1.Analysis of Potential Suitable Areas and Key Ecological Factors of Polygonatum odoratum Based on MaxEnt Model

ObjectiveThe potential suitable area for ecological planting, key ecological factors, and suitable range of Polygonatum odoratum in China were analyzed to provide theoretical and scientific guidance for the artificial planting of P. odoratum. MethodA total of 454 geographical distribution records of P. odoratum in China and 118 ecological factors were used in this study. The maximum entropy model (MaxEnt) was adopted to predict the suitable areas of P. odoratum. The key ecological factors and their suitable ranges were analyzed by the jackknife method, contribution rates of ecological factors, and response curves. ResultThe suitable areas of P. odoratum were mainly located in the northwest, north, and northeast of China, the highly suitable areas of which were concentrated in Shaanxi, Shanxi, Gansu, etc. Solar radiation in November (Srad11), precipitation in July (Prec7), percentage of evergreen/deciduous needleleaf trees (Class1), silt content (2-50 μm) mass fraction (SLTPPT), and annual average temperature (Bio1) were found to be the key ecological factors affecting the suitable distribution of P. odoratum in China. The cumulative contribution rate of solar radiation factors (31.29%)>vegetation factors (25.61%)>soil factors (19.52%)>precipitation factors (11.38%)>temperature factors (8.57%)>topography factors (3.63%). ConclusionIt is suggested to carry out ecological planting of P. odoratum mainly in Shaanxi (such as Baoji and Ankang Cities and Ningshan, Liuba, and Hua Counties), Gansu (such as Tianshui City, Gannan Tibetan Autonomous Prefecture, and Liangdang and Huating Counties), and Shanxi (such as Yangquan, Taiyuan, Fenyang, and Jinzhong Cities, as well as Xingxian County) of China. Solar radiation factors should be given priority in the planting process, followed by vegetation, soil, precipitation, temperature, and topography factors. The range of key ecological factors, namely Srad11, Prec7, Class1, SLTPPT, and Bio1 should be controlled within 8 095.21-10 334.98 (optimum 8 787.50) kJ·m-2·d-1, 109.99-223.60 (146.91) mm, 1.00%-9.45% (6.76%-10.68%), 41.73%-50.35% (46.53%), and 3.29-16.33 (13.57) °C, respectively.
2.Diffusion weighted imaging for assessment of efficacy and volume response after uterine artery embolization of uterine fibroids

Objective To observe the clinical and symptomatic improvement three months after uterine artery embolization(UAE),and to analyze the value of apparent diffusion coefficient(ADC)in MR diffusion weighted imaging(DWI)in assessing the response of fibroids volume after UAE.Methods A total of 40 patients with uterine fibroids were included.The volume changes of fibroids,clinical and symptomatic improvement before and after treatment were recorded,and the efficacy of UAE was comprehensively analyzed.All patients underwent MR DWI before UAE and were evaluated at three months postoperatively by outpatient MR follow-up,with fibroids vol-ume and ADC quantitative measurements were performed to compare the changes in ADC values of fibroids preoperatively and post-operatively at each b value.Pearson correlation analysis was performed to analyze the correlation between baseline ADC values and postoperative fibroids volume reduction.Regression analysis was performed to assess the relationship between ADC and fibroids volume reduction after UAE.And the receiver operating characteristic(ROC)curve were plotted to analyze the predictive value of ADC values for evaluating fibroids volume reduction of more than 30%after UAE.Results The patients'clinical symptoms was improved in the three months after surgery,the volume of fibroids was significantly reduced,and the life quality was improved,the difference was sta-tistically significant(P<0.05).There was no significant effect on ovarian function,hormone levels did not change significantly com-pared to before surgery,with no statistical significance(P>0.05).When b=50,1 000 s/mm2,the changes in ADC values before and after uterine fibroids treatment were not significant,with no statistical significance(P>0.05).However,the changes in ADC values before and after uterine fibroids treatment were significant when b=800 s/mm2 and the difference was statistically significant(P<0.05).Under the condition of b=800 s/mm2,Pearson correlation analysis showed ADC value had a positive correlation with postoperative uterine fibroids volume reduction rate(r=0.45,P<0.05),and the area under the curve(AUC)for ADC value to predict the reduction rate of uterine fibroids volume by more than 30%after UAE was 0.787.The cut-off value was 1.143 × 10-3 mm2/s,with sensitivity and specificity of 0.793 and 0.818,respectively.Conclusion UAE is more effective in treating uterine fibroids.The baseline ADC value of uterine fibroids correlated significantly with the volume reduction after UAE.The ADC value can be used to assess the volume response after UAE.
3.Effect of Modified Huangqi Gancaotang on Proliferation, Apoptosis, Invasion, Migration, and Epithelial-mesenchymal Transition of Non-small Cell Lung Cancer Cells Based on Wnt/β-catenin Pathway

