1.Regulation of DZIP1 Expression in CAFs by Jianpi Yangzheng Xiaozheng Formula and Its Impact on Mesenchymal Characteristics of Diffuse Gastric Cancer

ObjectiveTo establish a subcutaneous xenograft model of gastric cancer in nude mice and to identify differentially expressed genes (DEGs) affected by Jianpi Yangzheng Xiaozheng formula (JPYZXZ) in diffuse gastric cancer (DGC) using transcriptome sequencing. The study aims to identify potential key prognostic factors and preliminarily elucidate the therapeutic mechanism of JPYZXZ. MethodsSubcutaneous tumor tissues were collected from nude mice treated with JPYZXZ (6 g·kg-1)and from the untreated traditional Chinese medicine control group. High-throughput RNA sequencing was performed to extract and analyze total RNA and identify DEGs, followed by functional enrichment analysis. DEGs were intersected with cancer-associated fibroblast (CAF)-related genes identified from single-cell RNA sequencing (scRNA-seq) data. A Cox proportional hazards regression model (Cox model) was constructed to identify potential key targets, among which DAZ interacting protein 1 (DZIP1) was selected. The role of DZIP1 in DGC progression was further verified through in vitro and in vivo experiments. ResultsTranscriptome sequencing revealed that DEGs regulated by JPYZXZ were significantly enriched in biological processes such as focal adhesion, phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway, and vascular development (P<0.05). Intersection analysis with CAF marker genes identified ten potential common target genes. Cox regression analysis combined with the Cancer Genome Atlas (TCGA) dataset for stomach adenocarcinoma (STAD) confirmed that DZIP1 is an independent prognostic factor significantly associated with poor outcomes. Transcriptome and immunohistochemical analyses showed that DZIP1 expression was significantly higher in gastric cancer tissues than in adjacent tissues, while JPYZXZ treatment downregulated DZIP1 expression in a dose-dependent manner. Clinical data further demonstrated that serum DZIP1 levels in patients treated with JPYZXZ were significantly lower than those in the control group. ConclusionJPYZXZ may inhibit the malignant progression of DGC by downregulating DZIP1 expression, thereby suppressing CAF activation and epithelial-mesenchymal transition (EMT). These findings provide experimental evidence and identify DZIP1 as a potential therapeutic target in DGC treatment.

Obesity and related metabolic syndromes have been recognized as important disease risks,in which the role of adipokines cannot be ignored.Adiponectin(ADP)is one of the key adipokines with various beneficial effects,including improving glucose and lipid metabolism,enhancing insulin sensitivity,reducing oxidative stress and inflammation,promoting ceramides degradation,and stimulating adipose tissue vascularity.Based on those,it can serve as a positive regulator in many metabolic syndromes,such as type 2 diabetes(T2D),cardiovascular diseases,non-alcoholic fatty liver disease(NAFLD),sarcopenia,neurodegenerative diseases,and certain cancers.Therefore,a promising therapeutic approach for treating various metabolic diseases may involve elevating ADP levels or activating ADP receptors.The modulation of ADP genes,multimerization,and secretion covers the main processes of ADP generation,providing a comprehensive orientation for the development of more appropriate therapeutic strategies.In order to have a deeper understanding of ADP,this paper will provide an all-encompassing review of ADP.
4.Analysis of nosocomial infection after intermediate-high dose cytarabine consolidation theraphy in patients with acute myeloid leukemia

Objective:To investigate the clinical characteristics, influencing factors and prevention and treatment measures of nosocomial infection in patients with acute myeloid leukemia (AML) (non-acute promyelocytic leukemia) after applying intermediate-high dose cytarabine (Ara-C) chemotherapy.Methods:The clinical data of 80 patients with AML treated with intermediate-high dose Ara-C in the Second Hospital of Shanxi Medical University from March 2013 to January 2020 were analyzed retrospectively. The clinical features of nosocomial infection were summarized and the influencing factors of infection were analyzed by using multivariate logistic regression.Results:A total of 80 patients received 198 times of chemotherapy, and the infection rate was 72.7% (144/198). Infection sites mainly included respiratory tract infection, pulmonary infection, gastrointestinal infection. A total of 45 strains of pathogenic bacterias were detected, among which Gram negative bacilli accounted for 55.6% (25/45), Gram positive cocci accounted for 24.4% (11/45), fungi accounted for 8.9% (4/45) and viruses accounted for 11.1% (5/45). There were no significant differences in infection rate, hospitalization time, neutrophils recovery time and hospitalization expenses between the sterile laminar flow ward and the general ward (all P > 0.05). Multivariate logistic regression analysis showed that infection during induction chemotherapy was independent risk factor of infection ( OR = 5.076, 95% CI 1.978-13.022, P =0.001), and antibiotic prevention was independent protective factor of nosocomial infection ( OR = 0.332, 95% CI 0.136-0.803, P = 0.014). Conclusions:The infection rate of AML patients receiving intermediate-high dose Ara-C chemotherapy is high. During the treatment, we should be alert to the infection during induction chemotherapy and use antibiotics to prevent it in time. For patients undergoing intermediate-high dose Ara-C chemotherapy, strengthening the environmental cleanliness of general wards may achieve the same preventive effect as that of sterile laminar flow wards.
5.Myopia and its influencing factors among grade 4-6 pupils in Liaoning Province

