1.Correlation between music APP listening habits and depression tendency in college students based on SMOTEENN algorithm

Objective To investigate the influencing factors for tendency towards depression in college students having music listening habits with music APP,and develop a prediction model and further optimize it.Methods A total of 1 157 college students were subjected with convenient sampling and surveyed with questionaires between April and May 2023.Univariate analysis and logistic regression analysis were employed to identify the influencing factors.Then a prediction model was constructed based on these factors.SMOTEENN over-sampling algorithm was utilized to enhance the dataset and construct the prediction model.Results Logistic regression analysis revealed that female(OR=1.730,95%CI:1.257~2.396),senior grade(OR=2.649,95%CI:1.198~7.506),postgraduate grade(OR=2.041,95%CI:1.231~3.885),major in Science(OR=1.573,95%CI:1.052~2.350),listening for a duration of 0.5~2 h(OR=1.661,95%CI:1.011~2.695),music style of melancholy(OR=2.668,95%CI:1.701~4.226)and of nostalgia(OR=1.751,95%CI:1.086~2.837),and frequency of comments on 0~5%of songs(OR=2.938,95%CI:1.018~8.417)were independent risk factors for depressive tendency.Time since listening to music for 1~3 years(OR=0.547,95%CI:0.347~0.872),listening to music from 14:00 to 18:00(OR=0.375,95%CI:0.167~0.845)and 18:00 to 21:00(OR=0.313,95%CI:0.148~0.671),and preference for Chinese style songs(OR=0.711,95%CI:0.541~0.941)were independent protective factors.The logistic early warning model based on SMOTEENN algorithm demonstrated optimal predictive performance with an AUC value of 0.923.Conclusion Our constructed logistic regression model has identified 9 independent influencing factors associated with depression tendency among college students.The early warning model based on SMOTEENN algorithm can predict the depression tendency more accurately for college students.
2.Application of lower eyelid blepharoplasty by transconjunctival approach combined with micro-autologous fat transplantation in the rejuvenation of the infraorbital region

Objective:To observe the clinical effect of lower eyelid blepharoplasty combined with micro-autologous fat transplantation (MAFT) in the rejuvenation of the infraorbital regions.Method:A retrospective analysis was conducted on the clinical data of patients with lower eyelid pouch and tear trough treated with lower eyelid blepharoplasty combined with MAFT in the Department of Plastic Surgery of Affiliated Hospital of Nanjing University of Chinese Medicine from March 2020 to March 2022. Patients seeked medical attention due to lower eyelid pouch and tear trough, with or without skin laxity, and some patients had varying degrees of midface depression. After preoperative examination and evaluation of the patient’s infraorbital area and midfacial depression, appropriate individualized plans were developed.The patient underwent lower eyelid blepharoplasty by transconjunctival approach combined with tear trough MAFT. For patients with low or depressed midface, midface MAFT was also performed, and for patients with skin laxity, skin removal surgery was performed simultaneously. Complications were recorded after the operation. Preoperative and postoperative photos of more than 6 months were taken with the same postion and delivered to the same doctor to grade the patient’s Barton classification (0-3 grades, with higher grade indicating more obvious tear trough deformity). The improvement of tear trough deformity after surgery was assessed, and the global aesthetic improvement scale (GAIS) score was also assessed. The patients evaluated their improvement using the patients aesthetic improvement scale (PAIS) score (GAIS and PAIS are both 1-5 points, with higher scores indicating more significant postoperative improvement). Normal distribution metric data was represented by Mean±SD, and count data was represented by frequency and/or percentage; skewed distribution metric data was represented by M ( Q1, Q3), and compared by rank sum test. Results:A total of 69 patients were enrolled, including 9 males and 60 females, aged 25-45 years, with an average age of 36.7 years. Among them, 35 patients had midfacial depression, and 11 patients had skin laxity. (0.37±0.07) ml (0.2-0.6 ml) of micro-autologous fat was injected into the orbicularis oculi muscle on each side; (0.18±0.06) ml (0.1-0.2 ml) of micro-autologous fat was injected into the subcutaneous layer on each side; (3.80±0.98) ml (3-5 ml) of micro-autologous fat was injected into the deep medial cheek fat compartment and pyriform aperture fat compartment on each side. No serious complications occurred after the surgery. Postoperative follow-up lasted for 6-13 months, with an average of 8.7 months. The patient’s lower eyelid pouch, tear trough deformity, and depression in the midface have all improved, presenting a youthful and smooth appearance from the infraorbital area to the midface. The skin texture in the lower eyelid area had also improved. At the last follow-up, the Barton classification of the lacrimal groove decreased compared with that before surgery, and the difference was statistically significant [grade 0 (0, 1) vs. grade 2 (1, 2), Z=-7.62, P<0.001]. At the last follow-up, the PAIS score of 69 patients was 5 points in 49 cases (71%) and 4 points in 20 cases (29%); doctor’s GAIS score was 5 points in 38 cases (55%), 4 points in 27 cases (39%), and 3 points in 4 cases (6%) . Conclusion:For those patients with aging status of infraorbital region, like lower eyelid pouch and tear trough deformities, the combination of lower eyelid blepharoplasty through the transconjunctival approach and MAFT can achieve good therapeutic effects. This method is safe with a short recovery time.
3.Progress of Research on Intestinal Flora and its Metabolites in Ulcerative Colitis

