1.Therapeutic effect and mechanism of the topical preparation of baicalein on atopic dermatitis

To evaluate the therapeutic effect of baicalein topical preparation on atopic dermatitis, we first constructed two atopic dermatitis-like mouse models induced by calcipotriol and 1-fluoro-2,4-dinitrobenzene to assess their therapeutic effect with skin tissue staining and other experiments. It was found that topical preparation of baicalein could alleviate epidermal thickening of diseased skin tissues, repair damaged skin barrier proteins, and inhibit T helper 2 cells (Th2) infiltration and mast cell infiltration and activation in lesional sites. Cyberpharmacology was utilized to analyze whether baicalein could treat atopic dermatitis by interfering with multiple pathogenesis-associated pathways. Results indicated that baicalein reduced the mRNA levels of inflammatory factors and inhibited the phosphorylation of NF-κB p65 and STAT1 proteins in keratinocyte cells. Together, the topical preparation of baicalein may be effective in alleviating atopic dermatitis-like symptoms in mice by down-regulating the phosphorylation level of NF-κB in keratinocytes, thereby decreasing the expression of inflammatory factors in keratinocytes, which provides an idea and a theoretical basis for the topical preparation of baicalein for the treatment of inflammatory skin diseases such as atopic dermatitis.

The accumulation of fatigue during military operations may lead to decreased operational efficiency and non-combat attrition,which can impact combat effectiveness.On-site monitoring and evaluation of fatigue during military operations,as an important means to keep track of military operations and bring about quick changes in training,underlie the combat effectiveness of military personnel.Focusing on the on-site monitoring and evaluation methods of fatigue during military operations,this paper reviews the determinants of such fatigue as well as on-site monitoring and comprehensive evaluation methods so as to provide reference for accurate and efficient evaluation of fatigue during military operations and for early warning of such fatigue.
3.Study on reverse transsynaptic virus tracing targeting Shenmen (HT7) and heart in mice

Objective: To investigate the neural connections between Shenmen (HT7)-heart and the brain by observing the tracing viruses co-labeled brain nuclear groups after injection of the pseudorabies viruses (PRV), the reverse transsynaptic virus tracer carrying different fluorescent protein genes, into the myocardium and Shenmen (HT7) point, respectively.Methods: Pseudorabies virus 531 (PRV531) carrying the green fluorescent protein gene and pseudorabies virus 724 (PRV724) carrying the red fluorescent protein gene were injected into the left ventricular wall and Shenmen (HT7) point area of the left forelimb of six C57BL/6 mice, respectively. After 120 h, whole brain tissue was extracted under 4% paraformaldehyde perfusion to prepare brain sections. Neuronal co-labeling with the tracing viruses was observed under fluorescence microscopy. Results: Co-labeled signals from the mouse ventricular wall and Shenmen (HT7) point region were found at all levels of the mouse central nervous areas, such as the cerebral cortex, hypothalamus, midbrain, pons, and medulla oblongata. The number of co-labeled neurons was higher in the primary motor area, the hypothalamic paraventricular nucleus, the subceruleus nucleus, and the paramedian reticular nucleus. Conclusion: There is a neural connection between Shenmen (HT7), the heart, and the brain, which may be most closely related to the autonomic nervous system.
4.Ribitol⁃5⁃phosphate transferase fukutin inhibits the secretion of α⁃dystroglycan in HeLa cells

