1.Effect of cancer nodules on liver metastases after radical resection of colorectal cancer

		                        			Objective:To investigate the effect of cancer nodules on postoperative liver metastasis in patients undergoing radical resection of colorectal cancer.Methods:The clinicopathological data of 196 patients undergoing radical operation for colorectal cancer admitted to the Dongnan Hospital of Xiamen University from January 2019 to June 2020 were retrospectively analyzed. According to the pathological results, they were divided into cancer nodule group and non-cancer nodule group. Time and the number of liver metastasis during the follow-up period were analyzed. The relationship between cancer nodules and clinicopathological features was analyzed, and the influencing factors of postoperative liver metastasis in patients with colorectal cancer were analyzed by univariate and logistic multivaiate analysis. The receiver operator characteristic (ROC) curve was drawn to analyze the predictive effect of cancer nodule diameter on postoperative liver metastasis of colorectal cancer.Results:There were 50 patients accompanied by cancer nodules in 196 patients with colorectal cancer, the incidence rate was 25.5%, the diameter of cancer nodules was 0.4-1.8 cm, the median diameter was 1.0 cm, and there were 46 patients with liver metastasis, the incidence rate was 23.5%. There were statistically significant differences in carbohydrate antigen CA19-9 ( χ2=7.55, P=0.006), maximum tumor diameter ( χ2=12.23, P<0.001), T stage ( χ2=15.79, P<0.001), vascular thrombus ( χ2=12.79, P<0.001), lymph node metastasis ( χ2=16.18, P<0.001) between the cancer nodule group and the non-cancer nodule group. Univariate analysis showed that CA19-9 ( χ2=7.40, P=0.007), maximum tumor diameter ( χ2=8.23, P=0.004), T stage ( χ2=4.58, P=0.032), vascular cancer thrombus ( χ2=12.97, P<0.001), lymph node metastasis ( χ2=3.96, P=0.047), cancer nodules ( χ2=67.60, P<0.001), cancer nodule diameter ( t=4.28, P<0.001), and the number of cancer nodules ( χ2=68.77, P<0.001) were all influential factors for postoperative liver metastasis in colorectal cancer patients. Multivariate analysis showed that stage T 3-4 ( OR=4.62, 95% CI: 1.51-7.35, P=0.003), vascular cancer thrombus ( OR=5.42, 95% CI: 1.75-12.85, P<0.001), accompanied by cancer nodules ( OR=18.54, 95% CI: 7.45-39.73, P<0.001), the diameter of cancer nodules ( OR=13.20, 95% CI: 4.74-29.64, P<0.001), number of nodules [one cancer nodule ( OR=9.11, 95% CI: 2.24-27.34, P<0.001), two cancer nodules ( OR=6.36, 95% CI: 1.38-9.08, P<0.001) and three cancer nodules ( OR=5.00, 95% CI: 1.98-8.84, P<0.001) ] were independent influencing factors for postoperative liver metastasis in patients with colorectal cancer. ROC curve analysis showed that the best threshold of the diameter of cancer nodules for predicting liver metastasis was 1.1 cm [area under the curve (AUC) =0.764, 95% CI: 0.632-0.896, P=0.002). The incidence of liver metastasis in patients with cancer nodules diameter≥1.1 cm was 95.65% (22/23), higher than that in patients with cancer nodules diameter<1.1 cm (40.74%, 11/27), with a statistically significant difference ( χ2=16.69, P<0.001) . Conclusion:Patients with colorectal cancer with cancer nodules≥1.1 cm in diameter should be vigilant against postoperative liver metastasis.
		                        		
		                        		
		                        		
		                        	
2.Mechanism of miR-26a-5p/cAMP response element binding protein 1 molecular axis regulating osteogenic differentiation of adipose-derived mesenchymal stem cells.

		                        			OBJECTIVE:
		                        			To investigate the regulatory effects of miR-26a-5p on the osteogenic differentiation of adipose-derived mesenchymal stem cells (ADSCs) by regulating cAMP response element binding protein 1 (CREB1).
		                        		