ObjectiveTo investigate effect of lyophilized powder of modified Huangqi Gancaotang on proliferation, apoptosis, invasion, migration, and epithelial-mesenchymal transition of human non-small cell lung cancer cells (A549, PC9) and possible mechanism. MethodEffect of 1.0, 2.0, 4.0, 8.0, 12.0 g·L-1 modified Huangqi Gancaotang on the proliferation of non-small cell lung cancer cells was detected by cell counting kit-8 (CCK-8) assay. A549 and PC9 cells were classified into the blank group and the low-, medium-, and high-dose Huangqi Gancaotang groups (2.0, 4.0, 8.0 g·L-1). Plate cloning assay was used to examine the effect of modified Huangqi Gancaotang on cell cloning ability. Hoechst 33342 staining and flow cytometry were employed to detect the apoptosis, and scratch assay and Transwell migration assay were applied to examine cell migration and invasion abilities, respectively. Mammosphere assay was used to examine the sphere-forming ability of tumor cells, and real-time polymerase chain reaction (Real-time PCR) to detect the mRNA expression of stemness-related molecules octamer-binding transcription factor 4 (Oct-4), human sex-determining region Y-box 2 (Sox2), and homeobox transcription factor (Nanog) to assess cancer stem cell activity. The protein expression of B-cell lymphoma 2 (Bcl-2), Bcl-2-associated death promoter (Bad), Bcl-2-associated X protein (Bax), cleaved Caspase-3, Caspase-3, E-cadherin, N-cadherin, vimentin, matrix metalloproteinase-2 (MMP-2), β-catenin, c-Myc, Cyclin D1, and zinc-finger transcription factor (Slug) was determined by Western blot. ResultThe proliferation ability of A549 and PC9 cells was significantly inhibited after 24 h and 48 h treatment with 1.0, 2.0, 4.0, 8.0, and 12.0 g·L-1 lyophilized powder of modified Huangqi Gancaotang compared with that in the blank group and the inhibition was dose- and time-dependent (P<0.05, P<0.01). Compared with the blank group, the low-, medium-, and high-dose modified Huangqi Gancaotang suppressed the cloning ability of A549 and PC9 cells (P< 0.05, P<0.01), and high-dose modified Huangqi Gancaotang induced apoptosis of A549 and PC9 cells (P<0.01). In comparison with the blank group, the low-, medium-, and high-dose modified Huangqi Gancaotang inhibited the invasion and migration of A549 and PC9 cells (P<0.05, P<0.01). Compared with the blank group, the low-, medium-, and high-dose modified Huangqi Gancaotang significantly decreased volume of the microspheres of A549 cells and the mRNA expression of Oct-4, Sox2, and Nanog in A549 and PC9 cells (P<0.05, P<0.01). Compared with the blank group, the medium- and high-dose modified Huangqi Gancaotang down-regulated the expression of the anti-apoptotic protein Bcl-2 (P<0.05, P<0.01), up-regulated the expression of pro-apoptotic proteins Bad, Bax, and cleaved Caspase-3/Caspase-3 (P<0.05, P<0.01) in A549 and PC9 cells, decreased the expression of MMP-2, N-cadherin, and vimentin (P<0.05, P< 0.01), and raised the E-cadherin expression (P<0.05, P<0.01). Moreover, the medium-dose and high-dose modified Huangqi Gancaotang all reduced the expression of β-catenin, c-Myc, Cyclin D1, and Slug in A549 and PC9 cells (P<0.01). ConclusionModified Huangqi Gancaotang can inhibit the proliferation, invasion, migration, activity of cancer stem cells, and epithelial-mesenchymal transition of human non-small cell lung cancer (A549, PC9) cells and induce apoptosis, and the mechanism is the likelihood that it regulates Wnt/β-catenin signaling pathway.
4.Prenatal and postnatal MRI findings of fetal pericallosal lipoma with normal corpus callosum: a case report

