1.Relationship between self-disclosure and demoralization syndrome in patients with permanent enterostomy for colorectal cancer

Objective:To explore the impact of self-disclosure on demoralization syndrome in patients with permanent enterostomy for colorectal cancer, so as to provide reference for clinical nursing interventions for those patients.Methods:From January to May 2023, convenience sampling was used to select 207 patients with permanent enterostomy for colocrctal cancer of the Wound Ostomy Clinic of Laoshan Campus of the Affiliated Hospital of Qingdao University as the research subjects. A survey was conducted on patients using the General Information Questionnaire, Distress Disclosure Index, Social Support Rating Scale, and the Demoralization Scale Mandarin Version.Results:Among 207 patients with permanent enterostomy for colorectal cancer, the scores of self-disclosure, social support, and demoralization syndrome were 35.00 (26.00, 47.00) , 32.00 (26.00, 39.00) , and 35.00 (23.00, 47.00) , respectively. A total of 128 patients (61.8%) were in moderate demoralization, and 37 patients (17.9%) were in severe demoralization. Multiple linear regression analysis showed that place of residence, self-care of stoma, self-disclosure, and social support were the influencing factors for demoralization syndrome in patients with permanent enterostomy for colorectal cancer ( P<0.05) . Conclusions:Patients with permanent enterostomy for colorectal cancer have a high overall score of demoralization syndrome, and a high proportion of patients with moderate to severe demoralization. Medical and nursing staff should focus on patients in remote rural areas and those with poor self-care abilities to stomas, develop targeted intervention measures to improve patients' negative emotions, enhance their self-disclosure, and pay attention to strengthening social support to relieve their demoralization syndrome, thereby improving their quality of life.
2.Treatment experience of neck tumor surgeries involving carotid artery.

Objective:To investigate surgical treatment of carotid artery diseases in neck tumor surgery. Methods:A retrospective analysis of the clinical data on carotid artery treatment was conducted in the five cases of neck tumor surgeries treated at Department of Surgical Oncology, the First Peoples Hospital of Lanzhou from March 2010 to May 2020. Surgical methods, including carotid artery resection and ligation, tumor-involved artery resection and vascular reconstruction, and tumor peeling and carotid rupture repairing were used, respectively. Results:Five cases were successfully operated on. One case of carotid artery ligation was followed by intermittent dizziness and decreased contra-lateral limb strength after the surgery. The remaining patients exhibited no neurological complications. A patient with cervical low-grade myofibroblastoma developed into lung metastases 8 months after the surgery. Another patient with cervical lymph node metastases in papillary thyroid cancer developed into lung metastases 24 months after the surgery. Conclusion:Currently, surgical methods for clinical treatment of diseased carotid arteries include carotid artery resection and ligation, simple tumor peeling, tumor invasion artery resection and vascular reconstruction, and interventional therapy. Each surgical method has its own advantages and disadvantages. Therefore, the choice of treatment depends on the patient's specific conditions, physician's clinical experience, and the equipment available.

3.Research on the fingerprint of Fritillaria anhuiensis Bulbus by UPLC-Q-TOF-MS technique with chemical recognition mode

Objective:To establish the fingerprint and stoichiometric analysis mode of Ultra Performance Liquid Chromatography Tandem Quadrupole Time-of-Flight Mass Spectrometry (UPLC-Q-TOF-MS) of Fritillaria anhuiensis Bulbus, so as to provide reference for its quality evaluation and standard formulation. Methods:By setting the CORTECS C18 column at 4.6 mm×150 mm, 2.7 μm with the mobile phases of acetonitrile-0.1% formic acid and 10 mmol/L aqueous ammonium formate for gradient elution at a flow rate of 0.4 ml/min and an injection volume of 2.0 μl. The TCM fingerprint similarity evaluation system (2012 version) was used to evaluate 9 batches of Fritillaria anhuiensis Bulbus samples. By using Liquid Chromatography-Mass Spectrometry (LC-MS) technique to make quantity analysis and by combining cluster analysis, principal component analysis and orthogonal least squares-discriminant analysis to make overall quality evaluation. Results:The fingerprint profiles of different batches of Fritillaria anhuiensis Bulbus were established and 21 common peaks were identified, and 12 of them were initially identified. Cluster analysis, principal component analysis and orthogonal least squares-discriminant analysis were used to cluster the nine batches of Fritillaria anhuiensis Bulbus into three categories. Conclusion:The fingerprint established in this study combined with the chemical pattern recognition method are highly sensitive and specific, which could reflect the overall characteristics and differences of Fritillaria anhuiensis Bulbus, providing reference for the quality evaluation of Fritillaria anhuiensis Bulbus and standardization of it.
4.Comparative Study of Protective Effects of Atomization Inhalation and Intraperitoneal Injection of Edaravone on Smoke Inhalation Lung Injury Model Rats