		                        			METHODS:
		                        			The adipose tissues of four 3-4 weeks old female C57BL/6 mice were collected and the cells were isolated and cultured by digestion separation method. After morphological observation and identification by flow cytometry, the 3rd-generation cells were subjected to osteogenic differentiation induction. At 0, 3, 7, and 14 days after osteogenic differentiation induction, the calcium deposition was observed by alizarin red staining, ALP activity was detected, miR- 26a-5p and CREB1 mRNA expressions were examined by real-time fluorescence quantitative PCR, and CREB1 protein and its phosphorylation (phospho-CREB1, p-CREB1) level were measured by Western blot. After the binding sites between miR-26a-5p and CREB1 was predicted by the starBase database, HEK-293T cells were used to conduct a dual-luciferase reporter gene experiment to verify the targeting relationship (represented as luciferase activity after 48 hours of culture). Finally, miR-26a-p inhibitor (experimental group) and the corresponding negative control (control group) were transfected into ADSCs. Alizarin red staining, ALP activity, real-time fluorescent quantitative PCR (miR-26a-5p) and Western blot [CREB1, p-CREB1, Runt-related transcription factor 2 (RUNX2), and osteocalcin (OCN)] were performed at 7 and 14 days after osteogenic induction culture.
		                        		
		                        			RESULTS:
		                        			The cultured cells were identified as ADSCs. With the prolongation of osteogenic induction culture, the number of calcified nodules and ALP activity significantly increased ( P<0.05). The relative expression of miR-26a-5p in the cells gradually decreased, while the relative expressions of CREB1 mRNA and protein, as well as the relative expression of p-CREB1 protein were increased. The differences were significant between 7, 14 days and 0 day ( P<0.05). There was no significant difference in p-CREB1/CREB1 between different time points ( P>0.05). The starBase database predicted that miR-26a-5p and CREB1 had targeted binding sequences, and the dual-luciferase reporter gene experiment revealed that overexpression of miR-26a-5p significantly suppressed CREB1 wild-type luciferase activity ( P<0.05). After 7 and 14 days of osteogenic induction, compared with the control group, the number of calcified nodules, ALP activity, and relative expressions of CREB1, p-CREB1, OCN, and RUNX2 proteins in the experimental group significantly increased ( P<0.05). There was no significant difference in p-CREB1/CREB1 between the two groups ( P>0.05).
		                        		
		                        			CONCLUSION

3.Effects of Longshengzhi capsule on learning and memory ability of vascular dementia model rats and study on its metabolomics

		                        			OBJECTIVE To study the effect of Longsheng zhi capsule on lear ning and memory ability of vascular dementia model rats and explore its mechanism based on metabonomics. METHODS Totally 90 SD rats were randomly divided into sham operation group ,model group ,Dihydroergotoxin mesylate tablet group (positive control ,0.54 mg/kg),Longshengzhi capsule high-dose,medium-dose and low-dose groups (2.16,1.08 and 0.54 g/kg),with 15 rats in each group. In addition to sham operation group (only threading without ligation ),the vascular dementia model was prepared by permanent ligation of bilateral common carotid arteries in each group ,which was administered by gavage for 28 d. Morris water maze test was used to determine the learning and memory ability of rats ;hematoxylin eosin (HE)staining was used to observe the histopathological changes of hippocampus;the serum levels of superoxide dismutase (SOD),malondialdehyde(MDA)and glutathione peroxidase (GSH-Px) were detected ;the serum metabolic map was analyzed by ultra high performance liquid chromatography quadrupole time of flight tandem mass spectrometry (UPLC-Q-TOF/MS),the specific metabolites were screened by multivariate statistical analysis ,and the metabolic pathway was enriched and analyzed. RESULTS Morris water maze test showed that compared with model group ,the escape latency of rats in each administration group was significantly shortened ,the number of crossing the platform was significantly increased ,and the residence time in the target quadrant was significantly prolonged (P<0.01 or P< 0.05). The results of serum biochemical indexes showed that compared with model group , the s erum level of SOD increased significantly in Dihydroergotoxine mesylate tablet group and Longshengzhi capsule high-dose group ,the serum level of GSH-Px increased significantly while the MDA level decreased significantly in each administration group (P<0.05 or P<0.01). HE staining showed that Longshengzhi capsule could improve the histopathological damage of hippocampus in vascular dementia model rats. A total of 14 differential metabolites were screened and identified by UPLC-Q/TOF-MS and orthogonal partial least squares discriminant analysis model (VIP>1 and P<0.05). The results of metabolic pathway enrichment analysis showed that the metabolic pathways involved in vascular dementia in rats mainly included vitamin B 6 metabolism,fatty acid metabolism and steroid hormone biosynthesis. CONCLUSIONS Longshengzhi capsule can improve the learning and memory ability of rats caused by vascular dementia. Its effect may be related to improving the oxidative stress injury caused by lipid accumulation in the process of vascular dementia. 		                        			Chest trauma is one of the most common injuries. Venous thromboembolism (VTE) as a common complication of chest trauma seriously affects the quality of patients′ life and even leads to death. Although there are some consensus and guidelines on the prevention and treatment of VTE at home and abroad, the current literatures lack specificity considering the diagnosis, treatment and prevention of VTE in patients with chest trauma have their own characteristics, especially for those with blunt trauma. Accordingly, China Chest Injury Research Society and editorial board of Chinese Journal of Traumatology organized relevant domestic experts to jointly formulate the Chinese expert consensus on the diagnosis, treatment and prevention of chest trauma venous thromboembolism associated with chest trauma (2022 version). This consensus provides expert recommendations of different levels as academic guidance in terms of the characteristics, clinical manifestations, risk assessment, diagnosis, treatment, and prevention of chest trauma-related VTE, so as to offer a reference for clinical application.
		                        		
