The aim of this study was to investigate and evaluate the antitumor effects of a novel poly(ADP-ribose) polymerase (PARP) 1/2 inhibitor, YHP-836, in combination with temozolomide (TMZ) for the treatment of glioblastoma (GBM). The cytotoxicity of YHP-836 was tested alone or in combination with TMZ using MTT assay. Immunoblotting and flow cytometry were also employed to assess the combination activity of YHP-836 and TMZ in multiply GBM cell lines. Further, the antitumor activity of YHP-836 and TMZ was evaluated using subcutaneous and orthotopic mice xenograft tumor models. All procedures were approved by the Ethics Committee for Animal Experiments of the Institute of Materia Medica, Chinese Academy of Medical Sciences and Peking Union Medical College and conducted under the Guidelines for Animal Experiments of Peking Union Medical College. 2.Effect of Wuwei Xiaoduyin on NF-κB Signaling Pathway in Lipopolysaccharide-induced Rat Mesangial Cells

ObjectiveTo study the effect and mechanism of Wuwei Xiaoduyin in treating rat renal mesangial cells (HBZY-1) induced by lipopolysaccharide (LPS) through the nuclear factor-κB (NF-κB) signaling pathway. MethodRat HBZY-1 cells were randomly assigned into the normal group, model group, benazepril (50 μmol·L-1) group, and high- and low-dose (2.75 and 0.69 g·kg-1) Wuwei Xiaoduyin groups. The normal group, model group, and benazepril group were treated with 10% normal rat serum, and the Wuwei Xiaoduyin groups with 10% medicated serum. Except the normal group, the other four groups were treated with LPS (100 ng·mL-1) for modeling in vitro. The changes of cell morphology were observed under optical microscope. The expression of NF-κB p65 was detected by immunofluorescence (IF) method. Methyl thiazolyl tetrazolium (MTT) colorimetry was employed to detect cytotoxicity and cell proliferation. The levels of interleukin-1β (IL-1β), intercellular adhesion molecule-1 (ICAM-1), laminin (LN), and fibronectin (FN) in cell supernatant were determined by enzyme-linked immunosorbent assay (ELISA). The mRNA levels of IL-1β, FN, and NF-κB p65 were measured by real-time fluorescence quantitative PCR. The protein levels of phosphorylated inhibitor of NF-κB kinase β (p-IKKβ), phosphorylated NF-κB inhibitor (p-IκBα), and NF-κB p65 were determined by Western blot. ResultCompared with the normal group, the modeling increased cell proliferation (P<0.01), elevated the levels of IL-1β, ICAM-1, LN, and FN in cell supernatant (P<0.01), and up-regulated the mRNA levels of IL-1β, FN, and NF-κB p65 (P<0.01) and the protein levels of p-IKKβ, p-IκBα, and NF-κB p65 (P<0.01). Such changes were recovered by benazepril and Wuwei Xiaoduyin (P<0.05, P<0.01). ConclusionWuwei Xiaoduyin can mitigate the inflammatory injury of renal mesangial cells induced by LPS by inhibiting the NF-κB signaling pathway.

BACKGROUND:
Shenyankangfu Tablet (SYKFT) is a Chinese patent medicine that has been used widely to decrease proteinuria and the progression of chronic kidney disease.
OBJECTIVE:
This trial compared the efficacy and safety of SYKFT, for the control of proteinuria in primary glomerulonephritis patients, against the standard drug, losartan potassium.
DESIGN, SETTING, PARTICIPANTS AND INTERVENTION:
This was a multicenter, double-blind, randomized, controlled clinical trial. Primary glomerulonephritis patients, aged 18-70 years, with blood pressure ≤ 140/90 mmHg, estimated glomerular filtration rate (eGFR) ≥ 45 mL/min per 1.73 m
MAIN OUTCOME MEASURES:
The primary outcome was change in the 24-hour proteinuria level, after 48 weeks of treatment.
RESULTS:
A total of 735 participants were enrolled. The percent decline of urine protein quantification in the SYKFT group after 48 weeks was 8.78% ± 2.56% (P = 0.006) more than that in the losartan 50 mg group, which was 0.51% ± 2.54% (P = 1.000) less than that in the losartan 100 mg group. Compared with the losartan potassium 50 mg group, the SYKFT plus losartan potassium 50 mg group had a 13.39% ± 2.49% (P < 0.001) greater reduction in urine protein level. Compared with the losartan potassium 100 mg group, the SYKFT plus losartan potassium 100 mg group had a 9.77% ± 2.52% (P = 0.001) greater reduction in urine protein. With a superiority threshold of 15%, neither was statistically significant. eGFR, serum creatinine and serum albumin from the baseline did not change statistically significant. The average change in TCM syndrome score between the patients who took SYKFT (-3.00 [-6.00, -2.00]) and who did not take SYKFT (-2.00 [-5.00, 0]) was statistically significant (P = 0.003). No obvious adverse reactions were observed in any group.
CONCLUSION:
SYKFT decreased the proteinuria and improved the TCM syndrome scores of primary glomerulonephritis patients, with no change in the rate of decrease in the eGFR. SYKFT plus losartan potassium therapy decreased proteinuria more than losartan potassium therapy alone.

