1.Effect of slice thickness on reproducibility of radiomic features in urinary stones

Objective To explore the effect of CT slice thickness on the reproducibility of radiomic features in urinary stones.Methods A retrospective analysis was performed on 30 cases of stones diagnosed by urinary CT.Thin-slice(1 mm)images were reconstructed into two groups of images with thicknesses of 2 mm and 5 mm.Two radiologists conducted manual segmentation independently and then extracted features.The reproducibility of the radiomic features was calculated using the concordance correlation coefficient(CCC).Results Under different slice thickness conditions by the same radiologist,the average CCC of all groups was less than 0.85(P<0.05).For radiologist A,the proportion of stone features with CCC<0.85 were 50.13%,79.91%,and 82.38%in the 1 mm vs 2 mm group,2 mm vs 5 mm group,and 1 mm vs 5 mm group,respectively.Corresponding values for radiologist B were 44.55%,79.47%and 82.32%,respectively.Among the seven categories of radiomic features,the morphological features with CCC<0.85 was 100%in the 1 mm vs 5 mm group.Conclusion CT slice thickness significantly affected the reproducibility of radiomic features in urinary stones,with the greatest effect observed in morphological features.Thin-slice imaging demonstrated more stable reproducibility of stone characteristics.
2.Preoperative CT radiomics models for predicting composition of in vivo urinary calculi

Objective To observe the value of preoperative CT radiomics models for predicting composition of in vivo urinary calculi.Methods Totally 543 urolithiasis patients were retrospectively enrolled and divided into calcium oxalate monohydrate stone group(group A,n=373),anhydrous uric acid stone group(group B,n=86),carbonate apatite group(group C,n=30),ammonium urate stone group(group D,n=28)and ammonium magnesium phosphate hexahydrate stone group(group E,n=26)according to the composition of calculi,also divided into training set and test set at the ratio of 7:3.Radiomics features were extracted and screened based on plain CT images of urinary system.Five binary task models(model A-E corresponding to group A-E)and a quinary task model were constructed using least absolute shrinkage and selection operator algorithm for predicting the composition of calculi in vivo.Then receiver operating characteristic curves were drawn,and the area under the curves(AUC)were calculated to evaluate the predictive efficacy of binary task models,while the accuracy,precision,recall and F1 score were used to evaluate the predictive efficacy of the quinary task model.Results All binary task models had good efficacy for predicting the composition of urinary calculi in vivo,with AUC of 0.860-0.948 in training set and of 0.856-0.933 in test set.The accuracy,precision,recall and F1 score of the quinary task model for predicting the composition of in vivo urinary calculi was 82.25%,83.79%,46.23%and 0.596 in training set,respectively,while was 80.63%,75.26%,43.48%and 0.551 in test set,respectively.Conclusion Binary task radiomics models based on preoperative plain CT had good efficacy for predicting the composition of in vivo urinary calculi,while the quinary task radiomics model had high accuracy but relatively poor stability.
3.Establishment of A Quality Control Method for Simultaneous Determination of Multiple Active Components in Uygur Medicine of Nymphaeae Flos

ObjectiveTo develop a quality control method for the simultaneous determination of multiple active components in Nymphaeae Flos aiming at the problems of the single index for quality control and the relatively low overall quality control level. MethodUltra performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS)was used to identify and select the index components for quality control with the mobile phase of 0.1% formic acid aqueous solution(A)-acetonitrile(B)for gradient elution (0-2 min, 3%-8%B; 2-4 min, 8%-10%B; 4-13 min, 10%-15%B; 13-19 min, 15%-20%B; 19-26 min, 20%-45%B) at a flow rate of 0.4 mL·min-1, detection wavelength of 350 nm, electrospray ionization(ESI), negative ion scanning mode, ion source temperature of 120 ℃, scanning range of m/z 100-1 200, transmit collision energy of 6 eV for low-energy scanning and 25-50 eV for high-energy scanning. High performance liquid chromatography(HPLC)was used to establish the quality control method for the simultaneous determination of multi-index components with the mobile phase of 0.2% phosphoric acid aqueous solution(A)-acetonitrile(B) for gradient elution(0-30 min, 12%-15%B; 30-60 min, 15%-22%B; 60-90 min, 22%-40%B)and detection wavelength of 350 nm. The preparation method of the test solution for content determination was refluxing extraction for 60 min with 80 times the amount of 70% methanol. ResultBy comparing the retention time, ultraviolet absorption characteristics, MS and MS/MS spectrometric signals in the samples with the reference substances, 8 active components with high contents, including brevifolincarboxylic acid, ellagic acid, rutin, nicotiflorin, astragalin, quercetin, quercetin-3-methylether and kaempferol, were identified qualitatively from Nymphaeae Flos, which were selected as the index components for quality control. Under the established HPLC conditions, the above 8 components could be well separated(resolution>1.5), and showed good linearity(r=0.999 9)between the concentration ranges of 1.99-99.6, 1.76-176, 1.52-75.8, 3.60-180, 0.964-96.4, 1.18-118, 1.94-96.8, 1.04-104 mg·L-1 and the peak areas, respectively. The detection limits of them were 10-49 μg·L-1, and the limits of quantitation were 34-164 μg·L-1. The average recoveries were 97.12%-103.1% with the relative standard deviations (RSDs) were 1.1%-2.2%. ConclusionA quality control method for simultaneous determination of the multiple active components in Nymphaeae Flos have been developed, which is simple, accurate and reproducible, and it can provide a scientific basis for the formulation of quality standard of this herb and lay a research foundation for the transformation of Uygur hospital preparations containing Nymphaeae Flos into new drugs.
4.Application of narrative therapy theory implanted games into bowel preparation in children