		                        		
		                        		
		                        	
6.miR-219 regulates PRKCI expression in tongue squamous cell carcinoma

		                        			Objective The objective of this study were to investigate the effects of miR-219 on cell proliferation,invasion and metastasis and the correlation between PRKCI and miR-219 expression in tongue squamous cell carcinoma. Methods The lu-ciferase reporter gene assay was used to verify the predicted target gene. The expression of PRKCI in tongue squamous cell carcinoma cells overexpressed exogenous miR-219 was detected by qRT-PCR and Western blot. Finally,the reverse effects of PRKCI on the proliferation,clone formation,migration and invasion ability were examined in stable overexpressing miR-219 tongue squamous cell carcinoma(TSCC)cells by MTT assay,cell plate cloning assay,scratch assay and Transwell chamber assays. qRT-PCR assay was used to determine the expression of PRKCI gene and miR-219 in tongue squamous cell carcinoma tissues,and the relationship be-tween PRKCI and miR-219 was further analyzed. Results The bioinformatics analysis predicted that the downstream target gene of miR-219 was PRKCI. The double luciferase reporter gene assay showed that miR-219 was able to reduce the fluorescence activity of the wild type PRKCI reporter vector. In addition,qRT-PCR and Western blot also showed that miR-219 could down-regulate the expression of PRKCI in TSCC cells. MTT results showed that overexpression of PRKCI could reverse the inhibitory effect of miR-219 on the proliferation of TSCC cells,and further demonstrated that the overexpression of PRKCI could reverse the inhibitory effect of miR-219 on the proliferation, invasion and metastasis of TSCC cells by cell plate cloning, scratch and Transwell experiments. Conclusion MiR-219 plays a role in inhibiting tumor by directly targeting PRKCI and negatively regulating the expression of PRK-CI. miR-219 was negatively correlated with PRKCI expression in tongue squamous cell carcinoma.
		                        		
		                        		
		                        		
		                        	
7.Predictive Values of Child-Pugh Score and Albumin-bilirubin Score for Liver Injury and Prognosis After Stereotactic Ablative Radiotherapy in Patients With Hepatocellular Carcinoma

		                        			Background:Child-Pugh score(CPS)and albumin-bilirubin(ALBI)score have important predictive value for the assessment of survival of patients with hepatocellular carcinoma(HCC). Aims:To investigate the predictive values of CPS and ALBI score for radiation-induced liver disease(RILD)and long-term prognosis in patients with HCC receiving stereotactic ablative radiotherapy(SABR). Methods:A total of 152 HCC patients receiving SABR from June 2006 to June 2012 at Baoji Central Hospital were enrolled. CPS and ALBI score were determined,and the predictive values of CPS and ALBI score for RILD and prognosis were compared. Results:In 152 patients with HCC,CPS-A was noted in 119 patients, CPS-B in 33 patients;ALBI-1 was noted in 60 patients,ALBI-2 in 86 patients,ALBI-3 in 6 patients. The 5-year overall survival rate was significantly higher in CPS-A patients than in CPS-B patients(46.2% vs. 15.2%,P <0.001). Significant difference in 5-year overall survival rate was found among patients with ALBI-1,ALBI-2 and ALBI-3(P =0.002). CPS grade(HR=1.38,95% CI:1.18-1.94,P=0.008)and ALBI grade(HR=1.68,95% CI:1.32-2.03, P=0.003)were independent risk factors for 5-year survival of patients with HCC. RILD was found in 20 cases(13.8%) within 4 months after SABR. AUC of ALBI for prediction of RILD was significantly higher than that of CPS(0.784 vs. 0.611,P=0.028). Patients with CPS-A and ALBI< -2.76 were less likely to develop RILD after SABR(2.4%). 8.Experiment design and feasibility of BOLD and MRS multimodal fMRI in analysis of brain effect induced by acupuncture