Objective :
To investigate the effects of fukutin ( FKTN) which is a member of ribitol⁃5⁃phosphate transferases on HeLa cell cycle, apoptosis, migration and the secretion of α⁃dystroglycan(α⁃DG) .
Methods :
The plasmid pcDNA3. 1⁃α⁃DAG⁃HA was constructed. HeLa cells were transfected with plasmid pcDNA3. 1⁃FKTN ⁃ 3xFlag, then the total protein was extracted for Western blot to determine the expression level of FKTN. Cell cycle and apoptosis were measured by flow cytometry after the overexpression of FKTN. Following the overexpression of FKTN, wound⁃healing assay was performed to detect the cell migration rate, as the same time wheat germ agglutinin⁃agarose (WGA⁃agarose) was used to enrich α⁃DG in the cell cultures, then α⁃DG was detected by Western blot.
Results :
The eukaryotic expression plasmid pcDNA3. 1⁃α⁃DAG⁃HA was constructed successfully. FKTN could be overexpressed in HeLa cells. After the overexpression of FKTN,the percentage of S phase of cell cycle in the experimental group decreased (P < 0. 001) and apoptosis rate unchanged (P > 0. 05) when compared with the control group. There was no change in the cell migration rate of experimental group (P > 0. 05), but after the overexpression of FKTN, secretion of α⁃DG decreased when compared with control group.
Conclusion
Overexpressing FKTN arrests cell cycle and inhibits the secretion of α⁃DG in HeLa cells. Apoptosis and cell migration of HeLa cells are not affected by the overexpression of FKTN.
5.Effects of p100 Rb and p80 Rb overexpression on cell cycle and apoptosis of HEK 293T cells

Objective :
To investigate the effects of p100Rband p80Rbtruncated forms of retinoblastoma protein(Rb) on HEK 293T cell cycle and apoptosis.
Methods :
The eukaryotic expression plasmid pcDNA3. 1-p100Rb-FLAG and pcDNA3. 1-p80Rb-FLAG were constructed,plasmid was transfected into U2OS cells and immunofluorescence was performed to define the localization of p100Rband p80Rbin cells. Plasmid was also transfected into HEK 293T cells,and the total protein was extracted for Western blotting to detect the protein expression level. After overexpression of Rb,p100Rband p80Rbin HEK 293T cells,cell cycle and apoptosis were measured by flow cytometry.
Results:
The eukaryotic expression plasmid pcDNA3. 1-p100Rb-FLAG and pcDNA3. 1-p80Rb-FLAG were constructed successfully. p100Rbwas mainly located in the nucleus while p80Rbwas mainly located in the cytoplasm,in U2OS cells. Rb,p100Rband p80Rbcould be overexpressed in HEK 293T cells. Following overexpression of Rb,p100Rbin HEK 293T cells,the percentage of G1phase of cell cycle significantly increased compared with the control group,and p100Rband p80Rbinhibited apoptosis compared with control group(P<0. 05).
Conclusion
Overexpression of p100rb can inhibit the apoptosis of HEK 293T cells and block the cell cycle. Overexpression of p80rb can inhibit the apoptosis of HEK 293T cells,but has no effect on cell cycle.
6.The interaction of RACK1 with CLIC1 in cells

Objective :
To investigate the interaction between intracellular chloride ion protein 1(CLIC1) and activated protein kinase C receptor 1 ( RACK1) .
Methods :
Plasmids pcDNA3. 1⁃RACK1⁃HA and/or pcDNA3. 1 ⁃CLIC1⁃FLAG were transfected into HEK 293T cells, and pcDNA3. 1⁃RACK1⁃HA and/or pcDNA3. 1⁃CLIC1⁃FLAG were transfected into COS7. GST⁃pulldown and immunoprecipitation assays were performed to determine the interaction between CLIC1 and RACK1 in vivo and in vitro. The co⁃localization of CLIC1 and RACK1 was observed by indirect immunofluorescence assay.
Results :
Western blot confirmed that CLIC1 and RACK1 could be highly expressed in HEK 293T cells. GST⁃pulldown showed that RACK1 bound CLIC1 in vitro, and immunoprecipitation showed that CLIC1 and RACK1 interacted in vivo. Furthermore, indirect immunofluorescence assay showed CLIC1 co⁃localized with RACK1 .
Conclusion
Human CLIC1 protein interacts with RACK1 in vitro and in vivo.
7.Study on the mechanism of Dahuangfuzi decoction in improving intestinal motility disorder of rats with severe acute pancreatitis

