1.Protective Effects of Ademetionine 1,4-Butanedisulfonate on Radiation Injury

Objective This study aimed to investigate the protective effects of ademetionine 1,4-butanedisulfonate (SMT) against radiation injuries.Methods ICR mice were randomly divided into 7 groups,including normal control,irradiation-only,SMT administration-only,low-,medium-and high-dosages (250,500,1 000 mg·kg-1) of SMT pre-irradiation and high-dose of SMT post-irradiation in experimental groups.Blood and immunological experiments,organs index experiment and 30-day''s survival experiment were carried out to observe the protective effects of SMT on peripheral blood and immune system,organ index and the whole body injuries.Results Compared with irradiation-only group (4.23±1.16) ×109·L-1,the number of nucleated cells in bone marrow was (11.20±4.63) ×109·L-1 in the high dose of SMT pre-irradiation.The difference between two groups was significant.Compared with irradiation-only group (19.25±9.36),the colony forming unit-spleen was (39.00±7.57) in the high-dose SMT pre-irradiation group,there was a significant difference between the two groups.The index of liver,spleen,kidney and pancreas were significantly higher than those of the irradiation-only group in SMT administration groups.The survival rate of mice treated with SMT was increased,especially for the high dose group (46% lifted) when compared with irradiation-only group.Conclusion SMT can protect mice from radiation injuries.
2.Function and safety re-evaluation of Xuebijing injection, a traditional Chinese medicine injection

Objective To evaluate the tonic and health effects of Xuebijing injection,a traditional Chinese medicine injection,by means of corn low-nutritional mice model combined with benefit-damage index-general score (BDI-GS) evaluation system under a physiological dosage.Further,to investigate the scientific and practical benefits of the evaluation system in efficacy and safety re-evaluation of clinical traditional Chinese medicine injections.Methods Healthy ICR mice during growth period were randomly divided into blank control group and low,middle and high dose groups.All the mice were fed with corn diets,and treated by intraperitoneal injection once a day.The mice in the blank control group were injected with 0.4 ml 0.9% sodium chloride injection,and the mice in the low,medium and high dose groups were injected with 0.2,0.4 and 0.6 ml Xuebijing injection,respectively,for seven days.On the 8th day,the mice were killed,dissected,and took nine kinds of organs including heart,lung,thymus,spleen,pancreas,kidney,gonad and femur.The weight BDI,index BDI and the corresponding weight BDI general score (GSw) and index BDI general score (GSI) were calculated,and the serum biochemical parameters were measured.Results Under the physiological dose,Xuebijing injection shows good safety of continuous infusion,tonic and health effects,and slight damages on the rest of organs.More specifically,the values of weight BDI and index BDI were all higher than 9.0,and values of GSw and GSI were all higher than the basic value of 9.0.Furthermore,these parameters increased in a dose-dependent manner.In addition,the total protein level of (47.6 ± 4.30) g/L in the high dose groups was significantly higher than (43.18 ± 2.60) g/L in the blank control group (P<0.05).Conclusions The Xuebijing injection has a lower risk for conventional usage.This injection is safe under the physiological dosage and much safer in higher dosage designed in this study.The low-nutritional model combined with BDI-GS evaluation system can be used for re-evaluating the function and safety of clinical traditional Chinese medicine injections.
3.The effects of amelogenin on the migration,adhesion and proliferation of periodontal ligament stem cells

Objective:To evaluate the effects of amelogenin(AML)on the migration,adhesion and proliferation of periodontal liga-ment stem cells(PDLSCs).Methods:PDLSCs were cultured with AML at 0.25,50 amd 100 μg/ml respectively.The migration, adhesion and proliferation of the cells were examined by wound healing migration assay,transwell migration assay,attachment assay, MTT assay and cell counting,respectively.Results:AML induced the migration of PDLSCs in a dose-dependent manner(P <0.05), increased the adhesion and proliferation of PDLSCs(P <0.05).Conclusion:AML may promote the migration,adhesion and prolifer-ation of PDLSCs.
4.Computational Pharmacology of Xianhe Yanling Recipe in Anti-tumor Activity

Objective To explore the antitumor mechanism of xianhe yanling recipe( XHYLR ). Methods The mechanism of action of 241 compounds contained in the Chinese medicine, XHYLR was studied by using computational pharmacological. Methods including molecular docking,biotechnology network,chemical space technology and the prediction of absorption,distribution,metabolism,excretion and toxicity( ADME/T). Results Most of the compounds in XHYLR had good drug-like properties. Distribution of 241 compounds in chemical space also revealed that these compounds showed similar anti-tumor activity. It suggested that the prescription of XHYLR was reasonable. The phenomena that one molecular interacted with multiple target proteins and several molecules interact with one target protein were observed in XHYLR. For example,the COX-2 interacted with 40 micromolecules,and catechin interacted with five targets including COX-2. Conclusion These results are helpful for understanding the complicated mechanism of anti-tumor activity of XHYLR.
5.Autologous bone marrow stem cell mobilization transplantation versus surgical transplantation for treatment of spinal cord injury

