1.Attraction of different concentrations of glucose solution to Aedes albopictus, Culex pipiens pallens and their egg-laying behaviors

Objective:
To explore the effects of different concentrations of glucose solution on the survival of Aedes albopictus and Culex pipiens pallens larvae, the attraction to mosquitoes and egg-laying behaviors, so as to provide the reference for developing mosquito control technology based on sugar bait.
Methods:
White porcelain bowls were filled with 100 mL of 3%, 5%, 8%, 10% and 15% glucose solutions. Ten of fourth instar larvae of Aedes albopictus or Culex pipiens pallens were added to each bowl, and the survival of larvae was recorded after 2, 4, 6, 24, 48 and 72 hours. Egg-laying cups containing 5%, 8% and 15% glucose solution were put in mosquito cages containing fully blooded female mosquitoes of Aedes albopictus and Culex pipiens pallens (50 mosquitoes each), and the total number of eggs laid in 72 hours was observed. The analogous site room was filled with fully blooded and starved female mosquitoes of Aedes albopictus and Culex pipiens pallens (100 mosquitoes each), and simple mosquito control buckets containing 5% and 8% glucose solution and black sticky insect plates. The number of mosquitoes and eggs was observed after 6 days. All the above experiments were repeated 3 times using dechlorinated water as the control.
Results:
The 72 hour corrected mortality rates of Aedes albopictus and Culex pipiens pallens larvae gradually increased with the increase of glucose concentration. The glucose solution with 5% and higher concentrations was not suitable for mosquito larvae to survive. The attraction of egg-laying behaviors to Aedes albopictus and Culex pipiens pallens gradually decreased with the increase of glucose concentration. The effects were similar between 5% and 8% glucose solution, with the averages of 686.67 and 682.33 eggs for Aedes albopictus, and 3.00 and 2.33 egg rafts for Culex pipiens pallens. In analogous site room, there were 93.33, 105.00 and 130.33 adult mosquitoes captured on average in the control group, 5% and 8% glucose solution groups, respectively, with 8% glucose solution group more attractive to adult mosquitoes than the control group (F=3.283, P=0.030); there were 70.33, 55.33 and 63.00 Aedes albopictus eggs (eggs counts+larvae counts) on average, respectively, with statistically significant differences among the three groups (H=6.761, P=0.034).
Conclusion
Glucose solution with concentration of 5% or higher can effectively inhibit the survival of Aedes albopictus and Culex pipiens pallens larvae, and attractive to adult mosquitoes and egg-laying behavoirs.
2.Effect of the enhanced external counterpulsation combined with drug on social function and efficacy in patients with depressive episodes

ObjectiveTo observe the effect of the enhanced external counterpulsation (EECP) combined with drug on social function and efficacy in patients with depressive episodes, so as to provide references for the treatment of depressive episodes. MethodsA total of 66 hospitalized patients who was in hospital at department of psychiatry of the Second Xiangya Hospital of Central South University, met the criteria of Diagnostic and Statistical Manual of Mental Disorders,fifth edition(DSM-5) diagnosis of depressive episode or bipolar disorder depressive episode from May 2019 to March 2020 were included by simple random sampling. The participants were divided into study group (n=36) and control group (n=30) according to the random number table method. Both groups received conventional drug treatment, and the study group recieved the EECP intervention at same time. The Depression symptoms and social function were assessed before and after treatment by using Hamilton Depression Scale-24 item (HAMD-24) and Sheehan Disability Scale (SDS). Treatment efficacy of the two groups was compared. ResultsAfter the intervention, the HAMD-24 and SDS scores in both groups were lower than those before treatment, the differences were statistically significant (t=8.149, 5.791, 8.016, 3.488, P˂0.01). And the SDS score of the study group was siginficantly lower than that of the control group (t=-3.008, P<0.05). The total effective rate of treatment in the study group was higher than that of the control group, and the difference was statistically significant (90.63% vs. 63.33%, χ²=8.725, P˂0.05). ConclusionEECP therapy combined with drug has better efficacy on the patients with depressive episodes, and it can improve social function effectively.
3.Effects of a perioperative whole course composite thermal insulation strategy on complications of cesarean section, maternal coagulation function and serum inflammatory indexes

