1.Analysis of depression-like behavioral performances of mouse models of vitiligo

Objective:To observe and analyze depression-like behavioral performances of mouse models of vitiligo.Methods:Fifteen female C57BL/6 mice aged about 9 weeks were modeled for vitiligo. Whether the mouse models of vitiligo were successfully constructed or not was determined by macroscopy and full-thickness epidermal immunofluorescence staining of mouse tail tissues on day 23 after the start of the experiment; on day 8 (pre-modeling stage) and day 21 (early modeling stage), the elevated plus maze test and the open field test were used to evaluate the behavioral performances of the mice, including the number of entry into the open arms, percentages of time spent in the open arms, percentages of time spent in the central area and total distance traveled, aiming to assess whether depression-like behaviors were exhibited in the mouse models of vitiligo. To further clarify the degree of the impact of vitiligo modeling on the depression-like state in mice, 20 female C57BL/6 mice were equally divided into 2 groups: vitiligo modeling group and vitiligo modeling + chronic restraint stress group; the mice in the vitiligo modeling + chronic restraint stress group were subjected to chronic restraint stress on day 9, that is, these mice were placed in centrifuge tubes and restrained for about 6 hours every day for 28 consecutive days; on days 7, 22, 29 and 38 after the start of vitiligo modeling, the above-mentioned behavioral indicators were determined by the elevated plus maze test and open field test in the 2 groups. Repeated measurement data in a single group were compared before and after treatment by using paired t-test, and repeated measurement data at multiple time points were compared by using two-way repeated measures analysis of variance. Results:By macroscopy, the mice gradually developed well-defined white patches on the tail skin during vitiligo modeling, which were similar to the clinical manifestations of vitiligo patients; on day 23, full-thickness epidermal immunofluorescence staining of the mouse tail tissues was conducted and showed obvious infiltration of CD8 + T cells and a decrease in the number of Melan-A-positive epidermal melanocytes under a laser confocal microscope, which were consistent with typical pathological characteristics of vitiligo; based on the macroscopic results and immunofluorescence findings, a total of 12 mouse models of vitiligo were successfully constructed on day 23. The elevated plus maze test showed that the number of entry into the open arms and the percentages of time spent in the open arms were significantly lower in the 12 mouse models of vitiligo on day 21 (2.33 ± 1.78 times, 5.01% ± 5.27%, respectively) than in those on day 8 (10.75 ± 2.30 times, 29.20% ± 12.48%, t = 9.63, 6.36, respectively, both P < 0.001) ; the open field test showed that the percentages of time spent in the central area and total distance traveled were also significantly lower in the mouse models on day 21 (2.31% ± 1.53%, 2 518.31 ± 528.38 cm, respectively) than in those on day 8 (4.47% ± 2.65%, 3 533.45 ± 465.47 cm, t = 2.40, 5.47, P = 0.036, < 0.001, respectively). In the chronic restraint stress test, a total of 14 mouse models of vitiligo were successfully constructed on day 23, including 5 in the vitiligo modeling group and 9 in the vitiligo modeling + chronic restraint stress group. There were no significant differences in the number of entry into the open arms, percentages of time spent in the open arms, percentages of time spent in the central area, and total distance traveled between the vitiligo modeling group and the vitiligo modeling + chronic restraint stress group on days 7, 22, 29, and 38 ( F = 0.21, 0.20, 0.46, 2.35, P = 0.889, 0.893, 0.719, 0.134, respectively) ; moreover, all the above indicators significantly changed over time (all P < 0.001), except for the total distance traveled ( P = 0.422) . Conclusion:The mouse models of vitiligo developed depression-like behavior at the early modeling stage, and the degree of depression could not be further deepened by chronic restraint stress on the basis of vitiligo modeling.
2.Evaluation of the effect of contrast medium injection in CT enhanced examination of malignant tumor patients with standardized nursing procedure of totally implantable venous power port