4.Quality evaluation of Magnoliae Officinalis Cortex based on combinative method of fingerprint,quantitative analysis of multicomponents and chemometrics.

This study aims to establish a combinative method based on fingerprint,assay of multi-component and chemometrics for quality evaluation of Magnoliae Officinalis Cortex. Twenty batches of samples were determined by UPLC and a common mode of fingerprint was established. The similarities between fingerprints of 20 batches of samples were over 0. 90 and the common mode were evaluated. Eight components were identified as syringing, magnocurarine, magnoflorine, magnoloside B, magnoloside A, honokiol,magnolol,and piperitylmagnolol by comparison with reference substances and their content in samples were simultaneously determined.Based on the results,the fingerprint had good consistency between the same origin and minor diversity between the different sources.Piperitylmagnolol and peak 13 could be used as a distinction with the different sources. According to content of 8 components,Fisher discriminant analysis model was established and different source sample was classified pursuant to the discriminant fraction. It is indicated that simultaneous quantification of multi components coupled with chemometrics analysis could be a well-acceptable strategy to identify and evaluate the quality of Magnoliae Officinalis Cortex.

5.Surveillance of key parasitic diseases in Baise City from 2006 to 2015

Objective To understand and master the infection status of key parasitic diseases in Baise City from 2006 to 2015, so as to provide evidence for the development and adjustment of the control strategy for parasitic diseases. Methods The relevant information of the final evaluation of the key parasitic diseases in 12 counties (cities and districts) of Baise City from 2006 to 2015 was collected, and analyzed. Results During the period of 2006-2015, a total of 20 654 person-times were investigated for parasitic diseases in Baise City and 1 147 persons were infected, with an average infection rate of 5.55%. In 2006, the infection rate was the highest (28.67%, 362/1 254), while the lowest was in 2013 (2.08%, 44/2 113). The infection rate of Clonorchis sinensis was the highest, and the infection rate of Ascaris lumbricoides was the second. The infection rates of the males and females were 5.35% (631/11 795) and 5.82% (516/8 859) respectively, with no significant difference between them (χ2 = 2.175, P > 0.05). The highest infection rate existed in the 40-49 years group, the rural residents, the farmers and the crowd with the educational level of senior high middle school or above. Conclusions The infection rate of parasitic diseases presents a decline trend overall in Baise City from 2006 to 2013, however it rebounded in 2014 and 2015. The next step is to strengthen the prevention and control of parasitic diseases by promoting health education, regular insect repellent, and fecundity management.
6.Comprehensive treatment of adrenal cortical carcinoma.

OBJECTIVE:
To investigate the clinical and functional imaging examination and pathological features of adrenocortical carcinoma (ACC), in order to improve the diagnosis and treatment of ACC.
METHODS:
The clinical data of 93 patients with ACC were analyzed retrospectively. Their diagnosis, surgical treatment and follow-up of mitotane medcine therapy were madeaccording to clinical manifestations, adrenal endocrine function determination, imaging examination characteristics and histopathological results.
RESULTS:
Among the 93 patients, the age ranged from 11 to 76 years, with a median age of 48 years. The ratio of male to female was 1:1.2. Twenty-four hours urinary free cortisol (UFC) elevated in 86 cases, adrenocorticotropic hormone (ACTH) decreased in 88 cases, blood F rhythm disappeared in 82 cases, and 31 cases of aldosterone increased. Thirty-six cases of sexual hormone increased. Neuron specific enolase (NSE) increased in 27 cases. Insulin-like growth factor-1 (IGF-1) increased in 26 cases. Seventy-six cases of high-dose dexamethasone suppression test (HDDST) and low-dose dexamethasone supression test (LDDST) were not suppressed separately. There were 62 cases of hypertension, and typical Cushing manifestations in 81 cases. Blood glucose elevated in 54 cases. Hypokalemia was in 21 cases and androgen secretion increased in 36 cases. The maximum diameter of the tumor was 3-17 cm, with 6 cases of adrenal central vein, renal vein and inferior vena cava tumor thrombus. The recurrence time was 1.2-5.0 years after operation. Metastasis and recurrence were in 56 cases during the follow-up, lung metastasis in 13 cases, liver metastasis in 17 cases, retroperitoneal lymph node metastasis in 9 cases, lumbar metastasis in 7 cases, ovarian metastasis in 3 cases, abdominal wall and incision implantation in 4 cases. Fifteen cases with distant metastasis. Seventy-seven patients were treated with radical adrenalectomy, and the other patients were treated with renal and adrenal resection on the same side of the kidney. There were 5 cases of adrenal tumor with vena cava tumor thrombus in the removal of the tumor, and the other for the partial resection of the vena cava in 3 cases. Regarding the clinical stage, stage I was in 39 cases, stage II in 28 cases, stage III in 16 cases and stage IV in 10 cases. The patients were followed up for 8-69 months, and 43 patients survived more than 5 years.
CONCLUSION
Function imaging combined with clinical features and endocrine hormone levels have important roles in ACC early diagnosis. Radical excision is the only effective treatment. Adjuvant or adjuvant mitotane drug therapy can brused for the treatment of recurrence and metastasis ACC patients,. ACC is a tumor with high malignancy and poor prognosis.