The incidence of ulcerative colitis(UC)is on the rise globally,but its pathogenesis has not yet been clarified.Existing studies suggest that UC is closely related to genetic susceptibility,impaired intestinal epithelial barrier,abnormal immune response,intestinal flora dysbiosis and environmental factors.Intestinal flora has the functions of substance metabolism,nutrient absorption,energy conversion,immune regulation and maintenance of intestinal mucosal barrier,and has been proved to play an important role in UC.This article reviewed the progress of research on intestinal flora and its metabolites in UC,thus providing theoretical basis and new ideas for the diagnosis and treatment of UC.

5.Non-enterotoxin-producing Bacteroides fragilis inhibits TNF-α-induced inflammatory response in colonic epithelial cells by down-regulating the NF-κB signaling pathway

Objective:To investigate the mechanism by which non-enterotoxin-producing Bacteroides fragilis (NTBF) inhibits TNF-α-induced inflammatory responses in human normal colonic epithelial cells hcoEPIC, and explore new probiotic therapies for the prevention and treatment of colitis. Methods:The co-culture system of NTBF and hcoEPIC cells was established, and the adhesion and invasion ability of NTBF were detected, respectively. TNF-α was added to induce cellular inflammation after 4 h of co-culture of NTBF and hcoEPIC cells, and cell survival and apoptosis were detected by the CCK-8 assay and the AnnexinⅤ-FITC/PI assay respectively after 24 h. Key proteins of the NF-κB signalling pathway in hcoEPIC cells in different treatment groups were detected by Western blot and RT-qPCR, and the expression of downstream cytokines of this pathway incluing IL-1β, TNF-α and IL-10 were detected by ELISA. The effect of NTBF intervention on dextran sodium sulfate(DSS)-induced colitis mice was assessed by in vivo animal experiments. Results:NTBF adhered to hcoEPIC cells, and was non-toxic to the cells. Compared with control group, NTBF treatment alone did not affect cell survival and apoptosis of hcoEPIC cells ( P>0.05), but significantly reduced cell damage and apoptosis induced by TNF-α ( P<0.05); Compared with the TNF-α treatment alone group, the expression levels of p-NF-κB p65 and p-IκBα protein as well as NF-κB and IκBα mRNA were significantly reduced ( P<0.05); the production of IL-1β and TNF-α in the cell supernatant was reduced and the release of IL-10 was increased ( P<0.05). Animal experiments demonstrated that NTBF was indeed effective in alleviating DSS-induced colitis in ulcerative colitis model mice, which was mainly manifested by inhibiting weight loss, lowering DAI scores, improving colonic shortening, and attenuating colonic pathological damage in colitis-induced mice. Conclusions:NTBF may inhibit TNF-α-induced inflammatory responses in colonic epithelial cells by down-regulating the NF-κB pathway.
6.Clinical Efficacy of Qimai Qinlou Prescription in Treating Elderly Community-acquired Pneumonia (Non-severe) with Qi and Yin Deficiency and Phlegm-heat Obstructing Lung Syndrome