Objective:To explore the standardization of totally implantable venous power port of nursing process in CT enhancement and application effect of contrast medium injection, so as to provide a safer and more efficient way for contrast medium injection in CT enhanced examination for patients with malignant tumors.Methods:A non-randomized prospective study was conducted, 358 patients with malignant tumors were selected in Sun Yat-sen Memorial Hospital, Sun Yat-sen University who underwent CT enhanced examination from August 1, 2022 to July 31, 2023, 179 patients who had been implanted totally implantable venous power port were selected as the experimental group, and the standardized nursing procedure was given. The other 179 patients were the control group, using radiology routine high-pressure intravenous indwelling needle as the contrast medium access, with routine peripheral venous nursing process. The incidence of contrast medium extravasation during CT enhanced examination was observed and compared between the two groups.Results:All the patients were included. There were 85 males and 94 females, aged (55.50±11.72) years old in the control group. There were 83 males and 96 females, aged (54.50±12.24) years old in the experimental group. The incidence of contrast medium extravasation was 0 in the experimental group and 3.35%(6/179) in the control group. The difference between the two groups was statistically significant (Fisher exact probability, P<0.05). Conclusions:The application of standardized nursing procedure of totally implantable venous power port to the injection of contrast medium in CT enhanced examination of malignant tumor patients, can significantly reduce the incidence of contrast medium extravasation.
3.Immunological mechanisms in steatotic liver diseases: An overview and clinical perspectives

Steatotic liver diseases (SLD) are the principal worldwide cause of cirrhosis and end-stage liver cancer, affecting nearly a quarter of the global population. SLD includes metabolic dysfunction-associated alcoholic liver disease (MetALD) and metabolic dysfunction-associated steatotic liver disease (MASLD), resulting in asymptomatic liver steatosis, fibrosis, cirrhosis and associated complications. The immune processes include gut dysbiosis, adiposeliver organ crosstalk, hepatocyte death and immune cell-mediated inflammatory processes. Notably, various immune cells such as B cells, plasma cells, dendritic cells, conventional CD4+ and CD8+ T cells, innate-like T cells, platelets, neutrophils and macrophages play vital roles in the development of MetALD and MASLD. Immunological modulations targeting hepatocyte death, inflammatory reactions and gut microbiome include N-acetylcysteine, selonsertib, F-652, prednisone, pentoxifylline, anakinra, JKB-121, HA35, obeticholic acid, probiotics, prebiotics, antibiotics and fecal microbiota transplantation. Understanding the immunological mechanisms underlying SLD is crucial for advancing clinical therapeutic strategies.
4.Peer support experience of new graduated nurses during standardized training period: a qualitative study

Objective:To explore the peer support experience of newly graduated nurses during standardized training period.Methods:Using phenomenological qualitative research methods and purposive sampling, from April to December 2020, 32 new nurses who participated in the standardized training peer support program in the Affiliated Hospital of Yangzhou University were selected. Semi-structured interview was conducted and Claizzi content analysis method was used to analyze the data.Results:Peer support during the standardized training of new graduated nurses could be summarized into four major themes and eleven sub-themes. Theme 1: cognition and attitude (emotional security, adaptive guidance, recognition of the value of peer support, positive intention to support). Theme 2: peer support needs (department-related information needs; knowledge and skills experience needs; emotional communication needs; career growth support needs). Theme 3: peer support experience (adaptive behavior changes, cognitive and emotional changes, differences in perceived expectations of supporters and support effects).Conclusions:During the standardized training, we should attach importance to the important supporting role of peer support for new nurses. By building a peer support platform for new nurses and broadening experience exchange channels, we can enhance the positive experience of peer support, so as to promote the role adaptation and career growth of new nurses.
5.Effect of swallowing function training based on dynamic swallowing function assessment and aspiration risk classification in elderly patients with dysphagia