OBJECTIVE:To compare the protective effect of atomization inhalation and intraperitoneal injection of edaravone on acute lung injury in smoke inhalation lung injury model rats. METHODS :Thirty male SD rats were divided into normal control group(group A ),injury group (group B ),intraperitoneal injection group (group C ),low-dose aerosol inhalation group (group D),high-dose aerosol inhalation group (group E )according to random numble table ,with 6 rats in each group. Group B-E were placed in smoke generator containing pine sawdust to induce smoke inhalation lung injury model. In group A ,the operation was the same as above except that the pine sawdust was not placed. Thirty minutes after modeling ,group C were injected intraperitoneally with edaravone 18 mg/kg(every 70 min,4 times in total ). Group D and E inhaled edaravone 9,1.8 mg/kg(every 60 min,lasting for 10 min each time ,4 times in total ). The rats were treated by no means in group A and group B. Six hours after last medication,arterial blood gas analysis was performed ,and the lung wet to dry ratio (W/D)and water content of lung tissue were calculated. The levels of TNF-α,IL-6 and IL- 10 in serum were detected by double antibody ELISA. The contents of MDA ,MPO, SOD and Caspase- 3 in lung tissue were determined by ELISA and other methods. HE staining was used to observe the pathological changes of lung tissue. The apoptotic rate of cells in lung tissue were determined by TUNEL assay. RESULTS :No abnormality was found in lung tissue of group A ;in group B ,hemorrhage and edema were found in lung tissue ,alveolar structure was difficult to identify,and inflammatory cells and red blood cell infiltration were seen. Above symptoms of rats in group C-E were improved to different extent. Compared with group A ,PaO2/FiO2 and SOD content of lung tissue were decreased significantly in other groups (P<0.05);water content of lung tissue ,W/D,serum contents of TNF-α,IL-6 and IL- 10,the contents of MDA ,MPO and Caspase-3 in lung tissue ,apoptotic rate were increased significantly (P<0.05). Compared with group B ,PaO2/FiO2 and serum contents of IL- 10 were increased significantly in administration groups (P<0.05);water content of lung tissue ,W/D,serum contents of TNF-α and IL-6,the contents of MDA ,MPO and Caspase- 3 in lung tissue ,apoptotic rate were significantly decreased,in dose-dependent manner (P<0.05). CONCLUSIONS :Edaravone has a certain protective effect on smoke inhalation lung injury model rat. It can reduce the production and release of inflammatory mediators and/or cytokines ,reduce the peroxide damage and inhibit cell apoptosis in a dose-dependent manner. The effect of atomization inhalation is more obvious than that of intraperitoneal injection.
5.Study on microdosimetry for carbon ion radiotherapy

