2.Expression of transcription factor SOX12 in lung adenocarcinoma and its clinical significance.

OBJECTIVE:
To explore the expression patterns of transcription factor SOX12 in lung adenocarcinoma and its significance in the diagnosis and prognosis of the malignancy.
METHODS:
Large cancer genome databases were used to analyze SOX12 expression level in lung adenocarcinoma. Immunohistochemistry (IHC) and semiquantitative PCR were used to detect the expression of SOX12 in 36 specimens of lung adenocarcinoma tissues, 15 adjacent tissues and 21 normal lung tissues. The prognostic value of SOX12 in lung adenocarcinoma and lung squamous cell carcinoma were analyzed using Kaplan-Meier Plotter database, and the relationship between SOX12 expression and the overall survival (OS) and progression free survival (PPS) of the patients were analyzed.
RESULTS:
Analysis of TCGA database and GEO (GSE40419) database showed that SOX12 expression levels were significantly higher in in lung adenocarcinoma than in normal lung tissues ( < 0.001). The results of IHC and semiquantitative PCR revealed that SOX12 was expressed at significantly higher levels in lung adenocarcinoma than in normal lung tissues ( < 0.001). Kaplan-Meier survival analysis showed that patients with lung adenocarcinoma positive for SOX12 had a significantly shorter OS and PPS time than those negative for SOX12 ( < 0.05), but SOX12 positivity did not significantly affect OS and PPS of patients with lung squamous cell carcinoma.
CONCLUSIONS
High expression levels of SOX12 in lung adenocarcinoma are significantly associated with a poor OS of the patients, suggesting the value of SOX12 to assist in early diagnosis and prognostic evaluation of lung adenocarcinoma.

3.Rapid preimplantation genetic diagnosis of -thalassemia SEA deletion with blastocyst cell whole genome amplification and short fragment Gap-PCR method.

OBJECTIVETo develop a rapid preimplantation genetic diagnosis method for -thalassemia SEA deletion based on blastocyst cell whole genome amplification (WGA) combined with short fragment Gap-PCR.
METHODSUsing multiple displacement amplification (MDA) WGA technique, we established a double-fluorescent PCR system of the housekeeping genes GAPDH and β-actin for WGA quality testing, and a genotyping PCR system of mutant and normal short sequences for α-thalassemia SEA deletion. The sensitivity and accuracy of this method for diagnosis of -thalassemia SEA deletion were evaluated by detecting lymphocyte samples containing different cell numbers from carriers of SEA deletion. The applicability of this method was evaluated by testing of 12 blastocyst biopsy samples.
RESULTSDetection of lymphocyte samples with different cell numbers using the method developed in this study revealed no ADO in 3-cell samples, and the product quantity of WGA became stable for 4-cell samples. Genotyping of the 10 blastocyst biopsy samples with successful WGA showed a genotype of --/ in 5 samples and / in the other 5 samples, which were consistent with the verification results.
CONCLUSIONSThe method developed in this study is a complete testing process for 4-6 blastocyst biopsy cells to allow rapid, accurate, and cost-effective PGD genotyping of -thalassemia SEA deletion using short fragment gap-PCR.
4.Curative efficacy of batroxobin combine with ginkgo injection in treatment of Sudden deafness and its effects on Serum ET、NO、SOD、sVCAM-1

Objective To study the effect of batroxobin combined with Yinxingdamo injection on serum endothelin(ET),nitric oxide(NO),superoxide dismutase(SOD),vascular cell adhesion molecule-1(VEGF-1)in patients with sudden deafness(SVCAM-1),to explore the best treatment for patients with sudden deafness.Methods Ninety patients with sudden deafness from June 2014 to June 2015 were recruited as the subjects of this study.The patients in control group were treated with batroxobin and batroxobin and ginkgo dipyridol.The levels of serum ET,NO,SOD,sVCAM-1,time of symptom recovery and the curative effect were observed.Results After treatment,the levels of ET,NO and sVCAM-1 in the observation group were significantly lower than those in the control group [(64.28±5.72)pg/mL vs(67.36±6.31)pg/mL,(43.08±9.53)μmol/(93.24±11.25)NU/mL](P<0.05).The results showed that there was no significant difference between the two groups.The recovery time of tinnitus,auria and vertigo in the observation group were less than those in the control group [(3.86±1.02)d vs(5.97±1.34),(5.03±1.24)d vs(7.37±2.01)d,(8.09±2.10)d vs 9.07±2.37)d](P<0.05).The total effective rate in the observation group was better than that in the control group(93.33%vs 75.55%,P<0.05).Conclusion Batroxobin combined with ginkgo dipyridolum injection can decrease the level of ET,NO and sVCAM-1,improve the level of SOD,and improve the microcirculation of the inner ear.Compared with the single effect of Batroxobin More desirable,worthy of promotion.
5.Relationship between serum magnesium level with vascular calcification and mineral disorder in patients on continuous ambulatory peritoneal dialysis

