1.Effects of Shoutai Pills in Regulating Histone Modification on Endometrial Decidualization of Mice with Recurrent Spontaneous Abortion

Objective To observe the effect of Shoutai Pills on endometrial decidualization of mice with recurrent spontaneous abortion(RSA);To explore its possible mechanism in the treatment of RSA based on histone modification.Methods Totally 40 female CBA/J mice were divided into normal group,model group,Shoutai Pills low-dosage group(7.5 g/kg),Shoutai Pills high-dosage group(15 g/kg)and dydrogesterone group(3 mg/kg).The normal group were co housed with BALB/C male mice,while the other groups were co housed with DBA/2 male mice to establish an RSA mouse model.After modeling,the administration groups were given corresponding medication solution by gavage,while the normal group and model group were given equal volume of pure water by gavage for 10 consecutive days.The embryo condition was observed and the embryo loss rate was calculated,ELISA was used to detect serum prolactin(PRL)content,HE staining was used to observe the morphological changes of decidual tissue,RT-PCR was used to detect PRL mRNA expression in decidual tissue,Western blot was used to detect the protein expressions of H4ac,H3K27ac,H3K27me3.Results Compared with the normal group,the model group mice showed a significant increase in embryo loss rate,a significant decrease in serum PRL content,disordered arrangement of decidual cells,and extensive bleeding and necrosis;the expression of PRL mRNA and protein in decidual tissue significantly decreased,the protein expressions of H4ac and H3K27ac significantly decreased,while the expression of H3K27me3 protein significantly increased,with statistical significance(P<0.05).Compared with the model group,the embryo loss rate of Shoutai Pills low-and high-dosage groups and the dexamethasone group significantly decreased,the serum PRL content significantly increased,tightly arranged decidual cells,reduced necrosis,and intact glands;the expression of PRL mRNA and protein in decidual tissue of mice in Shoutai Pills high-dosage group and the dexamethasone group significantly increased,the protein expressions of H4ac and H3K27ac significantly increased,the expression of H3K27me3 protein significantly decreased,with statistical significance(P<0.05).Conclusion Shoutai Pills can promote endometrial decidualization in RSA mice,which is related to the changes of histone modification in endometrial stromal cells.
2.Research progress in the role of the medial prefrontal cortex layered structure in general anesthesia

The medial prefrontal cortex engages in various higher brain functions,including attention,transition of consciousness,and information integration,and is the central structure for the action of general anesthetics.In addition,it is considered both the end point for the bottom-up model of the layer-by-layer transmission of information and the starting point for the top-down modulation and integration of information.Classically,the medial prefrontal cortex can be divided into six layers of different functions in the vertical direction,and there are rich neural connections among the layers.In this review,we describe the laminar flow structures found in the medial prefrontal cortex,then discuss how general anesthetics change consciousness via the cortical laminar flow structures.To this aim,the article reviews the available evidence from current studies using rodents,primates,and human subjects that highlight the role of the medial prefrontal cortex in anesthesia.
3.Multifactor analysis of postoperative patency of microsurgical vasoepididymostomy

Objective To analyze the factors which may affect postoperative patency of microsurgical vasoepididymostomy (VE).Methods Ninety-four patients underwent VEs from September 2014 to June 2016 in the Department of Urology,Peking University Third Hospital,with average age of (30.7 ± 4.8) years,and body mass index (BMI) of (25.1 ± 3.0) kg/m2.Semen analyses were performed 1 month,3 months and 5 to 6 months after the operation.The following semen analyses were performed every 3-6 months thereafter.Patency was defined by finding sperms in twice or more analyses during the followup until August 2017.Patients were followed up by face-to-face or telephone interview.Seven factors (age,BMI,bilateral or unilateral anastomosis,anastomosis sites,the adjustment of anastomosis sites,motility and quantity of sperms found in epididymal fluid) were analyzed by Chi-square analysis and multifactor logistic regression analysis.Results Eighty-two patients were followed up (87.2%,82/94) while 12 patients were lost of follow-up.The mean follow-up time was 19 months.Sperms were found in the ejaculate in 59 patients postoperatively.The overall patency rate was 72.0% (59/82),and natural paternity rate was 32.8% (19/58).Patients ≤28 years old and those >28 years old had patency rates of 89.3% (25/28) and 63.0% (34/54,P =0.012),respectively.Patients with BMI <26.0 kg/m2 and BMI≥26.0 kg/m2 had patency rates of 80.4% (41/51) and 58.1% (18/31,P =0.029),respectively.Patency rate of bilateral surgery was 72.1% (44/61) and of unilateral surgery was 71.4% (44/62,P =0.727).Patency rate of caput anastomosis achieved 75.0% (15/20) and of corpus/caudal anastomosis was 71.0% (44/62,P =0.727).Patency rates of patients with and without adjustment of anastomosis sites were 77.8% (7/9) and 71.2% (52/73,P =0.680),respectively.Patency rates of a lot,a few,motile and seldom-motile sperms in epididymal fluid were 74.3% (55/74) vs.50.0% (4/8,P =0.146) and 70.0% (28/40) vs.73.8% (31/42,P =0.701),respectively.Multifactor logistic regression analysis showed that age was well associated with patency rate (OR=4.705,95%CI 1.181-18.742,P=0.028).Conclusions Age ≤28 years is an independent factor leading to higher patency rates.Patients with lower BMIs and younger could have higher patency rates.Factors of anastomosis sides,anastomosis sites,the adjustment of anastomosis sites,motility and quantity of sperms found in epididymal fluid showed no statistical difference in patency rates.
4.Effects of l-tetrahydropalmatine on conditioned place preference and expression of dopamine in the striatum of morphine dependent rats

