Hemophilia B is a genetic disorder caused by coagulation factor Ⅸ(FⅨ) deficiency, mainly manifesting as joint, muscle and deep tissue bleeding. Hemophilia pseudotumor is a mass formed by soft tissue liquefaction and necrosis caused by repeated bleeding. Most pseudotumors occur in the bone and muscle. We report a case of hemophilia B with pseudotumor formation in the pelvis and abdomen, where lesion location is relatively rare. After active and effective hemostasis, the patient's hematuria symptom gradually improved. This case suggests that early and timely hemostatic treatment is crucial for patients with hemophilia.

OBJECTIVES:
To investigate the effect of icariin (ICA) on early β-defensin-2 and T cell subsets in rats after tracheotomy.
METHODS:
A total of 54 SPF male Sprague-Dawley rats were randomly divided into a normal control group (group A), a model group (group B), and a model+ICA treatment group (group C), with 18 rats in each group. A tracheotomy intubation model of the B and C group was prepared. After 6 h of surgery, ICA intervention was given to group C. Groups A and B were given the same amount of normal saline. Lung tissue, alveolar lavage fluid and peripheral blood were taken at 24 h, 72 h and 168 h, respectively. The expression of rat β-defensin-2 mRNA in lung tissue was detected by RT-PCR. The content of β-defensin-2 in alveolar lavage fluid and peripheral blood serum was detected by ELISA. The content of peripheral blood T cell subsets (CD3, CD4, CD8) was detected by flow cytometry, and the ratio of CD4/CD8 was calculated.
RESULTS:
After tracheotomy, the levels of β-defensin-2 mRNA and β-defensin-2 in lung tissue from the group B were increased significantly at 24 h, then they were decreased gradually, and decreased most significantly at 168 h (<0.05). The content of β-defensin-2 in peripheral blood of group B decreased gradually, and the content of β-defensin-2 in 168 h was significantly lower than that in 24 h (<0.05), but there was no significant difference between group B and group A (>0.05). The level of CD3 T cells in peripheral blood was significantly lower than that in the group A (<0.05), but their was no significant difference in CD4 and CD8 T cells compared with group A (>0.05). After ICA intervention in group C: lung tissue, alveolar lavage fluid, peripheral blood serum β-defensin-2 content, and peripheral blood CD3 and CD4 T cell levels were gradually increased, significantly higher than those in the group B (<0.05). CD8 T cell level was significantly lower than that in the group A at 24 h (<0.05), the CD4/CD8 ratio was significantly higher at 168 h than those in the group A or B (both <0.01).
CONCLUSIONS
ICA can improve the early lung immune function in rats with tracheotomy, which might be related to up-regulation of β-defensin-2 in lung tissue and alveolar lavage fluid, concomitant with increases in CD3 and CD4 T cells and CD4/CD8 ratio in peripheral blood while reduction in CD8 cells.

Aging increases the risk of various diseases. The main goal of aging research is to find therapies that attenuate aging and alleviate aging-related diseases. In this study, we screened a natural product library for geroprotective compounds using Werner syndrome (WS) human mesenchymal stem cells (hMSCs), a premature aging model that we recently established. Ten candidate compounds were identified and quercetin was investigated in detail due to its leading effects. Mechanistic studies revealed that quercetin alleviated senescence via the enhancement of cell proliferation and restoration of heterochromatin architecture in WS hMSCs. RNA-sequencing analysis revealed the transcriptional commonalities and differences in the geroprotective effects by quercetin and Vitamin C. Besides WS hMSCs, quercetin also attenuated cellular senescence in Hutchinson-Gilford progeria syndrome (HGPS) and physiological-aging hMSCs. Taken together, our study identifies quercetin as a geroprotective agent against accelerated and natural aging in hMSCs, providing a potential therapeutic intervention for treating age-associated disorders.

Objective To investigate the current situation of maternal breastfeeding after the childbirth in Shanghai and analyze its influencing factors, so as to improve breastfeeding. Methods In this study, longitudinal follow-up and questionnaire designed based on the hospital were carried out on pregnant women at the outpatient department in eight baby-friendly hospitals of Shanghai from October 2015 to January 2016. The questionnaires included demographic characteristics and their prospective feeding method after the childbirth. Pregnant women were followed up until the delivery was finished. At the time of hospitalization discharge, they were given the 2nd questionnaire containing the basic situation after the delivery and the actual breastfeeding behavior situation. Results The rates of pregnant women with the intention of breastfeeding and exclusive breastfeeding during the third trimester were 82.5% and 82.3% respectively. After the delivery,56.4% (399/707) of mothers were breastfeeding. Binary logistic regression analysis showed that, for primipara group, breastfeeding behavior after the delivery was related to five factors:the breastfeeding intention in third trimester, age, household register, the time of onset of lactation and whether the baby was in the care unit;for multiparas group, breastfeeding behavior was related to two factors:the time of onset of lactation and whether the baby was in the care unit. Conclusions Health care workers should attach importance to the promotion of breastfeeding during pregnancy for primipara group and pay attention to the health education of the mothers with registeredpermanent residence of Shanghai. During the hospitalization, health care workers should strictly implement the "early sucking, early contact" as the baby-friendly hospital required, strengthen the breastfeeding knowledge and skill training for mothers whose babies are in the maternal care unit, ensure the normal lactation of mothers without their babies so as to improve the early breast-feeding rate after the delivery.