Objective To explore the relationship between serum magnesium level,abdominal aortic calcification(AAC) and mineral metabolism in continuous ambulatory peritoneal dialysispatients.Methods Sixty-two cases patients who were on Continuous Ambulatory Peritoneal Dialysis in Pritoneal Dialysis Center of Qingdao Municipal Hospital were enrolled prospectively.The AAC was detected by abdomen lateral plain radiographs,and the AAC score(AACs) was calculated.According to the AACs,the patients were divided into group A(AACs≤4 points) and group B(from >4 point to ≤15 point) and group C(AACs>15 point).And collected the patient′s clinical data and blood biochemical indicators.The serum magnesium levels of 3 groups were observed,and explored the relationship of serum magnesium and intact parathyroid hormone(iPTH),serum calcium,serum phosphorus.Results There were 41 cases of vascular calcification in 62 cases maintenance ambulatory peritoneal dialysis patients,and its incidence was 66.13%.Group levels of serum magnesium were in group A was (1.00±0.18) mmol/L,(0.89±0.10) mmol/L of group B,and (0.75±0.13) mmol/L of group C,the mean differences were significant(F=11.99,P<0.05),and the group A was significantly higher than the group B and C,the differences were significant(P<0.05),the group B was significantly higher than the group C,the difference was significant(P<0.05).There was negatively correlation between serum magnesium and iPTH(r=-0.288,P<0.05),serum phosphorus(r=-0.272,P<0.05) and AACs(r=-0.610,P<0.01).While there was no significant correlation of serum calcium(r=0.174,P>0.05).Conclusion Vascular is the calcification frequent complications in continuous ambulatory peritoneal dialysis patients.The serum level of serum magnesium may be correlated with the AAC,iPTH,serum phosphorus.
6. Effect analysis of anterior cervical operation for severe cervical kyphosis

Objective:
To determine the feasibility and safety of anterior cervical decompression and fusion in severe cervical kyphosis treatment.
Methods:
Totally 29 patients with severe cervical kyphosis(Cobb angle>50°) underwent anterior cervical decompression and fusion from June 2008 to May 2016 were studied retrospectively. There were 19 males and 10 females. The average age was 32.6 years ranging from 14 to 53 years. According to the etiology, 12 patients had iatrogenic deformity (11 had post-laminectomy cervical kyphosis, 1 had kyphosis due to anterior graft subsidence), 5 had neurofibromatosis, 4 had infective kyphosis, 8 had idiopathic cervical kyphosis. The curvature of cervical angle was measured by two-line Cobb method. The severity of cervical kyphosis was evaluated by kyphosis index (KI). Parameters including kyphosis levels, the apex of the kyphosis, C2-7 sagittal vertical axis(SVA) and T1 slope were also measured on lateral radiographs in the neutral position in each patient. The pre- and post-operative Japanese Orthopaedic Association(JOA) scores, visual analogue scale (VAS) of neek pain, neck disability index (NDI) and cervical alignment were compared. All patients were treated by skull traction. Motor evoked potential and somatosensory evoked potential were applied intraoperation as the spinal cord monitor.
Results:
Skull traction was performed for an average of 6.3 days. The mean vertebral number in kyphotic region was 4.7. The average operation time was 155 minutes and blood loss was 135 ml. The preoperative C2-7Cobb angle was 46.6°±18.1° in average. It was reduced to 11.4°±6.4° in average after operation. The Cobb angle of operation region was 72.9°±19.6° in average before operation. It was reduced to 11.2°±6.4° in average after operation. The kyphosis region correction rate was 84.6%. The mean preoperative C2-7SVA changed from (3.8±14.6) mm to (12.6±7.8) mm postoperatively. The mean preoperative T1 slope changed from -10.6°±16.4° to 7.1°±14.9° postoperatively. The average postoperative C2-7 Cobb angle, Cobb angle of kyphosis region, KI, C2-7 SVA and T1 slope changed significantly compared with preoperation (
7.Modified (Wu's) esophagectomy for a huge thoracic esophageal squamous cell carcinoma 18.3 cm in length.