We describe the prenatal diagnosis and postnatal follow-up of a fetus with a pericallosal lipoma diagnosed by MRI. The pregnant woman was admitted to Zhongnan Hospital of Wuhan University in February 2019. MRI examinations with multiple sequences and imaging methods were performed prenatally at 31 +6 and 38 +3 weeks of gestation, and at two days and three months postnatally. The MRI images showed that the corpus callosum was normal and the lesion was located around the corpus callosum, with high signal intensity on T1 weighted imaging, medium-to-high signal intensity on T2 weighted imaging, and low signal intensity on fat suppression sequence. No abnormal blood vessels were found on susceptibility-weighted imaging and time-of-flight magnetic resonance angiography sequences. Based on the prenatal and postnatal MRI findings, the tumor was clinically diagnosed as a pericallosal lipoma.
5. Exploration and analysis of the management mode of a cabin hospital during the outbreak of COVID-19

Since the outbreak of COVID-19 in Wuhan, cabin hospitals have played an important role in preventing the spread of the epidemic and admitting all the patients and suspected ones. As one of the first three cabin hospitals in Wuhan, Jianghan cabin hospital under care of Wuhan Union Hospital has efficiently fulfilled its purpose of admitting patients of mild symptoms. Measures taken by the cabin hospital include clearing its positioning, optimizing its layout, establishing organization structure, setting up the rules and regulations, and strengthening the prevention and control of hospital infection. This article briefly summed up the operation and management practices of the cabin hospital, and analyzed its management difficulties, hence putting forwards suggestions on medical emergency management system of China, for references of hospitals and authorities in charge.
6.Acupuncture artificial menstrual cycle method for abnormal uterine bleeding-ovulatory dysfunction (spleen deficiency syndrome).

OBJECTIVE:
To compare the clinical efficacy between acupuncture artificial menstrual cycle method and medication artificial menstrual cycle method for abnormal uterine bleeding-ovulatory (AUB-O) dysfunction (spleen deficiency syndrome).
METHODS:
Sixty patients were randomly assigned into an observation group and a control group, 30 cases in each one. 3 cases dropped out in the observation group and 27 cases were included into analysis; 2 cases dropped out in the control group and 28 cases were included into analysis. The patients in the observation group were treated with acupuncture artificial menstrual cycle method. The acupoints were selected according to different stages of menstrual cycle. The acupuncture was given for 30 min per treatment, once every other day; no treatment was given during menstrual period. Xuehai (SP 10), Sanyinjiao (SP 6), Taixi (KI 3), Guanyuan (CV 4), Qihai (CV 6), Zusanli (ST 36) and Zhangmen (LR 13) were selected after menstrual period. Taichong (LR 3), Hegu (LI 4), Yaoyangguan (GV 3), Geshu (BL 17), Ganshu (BL 18), Sanyinjiao (SP 6), Pishu (BL 20) were selected in ovulaton periods. Baihui (GV 20), Shenshu (BL 23), Yaoyangguan (GV 3), Geshu (BL 17), Sanyinjiao (SP 6), Zusanli (ST 36), Gongsun (SP 4) were selected before menstrual period. The patients in the control group were treated with medication artificial menstrual cycle method. Femoston (estradiol tablets/estradiol dydrogesterone tablets) was taken orally. The white tablets were estradiol (1 mg), while the gray tablets were estradiol (1 mg) and dydrogesterone (10 mg). The Femoston was taken once a day, white tablets for the first 14 days, then gray tablets for the last 14 days. The 28-day treatment was taken as one session, and one cycle menstrual was taken as one treatment course, and totally 3 courses were given in the two groups. The clinical symptom score was observed before and after treatment as well as in follow-up visit (3 months after treatment). The clinical efficacy was evaluated based on the total effective rate, the marked effective rate of main symptoms (including menstrual cycle, menstrual period, menstrual volume) and basal body temperature (BBT).
RESULTS:
① Compared before treatment, the clinical symptom score in the two groups was decreased after treatment and in the follow-up visit (<0.05). Compared with the control group, the clinical symptom score after treatment as well as the difference of that before and after treatment had no significant difference in the observation group (>0.05). Compared with the control group, the clinical symptom score in the follow-up visit as well as the difference of that between pre-treatment and follow-up visit had significant difference in the observation group (<0.05, <0.01). ② The total effective rate was 81.5% (22/27) in the observation group and 85.7% (24/28) in the control group, without significant difference between the two groups (>0.05). ③ After treatment, the marked effective rate of menstrual cycle, menstrual period, menstrual volume had no significant difference between the two groups (>0.05); during the follow-up visit, the marked effective rate of menstrual cycle in the observation group was superior to that in the control group (<0.05), but that of menstrual period and menstrual volume had no significant difference between the two groups (>0.05). ④ After treatment, the diphasic curve rate of BBT had no significant difference between the two groups (>0.05); during the follow-up visit, the diphasic curve rate of BBT in the observation group was higher than that in the control group (<0.05).
CONCLUSION
Acupuncture artificial menstrual cycle method has superior efficacy for AUB-O dysfunction (spleen deficiency syndrome), which has similar efficacy with medication artificial menstrual cycle method. Moreover, acupuncture has advantages in regulating menstrual cycle and improving ovulation, and has a longer curative effect.

