide the nutritional intervention of children in the clinical practice ,by exploring the effect of over-nutrition in postnatal life on growth of the female SD rats.【Methods】We established the rat model of intra? uterine growth retardation(IUGR)through diet restriction in pregnant rats and selected female newborn rats that met the criteria as the experimental group(group I). Group I was randomly divided into control group(group IC)and small group(group IS). Fifty-eight female newborn rats with normal diet in pregnant period(group C)were also divided into control group(group CC)and small group(group CS). After 21-days-lactation,group IC and group CC were randomly subdivided into 4 rats per cage, and then fed with normal diet, while group IS and group CS were fed with high fat diet. We measured the body weight and the body length of each rat every week. Serum fasting insulin, fasting blood glucose,IGF-1,IGF- BP3 levels, and rat insulin sensitivity index(ISI)were measured at different growth and development stages.【Results】 The body weight and body length of group IS were higher than those of group IC(P<0.05);the same was true for group CS and group CC. In the different growth and development stages, the FINS,FBG,IGF- 1 and IGF- BP3 of group IS were higher than those of group IC(P < 0.05),the ISI was lower than that of group IC(P < 0.05). Besides, the FINS,FBG of group CS were higher than those of group CC and the ISI of group CS was lower than that of group CC in the 75 days after birth(P < 0.05).【Conclusions】We found that those small for gestational age infants can be added nutrition appropriately for rapid catch-up,but it is necessary to evaluate the growth index closely and adjust the nutrition on time. Children with normal intrauterine development also need to prevent over-nutrition.

BACKGROUNDOverexpression of G-protein coupled receptor 34 (GPR34) affects the progression and prognosis of human gastric adenocarcinoma, however, the role of GPR34 in gastric cancer development and progression has not been well-determined. The current study aimed to investigate the effect of GPR34 knockdown on the proliferation, migration, and apoptosis of HGC-27 gastric cancer cells and the underlying mechanisms.
METHODSThe expression of GPR34 in gastric cancer cell line HGC-27 was detected by quantitative real-time reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting. HGC-27 cells were employed to construct the stable GPR34 knockdown cell model in this study. Real-time RT-PCR and Western blotting were applied to validate the effect of short hairpin RNA (ShRNA) on the expression of GPR34 in HGC-27 gastric cells. The proliferation, migration of these cells were examined by Cell Counting Kit-8 and transwell. We also measured expression profile of PI3K/PDK1/AKT and ERK using Western blotting.
RESULTSThe ShRNA directed against GPR34 effectively inhibited both endogenous mRNA and protein expression levels of GPR34, and significantly down-regulated the expression of PIK3CB (P < 0.01), PIK3CD (P < 0.01), PDK1 (P < 0.01), phosphorylation of PDK1 (P < 0.01), Akt (P < 0.01), and ERK (P < 0.01). Furthermore, GPR34 knockdown resulted in an obvious reduction in HGC-27 cancer cell proliferation and migration activity (P < 0.01).
CONCLUSIONSGPR34 knockdown impairs the proliferation and migration of HGC-27 gastric cancer cells in vitro and provides a potential implication for therapy of gastric cancer.

Chinese Acupuncture & Moxibustion 2014;34(3):225-229
OBJECTIVETo compare efficacy differences between fire filiform needle combined with mild moxibustion and gabapentin combined with sham acupuncture for postherpetic neuralgia (PHN).
METHODSOne hundred cases of PHN were randomly divided into a needle group and a medicine group, 50 cases in each one. In the needle group, pricking method of fire filiform needle was given at the Ashi points, and then mild moxibustion was applied for 15 min. In the medicine group, the oral administration of gabapentin capsule and sham acupuncture at non-acupoints in the distal end of lesions were applied. The treatment was required for 21 days in both groups. The visual analogue score (VAS) was recorded before treatment and on the 1st day, 2nd day, 3rd day, 6th day, 9th day and 12th day of treatment. The most severity of pain within last 24 h, preset severity of pain, immediate analgesia effect and starting time of pain relief were observed, also the efficacy was assessed and improvement of symptoms was observed in the follow-up visit.
RESULTSThe total effective rate was 94.0% (47/50) in the fire filiform needle group, which was superior to 86.0% (43/50) in the medicine group (P < 0.05). Compared with medicine group, the VAS of the most severity of pain within last 24 h was obviously reduced after the 2nd treatment in the fire filiform needle group while that of present severity of pain was relieved after the 1st treatment (both P < 0.05). The immediate analgesia effect in the fire filiform needle group was obviously superior to that in the medicine group in the first three times of treatment (all P < 0.05). The average time of pain relief was (3.91 +/- 0.82) days in the fire filiform needle group, which was significantly earlier to (6.53 +/- 1.13) days in the medicine group (P < 0.05). 26 cases were cured in the fire filiform needle group in the follow-up visit, which was superior to 2 cases in the medicine group (P < 0.05). The improvement of VAS, pain range and sleep quality in the needle group were also superior to those in the medicine group (all P < 0.05). The direct medical cost in the fire filiform needle group was (232.32 +/- 48.108) yuan, which was significantly lower than (466.00 +/- 41.09) yuan in the medicine group (P < 0.05). There was only one case of adverse effect in the medicine group during the treatment.
CONCLUSIONThe fire filiform needle combined with mild moxibustion could obviously relieve the pain in PHN patients, which has superior immediate analgesia effect and pain relieving time compared with gabapentin, which also has less adverse effects and cheap cost.