Objective:To measure the microdosimetric spectrum of carbon ion beam and calculate the relative biological effect (RBE) distribution, so as to provide reference for radiotherapy microdosimetric research.Methods:A silicon on insulator (SOI) microdosimeter was used to measure the microdosimetric spectrum of 12C ion beam, at 260 MeV/u, provided by Lanzhou Heavy Ion Accelerator National Laboratory. The measured pulse amplitude spectrum was converted to obtain the dose distribution. The microdosimetric spectra and RBE values at different polymethyl methacrylate depths were measured by the combination of different thickness PMMA. Results:The microdosimetric spectra of 12C ion beam at 260 MeV/u were measured at different PMMA depths, and the relationship between dose line energy yD and RBE and different PMMA depths was also obtained. The measurement result showed that the RBE value reached a peak of 2.6 at the PMMA depth of 116.5 mm, and decreased rapidly after the Bragg peak. However, the RBE value was still 1.3 at the trailing point, which was about twice as much as the entrance of the flat zone. Conclusions:This paper provides basic data for carbon ion beam microdosimetric spectroscopy through measurement. The RBE value of 12C ion beam gradually rises and reaches a peak with the increase of PMMA depth. After the Bragg peak position, it drops rapidly, but the biological effect at the tail cannot be ignored. At the same time, it reflects the dose fraction caused by different line energy intervals, and provides a reference for assessing the risk of heavy ion therapy for secondary cancer.
6.Effect of recombinant human granulocyte/macrophage colonystimulating factor combined with nano-silver on deep burn degreen Ⅱ about inflammation

Objective To observe the effect of recombinant human granulocyte/macrophage colonystimulating factor (rhGM-CSF) combined with nano-silver as a treatment on deep burn degreen Ⅱ about inflammation .Methods The burn model was built with Wistar rats .They were randomly divided into four groups ,petrolatum treatment (group A ,n= 30) ,nano-silver treatment (group B ,n=30) ,rhGM-CSF treatment(group C ,n=30) ,and rhGM-CSF combined with nano-silver treatment(group D ,n=30) . observation the inflammatory reaction ,and culture bacteria on wound of the four groups at 1st ,4th ,7th ,10th ,14th ,21th day after treatment were made .The level of IL-2 and IL-8 were measured in serums with ELISA .Results The inflammatory reaction:group A>group B>group C>group D ;Bacterias were observed in group A ,group B/C and group D at 4th ,10th ,14th day respectively af-ter treatment .The number of bacterial growed in group D was less than in group A ,group B and group C .The numbers of bacterial growed in group B and group C were less than in group A .And after 10 ,14 ,21 days treatment ,there was significantly statistical difference(P<0 .05) .There was difference among groups in the levele of IL-2 and IL-8 ,which were the lowest in group D and the highest in group A .The level of IL-2 has no significantly statistical difference between every groupat 1st day and between B and C group at 4th day(P>0 .05) .After other days treatment ,there was significantly statistical difference(P<0 .05) .There was signifi-cantly statistical difference in every group at each day of the IL-8 levele except A and B group ,B and C group ,C and D group at 1st day (P<0 .05) .Conclusion rhGM-CSF combined with nano-silver treatment could alleviate inflammatory reaction ,and be better than rhGM-CSF or nano-silver alone .

Objective To observe the effect of recombinant human granulocyte/macrophage colonystimulating factor (rhGM-CSF) combined with nano-silver for deep burn degreen Ⅱ. Methods The burn model were done with Wistar rats. They were randomly divided into four groups , group A (n = 30): petrolatum treatment, group B(n = 30): nano-silver treatment, group C(n = 30): rhGM-CSF treatment, and group D(n =30): rhGM-CSF combined with nano-silver treatment. The healing rates of the four groups were observed on postburn day 1, 4, 7, 10, 14, 21. Meanwhile the levels of VEGF and EGF in serums were measured with ELISA. Results All groups started to heal on postburn day 10. Group A had inflammation obviously , and group D moderately. There were significant difference in the healing retes on postburn day 10 , 14, 21 between four groups (P < 0.05). The level of VEGF in group A peaked on postburn day 21 (25.76 ± 1.46)pg/mL, but the levels of VEGF in group B, group C and group D peaked on postburn day 14[(29.73 ± 1.58)pg/mL, (38.91 ± 2.38)pg/mL, (43.54 ± 1.28)pg/mL]. On postburn day 4, 7, 10, 14, 21, there were significant difference(P <0.05). The level of EGF peaked on postburn day 21 in all groups [(0.72 ± 0.14)ng/mL, (0.93 ± 0.13)ng/mL, (1.18 ± 0.16)ng/mL, (1.50 ± 0.15)ng/mL]. There were significant difference on postburn day 7, 10, 14, 21 between four groups (P < 0.05). Conclusions rhGM-CSF combined with nano-silver treatment could promote wound healing, and be better than rhGM-CSF and nano-silver singly.
8.Effect of Recombinant Human Granulocyte∕macrophage Colony_stimulating Factor and Nano_silver on Neovascularization in Healing Process of Skin with Deep II Degree Burn