Objective:To explore the mechanism of Dahuangfuzi decoction on intestinal motility disorder by observing its effect on serum motilin, Cajal interstitial cells and motilin receptor in rats with severe acute pancreatitis (SAP).Methods:Eighteen clean male Sprague-Dawley rats were randomly divided into the control group, SAP group and Dahuangfuzi group ( n=6 each group). The SAP rat model was prepared by retrogradely injected 4% sodium taurocholate into cholangiopancreatic duct. The rats in the SAP group were given 2 mL normal saline (37℃) enema at 12 and 24 h after operation. The rats in the Dahuangfuzi group was given 2 mL Dahuangfuzi decoction (37℃) enema at 12 and 24 h respectively. For the control group, the pancreas was exposed in the same way and then the abdomen was closed. Forty-eight h after operation, the abdominal aorta blood samples were taken for determination of serum endotoxin and amylase, and for detection of serum motilin by enzyme-linked immunosorbent assay (ELISA); the pathological changes of pancreas and ileum were observed by hematoxylin-eosin (HE) staining. The expression of c-kit and motilin receptor protein in ICC in ileum tissue was detected by immunohistochemistry. Results:Compared with the control group, the levels of serum endotoxin and amylase in the SAP group were significantly higher [(504.98±88.81) pg/mL vs. (17.76±5.01) pg/mL; (532.28±66.53) vs. (69.45±3.61) U/L, P<0.05], while the levels of serum motilin were significantly lower [(195.4±6.7) ng/L vs. (301±8.10) ng/L, P<0.05], and the scores of c-kit and motilin receptor protein were decreased ( P<0.05); compared with the SAP group, the levels of serum endotoxin and amylase in the Dahuangfuzi group were significantly reduced [(189.9±38.23) pg/mL vs. (504.98±88.81) pg/mL; (294.23±25.66) vs. (532.28±66.53) U/L, P<0.05], while the levels of serum motilin were significantly increased [(264.2±8.3) ng/L vs. (195.4±6.7) ng/L, P<0.05], and the scores of c-kit and motilin receptor protein were increased ( P<0.05). Conclusions:Dahuangfuzi decoction can improve the intestinal motility of SAP rats by promoting the secretion of motilin, increasing the activity of ICC cells and the expression of motilin receptor.
8.Application of virtual reality technology in thoracoscopic anatomical segmentectomy

Objective:To explore the application value of virtual reality technology in thoracoscopic anatomical segmentectomy.Methods:Eighty-four patients with early stage non-small cell lung cancer admitted to the First Hospital of Jiaxing from December 2017 to December 2018 were enrolled in the study.They were divided into observation group and control group according to the random digital table method, with 42 cases in each group.The observation group used virtual reality technology to construct a three-dimensional digital model, and performed preoperative evaluation and simulated surgical drills and intraoperative navigation on the three-dimensional digital model, based on the preoperative evaluation and simulated surgical drill results, combined with the specific actual situation during the operation, developed and implemented individualized thoracoscopic anatomical segmentectomy.Thoracoscopy anatomical segmentectomy was routinely performed in the control group.The operation time, intraoperative blood loss, intraoperative lymph node dissection number, intraoperative lymph node dissection, postoperative hospital stay, postoperative thoracic closed drainage tube, total postoperative drainage, total hospitalization, cost and incidence of postoperative complications were compared between the two groups.Results:The operation of both two groups was successfully completed, and no intraoperative thoracic surgery was performed during the operation.There was no perioperative death.The operation time, intraoperative blood loss, intraoperative lymph node dissection number, intraoperative lymph node dissection, postoperative hospital stay, postoperative thoracic closed drainage tube, total postoperative drainage, total hospitalization cost and the incidence of postoperative complications in the observation group were (100.98±26.51)min, (67.98±32.96)mL, (7.79±1.32), (11.98±4.69), (4.60±1.43)d, (2.86±0.81)d, (437.14±193.86)mL, (3.76±0.31)million, 9.52%(4/42), respectively, which in the control group were (114.88±24.26)min, (104.52±52.37)mL, (6.45±0.3), (8.31±1.94), (6.50±2.55)d, (4.00±2.25)d, (667.26±415.01)mL, (4.20±0.65)million, 26.19%(11/42), respectively, the differences between the two groups were statistically significant ( t=-2.208, -3.328, 5.916, 4.678, -4.221, -3.993, -3.265, -3.968, χ 2=3.977, all P<0.05). No local recurrence or distant metastasis was found during the follow-up period. Conclusion:Virtual reality technology can provide preoperative evaluation and simulated surgical exercises and intraoperative navigation for thoracoscopic anatomical segmentectomy, which can reduce the difficulty of surgery and improve the accuracy and safety of the operation.
9.Study on the doctor-patient interest demands satisfaction of the payment system reform in China's new rural cooperative medical care scheme