Objective:To investigate the effects of a perioperative whole course composite thermal insulation strategy on complications of cesarean section, maternal coagulation function and serum inflammatory indexes.Methods:A total of 250 pregnant women who were subjected to cesarean section in Zhoushan Hospital between June 2020 and August 2021 were included in this study. The 125 pregnant women who gave birth using a routine simple thermal insulation strategy from June to November 2020 were assigned to the routine simple thermal insulation group, and those who gave birth using a perioperative whole course composite thermal insulation strategy were assigned to whole course composite thermal insulation group. Two groups of pregnant women underwent cesarean section under subarachnoid block. Volume of intraoperative blood loss was recorded. The incidence of complications such as shivering and postoperative infection was calculated. Platelet count, prothrombin time, activated partial thromboplastin time, thrombin time measured before surgery and 48 hours after surgery were compared between the two groups. Peripheral blood white blood cell count, neutrophil count (N%), C-reactive protein, procalcitonin, interleukin-6 measured 48 hours after surgery were compared between the two groups.Results:Volume of intraoperative blood loss in the whole course composite thermal insulation group was significantly lower than that in the routine simple thermal insulation group [(393.84 ± 79.78) mL vs. (434.80 ± 123.49) mL, t = 3.11, P < 0.05). The incidence of shivering and postoperative infection in the whole course composite thermal insulation group was 10.4% (13/125) and 7.2% (9/125), respectively, which was significantly lower than that in the routine simple thermal insulation group [25.6% (32/125), 18.4% (23/125), χ 2 = 9.78, 7.02, both P < 0.05]. At 48 hours after surgery, prothrombin time, activated partial thromboplastin time, thrombin time in the whole course composite thermal insulation group were (10.28 ± 0.48) seconds, (26.97 ± 2.27) seconds, and (14.09 ± 1.36) seconds, respectively, which were significantly shorter than those in the routine simple thermal insulation group [(11.71 ± 0.27) seconds, (27.96 ± 2.25) seconds, (15.91 ± 1.09) seconds, t = 7.34, 3.43, 11.66, all P < 0.05]. At 48 hours after surgery, white blood cell count, neutrophil count, C-reactive protein, procalcitonin, and interleukin-6 in the whole course composite thermal insulation group were (10.38 ± 2.38) ×10 9/L,(0.79 ± 0.06), (52.79 ± 20.73) mg/L, (0.13±0.42) μg/L, and (55.73 ± 24.38) ng/L, respectively, which were significantly lower than those in the routine simple thermal insulation group [(12.24 ± 7.05) × 10 9/L, 0.81 ± 0.05, (65.38 ± 25.92) mg/L, (0.20 ± 0.97) μg/L, (76.22 ± 39.08) ng/L, t = 2.79, 2.92, 4.24, 8.12, 4.97, all P < 0.05]. Conclusion:Perioperative whole course composite thermal insulation strategy can improve the coagulation function of pregnant women who are subjected to cesarean section under subarachnoid block, reduce volume of intraoperative blood loss, and decrease incidence of shivering, inflammatory reaction, and postoperative infection.

Clinical genetic testing results are compiled into a standardized report by genetic specialists and provided to clinicians and patients (Should the patient be intellectually disabled or under 18,the report will be provided to his/her parents or legal guardians).The content of genetic testing report should conform to relevant guidelines,industry standards and consensus.The decisions of clinicians will be made based on the report and clinical indications.Genetic counselors should provide post-test counseling to clinicians and patients or their authorized family members.A mechanism of follow-up visit after the genetic testing should be established with informed consent.Data should be shared by clinical institutions and genome sequencing institutions.As findings upon follow-up visit can help with further evaluation of the results,genome sequencing institutions should regularly re-analyze historical and follow-up data,and the updated results should be shared with clinical institutions.All activities involving reporting,genetic counselling,follow-up visiting,and re-analyzing should follow the relevant guidelines and regulations.
5.Efficacy and safety of pazufloxacin mesylate eye drops for bacterial conjunctivitis:a multicenter randomized, double-masked, parallel controlled clinical trial