An esophageal squamous cell carcinoma measuring 18.3 cm in length and 5 cm in diameter was found in the mediastinum of a 53-year man. The patient underwent a modified 3-stage esophagectomy and an esophagogastrostomy at the cervical level (Wu's method). The operation was performed smoothly and the patient recovered uneventfully after the operation. The patient was followed up for 6 months after discharge and reported no difficulties in eating with improved quality of life. This case represents the world's longest esophageal cancer that had been surgically removed. Local advanced esophageal cancer should be removed immediately to prevent potential occurrence of esophageal obstruction, tracheoesophageal fistula or aorto-esophageal fistula.

8.Red blood cell distribution width combined with lipoprotein-associated phospholipase A2 detection for improving diagnostic accuracy of coronary artery stenosis in patients with coronary artery disease.

OBJECTIVETo study the association of red blood cell distribution width (RDW) and lipoprotein-associated phospholipase A2 (LP-PLA2) with the degree of coronary artery stenosis in patients with coronary artery disease (CAD) and the value of RDW combined with LP-PLA2 detection in accurate evaluation of coronary artery stenosis.
METHODSA total of 224 patients including 119 non-CAD cases and 105 CAD cases admitted in our hospital between June, 2013 and June, 2014 were enrolled in this study. The patients' baseline clinical data were collected and venous blood samples were obtained for detecting WBC, RDW-CV and LP-PLA2. The Gensini score of the CAD patients was calculated based on coronary angiographic findings.
RESULTSCompared with the non-CAD patients, CAD patients had significantly higher RDW-CV (P=0.009) and LP-PLA2 (P=0.004) levels. The CAD patients with high Gensini scores had also significantly higher RDW-CV (P=0.001) and LP-PLA2 (P<0.001) levels than those with low scores; RDW-CV and LP-PLA2 were significantly correlated with the Gensini score, and the area under curve of their combined detection was 0.931.
CONCLUSIONCombination of RDW and LP-PLA2 can improve the diagnostic accuracy of the degree of coronary artery stenosis in patients with CAD.

9.The Impact of Humanistic Spirit Cultivation on Nursing Students' Ethical Cognition

Objective:To investigate the impact of humanistic spirit cultivation on nursing students' ethics cog-nition. Method:Total 364 internship nursing students in 2010 were taken as the control group with the routine clini-cal teaching method, 381 in 2014 as the experimental group, not only a routine clinical teaching method but also to increase of their humanistic spirit culture. Results:Humanity spirit cultivation before and after practice nursing students in medical security system cognition difference is not significant, but in medical optimization, the patients right of informed consent, the life value differences of cognitive and ethical cognitive dimension score was statisti-cally significant ( P<0 . 05 ) . Conclusions:Humanistic spirit culture promote the practice of nursing students to raise the level of moral cognition foundation for humanity quality education and nursing ethics education, should further strengthen the diversification of internship training nursing students humanistic spirit.
10.Analgesic and Anti-inflammatory Effects of Different Compatibility of Gentianopsis Paludosa and Sophora Alopecuroide on Mice

Objective To compare analgesic and anti-inflammatory effects of Gentianopsis Paludosa and Sophora Alopecuroide of different kinds of compatibility on the mice.Methods Mice were randomly divided into normal control group, positive medicine group, and different kinds of compatibility groups, 10 mice in each group. Mice received gavage for successive 7 days, once a day. Glacial acetic acid twist body method and hot board to pain method were used to observe and compare analgesic effects of different kinds of compatibility. Xylene to ear swelling method and agar to granulation swollen method were used to observe and compare inflammatory effects of different kinds of compatibility.Results Compared with normal group, compound Gentianopsis Paludosa had obvious analgesic and anti-inflammatory effects (P<0.01). When the ratio of Gentianopsis Paludosa and Sophora Alopecuroide was 20:0.6 and 20:0.9, the analgesic and anti-inflammatory effects were the best.Conclusion Gentianopsis Paludosa and Sophora Alopecuroide have good analgesic and anti-inflammatory effects, among which, when the ratio of Gentianopsis Paludosa and Sophora Alopecuroide is 20:0.6, the best effects show.