Objective To observe effects of conditioned place preference (CPP) of morphine dependent rats,variation of dopamine neurotransmitter and its receptor 2 of the striatum in rats suffered from 1-tetrabydropalmatine(l-THP).Methods The CPP model was established by morphine injection in rats with a increasing dose for 10 days,with the initial dose of 10 mg · kg-1 and the final dose of 100 mg · kg-1,10 mg · kg-1 was increased each day,thus 100 nmg · kg-1 Was injected by day 10.Treatments with administration of 1-THP(3.76,1.88 and 0.94 mg/kg) were performed respectively for 6 days,and the effects of CPP for psychological dependence in these rats were observed.Striatum samples were taken out and their variable contents of dopamine neurotransmitter and its receptor 2 in striatum were detected by high performance liquid chromatography and immunohistochemisty,respectively.Results Compared with the NS treatment group,the time of animals treated with 1-THP (3.76 and 1.88 mg/kg) staying in drug-paired compartment were reduced (354 ± 58,373 ± 79) (P< 0.01) ;the raised of variation contents of dopamine neurotransmitter were reduced in striatum from those rats(5.49 ± 1.95,6.11 ± 1.05),while the expression level of dopamine receptor 2 was increased(0.08 ± 0.02,0.07 ± 0.03) (P < 0.01 or P <0.05).Conclusion Reversing dopamine neurotransmitter and its receptor 2 in striatum of morphine dependent rats may be one of the possible mechanisms that 1-THP effectively inhibit the effects of morphine CPP.
5.IRON ABSORBABILITY FROM DIFFERENT DIETS IN IRON DEFICIENT ANEMIC CHILDREN BY NEUTRON ACTIVATION ANALYSIS OF A STABLE ISOTOPE (~(58)Fe)

The neutron activation analysis of a stable isotope (58Fe)was used successfully in the study of iron absorption from different diets in children suffering from iron deficient anemia. The method was correct and convenient, and uninjured to human body. The principle, procedure and the control of errors were also discussed. The iron absorbability of the balance diet,soy protein diet and soy protein diet plus vitamin-C were (group average?SD) 12.0? 3.6%, 10.7?4.7%, and 18,1?3.6% respectively. According to the results and the animal experiments which were reported in another paper, we agreed that soy protein unfavor or reduce the bioavailability of iron from diet, which was made by International Nutritional Anemia Consultative Group (INACG) and others, but it is still a disputing question. The data of iron absorbability from diets in children suffering from iron deficient anemia and the method of neutron activation analysis of a stable isotope (58Fe) have not be seen up to now in China. This paper will be valuable for studing, preventing and treating iron deficient anemia in our country.
6.A STUDY OF THE RELATIVE BIOAVAILABILITY OF IRONFROM SOY FLOUR BY THE WAY OF HEMOGLOBINREGENERATION IN RATS SUFFERED FROMIRON DEFICIENCY ANEMIA

In order to clarify the disputing view-points in some publications, that the soy products are whether unfavourable or not to the bioavailability of diet iron, we estimated the relative bioavailability of the iron from soy flour by hemoglobin regeneration method in rats suffered from iron deficiency anemia. In this experiment the ferrous sulfate was used as a standard, wheat flour as a vehicle, and the pig blood powder as a reference substance. The iron content in the regeneration diet fed to rats of each group was 32 ppm in total, and among it 24ppm was originated from (1) soy flour, (2) soy flour plus wheat flour (half and half), (3) pig blood powder, (4) pig blood powder plus wheat flour (half and half), and (5) ferrous sulfate respectively.The experimental results showed that during the 5-week regeneration period the regenerated hemoglobin level of the rats of corresponding groups were (1) 6.6?1.6→9.4?1.7, (2) 7.0?0.9→10.4 ?1.5, (3) 6.4?1.6→ 11.4?1.4, (4) 6.1?0.6-11.1?1.9, and (5) 5.3?1.2-12.8 1.0, thereby the percentage of iron relative bioavailability of five groups were (1)45.4, (2)65.0, (3)74.0, (4)72.0, (5)100.0 respectively. The iron from soy flour showed a lowest bioavailability aalong the different diet sources in our exper-iment, and therefore the iron from soy products was unfavourable to prevent and treat the iron deficiency anemia. We also made some discussions and suggestions in this paper.
7.A STUDY OF INHIBITING EFFECT OF ORDINARY VEGETABLES ON THE FORMATION OF N-NITROSO COMPOUNDS

The inhibiting effect of 11 kinds of ordinary vegetables including eggplant, cucumber, Chinese cabbage, fresh kidney bean, tomato, paprika, spinach, carrot, onion and celery on the formation of N-nitroso compounds was studied. Most vegetables inhibited the formation of N-nitrosodimethylamine when am-inopyrine and nitrite added to the vegetable juice and citric acid buffer were incubated in a model system simulated stomach condition (pH 3.00, T 37℃) for 1 hr. The inhibiting rate related to the contents of ascorbic acid in the vegetables ranged from 32.70% to 97.61%. Onion, fresh kidney bean, spinach and eggplant inhibited the formation of N-nitrosodimethylamine effectively with the inhibiting rate of 77.91%, 73.40%, 57.48% and 30.75% respectively (the inhibiting effect of ascorbic acid contained in the vegetables had been excluded). The absorptioa of zinc in the fermented soybean weaning food was studied using methed of neutron activation analysis of a stable isotope 70Zn. The results indicated that the rate of zinc absorption (38.7%), activity of alkaline phosphatase in serum (102.3V/L) and the increment of weight (0.5kg) of the infants (7-18m) eating the soybean fermented weaning food were higher than those eating the nonfermented soybean weaning food (30.9%, 71.0V/L, 0.4kg) and the traditional grain weaning food (rice gruel and noodle) (27.5%, 61.5V/L, 0.4kg)
