Chinese Journal of Biologicals 2025;38(04):407-412
Objective To compare the immune responses in lung tissue of K18-hACE2 transgenic mice infected with SARS-CoV-2 prototype strain Prototype and its variants Delta and Omicron BF.7, so as to provide experimental evidence for the research and development of related drugs. Methods K18-hACE2 transgenic mice were infected with Prototype, Delta or Omicron BF.7 by nasal drip. On the 7th day after infection, the lung tissues of mice were taken aseptically for subsequent analyses. Viral RNA copy numbers and cytokine gene expression levels in the lung tissues were quantified by qPCR. Histopathological examination of lung tissue damage was performed using HE staining. Results The copy number of viral nucleic acid in lung tissue of mice in Delta group was significantly higher than that in Prototype and Omicron BF.7 groups(t = 5. 419-6. 908, each P < 0. 01), the expression levels of interferon γ(IFN γ)and interleukin-13(IL-13)in lung tissue were significantly higher than those in Prototype and Omicron BF.7 groups(t = 3. 990-7. 282, each P < 0. 05), and the expression levels of tumor necrosis factor-α(TNF-α),and IL-5 were significantly higher than those in Prototype group(t = 7. 108 and 2. 908,each P < 0. 05). In addition, obvious pathological changes related to virus infection were observed in lung tissue sections of mice in three groups. Conclusion Delta variant strain induced the strongest inflammatory response in lung tissue of K18-hACE2 transgenic mice with the highest pathogenicity, followed by prototype strain and Omicron BF.7 variant strain.

OBJECTIVE To investigate the effect of salidroside (Sal) on bone loss in obstructive sleep apnea syndrome(OSAS) rats by regulating the osteoprotegerin(OPG)/receptor activator of nuclear factor kappa-B ligand(RANKL) pathway. METHODS Rats were randomly divided into(12 rats/group) control group,OSAS group,Sal-L,Sal-M,and Sal-H groups(17.5,35,70 mg/kg). Except for the control group, all other groups were used to replicate the OSAS rat model through hypoxia and reoxygenation cycles. Bone density meters,three-point bending experiments, and Micro CT were applied to measure the bone density, biomechanics, and microstructural changes of the femur in rats. ELISA method was applied to detect serum levels of osteocalcin(BGP),alkaline phosphatase(ALP),and cross-linked carboxy-terminal telopeptide of type Ⅰ collagen(CTX-I). RT-PCR was applied to detect OPG and RANKL mRNA levels in the femur. Western blotting was applied to detect the expression of OPG/RANKL pathway proteins in the femur. RESULTS Compared with the control group, the bone density, maximum intensity, maximum load, trabecular bone volume fraction(Tb.BV/TV),trabecular number(Tb.N),trabecular thickness(Tb.Th),BGP,ALP,OPG mRNA and protein expression,OPG/RANKL ratio of rats in the OSAS group were decreased, the mRNA and protein expression of CTX-I and RANKL were increased(P<0.05). Compared with the OSAS group, the bone density, maximum intensity, maximum load,Tb.BV/TV,Tb.N,Tb.Th,BGP,ALP,OPG mRNA and protein expression,OPG/RANKL ratio of rats in the Sal-L,Sal-M,and Sal-H groups were increased sequentially, the mRNA and protein expression of CTX-I and RANKL were decreased sequentially, the above changes were most great in the Sal-H group(P<0.05). CONCLUSION Salidroside promotes bone formation and inhibits bone resorption by increasing OPG expression and decreasing RANKL expression, thereby reducing bone loss in OSAS rats.

Objective:To explore the relationship between the expression of long chain non coding ribonucleic acid (LncRNA) small nucleolar RNA host gene 1 (LncRNA SNHG1) and microRNA (miR)-143-3p in nasopharyngeal squamous cell carcinoma (HSCC) tissue and clinical pathological features and prognosis. Methods:A prospective selection was made from 97 HSCC patients admitted to the Handan Central Hospital from March 2016 to March 2018. Surgical resection of HSCC tissue and normal mucosa tissue adjacent to cancer were taken, and real-time fluorescence quantitative polymerase chain reaction (qRT-PCR) was used to detect the expression of LncRNA SNHG1 and miR-143-3p. The patient′s survival status was followed up after leaving the hospital. We compared the differences in the expression of LncRNA SNlHG1 and miR-143-3p in HSCC tissues with different clinical pathological parameters, analyzed the correlation between LncRNA SNHG1 and miR-143-3p expression, and the relationship between LncRNA SNHG1 and miR-143-3p expression and the prognosis of HSCC patients. Results:The expression of LncRNA SNHG1 in HSCC tissue was higher than that in normal mucosa tissue adjacent to cancer ( P<0.05), and the expression of miR-143-3p was lower than that in normal mucosa tissue adjacent to cancer ( P<0.05). The expression of LncRNA SNHG1 in cancer tissues of HSCC patients with tumor node metastasis (TNM) stage Ⅲ, low to medium differentiation, and lymph node metastasis was higher than that of HSCC patients with TNM stage Ⅰ-Ⅱ, high differentiation, and no lymph node metastasis (all P<0.05), and the expression of miR-143-3p was lower than that of HSCC patients with TNM stage Ⅰ-Ⅱ, high differentiation, and no lymph node metastasis (all P<0.05). The expression of LncRNA SNHG1 in HSCC tissues is negatively correlated with the expression of miR-143-3p ( r=-0.522, P<0.05). The 5-year cumulative survival rate of HSCC patients with high expression of LncRNA SNHG1 was lower than that of HSCC patients with low expression of LncRNA SNHG1 ( P<0.05), and the 5-year cumulative survival rate of HSCC patients with low expression of miR-143-3p was lower than that of HSCC patients with high expression of miR-143-3p ( P<0.05). Multivariate Cox regression analysis showed that TNM stage Ⅲ and high expression of LncRNA SNHG1 were risk factors for poor prognosis in HSCC patients (all P<0.05), while high expression of miR-143-3p was a protective factor ( P<0.05). Conclusions:The expression of LncRNA SNHG1 is upregulated and miR-143-3p is downregulated in HSCC tissues, with a negative correlation between the two, which is related to the malignant pathological characteristics and poor prognosis of HSCC.