Objective To study doctor-patient interest demands satisfaction and its influencing factors of the payment system reform of the new rural cooperative medical care scheme to provide reference for the reform. Methods Cross-sectional survey was conducted from September 2016 to February 2017. Multi-stage stratified random sampling was used in six counties of three provinces in the eastern, middle and western regions of China, and mathematical statistics was applied to analyze the data. Results The doctor-patient overall interest demands satisfaction was high, but the satisfaction was lower both with the income and ability improvement of medical staff and with the benefits of farmers. The influencing factors of the satisfaction of managers in medical institutions included the type of payment, educational level and work unit (P<0.05). The influencing factors of medical staff's satisfaction included the type of payment, work unit, and working years among others(P<0.05). The influencing factors of farmers'satisfaction included the type of payment and the average annual income, etc(P<0.05). Conclusions The core interest demands of both doctors and patients should be valued to enhance their satisfaction. Diseases related groups should be promoted and applied scientifically, and appropriately integrated with other methods of payment. Both doctors and patients'understanding of the payment reform should be improved by propaganda and training, to get their support and cooperation.
10.In vivo biocompatibility of nano-hydroxyapatite/polyphenylene sulfide composites

BACKGROUND:There is a lack of the research concerning the biocompatibility of nano-hydroxyapatite/polyphenylene sulfide (nHA/PPS) composites.OBJECTIVE:To evaluate the in vivo biocompatibility of nHA/PPS composites based on the completed research in vitro.METHODS:Systemic toxicity test:Sprague-Dawley rats were given the intraperitoneal injection of nHA/PPS extract or normal saline.The general situation,body mass and the histological changes of the liver and kidney were observed at 72 hours after injection.Delayed type hypersensitivity test:nHA/PPS extract or normal saline was injected subcutaneously into the back of the rats.Afterwards,skin irritation symptoms were observed at 72 hours.Local reaction experiment:nHA/PPS composites and polyethylene were respectively implanted into the back of the rats.The pathological changes of the implanted materials and their surrounding tissues were observed at 15 and 30 days after implantation.RESULTS AND CONCLUSION:(1) The rats were in good situation after nHA/PPS injection;the body mass increased steadily,which showed no significant difference from the control group (P < 0.05);the morphology and color of the liver and kidney were normal,and the systemic toxicity of the composite materials was normal according to the degree of toxicity classification.(2) There were no obvious skin irritation symptoms after the subcutaneous injection of nHA/PPS composites,and the primary irritation index was less than 0.4,suggesting a low hypersensitivity.After implantation of nHA/PPS composites,there was no obvious degradation,absorption and rejection,and both the degree of inflammatory reaction (15 days ≤ level Ⅲ,30 days ≤ level Ⅱ) and the thickness of fibrous capsule (15 days ≤ level Ⅲ,30 days ≤ level Ⅱ) revealed the good biocompatibility of the composites.These results suggest that the nHA/PPS composites hold an excellent biocompatibility in vivo.