Objective:
To investigate visual acuity of pupils of grade 4-6 in Liaoning Province in 2019 and to analyze its influencing factors, and to provide the scientific basis for myopia prevention and intervention.
Methods:
A total of 16 716 students of grade 4-6 in 14 cities of Liaoning Province were selected by multi-stage stratified random cluster sampling, and the long-range visual acuity and refraction was evaluated and a questionnaire survey was conducted. Logistic regression analysis was used to analyze the in-fluencing factors of myopia in primary school students.
Results:
The myopic rate of grade 4-6 pupils in Liaoning Province was 49.17%. Multiple Logistic regression analysis found that the main factors affecting vision included urban and rural division, sex, recess, restriction of electronic products usage from parents, and heredity(OR=0.93, 1.29, 0.90, 0.82, 3.12, 1.61, 1.64, P<0.05). Among them, rural areas, outdoor activity during recess and restriction of electronic product usage from parents was associated with lower risk of myopia, in contrast, being girl and parental myopia was associated with higher risk of myopia.
Conclusion
The incidence of myopia among primary school students in Liaoning Province is relatively high, exterting high pressure on prevention and control. In order to prevent myopia in primary school students, Composite interventions should be developed including change students’ lear-ning style, eliminate unhealthy living habits and targeted propaganda and education.
6.Prevalence of depression and its influencing factors among middle school students in Liaoning Province

Objective:
To investigate the prevalence of depression of middle school students in Liaoning Province in 2019 and analyze its influencing factors, to provide corresponding strategies and measures for depression prevention and control.
Methods:
By using the method of multi-stage stratified random cluster sampling, the 32 909 students of grade 1-3 of junior middle school, senior high school and vocational high school were selected from 14 cities in Liaoning province, and were investigated with Center for Epidermiologic Studies Depression Scale (CES-D) psychological scale.
Results:
The depression score of middle school students in Liaoning Province was (12.66±9.53), and the incidence of depression was 19.5%. Analysis of variance and multiple regression analysis showed that the main factors influencing the occurrence of depression were grade, bullying on campus, internet addiction, being beaten and scolded by parents, drinking, gender, Leave of absence from school, smoking, serious injury and economic classification (P<0.05).
Conclusion
Depression of middle school students in Liaoning Province is not optimistic. Effective prevention and control measures should be developed and implemented to the occurrence of adverse factors, so as to reduce the occurrence of depression symptoms.
7.A survey of nurses' knowledge and attitude towards cancer-related fatigue

Objective Cancer-related fatigue (CRF) is a key in the management of cancer patients' clinic syptoms.This article investigated the status quo of nurses' knowledge and attitude towards CFR.Methods THe method of cross-sectional survey and questionnaire was used to investigate the knowledge and attitude towards CRF among nurses from related departments of three Grade III hospitals in Nanjing.Results 142 nurses answered the questionnaire.The average correct rate was 76.25%, among which nurses from the oncology department had better congition rate than nurses from other medical and surgical departments (84.3%, 75.98%, 79.57%) , representing significant difference (P<0.05).64.79% of the nurses found the relatives of cancer patients and nurses often fail to understand cancer patients;complaint of fatigue, 76.76% of nurses assumed there is lack of communication in fatigue between patients and medical staff.94.36% of nurses agreed medical institutions should strengthen the management of CRF.Conclusion At present, the clinical nurses have inadequate knowledge about CRF, which should be enhanced in future work.
8.Programmed necrosis: a new target for ischemia reperfusion injury.

Recent years, the researchers have found a new type of cell death, referred to programmed necrosis or necroptosis, which involves the death receptor and the ligand binds and is initiated under the inhibition of apoptosis pathway. Programmed necrosis possesses the morphological features of typical necrosis accompanied by inflammation. The receptor interacting protein kinase 1/3(RIPK1/3) can be inhibited by the specific inhibitors, such as necrostatin-1. RIPK1/3 could regulate programmed necrosis and play a key role in the process. The significance of programmed necrosis in ischemia-reperfusion injury (IRI) has been attracted great attention at present. Simultaneously, a series of studies have found it also involves in the IRI of heart, kidney, brain and retina.

9.Quality of donated liver from citizen passed away.

International norms of organ transplantation mainly depends on the organ donation after the death of citizens. Numbers of studies on the quality of donated livers were conducted to reduce the early postoperative primary dysfunctions and relevant complications. The donations is based on donation after brain death (DBD) and donation after cardiac death (DCD). Hence, the differences in the quality of liver from DBD and DCD are the focus.

10.Genetic instability in cancer tissues analyzed by random amplified polymorphic DNA PCR.

OBJECTIVETo detect DNA and chromosomes instabilities during the progression of tumors and screen new molecular markers coupled to putative or unknown oncogenes and/or tumor suppressor genes.
METHODSFive kinds of tumors, in a total of 128 specimens, were analyzed by random amplified polymorphic DNA (RAPD) PCR. Bands representing instabilities were recovered, purified, and cloned, labeled as probes for Southern and Northern blot analysis and DNA sequencing.
RESULTSSample 5 and 3 of the gastric cancer tissues showed the highest genomic changes and the average detectability in five cancers was up to at least 40% (42.2% - 49.4%). There were significant differences in the ability of each primer to detect genomic instability, which ranged from 27% (primer 8) to 68% (primer 2). Band B is a single copy fragment, and was found to be an allelic loss in gastric and colon cancers. It is a novel sequence and was registered in GenBank with Accession Number AF151005. Further analysis revealed that it might be part of a cis-regulatory element of a new tumor suppressor gene, containing a promoter of cis-action "CACA" box, an enhancer of "GATA" family and a start codon.
CONCLUSIONSIt was impossible or difficult to get great achievements for cancer treatments with the procedure of gene therapy only to one oncogene or one tumor suppressor gene because the extensive DNA variations occurred during the progression of tumor. RAPD assay connected with other techniques was a good tool for the detection of genomic instabilities and direct screening of some new molecular markers related to tumor suppressor genes or oncogenes.