Objective:To explore the intervention effect of swallowing function training based on swallowing function assessment and aspiration risk classification in elderly patients with dysphagia.Methods:From January 2018 to December 2019, convenience sampling was used to select patients with dysphagia admitted to the Department of Geriatrics of the First Affiliated Hospital of Zhengzhou University. The patients were divided into a control group and an observation group according to the hospital stay. The patients hospitalized from January to December 2018 were in the control group, and the patients hospitalized from January to December 2019 were in the observation group. The control group carried out routine nursing for swallowing dysfunction, and the observation group implemented swallowing training based on dynamic swallowing function assessment and aspiration risk classification for 6 weeks. The Standardized Swallowing Assessment (SSA) score and aspiration risk grade, Deglutition Handicap Index (DHI) score, and the incidence of aspiration and aspiration pneumonia were compared between the two groups.Results:Among the 185 patients, 3 cases were eliminated without completing all training. There were 83 cases in the control group and 99 cases in the observation group. The SSA score and aspiration symptom scores of DHI of the control group after intervention were higher than those before intervention, and the differences were statistically significant ( P<0.05) . After the intervention, the SSA score, total score and scores of all dimensions of DHI of the observation group were lower than those before and of the control group after intervention, and the differences were statistically significant ( P<0.05) . After the intervention, the aspiration risk classification of the two groups was compared, and the difference was statistically significant ( Z=9.143, P<0.001) . The aspiration rate and the incidence of aspiration pneumonia in the observation group were 13.13% (13/99) and 7.07% (7/99) respectively, and the aspiration rate and the incidence of aspiration pneumonia in the control group were 25.30% (21/83) and 18.07% (15/83) , the differences were statistically significant ( P<0.05) . Conclusions:Swallowing function training based on dynamic swallowing function assessment and aspiration risk classification can effectively improve the swallowing function and quality of life of elderly patients with dysphagia, and reduce the incidence of aspiration and aspiration pneumonia.
6.Simultaneous Determination of 9 Components in Huichun yuzi Granules by LC-MS/MS

OBJECTIVE:To establish the method for simultaneous determination of 9 components in Huichun yuzi granules. METHODS:LC-MS/MS method was adopted. The determination was performed on Shim-pack XR-ODS C18column with column temperature of 30 ℃. The sample size was 5 μL,and ion source as electrospray ion source;MRM mode was adopted. The acetonitrile-water was used as mobile phase for ferulic acid,rutin,paeonol,icariin and schisandrin(gradient elution);positive ion mode monitoring was conducted. The methanol-0.1% fomic acid water was used as mobile phase for naringin,verbascoside, amygdalin and protocatechuic acid(gradient elution);negative ion mode monitoring was conducted. RESULTS:The linear ranges of ferulic acid,rutin,paeonol,icariin,schisandrin,naringin,verbascoside,amygdalin and protocatechuic acid were 0.499 5-500, 25-1 000,0.245 9-250,5.185 1-1 000,0.981 9-1 000,0.248 1-125,2.510 4-250,10-2 500,4.999 7-1 000 ng/mL(r≥0.997 8), respectively. The limits of detection were 0.075,0.30,0.072,0.015,0.015,0.075,0.15,0.30,0.15 ng/mL,respectively;the limits of quantitation were 0.25,1.00,0.24,0.51,0.49,0.25,0.50,0.99,0.49 ng/mL,respectively. The recoveries rate were 91.39%-103.56%(RSD=1.03%-3.67%,n=6).RSDs of stability test ranged 1.4%-3.4%(n=6)within 48 h at room temperature. CONCLUSIONS:The method is rapid,sensitive and reproducible,and can be used for content determination of 9 components in Huichun yuzi granules.It can be used for quality control of Huichun yuzi granules.
7.Fall risk assessment among surgical elderly during perioperative period

Objective To address the fall risks in surgical patients during different time of perioperative period, and explore the best time of risk assessment, and its influencing factors. Methods A total of 250 elderly, who had selective operation and were chosen by a convenient sampling method, received fall risk assessment by Johns Hopkins fall risk assessment tool. We assessed the fall risk factors once a day from the first day of hospitalization until the seven day after operation. Results The fall risk increased 1 d before operation, and it climbed the highest score on the operation day (13. 92 ± 1. 94) with statistical significant compared with fall risk of other day (P<0. 01). The average score of fall risk were (12. 87 ± 1. 42), (11. 98 ± 2. 15) in two days after operation having high fall risk. From 3 d to 7 d after operation, the score of fall risk maintained a medium level. By correlation analysis, it presented the Pearson coefficient was >0. 5, for medicine and nursing facilities of 3 d after operation, and the movement of Pearson coefficient was >0. 5 during the 2 d to 7 d after operation. The further outcome of regression analysis shew that medicine, movement, patient′s nursing facilities and the fall history were high risk factors during perioperative period. Conclusions The fall risk increases on one day before operation and reach peak when patients come back unit after operation. After 7 d operation, the score of fall risk decreases progressively, but it still stays at the medium level. Different time exists different high fall risk factors, and it indicates we should strengthen fall risk assessment and prevention at 1 d, day coming back unit after operation and 7 d after operation to prevent fall incidences.
8.Determination of nine steroidal saponins in Paris polyphylla from different areas of Guizhou province by HPLC.