7.Inhibition of SKP2 Sensitizes Bromocriptine-Induced Apoptosis in Human Prolactinoma Cells.

PURPOSE: Prolactinoma (prolactin-secreting pituitary adenoma) is one of the most common estrogen-related functional pituitary tumors. As an agonist of the dopamine D2 receptor, bromocriptine is used widely to inhibit prolactinoma progression. On the other hand, it is not always effective in clinical application. Although a dopamine D2 receptor deficiency contributes to the impaired efficiency of bromocriptine therapy to some extent, it is unknown whether there some other underlying mechanisms leading to bromocriptine resistance in prolactinoma treatment. That is the main point addressed in this project. MATERIALS AND METHODS: Human prolactinoma samples were used to analyze the S-phase kinase associated protein 2 (SKP2) expression level. Nutlin-3/adriamycin/cisplatin-treated GH3 and MMQ cells were used to analyze apoptosis in SKP2 overexpression or knockdown cells. SKP2 expression and the interaction partners of SKP2 were also detected after a bromocriptine treatment in 293T. Apoptosis was analyzed in C25 and bromocriptine-treated GH3 cells. RESULTS: Compared to normal pituitary samples, most prolactinoma samples exhibit higher levels of SKP2 expression, which could inhibit apoptosis in a p53-dependent manner. In addition, the bromocriptine treatment prolonged the half-life of SKP2 and resulted in SKP2 overexpression to a greater extent, which in turn compromised its pro-apoptotic effect. As a result, the bromocriptine treatment combined with C25 (a SKP2 inhibitor) led to the maximal apoptosis of human prolactinoma cells. CONCLUSION: These findings indicated that SKP2 inhibition sensitized the prolactinoma cells to bromocriptine and helped promote apoptosis. Moreover, a combined treatment of bromocriptine and C25 may contribute to the maximal apoptosis of human prolactinoma cells.

8.Clinical trial of gastroesophageal reflux disease with the disharmony between liver and stomach syndrome treated with acupuncture regulatingbased on the compatibility of the five meridians.