Objective:To investigate the effect of applying narrative therapy theory into game on improving the quality of bowel preparation in children, and to provide a basis for selecting nursing intervention methods before bowel preparation.Methods:This study was a quasi experimental study. Totally 62 children who took polyethylene glycol electrolyte dispersion for bowel preparation in Gansu Maternal and Child Health Hospital from January to December 2021 were included in this retrospective study. They were divided into control group with 28 cases and experimental group with 34 cases according to random number table method. The control group was given conventional bowel preparation and medication guidance, and the experimental group was given the intervention during bowel preparation by applying narrative therapy theory implanted games. The bowel preparation adequacy rate, complete medication taking rate and parental satisfaction of the two groups were observed and compared.Results:The bowel preparation adequacy rate and complete medication taking rate in the experimental group were 94.12% (32/34) and 52.94% (18/34), respectively, which were higher than 46.43% (13/28) and 10.71% (3/28) in the control group, and the differences were statistically significant ( χ2 = 15.23, 10.41, both P<0.01). According to the average BBPS score, the experimental group had better intestinal cleanliness compared to the control group (7.65 ± 1.07 vs 6.07 ± 1.41, t = -4.87, P<0.01), the difference was statistically significant. 97.06% (33/34) of the parents in the experimental group expressed satisfaction, which was higher than 64.29% (18/28) in the control group, and the difference was statistically significant ( χ2 = 12.74, P<0.05). Conclusions:By applying the narrative therapy theory implanted games, the complete taking of high-dose bowel cleasing agent can be promoted and the quality of bowel preparation can be improved, which is worth suggesting as a nursing intervention method for bowel preparation.
5.Acute Toxicity Test of Water Extracts from Unprocessed and Different Processed Products of Psoraleae Fructus in Mice

OBJECTIVE To compare the acute toxicity characteristics and differences of water extracts from unprocessed and different processed products of Psoraleae Fructus in mice. METHODS Kunming mice were divided into 36 groups, including Psoraleae Fructus raw product group, stir-fried group, salt-baked group, Leigong group, and wine soaked group, as well as control group. Each group of mice was given a single intragastric administration of 0.04 mL·g-1 and observed for 14 d. The body weight, serum biochemical indices, and mortality of the mice were detected, and the LD50 value was calculated to study the toxicity differences of Psoraleae Fructus raw product and different processed products. RESULTS There was no statistically significant difference in the body weight of mice before administration in different groups. On the first day of administration, some mice in the administration group showed a certain decrease in body weight. The LD50 values of the Psoraleae Fructus raw product group, stir-fried group, salt-baked group, Leigong group, and wine soaked group were 63.20, 56.92, 51.95, 88.61, 59.02 g·kg-1, respectively. Compared with the control group, the serum ALT and TBA levels in the male mice in the Psoraleae Fructus raw product group were significantly increased; the serum ALT, AST, and TBA levels in the stir-fried group were significantly increased, but the ALP level was not statistically significant; the serum ALP, ALT, AST, and TBA levels in the salt-baked group, Leigong group, and wine soaked group were significantly increased. Compared with the control group, the serum ALP, ALT, AST, and TBA levels in female mice in the Psoraleae Fructus raw product group, salt-baked group, Leigong group, and wine soaked group were significantly increased; the serum ALT, AST, and TBA levels in the stir-fried group were significantly increased; all groups had significantly decreased TP levels. Pathological results showed that there was no abnormality in the liver of mice in the control group; the liver of mice in the Psoraleae Fructus raw product group, stir-fried group, salt-baked group, Leigong group, and wine soaked group had pathological changes such as vacuolar degeneration of liver cells, glycogen degeneration of liver cells, loose cytoplasm of liver cells, and partial central hepatocellular hypertrophy; the degree of liver damage in mice caused by different processed products compared with the raw product group: raw product group > wine soaked group > stir-fried group > salt-baked group > Leigong group. Among them, the wine soaking method caused the highest degree of liver damage, while the Leigong method caused the least. CONCLUSION Psoraleae Fructus has different toxicities after being processed by different methods. According to LD50, the toxicity of Leigong method processed products is significantly reduced.
6.Mechanism study on miRNA- 18a and miRNA- 4802 suppressing drug resistance of lymphoma cells via inhibiting autophagy

