		                        			 Hepatocellular carcinoma (HCC) is a leading cause of cancer-associated death globally. Liver transplantation (LT) has emerged as a key treatment for patients with HCC, and the Milan criteria have been adopted as the cornerstone of the selection policy. To allow more patients to benefit from LT, a number of expanded criteria have been proposed, many of which use radiologic morphological characteristics with larger and more tumors as surrogates to predict outcomes. Other groups developed indices incorporating biological variables and dynamic markers of response to locoregional treatment. These expanded selection criteria achieved satisfactory results with limited liver supplies. In addition, a number of prognostic models have been developed using clinicopathological characteristics, imaging radiomics features, genetic data, and advanced techniques such as artificial intelligence. These models could improve prognostic estimation, establish surveillance strategies, and bolster long-term outcomes in patients with HCC. 5.Protein expression and clinical significance of KCTD8 gene in ductal carcinoma of the breast

		                        			Objective:To investigate the expression of KCTD8 gene in breast ductal carcinoma and its correlation with clinical factors and prognosis.Methods:Immunohistochemistry technology(IHC)were employed to detect protein expression levels of KCTD8 in 27 pairs of breast ductal carci-noma and its paired adjacent tissues.Analyzing the correlation between changes in KCTD8 expres-sion of protein and clinical factors using statistical techniques.RNA expression and methylation data of breast cancer(including intraductal cancer)were analysed from TCGA database.Result:The pro-tein expression of KCTD8 gene in 27 pairs of breast ductal carcinoma tissues showed a decreasing trend compared to adjacent tissues(P<0.05),and the decreased expression level of protein was cor-related with the tumor size of patients(P<0.05).The analysis results of the TCGA database indicate that the expression and hypemethylation of KCTD 8 gene in breast cancer(including intraductal can-cer)tissues affected the prognosis of patients.Conclusion:The reduced protein expression level of KCTD8 gene in breast ductal carcinoma may be involved in the development and affect the prog-nosis of patients.
		                        		
		                        		
		                        		
		                        	
6.Analysis of the efficacy and prognosis of radiotherapy in acute leukemia with extramedullary infiltration

		                        			Objective To investigate the clinical characteristics,treatment methods,and prognosis of a-cute leukemia patients with extramedullary infiltration.Methods The clinical characteristics and treatment methods of 47 acute leukemia patients with extramedullary infiltration admitted to the Affiliated Hospital of Guizhou Medical University from April 2014 to April 2023 were retrospectively analyzed.Subgroup analysis was performed according to whether there was extramedullary infiltration before transplantation,and whether there was isolated extramedullary recurrence after transplantation.Based on this analysis,the patients were di-vided into the pre-transplantation radiotherapy group and pre-transplantation non-radiotherapy group,the post-transplantation radiotherapy group and post-transplantation non-radiotherapy group.According to the treatment methods of central nervous system leukemia(CNSL),the patients were divided into the intrathecal injection group(n=12)and combination of intrathecal injection and radiotherapy group(n=13).The local remission situation,survival duration,and toxic and side effects of radiotherapy and chemotherapy were com-pared.Results For acute leukemia patients with extramedullary infiltration,the overall survival time(OS)in the radiotherapy group was better than that in the non-radiotherapy group(median OS:706 d vs.151 d,P=0.015).Subgroup analysis showed that the OS of the pre-transplantation radiotherapy group was better than that of the pre-transplantation non-radiotherapy group(median OS:592 d vs.386 d,P=0.035).For CNSL,the combination of intrathecal injection and radiotherapy group had a better OS than the intrathecal injection group(median OS:547 d vs.388 d,P=0.045).The event-free survival time(EFS)of the radiotherapy group was better than that of the non-radiotherapy group(median EFS:175 d vs.50 d,P=0.005).The COX pro-portional-hazards model showed that treatment with or without radiotherapy had a significant impact on the OS of acute leukemia patients with extramedullary infiltration.The risk of death in the pre-transplantation non-radiotherapy group was 2.231 times higher than that in the pre-transplantation radiotherapy group(HR=3.231,95%CI:1.021-10.227,P=0.046).Compared with the non-radiotherapy group,the radiother-apy group had a higher local remission and a lower risk of haematological toxicity,infection,and haemorrhage.Conclusion Radiotherapy can rapidly alleviate the local symptoms of acute leukemia complicated with extr-amedullary infiltration,prolong the survival time of these patients,and reduce the risk of hematologic toxicity,infection,and haemorrhage.

		                        			Objective To explore the correlation between endoscopic ultrasound features and immunohistochemical results of gastric stromal tumors at different risk levels,and provide reference for clinical diagnosis and treatment.Methods Retrospective collection of relevant indicators of gastric stromal tumor patients who received concurrent surgical treatment at Lanzhou University Second Hospital from January 2018 to July 2023,and analysis of differences in risk levels.Results The average age of onset of gastric stromal tumors was(56.02±10.94)years old,and the initial symptom of black stool was significant for different grades of gastric SMTs.Patients with different risk levels also received significant differences in treatment methods(P<0.05).There was no difference in the classification of risk levels among the four groups of extremely low risk,low risk,medium risk,and high risk in terms of location,origin level,boundary,echo,presence or absence of cystic changes,calcification,or blood flow signals(P<0.05);Tumor size,growth pattern,shape,internal echo uniformity,and appearance are significant factors for postoperative risk grading of gastric stromal tumors(P<0.05).Multivariate logistic regression showed that tumor size is an independent influencing factor(P<0.05).The immunohistochemical results showed that the positivity of DOG-1,CD34,SMA,and Desmin had no effect on the postoperative risk grading of gastric SMTs(P<0.05);The positive expression of Vimentin,Ki-67 index,and mitosis were significantly correlated with risk(P<0.05).Conclusion The different invasion grades of gastric stromal tumors are related to the size,shape,and echo of the lesion.Endoscopic ultrasound can effectively diagnose gastric stromal tumors,and combining with immunohistochemical related indicators can better evaluate the overall condition of patients with gastric stromal tumors,providing reference for clinical diagnosis and treatment.
		                        		