Objective To compare the clinical efficacy and safety following the topical application of pazufloxacin mesylate eye drops with levofloxacin eye drops (LOFX) for bacterial conjunctivitis.Methods A multicenter,randomized,investigator-masked,parallel-controlled clinical trial was performed.Five hundred and twenty eyes of 520 patients with bacterial conjunctivitis were enrolled from March to October 2008 in seven ophthalmic centers in China.The patients were randomized into trial group and control group in 3 ∶ 1 ratio with the 390 eyes in the trial group and 130 eyes in the control group.Pazufloxacin mesylate eye drops was topically used 4 times per day for consecutively 7-14 days in the trial group,and levofloxacin eye drops was used in the same way in the control group.Microbiological cultures for conjunctiva sac secretions and drug sensitive test were carried out before and at the end of the administration of eye drops.The signs and symptoms were observed and scored before treatment and 0,3,7 and 14days after treatment.The adverse events following the administration of the eye drops were evaluated and compared.Results The intention to treat analysis (ITT) showed that the cure rate was 59.38% in the trial group and 60.47% in the control group,with the effective rate 88.80% and 86.05%,respectively,with an insignificant difference between the two groups (x2 =0.12,P =0.72).The clinically evaluable analysis (CE) exhibited that the cure rates were 63.48% in the trial group and 63.87% in the control group,with the effective rate 92.46% and 88.24%,whichwas not significantly different between them (x2 =0.54,P=0.46).The modified-ITT analysis (mITT) showed that the cure rates were 60.57% in the trial group and 62.07% in the control group,with the effective rate 90.32% and 88.51%,without significant difference between the two groups (P>0.05).Based on microbiologically evaluable analysis (ME),the clinical cure rates were 63.71% and 63.41% in the trial group and control group,and the effective rates were 93.44% and 90.24%,respectively.There was no significantly difference between the both groups (P>0.05).In the trial group and control group,the bacterial eradication rate was 89.42% and 90.80% based on ITT,90.11% and 92.77% based on CE,respectively.There was no significant difference in incidences of adverse events following the administration of the drug between the trial group and control group,including ocular tolerance,burning sensation,pricking and itching (P =0.34).Conclusions The effectiveness and adverse response were resemble between Pazufloxacin mesylate eye drops and LOFV following the topical application for bacterial conjunctivitis,which indicate that Pazufloxacin mesylate eye drops is effective and safe.
6.Application of DNA barcoding to identification of rodents in Zhejiang Province

Objective:
To apply DNA barcoding to identifying the rodents in Zhejiang Province.
Methods :
Rodents were captured from Jiashan,Longyou,Yunhe and Ninghai counties in Zhejiang Province. The DNA was extracted from ears of rodent samples,and was amplified and sequenced with mitochondrial cytochrome C oxidase subunit I(COI)genes. The obtained sequences were compared with the related sequences in GenBank,and neighbour-joining evolutionary tree was constructed. Then the results by DNA barcoding and by morphological identification were compared.
Results :
A total of 22 COI gene samples were amplified. The evolutionary tree constructed by 18 samples was consistent with the morphological identification results and 4 samples were different:Suncus murinus should be Crocidura lasiura,infant rats of Rattus losea and Rattus tanezumi was re-identified as Rattus rattus,infant rats of Microtus fortis(sample number:NH-1)needs further identification.
Conclusion
DNA barcoding can effectively correct the errors of morphological identification,thus combining the two methods could improve the accuracy of rodent identification.
7.Timing and methods of indwelling catheterization for general anesthesia male surgical patients

Objective To explore the optimum time and method of indwelling urethral catheterization for male patients undergoing general anesthesia.Methods A total of 90 male patients undergoing elective general anesthesia were randomly divided into three groups.Group A (n =30) received urethral injection of lidocaine glue from the urethra before induction of general anesthesia and then carried out urethral catheterization,group B conducted urethral catheterization after induction of general anesthesia,and group C was injected with lidocaine gel before catheterization after general anesthesia induction.The changes of heart rate and blood pressure were compared between the three groups before urethral catheterization,and urethral irritation and restlessness score when urethral catheterization,awaken time of anesthesia,after returning to the ward were compared in the three groups.Results Group A had higher preoperative and postoperative heart rate and blood pressure than operation before (P < 0.05),but there was no significant difference between group B and group C after urethral catheterization (P > 0.05).Urethral irritation in group A was more obvious,but there was no stimulation in group B and group C,and was significantly higher in group A than that in groups B and C,was higher in group B in awaken period of anesthesia and after returning to ward,than groups A and C (P < 0.05).The restlessness score in group B was significantly higher than that in groups A and C (P < 0.05),but no significant difference between group A and group C was observed (P > 0.05).Conclusion After general anesthesia induction,urethral injection of lidocaine glue before indwelling urethral catheterization can effectively alleviate blood pressure and heart rate increase due to male patients'privacy,and urethral stimulation,and reduce incidence of restlessness of male patients with general anesthesia.