7.Final evaluation of medium-and long-term program of schistosomiasis con-trol in Baise City

Objective To evaluate the monitoring results of medium-and long-term program of schistosomiasis control in Baise City,so as to provide the reference for the elimination of schistosomiasis.Methods The data of schistosomiasis control in Baise City from 2004 to 2015 were collected and analyzed.Results By the end of 2015,the total number of regular screening serum tests for schistosomiasis in the planning period was 10 244 person-times,with 649 positive cases.The number of feces tests was 2 158 person-times in the permanent resident population,and the number was 2 683 person-times in the floating popu-lation.The Oncomelania hupensis snail survey area was 150.04 hm2,and the accumulated snail control area was 2.03 hm2.No schistosomiasis patients or schistosome-infected snails were found.Conclusion The effect of medium-and long-term program of schistosomiasis control is effective in Baise City,and the criterion of schistosomiasis elimination has been achieved.
8.Treatment and nursing of a cysticercosis patient with multiple foci

Objective To analyze the relevant data,including medical history,treatment and nursing of one cysticercosis patient with multiple foci in Baise City,so as to provide the evidence for improving the treatment and nursing of cysticercosis in the future.Methods The data of diet history,diagnosis,treatment and nursing of the patient were collected and analyzed.Re-sults The patient was a permanent resident in a cysticercosis high endemic area and had a definite history of eating measly pork. The combination of clinical manifestations and auxiliary examinations of the patient confirmed that he was suffered from cysticercosis with multiple foci.After the patient was firstly administered the antiparasitic treatment of cysticercosis(praziquan-tel),he had status epilepticus.However,with the close cooperation between physicians and nurses,his illness obviously im-proved and 35 days later,he discharged.Conclusion The physicians and nurses should pay much attention to the changes of illness of the patients with cysticercosis,especially the cysticercosis patients with multiple foci in the antiparasitic treatment of cysticercosis,so as to avoid cerebral herniation due to the increased intracranial pressure.
9.Investigation of a case of foreign imported falciparum malaria in Xilin County,Baise City

Objective To investigate the epidemiology,diagnosis and treatment of an imported falciparum malaria patient in Xilin County,Baise City,so as to provide the reference for improving the diagnosis and treatment of falciparum malaria pa-tients in the future. Methods The epidemiological and clinical data were collected and analyzed. Results The patient had lived in Africa where the malaria was epidemic. The disease attacked him after his coming back home from abroad. The blood test for Plasmodium falciparum was positive. In Xilin County,no local Plasmodium infection was found from 2004 to 2016,and therefore,we concluded that this case was overseas imported. Conclusion The monitoring of overseas returnees in Xilin Coun-ty should be strengthened to timely diagnose and treat the imported cases of malaria.
10.Analysis of electric shock protection requirements for IVD diagnostic equipment in IEC 61010-1:2010

Objective To study the electric shock protection requirements of in vitro diagnostic equipment and to help manufacturers understand the relevant requirements in order to design products reasonably. Methods The requirements of IEC 61010-1:2010 were analyzed, the electric shock protection measures were explored, and the principles for protecting ground impedance against electric shock were described. An example was taken to study the electric shock protection measures for the sampling needle of the automatic bio-chemical analyzer, so that the existence of electric shock hazard could be determined. Results The sampling needle proved its rationality in structure design by avoiding the risk for being electriferous in case of failed basic insulation, although the requirements for protective earthing, double insulation or reinforced insulation were not met efficiently. Conclusion The requirements for electric shock protection can be determined based on comprehensive understanding of the standard, determination of electriferous components of the equipment as well as analysis on electric shock protection measures.