ObjectiveTo evaluate the clinical efficacy of Qimai Qinlou prescription in the treatment of elderly community-acquired pneumonia (CAP) (non-severe) with Qi and Yin deficiency and phlegm-heat obstructing lung syndrome and its impact on immune-inflammatory factors. MethodA total of 120 eligible patients were randomly divided into an observation group (60 cases) and a control group (60 cases). Both groups received intravenous cefoxitin sodium. In addition, the observation group received oral Qimai Qinlou prescription, while the control group received an oral placebo simulating Qimai Qinlou prescription. The treatment course was 14 days. The disappearance time of major clinical symptoms and signs was recorded. Traditional Chinese medicine (TCM) syndrome scores and the Clinical Research Outcome (CAP-CRO) scale scores for pneumonia of the two groups were compared. Chest computed tomography (CT) scans were performed, and peripheral blood levels of procalcitonin (PCT), interleukin-6 (IL-6), serum amyloid A (SAA), high-sensitivity C-reactive protein (hs-CRP), CD4+, CD8+, and CD4+/CD8+ were measured. The conversion rate to severe condition during hospitalization, readmission rate within 30 days after discharge, and safety evaluation were recorded. ResultAfter treatment, the observation group showed significantly shorter time of fever, cough, expectoration, and disappearance time of lung moist rales than the control group (P<0.01). The TCM syndrome scores, CAP-CRO scores in all dimensions, and total scores in both groups were significantly reduced as compared with those before treatment (P<0.01). After treatment, the observation group had a more significant reduction than the control group (P<0.01). The levels of PCT, IL-6, SAA, and hs-CRP in both groups were significantly reduced as compared with those before treatment (P<0.01). After treatment, the observation group showed a more significant reduction than the control group (P<0.01). There was no statistically significant difference in the changes of CD4+, CD8+, and CD4+/CD8+ in the control group before and after treatment. However, in the observation group, CD4+ and CD4+/CD8+ levels significantly increased (P<0.01), while CD8+ level significantly decreased (P<0.01) after treatment. After treatment, CD4+ and CD4+/CD8+ in the observation group significantly increased (P<0.01), and CD8+ significantly decreased as compared with those in the control group (P<0.01). At 7, 10, and 14 days after treatment, the curative rates in the observation group were 53.33% (32/60), 85.00% (51/60), and 91.67% (55/60), respectively, which were higher than 31.67% (19/60), 61.67% (37/60), and 68.33% (41/60) in the control group (χ2=5.763, 8.352, 10.208, P<0.05). After treatment, the total effective rate of CT scanning in the observation group was 93.33% (56/60), higher than 80.00% (48/60) in the control group (χ2=4.615, P<0.05). The conversion rate to severe condition during hospitalization in the observation group was 3.33% (2/60), lower than 15.00% (9/60) in the control group (χ2=4.904, P<0.05). The readmission rate within 30 days after discharge in the observation group was 8.33% (5/60), lower than 23.33% (14/60) in the control group (χ2=5.065, P<0.05). No serious adverse drug reactions were observed in either group during the treatment period. ConclusionQimai Qinlou prescription can enhance immune function, alleviate inflammatory reactions, significantly relieve clinical symptoms, shorten the duration of the disease, improve the curative rate and CT scanning efficacy, prevent disease progression, reduce the readmission rate in the short term, and is clinically safe for the treatment of elderly patients with non-severe CAP with Qi and Yin deficiency and phlegm-heat obstructing lung syndrome. It is worthy of further research and application.
7.A behavioral model of heroin addiction in mice based on associative learning mechanism

