1.Association of sexually transmitted infection with semen quality in men from couples with primary and secondary infertility.

This study aims to compare the prevalence of sexually transmitted infections (STIs) with semen quality in men from couples with primary and secondary infertility. Semen samples were collected from 133 men who requested fertility evaluation. Seminal tract infection with Ureaplasma spp. (UU), Mycoplasma hominis (MH), Mycoplasma genitalium (MG), Chlamydia trachomatis (CT), Neisseria gonorrhoeae (NG), and herpes simplex virus-2 (HSV-2) was assessed by PCR-based diagnostic assays. Among all patients, the prevalence of STIs was higher in men from couples with primary infertility than that in men from couples with secondary infertility (39.7% vs 21.7%, P = 0.03). The prevalence of UU was 28.8% and 13.3% in men from couples with primary and secondary infertility, respectively. Men from couples with primary infertility were more likely to be positive for UU than men from couples with secondary infertility (P = 0.04). Regarding the UU subtype, the prevalence of Ureaplasma urealyticum (Uuu) and Ureaplasma parvum (Uup; including Uup1, Uup3, Uup6, and Uup14) did not differ between the two groups. No associations between the prevalence rates of MH, MG, and CT were found in men from either infertility group. A lower sperm concentration was associated with STI pathogen positivity in men with primary infertility according to the crude model (P = 0.04). The crude and adjusted models showed that semen volume (both P = 0.03) and semen leukocyte count (both P = 0.02) were independently associated with secondary infertility. These findings suggest the importance of classifying the type of infertility during routine diagnosis of seminal tract infections.

2.MMS21 Maintains the Stability of Complex MCM2-7 to Promote Hepatocellular Carcinoma Proliferation

ObjectiveTo investigate the expression level of MMS21 and its functions in hepatocellular carcinoma. MethodsThe GEPIA2 and CPATC databases were used to analyze the mRNA and protein levels of MMS21 in adjacent and carcinoma tissues; the endogenous expression levels of MMS21 were reduced individually by two siRNAs. Meanwhile, flow cytometry was used to detect the distribution ratio of cell cycle of hepatocellular carcinoma cells. BrdU staining and DNA fiber assay were employed to determine cells proliferation rate, and colony assay was used to evaluate the ability of cell proliferation; the MYC tagged MMS21 was cloned and transfected into SK-hep1 to generate MMS21 stably overexpressed cells. Co-IP combined with mass spectrometry were used to identify MMS21-interacting proteome and map interactive pathway; Western Blot (WB) was performed to determine the effect of MMS21 towards cell cycle-related proteins. ResultsCompared with adjacent tissues ( n = 160 ), MMS21 was significantly higher expressed in cancerous tissues ( n = 369;P < 0.05) and its high expression was associated with poor prognosis ( P < 0.01 ) in hepatocellular carcinoma; Knockdown of endogenous MMS21 by two siRNAs led to the increase of the proportion of cells in G0/G1 phase (P < 0.001; P < 0.01), and the decrease of the ratio of cells in S (P < 0.01; P < 0.05), G2/M (P < 0.05; P < 0.05) phase. Moreover, depletion of endogenous MMS21 greatly attenuated the speed of DNA replication (P < 0.000 1; P < 0.000 1) and the rate of cell proliferation (P < 0.01; P < 0.001). Mass spectrometry analysis identified the total of 641 high-confidence proteins for MMS21, and interactive pathway analysis showed that these binding proteins were highly correlated with cell cycle-related genes. The results of Western Blot further confirmed this conclusion, and depletion of endogenous MMS21 markedly reduced the protein levels of Microchromosome maintenance protein family (MCMs). ConclusionsMMS21 is highly expressed in hepatocellular carcinoma and positively correlated with the poor prognosis. MMS21 is closely related to the proliferation of Hepatocellular Carcinoma and regulated the stability of cell cycle related proteins and MCM family.
3.Clinical analysis of a family with familial hemangioblastoma

Objective To analyze the clinical and imaging features of a family(3 patients)with familial hemangioblastoma,and their diagnosis and prognosis.Methods The detailed data about clinical and imaging features of all patients diagnosed as familial hemangioblastoma,admitted to our hospital from October 2004 to May 2010,were analyzed,and the lesions of other regions,besides the tumor lesion,were observed.Results No lesions of other regions were noted in these 3 patients.Cranial MRI showed that 2 had cystic and solid tumor and 1 had solid tumor;,total removal was performed on these patients under microscope; regular follow-up was given and no recurrence was noted.Conclusion Familiar hemangioblastoma is serious hereditary disease; and MRI is the most important detective method; microsurgical operation is the most important therapy.Early diagnosis and treatment should be given to the patients with familiar hemangioblastoma due to its high recunence rate,having difficulty in operation and its trend to combining with other lesions of the other parts.
4.Evaluating the quality of niuhuangjiedu tablets by the systematic quantified fingerprint method.

The HPLC fingerprints of Niuhuangjiedu tablets (NHJDT) were established and assessed by systematic quantified fingerprint method. The chromatographic fingerprints (CFPs) were determined by reversed-phase HPLC, in which the CFPs containing 53 co-possessing peaks were marked by choosing baicalin peak as the referential peak. Fifteen batches of NHJDT were identified with hierarchical clustering analysis by using macro qualitative similarities and macro quantitative similarities as variables. According to the results of classification, the referential fingerprint (RFP) was synthesized from 10 batches of NHJDTs. Taking the RFP for the qualified model, the whole 15 batches of NHJDTs were evaluated by the systematic quantified fingerprint method. Among the 15 batches of NHJDT, 9 batches were completely qualified, the relative variance of leveling coefficient of 1 batch was obviously higher while the contents of other 5 batches were lower. The systematic quantified fingerprint method closely integrates the systematic fingerprints with the macro qualitative similarities and macro quantitative similarities to perform the entire qualification and quantitation analyses, which can briefly and effectively assess the quality of Chinese traditional medicine.