OBJECTIVETo observe the clinical effects of the acupuncture regulatingbased on the compatibility of the five meridians and common western medication for gastroesophageal reflux disease with the disharmony between liver and stomach.
METHODSSixty patients were divided into two groups according to non-erosive reflux disease (NERD) and reflux esophagitis (RE). Those in the two groups were randomly assigned into an observation group and a control group. There were 57 cases included (28 in the observation group with 2 dropping, 29 in the control group with 1 dropping). Based on the life care, patients in the observation group were treated with 30 min acupuncture regulatingbased on the compatibility of the five meridians at Zusanli (ST 36), Chongyang (ST 42), Weishu (BL 21), Zhongwan (CV 12), Xingjian (LR 2), Shaofu (HT 8), Dadun (LR 1), Taichong (LR 3), Ganshu (BL 18), Qimen (LR 14), Danzhong (CV 17), Zhongting (CV 16), three times a week. Patients in the control group were treated with rabeprazole orally. All the treatment was given for consecutive 8 weeks. The clinical symptom score was observed before and after treatment and 4 weeks after treatment. The effects were evaluated from 3 aspects, including the main symptoms, esophagitis improvement under gastroscope and total clinical symptoms.
RESULTSAfter treatment and at follow-up, the symptom scores decreased in the two groups compared with those before treatment (all<0.05), with better improvement in the observation group at follow-up (<0.05). The symptom scores after treatment of the two types in the two groups decreased compared with those before treatment (all<0.05). The total effective rate of heartburn after treatment in the observation group was 82.1% (23/28), which was lower than 93.1% (27/29) of the control group (<0.05). The total effective rate of belching after treatment in the observation group was 94.1% (16/17), which was better than 75.0% (15/20) in the control group (<0.05). The total effective rate of the esophagitis improvement under gastroscope after treatment in the observation group was 75.0% (12/16), and that in the control group was 82.4% (14/17), without statistical significance (>0.05). The total effective rate of the clinical symptoms after treatment in the observation group was 82.1% (23/28), and that in the control group was 86.2% (25/29), without statistical significance (>0.05).
CONCLUSIONAcupuncture regulatingbased on compatibility of the five meridians can effectively improve clinical symptoms and inflammation under gastroscope for gastroesophageal reflux disease with the disharmony between liver and stomach. This treatment has a longer curative effect and a greater alleviation of belching.
10.Transplantation of human umbilical cord mesenchymal stem cells in the treatment of systemic lupus erythematosus

BACKGROUND:Systemic lupus erythematosus is an autoimmune disease characterized as an emergence of a variety of autoantibodies in serum and multi-system and multi-organ lesions. Currently, there is a lack of effective treatment options, and umbilical cord mesenchymal stem cel s are a promising therapy for systemic lupus erythematosus based on cel biological roles. OBJECTIVE:To observe the therapeutic efficacy of human umbilical cord mesenchymal stem cel transplantation in the treatment of systemic lupus erythematosus in mice. METHODS:Human umbilical cord mesenchymal stem cel s were isolated and cultured fol owed by labeling with DiR fluorescence. Experimental mice were divided into normal control group (C57BL mice), model control group (C57BL/lpr mice), low-, medium-and high-dose umbilical cord mesenchymal stem cel therapy groups (C57BL/lpr mice), with 10 mice in each group. Mice in the low-, medium-and high-dose groups were respectively injected 0.5×106, 1×106, 2×106 human umbilical cord mesenchymal stem cel s, once a week, for 3 consecutive weeks. At the end of treatment, blood samples were col ected to measure antinuclear antibody, anti-histone antibody, anti-double stranded DNA antibody changes;OPG and Foxp3 gene expression changes were detected by quantitative PCR method. RESULTS AND CONCLUSION:After treatment, the levels of anti-nuclear antibodies, anti-histone antibodies and anti-double stranded DNA antibodies in the peripheral blood of mice were al declined in the low-, medium-and high-dose groups, while the number of peripheral blood CD4+CD25+T cel s was significantly elevated. OPG and Foxp3 gene expression was also increased dramatical y in the low-, medium-and high-dose groups, which was similar to that in the normal control group and significantly different from that in the model control group (P<0.01). Experimental findings demonstrate that after transplantation of human umbilical cord mesenchymal stem cel s, al relevant indicators in C57BL/lpr mice recovered to the normal levels, and the high-dose treatment group had the most obvious effect.