Objective :
To establish a laboratory model of heroin addiction in C57BL /6 mice based on associative learning mechanisms.
Methods:
The black box and white box were selected as the memory training environment, and three behavioral training paradigms were studied : ① Pavlovian conditional position preference ( CPP) training paradigm,mice were placed in the white box for memory reinforcement training for 30 min after intraperitoneal injection of 0. 1 ml of corresponding concentrations (5. 0,10. 0,20. 0 mg / kg) of heroin at 9 :00 a.m.,and 24 h later 0. 1 ml of 0. 9% NaCl solution was injected intraperitoneally into the black box for training,and after the training,the mice were tested for their memory preference for the black and white boxes (movement time of different boxes) . ② A naloxone conditional position aversion ( CPA) training paradigm was conductedbased on the results of the CPP training paradigm. ③ Behavioral sensitization training paradigm,heroin addiction rating scale was established based on the statistical results of 3 behavioral experiments and the lethality of experimental animal disease mice after drug administration.Three different doses of heroin (5. 0,10. 0,20. 0 mg / kg) were selected to induce heroin addiction,and the most appropriate heroin concentration was selected by the results on the rating scale.
Results:
In the CPP training paradigm,CPP was observed in all heroin groups (P<0. 05,P<0. 001,P<0. 05) .In the CPA training paradigm,the CPA induction rate was highest in the 10. 0 mg / kg heroin group compared to the control group (P <0. 01 ) .In the behavioral sensitization training paradigm ,all heroin groups caused behavioral sensitization changes (P<0. 001) ; but the 5. 0 and 10. 0 mg / kg heroin groups did not cause animal mortality.Overall,the 10. 0 mg / kg heroin group had the highest dose score on the rating scale.It could be used as a concentration to establish a stable experimental animal model of heroin addiction.
Conclusion
The study was effective in establishing a heroin addiction model in mice,and it was suitable for modeling drug concentration screening,with high animal survival rate and simple and practical.The combined learning mechanism can effectively shorten the model establishment period.
8.Establishing a rat model of preeclampsia in early pregnancy and observing its behavior and cognitive effects on the offspring

Objective:
To establish a rat model of preeclampsia (PE) in early pregnancy and to observe the changes in phenotype,pregnancy outcome and cognitive ability of offspring.
Methods :
The pregnant rats were randomly divided into model group and control group.Ultra-low dose lipopolysaccharide (LPS) (0. 5 μg / kg) and an equal volume of normal saline were injected into the tail vein of pregnant rats on the fifth day of pregnancy.The levels of blood pressure ,12-hour urinary protein ,peripheral blood coagulation factors and placental cytokines in the two groups were measured.Furthermore,placental pathology,pregnancy outcomes,and cognitive abilities of offspring were observed.
Results:
Blood pressure and urinary protein levels of model group were significantly higher than those of control group levels.Compared with the control group,the levels of platelet and antithrombin Ⅲ (AT Ⅲ) in the peripheral blood of pregnant rats in the model group were lower than those in the control group,while D-dimer was higher than that in the control group,the weight of the fetus and placenta in the model group decreased (P <0. 001) ,the expression levels of interleukin ( IL) -6,tumor necrosis factor α ( TNF-α) and interferon gamma (INF-γ) in peripheral blood increased,while the expression level of transforming growth factor β1 (TGF-β1) decreased(P<0. 001) .The water maze test showed that the latency of the offspring of the model group to the plat- form was longer than that of the control group (P<0. 05) ,while the frequency of crossing the platform quadrant and the time of staying in the platform quadrant of the model group were lower than those of the control group (P < 0. 05 ) .HE and PAS staining showed that there were infiltration of inflammatory cells in the basal layer of placenta, obvious decrease of blood vessels in labyrinthine area,slight edema of renal interstitium and degeneration of local renal tubular epithelial cells in the model group,while there were no above pathological changes in placenta and kidney in the control group.
Conclusion
A single injection of LPS in early pregnancy can successfully induce PE- related symptoms and adverse pregnancy outcomes such as fetal growth restriction and lead to the decline of cogni- tive ability of offspring.
9.Clinical observation on adverse reaction after vaccination of inactivated 2019-nCoV vaccine in population aged 3 years and above