Methods DeeP II degree burn Wistar rat model was established. The Pathological changes of wound of the three grouPs were obserVed 1,4,7,10,14 and 21 days after the treatment. The concentration of VEGF in serums was measured with ELISA. The leVels of HIF_1α mRNA exPression were detected by RT_PCR. Results On day 10, neoVascularization deVeloPed in grouPs A, B and C. Healing rate of the wound was the highest in grouP C, lowest in grouP A, with significant differences among the three grouPs on day 14 and day 21 (P<0. 05). VEGF leVel Peaked on day 14 in grouP A,and on day 10 in grouPs B and C. There were significant differences in VEGF leVel on day 1 between grouP A and grouPs B,C,on day 7 between grouP A and grouP C,on day 10,14 and 21 among the three grouPs (P<0. 05),but no significant differences on day 4 among the three grouPs. Conclusion rhGM_CSF and nano_silVer treatment can accelerate neoVascularization of wound,and rhGM_CSF is better than nano_silVer.
9.Promoter methylation of Wilms' tumor gene on the X- chromosome in gastric cancer.

OBJECTIVETo investigate the changes in methylation levels of the promoters of the tumor suppressor gene Wilms' tumor gene on the X-chromosome (WTX) and its possible role in gastric cancer.
METHODSWTX promoter methylation levels were detected in 20 pairs of specimens of gastric cancer and matched normal tissues and in 3 gastric cancer cell lines (MGC803, SCG7901, and BGC823) using the Sequenom MassARRAY quantitative analysis system. The gastric cancer cell line BGC823 was treated with 5-aza-2'-deoxycytidine (5-aza-dC) for demethylation and the changes in the level of WTX promoter methylation were investigated.
RESULTSWTX promoter methylation levels were very low and showed no significant differences among normal gastric tissues, gastric cancer tissues and the 3 gastric cancer cell lines. In BGC823 cells, treatment with 5-aza-dC did not obviously affect the promoter methylation levels of WTX.
CONCLUSIONHigh methylation levels of WTX promoters are rare in gastric cancer.

10.Study on compatibility of traditional Chinese medicine active constituents with antioxidant activity.

OBJECTIVETo find a research method suitable to multi-component multi-index composition-activity relationship of traditional Chinese medicines (TCMs) with the antioxidant activity of four compatible active constituents of TCMs as the subject.
METHODLARS-based regression algorithm and comprehensive weight coefficient method were adopted to study in vitro clearance of DPPH and polyaromatic hydrocarbon with different doses of four compatible active constituents--total flavonoids of Glycyrrhizae Radix et Rhizoma (TFG), Ginkgo Folium extract (GBE), total flavonoids of Epimedii Folium (TFE) and astragaloside (AST) according to the comprehensive efficacy assessment procedures, that is test design, efficacy test and mathematical modeling (model verification).
RESULTLARS and comprehensive weight coefficient method was adopted to assess the optimal comprehensive efficacy of clearing DPPH and polyaromatic hydrocarbon. The results showed that the optimal dose combination of compatible constituents is TFG-GBE-TFE-AST 1: 0.2545: 0.007 6:0.011 5.
CONCLUSIONThe above compatibility of TCMs constituents can effectively clear polyaromatic hydrocarbon and DPPH with a good antioxidation. The uniform design adopted in this experiment, in combination of synthetic weight method, is suitable to the data analysis of "non-linear and small-sample" biotic experiment and the study on screening and assessment of compatibility of TCMs constituents.