OBJECTIVETo compare steroidal saponins-main active ingredients of Paris polyphylla from different areas in Guizhou province, in order to provided basis for further proving Guizhou province to be the planting base of P. polyphylla.
METHODThe contents of nine steroidal saponins, namely paris-VII, (25R)-5-en-spirost-3beta,17alpha-diol-3-O-alpha-L-rhamnopyranosyl-(1-->2) [alpha-L-rhamnopyranosyl(1-->4)]-beta-D-glycopyanoside (PGRR), paris-H, paris-VI, paris-II , paris-III, gracillin, paris-I and paris-V of P. polyphylla from eight areas of Guizhou province were determined by HPLC-UV.
RESULTFive steroidal saponins-paris-VII, PGRR, paris-H, paris-VI and gracillin were detected in all the drugs, which provided basis for distinguishing P. polyphylla. P. polyphylla from Dushan showed the highest content of steroidal saponins (9.62%), followed by Tongren (6.39%), and the lowest was Zunyi (0.99%).
CONCLUSIONSaponins from different areas of Guizhou province show significant difference in variety and content, therefore Dushan is suitable for planting P. polyphylla.

9.Chemical constituents from herbs of Paris verticillata.

OBJECTIVETo study the chemical constituents in herbs of Paris verticillata.
METHODThe compounds were isolated by column chromatography with silica gel and purified by Sephadex LH-20 and RP-HPLC. The structures were identified by means of NMR analysis.
RESULTNine compounds were isolated from the EtOAc extract and the n-BuOH extract of P. verticillata. Their structures were identified as beta-sitosterol (1), stigmasterol (2), daucosterol (3), beta-ecdysterone (4), 4-hydroxymethyl-gamma-butyrolactone (5), diosgenin-3-O-alpha-L-arabinofuranosyl (1-->4)-[alpha-L-rhamnopyranosyl (1-->2)]-beta-D-glycopyranoside (6), pennogenin-3-O-alpha-L-arabinofuranosyl(1-->4)-beta-D-glycopyranoside (7), pennogenin-3-O-alpha-L-arabinofuranosyl (1-->4)-[alpha-L-rhamnopy-ranosyl (1-->2)]-beta-D-glycopyranoside (8), and pennogenin-3-O-alpha-L-rhamno-pyranosyl (1-->4)-alpha-L-rhamnopyranosyl (1--4)-[alpha-L-rhamnopyranosyl (1-->2)]-beta-D-glycopyranoside (9).
CONCLUSIONCompounds 1-9 are isolated from P. verticillata for the first time, and compounds 3, 5 are isolated from the genus Paris for the first time. The compounds 6-9 showed certain inhibition activeness of LA-795 cells, especially, the effects of compounds 6, 8 and 9 were more significant.

10.Construction and characterization of RNAi lentiviral vector targeting rat CD80 gene

Objective:To construct a RNAi lentiviral vector targeting rat CD80 gene and detect its effect of gene silencing in NRK and IEC6 cells.Methods:The effective sequence of siRNA targeting rat CD80 gene was confirmed in our previous work.Oligo-DNA fragment containing short hairpin frame was synthesized and reannealed,and then cloned into pGCSIL-GFP lentiviral expression vector.PCR and sequencing analysis were made for verifying the positive clones.The virus packaging plasmids were transfected into 293T cells to harvest shRNA lentivirus.After infection in NRK and IEC6 cells,Real-time PCR was performed to determine the expressing level of CD80.Results:PCR and sequencing revealed that shRNA plasmids was correctly constructed.Virus with a titer of 4×10~8 TU/ml was successfully packaged.CD80 expression in NRK and IEC6 cells could be knockdown by virus infection as characterized by 66.9% and 63.5% decrease of CD80 mRNA in NRK and IEC6 cells respectively,compared with negative control lentivirus.Conclusion:The recombinant lentiviral shRNA expressing vector targeting rat CD80 gene has been successfully constructed and packaged.CD80 mRNA could be down-regulated availably in NRK and IEC6 cells.