Objective:To evaluate the safety of mass vaccination of inactivated 2019-nCoV vaccine in population aged ≥3 years in Guizhou Province.Methods:The open-label study was conducted in eligible volunteers in Yanhe County of Guizhou from June 2021 to July in 2022. In the study, two doses of the inactivated 2019-nCoV vaccine were given at (0, 28) days according to the immunization schedule. The information about adverse reaction (AR) within 30 minutes and during 0-28 days after vaccination were collected, and the incidence of AR was analyzed by age, doses, and health status.Results:The overall incidence of AR was 1.51% (294/19 458), all ARs, mainly pain at injection site, occurred within 7 days after the vaccination, the AR grade was 1-2. The incidence of AR was 1.01% in age group 3- years (58/5 721), 2.44% in age group 18- years (220/9 017), and 0.34% in age group ≥60 years (16/4 720). The differences were significant ( P<0.001). The incidence of AR after the first dose vaccination (1.20%, 233/19 458) was significantly higher than that after the second dose (0.37%, 61/16 368), the difference was significant ( P<0.001). In the elderly aged ≥60 years, the incidence of AR was 0.36% (9/2 520) in healthy group and 0.32% (7/2 200) in group with underlying medical conditions, the differences were not significant ( P=0.818). Conclusion:The domestic inactivated 2019-nCoV vaccine showed good safety in mass vaccination in population aged ≥3 years, including the elderly in both healthy group and group with underlying medical conditions.
10.A study of the clinical curative effect of nucleos(t)ide analogues treated to pegylated interferon-α add-on therapy in patients with chronic hepatitis B

Objective:To investigate the hepatitis B surface antigen (HBsAg) clearance condition and its predictive factors after treatment with nucleos(t)ide analogues to pegylated interferon-α add-on therapy in patients with chronic hepatitis B.Methods:Patients with chronic hepatitis B who visited the First Affiliated Hospital of Zhengzhou University from 2018~2019 were prospectively enrolled. HBsAg≤ 1500 IU/mL, hepatitis B e antigen-negative, HBV DNA undetectable, received antiviral treatment with nucleos(t)ide analogues for at least one year, and pegylated interferon-α add-on therapy for 48 weeks were included. The primary endpoint of study was to determine the proportion of HBsAg clearance at 72 weeks. Concurrently, the predictive factors for HBsAg clearance were analyzed. Quantitative and qualitative data were analyzed using a t-test or non-parametric test and a Fisher's exact test.Results:A total of 38 cases were included in this study, of which 13 cases obtained HBsAg clearance at 48 weeks of therapy and another six cases obtained HBsAg clearance throughout the extended treatment period of 72 weeks, accounting for 50.00% of all enrolled patients. There was a significant difference in HBsAg dynamics between the HBsAg clearance group and the non-clearance group (P < 0.05). Univariate logistic regression analysis showed that patients' age, baseline, 12-and 24-week HBsAg levels, and early HBsAg reduction were predictive factors for HBsAg clearance at 72 weeks of treatment. Multivariate logistic regression analysis showed that age (OR = 1.311; P = 0.016; 95% confidence interval: 1.051~1.635) and HBsAg levels at 24 weeks of treatment (OR = 4.481; P = 0.004; 95% confidence interval: 1.634~12.290) were independent predictors for HBsAg clearance.Conclusion:Hepatitis B e antigen-negative, nucleos(t)ide analogue treated, HBsAg ≤ 1500 IU/mL, and HBV DNA undetectable, peg-IFNα add-on treatment for 48 weeks could promote HBsAg clearance in patients with chronic hepatitis B. Six of the sixteen cases (37.50%) who did not obtain HBsAg clearance at week 48 did so with the course of therapy extended to week 72. Hence, the optimal individualized treatment strategy should be customized according to the predictors rather than the fixed 48-week course. Age (≤ 38), baseline HBsAg level (≤2.86 log 10IU/ml), HBsAg level at 24 weeks (≤ 0.92 log 10IU/ml), and 12-week HBsAg decrease from baseline (≥ 0.67 log 10IU/ml) indicate that patients are highly likely to obtain HBsAg clearance at the 72 weeks of combination therapy, in which the combined indicator based on HBsAg level ≤0.92 log 10IU/ml at 24 weeks will identify 85.0% to 100.0% of patients with HBsAg clearance.