OBJECTIVE To stud y the regulation mechanism of miRNA- 18a and miRNA-4802 on drug resistance of lymphoma cells via autophagy. METHODS Using human burkitt ’s lymphoma cell Daudi and human mantle cell lymphoma cell JeKo- 1 as the research objects ,adriamycin(ADR)and vincristine (VCR)as experimental drugs. After treatment of ADR and VCR ,relative cell viability was detected with CCK- 8 kit;the expression of apoptosis marker protein activated cleaved caspase- 9 and cleaved caspase-6 were detected by Western blot assay. The drug resistances of the two cells to ADR and VCR were investigated. The difference of autophagy activity between the two kinds of cells by expression detection of autophagy related proteins LC 3-Ⅱ and p62,autophagy flow experiment and transmission electron microscope observation. Fluorescence quantitative polymerase chain reaction was used to investigate the expression differences of miRNA- 18a and miRNA- 4802,ULK1 and ATG 7 mRNA in the two cells,and to detect the expression differences of ULK 1 and ATG 7 proteins. Taking JeKo- 1 cells as the research object ,the changes of autophagy activity and drug resistance were investigated after treatment with endogenous miRNAs (miRNA-18a mimics , miRNA-4802 mimics)of two simulated organisms. RESULTS After ADR and VCR treatment ,compared with Daudi cells ,JeKo-1 cells had stronger drug resistance and autophagy activity. The expression of miRNA- 18a and miRNA- 4802 in JeKo- 1 cells were significantly lower than Daudi cells ,mRNA and protein expression of ULK 1 and ATG 7 were significantly higher than those of Daudi cells (P<0.001). After treatment of miRNA- 18a mimics and miRNA- 4802 mimics,the autophagy activities and drug resistances of JeKo- 1 cells were decreased significantly. CONCLUSIONS miRNA-18a and miRNA- 4802 can decrease drug resistances of lymphoma cells to ADR and VCR by reducing 2 the expression of autophagy-initiating genes ULK1 and ATG7, and inhibiting the autophagy activity of lymphoma cells.
7.Relationship between GNRI and perioperative rehabilitation of elderly patients with gastric cancer

Objective :
To explore the relationship between geriatric nutritional risk index( GNRI) and perioperative nutritional status,postoperative recovery and complications in elderly patients with gastric cancer.
Methods :
In this retrospective study,212 elderly patients ( aged ≥60 years ) with gastric cancer who underwent gastrectomy were recruited.GNRI was used to retrospectively assess the patients' preoperative nutritional status ,and analyze the relationship between GNRI and perioperative nutritional status,postoperative recovery and complications.The ROC curve was applied to explore the value of GNRI in predicting postoperative complications.
Results :
The inci- dence of preoperative nutritional risk in elderly patients undergoing gastric cancer surgery was 45. 07%.Compared with the patients whose GNRI>98 points,the patients whose GNRI≤98 points had different degrees of decrease in serum total protein,albumin,prealbumin,hemoglobin and lymphocyte counts before surgery,day 1 and day 5-8 after surgery (P <0. 05) .The patients whose GNRI <92 points had longer postoperative hospital stay than those with GNRI>98 points (P<0. 05) .With the decrease of GNRI scores,the incidence of complications showed an upward trend(P<0. 001) .The multivariate analysis of the relationship between GNRI and postoperative complica- tions showed that TNM staging of III -IV and GNRI <92 points were independent risk factors for complications. GNRI had a good predictive value for the occurrence of complications (AUC = 0. 639,95% CI : 0. 570-0. 703,P = 0. 001,Cut-off value : 92. 21) .
Conclusion
GNRI can be used for preoperative nutritional assessment for eld- erly gastric cancer patients.Patients with GNRI<92. 21 points should be actively given nutritional therapy to im- prove perioperative nutritional status,speed up postoperative recovery,and reduce the occurrence of complications.
8.Influence of low-dose occupational radiation exposure on peripheral blood cells in radiation workers