		                        			Objective To establish the methodology of combining BOLD and 1H-MRS for investigating correlation between the deactivation in medial prefrontal cortex (MPFC) and gamma-aminobutyric acid (GABA) concentration by acupuncture at LI4 (Point Hegu),and to optimize the experimental technique and procedure.Methods Twenty healthy adult volunteers were enrolled.During fMRI-BOLD scanning,each subject received acupuncture at right LI4 (Point Hegu).MRS scanning was based on MEGA-PRESS sequence,and ROIs were located at bilateral MPFC.The task BOLD fMRI was block design,including 3 stimulations (30 s) with 2 intervals (2 min).Then MRS scanning was performed before and after BOLD.The quantitative values of the BOLD positive and negative activations (Pm) and GABA concentrations were calculated.Results All 20 subjects completed BOLD fMRI scanning,and met the postprocessing requirements.MRS images of 9 subjects with good image quality were included in analysis.Among all 20 subjects,positive activation (Pm=1.17± 0.16) was observed in 9,while negative activation (Pm =-1.31 ± 0.17) was observed in 11 subjects.The GABA average values before and after the acupuncture were (19.93 ±1.04) nmol/L and (20.04±0.81)nmol/L,respectively,and the average amplitude between post-and pre-acupuncture was (0.11 ± 1.60)nmol/L.Conclusion The success rate of this method for quantitative study of brain function established multimodal-functional (BOLD-fMRI and MRS) was acceptable,and the multimodal brain function changes as well as the quantitative values were observed in the brain region during acupuncture.Combined BOLD and MRS quantitative method is feasible for testing acupuncture response in the brain.
		                        		
		                        		
		                        		
		                        	
9.C-reactive protein/albumin ratio predicting early death in patients with acute ischemic stroke

		                        			Objective To investigate the predictive value of C-reactive protein/albumin ratio (CAR) for 30 d survival status in patients with acute ischemic stroke.Methods Patients with acute ischemic stroke admitted to the Neurological Intensive Care Unit (NICU),Nanfang Hospital,Southern Medical University were selected from 2013 to 2016.They were divided into a survival group and a death group according to the 30 d survival status.The clinical data of both groups were compared and analyzed.Multivariate logistic regression analysis was used to determine the independent risk factors for 30 d survival status.The predictive value of the variables was analyzed using the receiver operating characteristic (ROC) curve.Results A total of 236 patients were enrolled in the study,including 64 (27.12%) in the death group and 172 (72.88%) in the survival group.The baseline National Institutes of Health Stroke Scale score,procalcitonin,C-reactive protein,CAR,and onset to NICU time in patients of the survival group were significantly lower or shorter than those of the death group,and the serum albumin level of the survival group was higher than that of the death group (all P <0.05).Pearson's correlation analysis showed that C-reactive protein (r =0.647,P < 0.001),CAR (r =0.632,P < 0.001),and onset to NICU time (r =0.596,P < 0.001) were closely associated with the 30 d survival status in patients with acute ischemic stroke.Multivariate logistic regression analysis showed that CAR was an independent risk factor for 30 d mortality in patients with acute ischemic stroke (odds ratio 1.895,95% confidence interval 1.573-2.282;P < 0.001).ROC curve analysis showed that the area under the curve of CAR was 0.873 (95% confidence interval 0.815-0.931),the optimal cut-off value was 2.197,the sensitivity of predicting 30 d death risk was 82.8%,and the specificity was 87.8%.Conclusion CAR is an independent risk factor for 30 d death in patients with acute ischemic stroke and can be used for 30 d survival assessment in patients with acute ischemic stroke.
		                        		
		                        		
		                        		
		                        	
10. Impact of intensified maintenance therapy on the prognosis of children and adolescents with advanced lymphoblastic lymphoma

		                        			To investigate the impact of intensified maintenance therapy on the prognosis of children and adolescents with advanced lymphoblastic lymphoma (LBL) .
		                        		
		                        			Methods:
		                        			Retrospective analysis on the treatment results of children and adolescents with stage Ⅲ and stage Ⅳ LBL who underwent BFM-NHL-90/-95 regimen without prophylactic radiotherapy. The intensified therapy group included the patients admitted from 1998 to 2005, while others were classified as the non-intensified therapy group. Patients in the intensified therapy group were intravenously treated with "etoposide phosphate plus cytrarabine" and high-dose methotrexate alternately per 2.5-3 months in addition to the oral chemotherapy with 6-mercaptopurine and methotrexate during the maintenance phase.
