1.Determination of diosgenin and ruscogenin in Radix Ophiopogonis by nonaqueous capillary electrophoresis.

Nonaqueous capillary electrophoresis is used for the determination of the contents of diosgenin and ruscogenin in Radix Ophiopogonis. The operating buffer was composed of 20 mmol x L(-1) Na2B4O7-HCl (pH 7.61) in 70% methanol. The applied voltage was 25 kV and detection potential was at +0.70 V. With these conditions, the components were successfully separated. The content of diosgenin in Radix Ophiopogonis was 0.018 mg x g(-1) and ruscogenin was 0.008 mg x g(-1). The average recoveries of diosgenin and ruscogenin were 102% and 99.2%, respectively. A new method of the quality control of diosgenin and ruscogenin in Radix Ophiopogonis is provided.
2.Determination of genistein and quercetin in Huaijiao Pills by RP-HPLC
Chinese Traditional Patent Medicine 1992;0(07):-
AIM To determine the contents of the two free-and conjugated quercetin and genistein in Huaijiao Pills (Fructus Sohorae,Radix Sanguisorbae,Radix Scutellariae,etc.).by RP-HPLC. METHODS HPLC was employed with a column of Diamonsil ODS-C_ 18 (4.6 mm?150 mm,5.0 ?m) and a mobile phase of MeOH-H_2O-HAc(2∶ 3∶ 0.25),the detection wavelength being at 254 nm and the column temperature being at 30?C . RESULTS Nice linear correlation existed, when quercetin was within 0.204 8-1.229 ?g and genistein within 0.532 5-3.192 ?g.The correlation coefficient of each group was 0.999 1 and 0.999 8 respectively.The average recoveries of quercetin and genistein were 99.0 % and 99.1 % ,respectively.The RSDs were 1.02 % and 0.40 % ( n =5),respectively. CONCLUSION With this method,quercetin and genistein in Huaijiao Pills could be determined simultaneously and applicable to the quality control of Huaijiao Pill.