BACKGROUND:The transplanted bone marrow stem cells (BMSCs) survive,migrate to the injury site and differentiate into neurons and astrocytes.Transplantation of BMSCs following spinal cord injury (SCI) may improve the recovery of spinal function and may be an effect way for treating spinal cord injury.The treatment of spinal cord injury by autologous BMSCs mobilization and surgical transplantation has a wide prospect of clinic application.However,it is still unclear whether outcomes and mechanisms of them are different.OBJECTIVE:To compare the efficacy of treating SCI in rats by mobilization and surgical transplantation of autologous BMSCs,and assess the two methods by qualitative indexes.DESIGN,TIME AND SETTING:A randomized controlled animal experiment was conducted at the Henan Province People's Hospital from June 2007 to April 2008.MATERIALS:A total of 90 healthy Sprague Dawley rats aged 10 wees old,half male and half female,weighing (240±10) g,were subjected to make spinal cord injury models.MATHODS:After 3 days injection with bromodeoxyuridine 50 mg/kg per day,BMSCs were isolated from bone marrow of healthy rats.All the rats were underwent spinal cord injury by NYU impactor.All 90 rats were randomly divided into 3 groups,with 30 for each group.Rats in the mobilization group were subcutaneously injected with grenulocyte-colony stimulating factor for 7 days,20 mg/kg per day.In surgical transplantation group,a total of 0.3 mL (1×10~(10)/L) BMSCs were transplanted into injured area of spinal cord.Rats in control group were given the same volume of saline (0.3 mL) into injured area of spinal cord.All the rats were injected with 50 mg/kg bromodeoxyuridine daily at day 3 before surgery for 10 days in each group.MAIN OUTCOME MEASURES:The Basso-Beattie-Brasnahan (BBB) locomotor score was used to evaluate functional recovery in rat hindlima at 3 days,1,2,4 and 8 weeks after SCI.Motion evoked potential (MEP) and somatosensory evoked potential (SEP) tests were performed to defect the neural pathway so as to evaluate recovery of injured spinal cord.The cell structural changes and the expression and distribution of Brdu,glial fibrillary acidic protein (GFAP),and neuron specific enolase (NSE) were observed by pathological and immunohistochemical methods.and 8 weeks after injury (P < 0.05),but there was no significant difference between the mobilization group and surgical was increased in the mobilization group and surgical transplantation group compared with the control group (P < 0.05-0.01),but Histopathology showed that there were less empty,necrosis and GFAP-positive glial scar tissue,more Brdu-positive cells and NSE-positive cells in the mobilization group and surgical transplantation group than in the control group.CONCLUSION:Autologous BMSCs mobilization transplantation and surgery transplantation could significantly reduce the degree of SCI,and promote the recovery of the spinal cord function after injury for treating SCI.Compared between the two,mobilization of autologous BMSCs are more convenient,non-invasive,more likely to seize the opportunity to treatment.
6.Determination of the Related Substances in Aosaibos Crude Drug by HPLC

OBJECTIVE: To determine the contents of the related substances in Aosaibos crude drug by HPLC.METHODS: The determination was carried out on a SunFireTM RP C18 column with mobile phase consisted of phosphate buffer solution(pH 3.05)-acetonitrile(8∶2) at a flow rate of 1 mL?min-1.The detection wavelength was set at 220 nm;The sample size was 20 ?L and column temperature was set at room temperature.RESULTS: The main peak and the impurity peaks of 3 batchs of Aosaibos crude drug were well-separated.The contents of the related substances were all lower than 1.19%.CONCLUSION: The determination method is simple,sensitive and applicable for the content determination of the related substances of Aosaibos.
7.A Research on Dactylogram of She Nationality Juveniles

Objective To find out the distribution of the normal rating dactylogram of the juveniles of the She Nationality. Methods According to living body measure need, Prussian blue was used. Result Each type of dactylogram in number was as follows: LU>WS>WD>AT>AS. Ridgs count of each finger in quantitative order:thumb>ring finger>middle finger>index finger>little finger. Conclusion The rate of appearance of normal finger prints in various nationalities and regions has obvious difference, the normal rating of the dactylogram of the juveniles of the She Nationality differs from that of the local juveniles of the Han nationality.
8.Identification of the Fruit of Terminalia chebula from Its Comfusable Varieties by RAPD Analysis

Different varieties of Terminalia chebula Retz. were identified by DNA finger prints, andat the same time, some problems encountered during the application of RAPD for the identification ofTCM were studied. The polymorphic features of DNA finger prints of different species were compared.Results of the study showed that they were differently related in heredity which can provide a basis for thedifferentiation of one species from the other. Means for the solution of problems arised during RAPD ap-plication were suggested. It was concluded that RAPD is an effective method to identify T. chebula from itsconfusable varieties.
9.Study on Quality Standard of Jinyou Tankeqing Tablets

Objective To study the quality standard of Jinyou Tankeqing Tablets (JTT). Methods Total flavonoids in JTT were identified by TLC. Naringin content in JTT was determined by HPLC. Results The relevant spots of total flavonoids in JTT can be identified by TLC. The content of naringin in JTT can be determined by HPLC. The linearity of naringin was good in the range of 2~20 ?g (r= 0.999 9). The average recovery of naringin was 101.29 %with RSD = 2.58 %. Conclusion The established quality standard is simple,feasible and repeatable,and can be used for the quality supervisory of JTT.
10.Studies on Anti-inflammatory Mechanism of Sinomenine

【Objective】The effect of sinomenine on human cyclo-oxygenase(COX) and interleukin-1?(IL-1?) was observed to explore the anti-inflammatory mechanism of sinomenine.【Methods】Peripheral blood monocytes(PBMC) were isolated and incubated with sinomenine.After incubation,total RNA from PBMC was extracted and the effect of sinomenine on COX and IL-1? expression with or without the stimulation of lipopolysaccharide(LPS) was observed by the method of semi-quantitative reverse transcription polymerase chain reaction(RT-PCR).【Results】Sinomenine had no effect on COX-1 gene expression,had a slight inhibitory effect on COX-2 gene expression and had a strong inhibitory effect on IL-1? gene expression.【Conclusion】 The anti-inflammatory mechanism may be related to the inhibition of inflammatory cytokines.