9.Synergistic antitumor activity of chemotherapy and immunotherapy in the treatment of established rat solid tumors

Objective To explore immunotherapy effective combined with active immunotherapy in different time according rats bearing-tumor after paclitaxel and carboplatin chemotherapy, and to identify the optimization time and strategy of vaccine and seek rational chemo-immunotherapy strategies in ovarian cancer treatment. Methods The dynamic immunocytes number and function in established tumor treated with paclitaxel and carboplatin chemotherapy were investigated. The changes of established tumor volume and immune function of different groups were observed according to combining different time after chemotherapy and vaccine. Results Lymphopenia was observed and the number of lymphocyte subset decreased remarkably on the 6th day, but all cells were found almost recovered on 15th day after chemotherapy. There is the process of immune-enhancing from post-chemotherapy 6 day to 10 day and reversal of immune suppression temporary. The combination post-chemotherapy 6 day with CTL caused a significantly delayed tumor growth in both tumor models and induced significant the proliferation of T lymphocyte by [H] 3 releasing. The number of CD8+T cell is the highest, but the expression of Tr cell was lowest in the group of post-chemotherapy 6 day with CTL. Furthermore, the ability of CD8+T secretion IFN-γ is the most in the post-chemotherapy 6 day with immunotherapy groups. Conclusion Combinational paclitaxel and carboplatin chemotherapy has synergistic effects with active immunotherapy boosting against tumor during window periods, where 6 days after chemotherapy with the most decreased number of lymphocytes in the animal periphery might represent the optimal checkpoint for the immune therapy against tumors. Therefore, monitoring the immune status of tumor patients might become one of the important prerequisites for the effective immune therapy when designing the comprehensive therapeutic strategies.
10.The dynamic changes in the numbers and functions of CD8~+T cells and NK cells in advanced ovarian cancer patients undergoing first line chemotherapy

Objective: The purpose of this study is to investigate the dynamic changes in the numbers and functions of CD8~+T cells and NK cells in patients with advanced ovarian cancer undergoing first line chemotherapy,so as to identify whether there was a "window period" of anti-tumor immune suppression reverse after chemotherapy.Methods: Peripheral blood samples from each ovarian cancer patient were obtained before (S_0) and at day 5-7 (S_1),day 12-14 (S_2) and day 25-28 (S_3) after Chemotherapy in 13 patients.The numbers and proportions of CD3~+,CD4~+,CD8~+ and nature killer (NK) cells were analyzed by flow cytometry technique.The percentages of specific IFN-γ-secreting CD8~+ cells were also calculated after that peripheral lymphocytes had been stimulated with self tumor lysates.Cytotoxicity of NK cells against K562 cells was detected by LDH releasing assay.Results: The numbers of CD3~+,CD4~+,CD8~+T cells and NK cells reduced to the lowest on S1.Compared to those of the control group,and the percentages of IFN-γ-secreting CD8~+T cells were remarkably higher on S_1,S_2 and S_3 when CD8~+T cells were stimulated with autologous tumor antigen,and the percentage of CD8~+IFN-γ~+ cell reached the highest on S2.No significant differences of NK cell cytotoxicity against K562 cells were found on S_1,S_2 and S_3 compared to S0.Conclusion: Paclitaxel and carboplatin induce lymphopenia,which triggers the temporary immune reconstitution.During immune reconstitution the enhanced priming of CD8~+T cell response by autologons tumor antigen is found while the function of NK cells does not change significantly.It probably turns out that the" window period"during immune reconstitution offers a best opportunity for cancer immunotherapy.