		                        		
		                        		
		                        	
8.Exploring the Effect of Shuwei Decoction (舒胃汤) for Rats with Functional Dyspepsia of Liver-Depression and Spleen-Deficiency Syndrome Model on the Ultrastructure of ENS-PDGFRα+ Cell-SMC Network in Gastric Sinusoidal Tissues

		                        			ObjectiveBased on the ultrastructure of enteric nervous system (ENS)-platelet-derived growth factor receptor α positive (PDGFRα+) cell - smooth muscle cell (SMC) network, the effect and mechanism of Shuwei Decoction (舒胃汤) for rats with functional dyspepsia (FD) of liver-depression and spleen-deficiency syndrome were explored. MethodsFifty SD rats were divided into blank group, model group, low-, medium- and high-dose Shuwei Decoction groups randomly, with 10 rats in each group. The blank group was not set up, and the other 4 groups were set up with the modified composite cause (chronic restraint stress + tail irritation + excessive fatigue + diet disorder) for 21 days, resulting in FD model rats with liver-depression and spleen-deficiency syndrome. After successful modelling, rats in the low-, medium- and high-dose groups were given 3.78, 7.56 and 15.12 g/(kg·d) of Shuwei Decoction by gavage, respectively, while rats in the blank group and the model group were given 10 ml/(kg·d) of saline by gavage; all of them were given gavage once a day for 14 days. Body mass increase were collected before and after gavage, and compared after the experiment; the elevated cross maze experiment was conducted to observe the percentage of staying time and the percentage of times entering the open arm of rats; the gastric emptying rate and small intestinal propulsion rate were measured by black semi-solid paste; HE staining was used to observe histopathology of the gastric antrum; the Western Blotting and RT-qPCR were used to detect the expression levels of PDGFRα, small conductance calcium activated potassium (SK3), and purinergic G protein-coupled receptor P2Y1 (P2Y1 R), and mRNA expression in gastric antrum; immunofluorescence double staining was used to detect the co-localization of PDGFRα with receptor tyrosine kinase (C-kit), protein gene product 9.5 (PGP9.5), Sk3, and smooth muscle cells (SMCs) in gastric sinusoidal tissues; and transmission electron microscopy was used to observe the ultrastructure of the ENS-PDGFRα+ cell-SMC network. ResultsHE staining results showed that the mucosal folds of gastric tissue were clear in all groups, no organic lesions such as bleeding, ulceration and swelling were observed, and the arrangement of gastric glands in the lamina propria of gastric sinusoidal tissues in model group and low-dose Shuwei Decoction group was disordered. The arrangement of gastric glands in lamina propria in medium- and high-dose Shuwei Decoction groups was more orderly, and the lesion degree was less than that in model group and low-dose Shuwei Decoction group. Compared with the blank group, the growth value of body mass, the percentage of open arm retention time, the percentage of times entering the open arm, the small intestine propulsion rate, and the gastric empty rate of rats in the model group significantly decreased after gavage; the expressions of PDGFRα, Sk3, P2Y1 protein and mRNA in gastric sinusoidal tissues decreased; the co-localization expression of PDGFRα with C-kit, Sk3, SMC and PGP9.5 significantly decreased (P<0.05 or P<0.01); the ultrastructure of ENS-PDGFRα+ cell-SMC network was damaged without gap junction. Compared with model group and low-dose Shuwei Decoction group, the growth value of body mass, the percentage of open arm retention time, the percentage of times entering the open arm, the small intestine propulsion rate, and the gastric empting rate of rats in medium- and high-dose Shuwei Decoction groups increased after gavage; the expressions of PDGFRα, Sk3, P2Y1 protein and mRNA in gastric sinusoidal tissues increased; the co-localization expression of PDGFRα with C-kit, Sk3, SMC and PGP9.5 significantly increased (P<0.05 or P<0.01); the ultrastructure of ENS-PDGFRα+ cell-SMC network was intact. Compared with medium-dose Shuwei Decoction group, the protein expressions of PDGFRα, Sk3 and P2Y1, the mRNA expressions of PDGFRα and Sk3, and the co-localization expressions of PDGFRα and C-kit, Sk3, SMC, and PGP9.5 in high-dose Shuwei Decoction group increased (P<0.05 or P<0.01). ConclusionShuwei Decoction can improve the pathological lesion of the gastric antrum in the FD model rats with liver-depression and spleen-deficiency syndrome and increase the gastric emptying rate as well as the small intestinal propusion rate. 		                        			 This experiment aims to study the taste-masking effects of different kinds of corrigent used individually and in combination on ibuprofen oral solution, in order to optimize the taste-masking formulation. Firstly, a wide range of corrigent and the mass fractions were extensively screened using electronic tongue technology. Subsequently, a combination of sensory evaluation, analytic hierarchy process (AHP)-fuzzy mathematics evaluation, and Box-Behnken experimental design were employed to comprehensively assess the taste-masking effects of different combinations of corrigent on ibuprofen oral solution, optimize the taste-masking formulation, and validate the results. The study received ethical approval from the Review Committee of the Beijing University of Chinese Medicine (ethical code: 2024BZYLL0102). The results showed that corrigent fractions and types were screened separately through single-factor experiments.