5.Effects of ligustrazine on extracellular acetylcholine levels in rat brain dialysate.

Using brain microdialysis and LC-MS/MS to detect acetylcholine in rat brain to investigate the effects of ligustrazine. A liquid chromatography-tandem mass spectrometry method has been developed and validated for the determination of acetylcholine in rat brain dialysate sampling by microdialysis. The results indicated that ligustrazine administration by subcutaneous injection significantly increased Ach release in rat medial prefrontal cortex and nucleus accumbens in a dose-related manner. The drug' s effect on Ach release in rat brain could be directly detected by microdialysis combined with HPLC-MS/MS and this method is selective and sensitive.

6.RP-HPLC fingerprint for quality assessment of Radix Angelicae dahuricae.

AIMTo establish the RP-HPLC fingerprint analysis for the quality control of Radix Angelicae dahuricae.
METHODSHPLC fingerprint analysis method of Radix Angelicae dahuricae was developed. Kromasil C18 column (250 mm x 4.6 mm ID, 5 microm) was used, with mixture of acetonitrile and water as mobile phase in a gradient mode. The flow rate was 1.0 mL x min(-1). The wavelength of measurement was 254 nm. Twenty-one batches of Radix Angelicae dahuricae were determined.
RESULTSThe 21 samples were classified as 4 clusters by cluster analysis and the 11 superior in producing area samples were confirmed to establish the mutual model. The samples' quality was assessed by Similarity Evaluation System for Chromatographic Fingerprint of TCM 2004.
CONCLUSIONThe method can be used to identify and evaluate the quality of Radix Angelicae dahuricae conveniently.

7.Function of endothelin-1 in neurogenic pulmonary edema.

AIMTo investigate the role of endothelin-1 in the pathogenesis of neurogenetic pulmonary edema.
METHODSThe levels of endothelin-1 in plasma and lung were measured in rats which suffered from diffuse brain injury on Marmarous' model. The changes of endothelin-1 in the lungs were also detected using an immunohistochemical method.
RESULTSAfter heavy diffuse brain injury in rats, the levels of endothelin-1 in plasma and lung began increasing at 1 hour, and peaked at 6 hour. Though a little declining at 24 hour, it maintained a higher level within 48 hours (P < 0.05). Pulmonary pathology showed that after brain injury there were congestion, swelling in pulmonary microvessels with broadened pulmonary interstitial tissue, and leucocyte infiltration was dominated by neutrophils and monocytes from 1 hour on, which peaked at 6 hour. More serious congestion, swelling and protein effusion in pulmonary alveoli were observed at both 24 h and 48 h. Immunohistochemically, endothelin-1 had more significant expression and higher levels of OD in the experimental groups than that in the control's, the most significance of which was at 6 hour.
CONCLUSIONThe inflammatory injury mechanism caused by endothelin-1 may play an important role in neurogenic pulmonary edema.

8.Stuides on evaluation of Semen Armeniacae amarum.

OBJECTIVETo systematically evaluate the quality of Semen Armeniacae Amarum.
METHODThe amygdalin content in the samples of Semen Armeniacae Amarum from different markets was analysed by HPLC. Some physical and chemical tests were done to investigate their quality.
RESULTSome significant differences in amygdalin contents and some physical and chemical parameters were shown between the crude drugs and the processed ones. There are few changes in amygdalin contents with the storage in 17 years.
CONCLUSIONThe processing conditions are the main factors that affect the quality of Semen Armeniacae Amarum. Furthermore, a systematic evaluation of the quality of its samples from the different markets has been established by a radar diagram based on the following five parameters: amygdalin contents, the lipid content, ethanol-soluble extract, total ash and acid-insoluble ash.

9.Analysis of Chemical Components and Antifungal Activity of Extraction from Conidia of Trichoderma viride LTR-2

To study the chemical components and the antifungal activity of extraction from conidia of Trichoderma viride LTR-2.The extraction were obtained by distilling with Methylene dichloride from conidia of Trichoderma viride LTR-2 cultured on wheat bran solid matrix.Antifungal activity were determined by mycelium growth method.The chemical components of the extraction were analysed by GC-MS,the relative components in the extraction were determined by area normalization.The extraction not only have broad-spectrum control,showed antibiosis against eleven different plant fungal pathogens in PDA dish,such as Rhizoctonia solani,Alternaria brassica,Verticillium dahliae,Macrophoma kawatsukai,Fusarium moniliforme,Botrytis cinerea,Rhizoctonia cerealis,Fusarium oxysporum f.sp.vasinfectum,Bipolaris sorokinana,Fusarium graminearum,Alternaria.mali,but also have high inhibitory effect,and had 89.3% suppressive rate to Rhizoctonia cerealis.About sixty components were separated and identified by GC-MS,majority components were Hydrocarbon,the number of the Hydrocarbon were fourty-three kinds.Ergosterol was the major chemical components of the extract,and has 41.90% content.Other components comprised:Ketone,Organic acid,Alcohol,Ene,et al.Conclusion:The extraction from conidia of Trichoderma viride LTR-2 have antifungal activity.The extration comprised 2H-Pyran-2-one,5,6-dihydro-6-pentyl,it has 2.35% content.reference others literature,2H-Pyran-2-one,5,6-dihydro-6-pentyl may be the suppressive component of the extration.