Objective:To investigate the effect of low-dose ionizing radiation on blood cell parameters of radiation workers.Methods:A total of 124 staff members engaged in radiology were selected into the observation group, and they were divided into 4 subgroups of physicians, physicists, technicians, and maintainer according to their jobs. A total of 130 non-radiation-related staff members from the same hospital were selected into the control group. Blood cell parameters of peripheral blood of all subjects from 2016 to 2019 were collected, and the differences in blood cell parameters between the radiation group and the control group as well as 4 subgroups of the control group were analyzed and compared, and the correlation between the differences in blood cell parameters and the cumulative radiation dose was compared.Results:Compared with the control group, the white blood cell count, neutrophil count, red blood cell count and hemoglobin count in the observation group were lower than those in the control group (all P<0.05). There are no significant differences in cumulative radiation dose among different types of work (all P>0.05). Correlation analysis showed that the blood cell parameters of peripheral blood cells were not significantly correlated with the cumulative radiation dose. The blood cell count changes after 4-year low-dose ionizing radiation between the physicist group, the technician group and the maintainer sub-group were significantly different (all P<0.05), but the above differences were not related to the cumulative radiation dose (all P>0.05). Conclusions:Under the same exposure and protection conditions, the blood cell counts of different radiation-related workers are not significantly different, and the long-term cumulative radiation dose has no significant correlation with blood cell parameters. Therefore, peripheral blood cell parameters can no longer be used as a good indicator to reflect radiation damage, and it is urgent to find more convenient, intuitive and sensitive indicators of radiation damage.
9.Effects of Butin in Vernohia anthelmintica on Proliferation of Human Immortal Keratinocyte Cell Strain HaCaT and Cell Secretory Factors

OBJECTIVE:To study the effects of butin in Vernohia anthelmintica(VW)on proliferation of human immortal ke-ratinocyte cell strain HaCaT and cell secretory factors,and explore the mechanism of butin in VW in the treatment of vitiligo. METHODS:MTT method was used to determine the survival rate of HaCaT cells cultured by 0 (blank control),0.1,0.5,1.0, 5.0,10.0 μg/mL of butin for 48 h. Enzyme-linked immunosorbent assay was used to determine the contents of cell secretory factors as endothelin 1 (ET-1),ET-3,melanocyte stimulating hormone (MSH),stem cell factor (SCF),basic fibroblast growth factor (bFGF)in culture medium after HaCaT cells were cultured by 0.5,1.0,5.0 μg/mL of butin for 48 h. RESULTS:Compared with blank control,cell survival rate was increased to varying degrees after cultured by 0.1-5.0μg/mL of butin for 48 h,while decreased after cultured by 10.0 μg/mL of butin. Contents of ET-1,SCF,bFGF in culture medium were significantly increased after cultured by 0.5,1.0,5.0μg/mL of butin for 48 h(P<0.01);and contents of ET-3,MSH in culture medium were significantly increased af-ter cultured by 1.0,5.0 μg/mL of butin for 48 h(P<0.01). CONCLUSIONS:Butin can promote the proliferation of HaCaT cells, the mechanism may be associated with promoting the secretion of cell secretory factors of ET-1,ET-3,MSH,SCF,bFGF.
10.Study on the in vitro Transdermal Absorption Properties of Galangin

OBJECTIVE:To investigate the in vitro transdermal absorption properties of galangin and effects of different pene-tration enhancers on its transdermal behaviors,and provide reference for developing skin preparations using galangin as APIs in the treatment of vitiligo. METHODS:HPLC was used to determine the galangin content. Using cumulative permeation rate (Q) and the transdermal rate(J)of galangin as indexes,the effect of absorption of receiving solution [20%,40% polyethylene glycol 400 (PEG400)solution and 30% ethanol solution] and rotating rate(200,300,400 r/min)on galangin in complete skin of mice were investigated,as well as the azone(1%,3%,5%)and propylene glycol(10%,20%,40%)alone or combination on its penetra-tion promotion. And the transdermal properties of galangin in complete skin,exfoliating skin,dermis skin of rats and mice were de-tected. RESULTS:The best permeability of complete skin of mice showed in 40% PEG400 solution at rotating speed of 300 r/min with 5% azone alone,J was 3.2570 μg/(cm2·h). Js of complete skin,exfoliating skin,dermis skin of mice were 2.7199,34.016, 33.874 μg/(cm2·h),respectively;and those of rats were 0.4996,9.5124,17.406 μg/(cm2·h). CONCLUSIONS:Galangin can penetrate the complete skin of mice and rats,however,the penetration quantity is far lower than exfoliating skin and dermis skin.
